Time courses for ingestion, retention and release via feces of microbial food was investigated using 2 bivalves with different feeding strategies, Potamocorbula amurensis, and Macoma balthica. The results showed 2 pathways for the uptake of food material in these clams. The first is represented by an initial label pulse in the feces. The second pathway operates over longer time periods. Inert 51Cr-labeled beads were used to determine time frames for these pathways. The first pathway, involving extracellular digestion and intestinal uptake, is relatively inefficient in the digestion of bacterial cells by P. amurensis but more efficient in …

Dynamics Of Bacterioplankton Abundance And Production In Seagrass Communities Of A Hypersaline Lagoon, Gerardo Chin-Leo, Ronald Benner

The significance of bacterioplankton in the flow of carbon and energy and in trophic dynamics of the upper Laguna Madre, Texas (USA), was estimated by measuring bacterioplankton abundance and production over an 18 mo period and over several diel cycles. Bacterioplankton production was estimated from incorporation rates of thymidine (DNA synthesis) and leucine (protein synthesis). These independent inhces of bacterial growth were generally in agreement and yielded nearly identical annual estimates of bacterial production (25.24 g C m-2 yr-1 based on thymidine and 25.12 g C m-2 yr-1based on leucine). Glycation, oxidation, and nonenzymatic browning of protein have all been implicated in the development of diabetic complications. The initial product of glycation of protein, fructoselysine (FL), undergoes further reactions, yielding a complex mixture of browning products, including the fluorescent lysine-arginine cross-link, pentosidine. Alternatively, FL may be cleaved oxidatively to form N(epsilon)-(carboxymethyl)lysine (CML), while glycated hydroxylysine, an amino-acid unique to collagen, may yield N(epsilon)-(carboxymethyl)hydroxylysine (CMhL). The floral fragrance glands (osmophores) of 18 species of Stanhopea and Sievekingia were examined through a series of developmental studies at light and electron microscope levels including late bud stages through postanthesis. Various characters were identified to be of potential systematic value and were recorded for each species. These characters included: texture of the osmophore surface, number of distinct cell layers comprising the osmophore, nature of lipid inclusions in osmophore cells, and presence or absence of plastoglobuli in osmophore amyloplasts. These characters were combined with traditional features of floral lip morphology for cladistic analysis. Sievekingia was the postulated outgroup. Stanhopea …

Specific and sensitive amplification of major outer membrane protein (MOMP) gene DNA sequences of Chlamydia psittaci was achieved in a two-step polymerase chain reaction. First, oligonucleotide primers specific for 5' and 3' nontranslated regulatory regions of the MOMP gene were used in a polymerase chain reaction to amplify a DNA fragment of approximately 1,400 bp. A portion of this DNA fragment was amplified in a second reaction using a degenerate oligonucleotide primer specific for a DNA sequence contained within the 1,400-bp DNA fragment and one of the first-step primers. The entire nucleotide sequences of the spike glycoprotein (S) genes of the highly virulent bovine coronavirus (BCV) strain BCV-LY138, the avirulent BCV-L9 and related Norden Vaccine (BCV-Vaccine) strains were determined using the polymerase chain reaction (PCR) to amplify cDNAs obtained by reverse transcription of viral RNA, and to produce single strand cDNAs for DNA sequencing. The S gene sequences of these viral strains were compared with those of recently published strains BCV-Mebus, BCV-Quebec, and BCV-F15. An open reading frame of 4092 nucleotides, encoding a protein of 1363 amino acid residues, was found in all six strains. Frameshifts and insertions or …

Monoclonal antibodies (MAbs) against two major structural proteins of the cell-adapted Mebus strain of bovine coronavirus (BCV-L9) were produced and characterized. Seven MAbs reacted with the peplomeric glycoprotein, gp100/S, while three MAbs reacted with the nucleoprotein p53/N in Western blot analysis of BCV polypeptides. MAbs to gp100/S reacted with discontinuous epitopes of gp100/S in Westerns under mild but not under standard denaturing conditions. In contrast, MAbs to p53/N reacted in both types of Westerns, and those epitopes were thus continuous. MAbs to p53/N failed to neutralize BCV infectivity, while 4 MAbs to gp100/S neutralized BCV effectively. Cross reactivity of MAbs …

