Purification And Characterization Of A Gamma-Like Dna-Polymerase From Chenopodium Album L., K. Meißner, Sabine Heinhorst, Gordon C. Cannon, T. Börner

A DNA polymerase activity from mitochondria of the dicotyledonous angiosperm Chenopodium album L. was purified almost 9000 fold by successive column chromatography steps on DEAE cellulose, heparin agarose and ssDNA cellulose. The enzyme was characterized as a gamma-class polymerase, based on its resistance to inhibitors of the nuclear DNA polymerase alpha and its preference for poly(rA).(dT)12-18 over activated DNA in vitro. The molecular weight was estimated to be 80,000 - 90,000. A 3' to 5' exonuclease activity was found to be tightly associated with the DNA polymerase activity through all purification steps. Three major policies underlie tort liability: deterrence, compensation, and corrective justice. A primary justification for proportional liability is its alleged superiority in advancing the tort policy of deterrence. This Article demonstrates a significant flaw in this claim by showing that the use of tort liability in multiple cause cases involving statistical evidence in fact serves the policy of deterrence quite poorly.

Glycation, oxidation, and browning of proteins have all been implicated in the development of diabetic complications. We measured the initial Amadori adduct, fructoselysine (FL); two Maillard products, N epsilon-(carboxymethyl) lysine (CML) and pentosidine; and fluorescence (excitation = 328 nm, emission = 378 nm) in skin collagen from 39 type 1 diabetic patients (aged 41.5 +/- 15.3 [17-73] yr; duration of diabetes 17.9 +/- 11.5 [0-46] yr, [mean +/- SD, range]). This article contributes to attempts on the part of Quechua scholars to understand the evidential system of this language family, and thereby paves the way for a more complex understanding of Quechua speakers' language and culture. The author opposes the position that the most general meaning of the -mi suffix is to indicate a direct or first-hand experience; and she holds that specific claims about Quechua speakers' epistemological orientations, based on such an analysis, cannot be supported. Evidence from speakers' use of -mi indicates that it encodes two paradigmatic contrasts: one is status-like or modal, the other evidential. The patterning …

Pine plantations on Typic Quartzipsamments in East Texas are difficult to establish. Forest management options following clearcutting are limited. A eight year regeneration study of the growth and survival of loblolly, Pinus tuedu, L. shortleaf, P. echinutu Mill., slash, P. elliofii Engelm and longleaf pines P. palustris Mill. was conducted to determine optimum tree species and treatments for reforestation; and to recommend practical alternative land uses and management strategies for Typic Quartzipsamrnents. During spring and summer of 1985 and 1986, we investigated activity patterns and habitat use of female northern bobwhite (Colinus virginianus) on 2 sites in south Texas. One site had been subjected to a short duration grazing (SDG) system and the other to a continuous grazing (CG) system. Nineteen females were radio-tagged in 1985 and 28 in 1986. Rainfall was above average in 1985 and below average in 1986; as a result, herbaceous ground cover was more dense in 1985 than in 1986. DNA sequences coding for 81% of the ompA gene from 24 chlamydial strains, representing all chlamydial species, were determined from DNA amplified by polymerase chain reactions. Chlamydial strains of serovars and strains with similar chromosomal restriction fragment length polymorphism had identical ompA DNA sequences. The ompA sequences were segregated into 23 different ompA alleles and aligned with each other, and phylogenetic relationships among them were inferred by neighbor-joining and maximum parsimony analyses. The neighbor-joining method produced a single phylogram which was rooted at the branch between two major clusters. One cluster included all Chlamydia trachomatis ompA alleles (trachoma group). The …

Glycine And Alanine Supplementation Of Culture Medium Enhances Development Of In Vitro Matured And Fertilized Cattle Embryos, K. Moore, K. R. Bondioli

One-cell cattle embryos were prepared by in vitro oocyte maturation and fertilization (IVM/IVF) and cultured with or without oviductal cells. Embryos were evaluated after 7 days in culture to determine the percentage developing from the 1-cell stage to the morula or blastocyst stage. The combination of glycine (2 mM) and alanine (1 mM) with oviductal cells (experiment 1) improved embryo development over that in control culture (29 vs. 13%; p < 0.05). An optimum response was obtained with 10 mM glycine and 1 mM alanine in coculture (experiments 2 and 3). In experiment 4, the effects of glycine (0 or 10 mM), alanine (0 or 1 mM), and the presence or absence of oviductal cells were tested. In the absence of oviductal cells, the addition of glycine, alanine, or glycine and alanine combined improved embryonic development over that in control medium (45, 33, 42 vs. 24%, p < 0.001, p < 0.05, p < 0.001, respectively). However, the effect of the combination of glycine and alanine was not different from that of glycine alone when oviductal cells were absent (42 vs. 45%, p > 0.10). Twenty-four high producing (36.7 kg/d, initially) Holstein cows in early lactation were used in a 63-d lactation trial to study the effects of low (28%), medium (31%). and high (37%) nonstructural carbohydrate concentrations in diets with added fat. Diets with isonitrogenous (17% CP) and fed as TMR with 41% sorghum silage. Nonstructural carbohydrate concentrations were altered by substitution of corn gluten feed and soybean hulls (50:50) for wheat and corn in the concentrate portion of the diet. Fat was added at 3% of DM in the form of yellow grease. Production of milk (38.5, 38.9, and 40.4 kg/d) and 3.5% …

A major objective of the Advanced Combustion Engineering Research Center (ACERC) is the development of comprehensive combustion models to help in the solution of critical national combustion problems. Computer models incorporate research and technology results from center projects and from external research programs. The synergistic integration of scientific knowledge that is expected from the NSF engineering research centers is demonstrated to a great extent at ACERC by the development of these software tools. The transfer of technology from ACERC to industry is also accomplished in part by the implementation of the models at industrial firms. The effort to develop such …

In a Terrebonne Basin (Louisiana) marsh, Cs dating indicated that vertical accretion (0.98cm yr ) was extremely rapid relative to other marshes, but insufficient to counter submergence (1.38cm yr ). Mineral and organic matter accumulation were similar to that in other marshes. This marsh is threatened by a positive feedback loop of plant flooding stress and inadequate vertical accretion. Inadequate plant growth limits vertical accretion, which further increases flooding and decreases plant production. Thus plant production partly determines the degree of submergence some coastal and estuarine marshes will tolerate. -from Authors 137 -1 -1

Dna Replication In Chloroplasts, Sabine Heinhorst, Gordon C. Cannon

Chloroplasts contain multiple copies of a DNA molecule (the plastome) that encodes many of the gene products required to perform photosynthesis. The plastome is replicated by nuclear-encoded proteins and its copy number seems to be highly regulated by the cell in a tissue-specific and developmental manner. Our understanding of the biochemical mechanism by which the plastome is replicated and the molecular basis for its regulation is limited. In this commentary we review our present understanding of chloroplast DNA replication and examine current efforts to elucidate its mechanism at a molecular level.

In this article, Professor Peter A. Alces investigates the tension that exists between the independent and interdependent nature of contractual relations arising in suretyship agreements and letter of credit transactions. This discussion is particularly timely as the American Law Institute is currently revising both the Restatement of the Law of Suretyship and Article 5 of the Uniform Commercial Code, "Letters of Credit." This article discerns a basic incongruity between the two revisions' treatment of interrelated multiple party rights and discusses the consequences that this incongruity can be expected to have upon commercial transactions.