Various mechanisms have been suggested for the quenching of Cd ion activity in plant vacuoles. These include solution complexation with organic acids and sulfhydryl-containing peptides and precipitation as sulfides. Because direct experimental support for these mechanisms is lacking and difficult to obtain, we have used a computer model to evaluate the quenching role of possible organic and inorganic ligands of tobacco cultured cells exposed to Cd. Results of this thermodynamic evaluation, which assumes that a chemical equilibrium state is met in the vacuole, support the conclusion that sulfhydryl-containing peptides and certain organic acids may form soluble Cd complexes. Acrosomal integrity of bovine spermatozoa was evaluated using naphthol yellow S-erythrosine B stain with bright field microscopy, .2% glutaraldehyde fixation with Nomarski optics, and live wet smears with Nomarski optics to compare the evaluation techniques. Four ejaculates per two Holstein bulls were enriched through a Percoll gradient, capacitated with heparin, and acrosome-reacted with lysophosphatidylcholine to prepare spermatozoa for in vitro fertilization. Spermatozoa samples were taken after each preparatory step, and percentage of intact acrosomes was evaluated with the stain, .2% glutaraldehyde, and live wet smear. Changes in the activites of S-adcnosylmethionine (SAM) synthetase (methionine adenosyltransferase, EC 2.5.1.6.) and SAM decarboxylase (EC 4.1.1.50) were studied in carrot (Daucus carota) cell cultures in response to 2,4-dichlorophenoxyacetic acid (2,4-D) and several inhibitors of polyamine biosynthesis. Activity of SAM synthetase increased significantly during the first 5 days of culture under all conditions, maximum activity being observed in thc presence of 2,4-D. DL-α-Difluoromethylarginine (DFMA), I,L-α-difluoroinethylornithine (DFMO) and cyclohexylammonium sulfatc (CHAS) had little effect on the activity of this enzyme. The enzyme activity was lower than the controls in the presence of methylglyoxal bis(guany1hydrazone) (MGBG). The activity of SAM dccarboxylasc, which …

A DNA polymerase has been highly purified from Anacystis nidulans R2. Electrophoretic analysis in sodium dodecyl sulfate-polyacrylamide gels revealed that the final fraction contains three bands of Mr 107,000, 93,000, and 51,000, respectively. Analysis of purified DNA polymerase activity in situ indicates that of the three polypeptides the Mr 107,000 species has the catalytic activities. The native molecular weight of the enzyme was estimated by glycerol gradient sedimentation to be 100,000. The enzyme has an absolute requirement for a divalent cation. Mg2+ can be replaced with Mn2+, but the DNA polymerase is less active. Potassium …

The relations of systolic and diastolic blood pressures to alcohol intake and exercise tolerance levels in 15,612 men and 3,855 women were investigated. Alcohol intake was assessed by questionnaire and stratified into seven levels for men and six for women according to the ounces of ethanol consumed per week. Exercise tolerance was determined by maximal treadmill exercise testing and was categorized into six age-specific by sex-specific levels. Both systolic and diastolic blood pressure were significantly related to both alcohol intake and exercise tolerance levels in both men and women. The United States military presence in Central and South America is under attack, both literally and politically, as bombings and sniper fire directed against U.S. military personnel characterize the low-intensity conflict (LIC) environment in certain parts of Latin America. The process of contracting support for U.S. forces in a LIC environment is oriented toward sustaining operations in the field, as opposed to contracting for mobilization or surge production. A significant characteristic of this field support is that a contracting officer (KO) deploys overseas with the supported military unit. In this experiment we have identified and partially characterized the immunosuppressive activity of preimplantation horse conceptus-conditioned medium (HCCM). Horse conceptuses were nonsurgically flushed from mares at Days 9-10 (n = 6), 15-16 (n = 3), and 25-26 (n = 3). After incubating the conceptuses for 24 h in RPMI-1640 supplemented with 15% fetal calf serum (FCS) and 1% penicillin/streptomycin, HCCM was obtained from cultures and tested for immunosuppressive activity in lymphocyte proliferation assays. Chloroplast And Mitochondrial: Dna Polymerases From Cultured Soybean Cells, Sabine Heinhorst, Gordon C. Cannon, Arthur Weissbach

DNA polymerases were purified from chloroplasts and mitochondria of cultured Glycine max cells. The chloroplast enzyme exists in two forms which are indistinguishable from each other biochemically. All three organellar enzymes have an estimated molecular weight of 85,000 to 90,000 and prefer poly(rA)dT12-18 over activated DNA as a template in vitro. Maximum activity of the chloroplast and mitochondrial DNA polymerases requires KCI and a reducing agent, and the enzymes are completely resistant to inhibitors of DNA polymerase a. The causes of the extensive (0.86%/yr; 288,414 ha/yr) and well documented dramatic and accelerating rate of coastal wetland loss in the northern Gulf of Mexico were investigated by an interdisciplinary university research team to discern the role of outer continental shelf development. The landscape changes and potential causal agents are emphasized herein. Natural driving factors include sea level rise and geological com- paction, which appear to remain constant this century, and sediment supply from the Mississippi River which has declined by 50%,since the 1950s. Antimicrobial Susceptibilities Of Staphylococcal Species Isolated From Mammary Glands Of Unbred And Primigravid Dairy Heifers, P. Trinidad, S. C. Nickerson, D. G. Luther

Staphylococcal isolates from teat canal keratin and mammary secretion samples of unbred and primigravid Jersey heifers were tested in vitro for susceptibility to 12 antimicrobial agents. More than 92% of the 311 isolates were susceptible to all antimicrobial agents tested. Staphylococci other than Staphylococcus aureus demonstrated an overall susceptibility of 98.3% to all antibiotics, and Staphylococcus aureus demonstrated a 97% susceptibility. Across all staphylococcal species, susceptibility of isolates from secretion samples was 98.1%, and susceptibility of isolates from teat canal keratin samples was 93.1%.
