This paper addresses the long-standing problem of the unavoidable gaps that arise when expressing the intersection of two NURBS surfaces using conventional trimmed-NURBS representation. The solution converts each trimmed NURBS into an untrimmed T-Spline, and then merges the untrimmed T-Splines into a single, watertight model. The solution enables watertight fillets of NURBS models, as well as arbitrary feature curves that do not have to follow isoparameter curves. The resulting T-Spline representation can be exported without error as a collection of NURBS surfaces.

Background: Rickettsia felis is a flea-associated rickettsial pathogen recurrently identified in both colonized and wild-caught cat fleas, Ctenocephalides felis. We hypothesized that within colonized fleas, the intimate relationship between R. felis and C. felis allows for the coordination of rickettsial replication and metabolically active periods during flea bloodmeal acquisition and oogenesis. Methodology/Principal Findings: a quantitative real-time PCR assay was developed to quantify R. felis in actively feeding R. felis-infected fleas. In three separate trials, fleas were allowed to feed on cats, and a mean of 3.9 × 106 R. felis 17-kDa gene copies was detected for each flea. A distinct …

In monogamous species that exhibit extensive biparental investment, such as termites, both sexes are predicted to be selective when choosing a mate. Size-related traits are expected to be important in partner selection because the fat reserves of the colony founders sustain the incipient colony. Partner relatedness and heterozygosity determine the degree of inbreeding and genetic diversity within the colony and may thus also influence partner selection. To test these predictions, we investigated whether phenotypic and genetic traits influence mate choice and/or competitive advantage during pair formation of Formosan subterranean termites, Coptotermes formosanus (Isoptera: Rhinotermitidae). Background. The NV1020 oncolytic herpes simplex virus type-1 has shown significant promise for the treatment of many different types of tumors in experimental animal models and human trials. Previously, we described the construction and use of the NV1020-like virus OncSyn to treat human breast tumors implanted in nude mice. The syncytial mutation gKsyn1 (Ala-to-Val at position 40) was introduced into the OncSyn viral genome cloned into a bacterial artificial chromosome using double-red mutagenesis in E. coli to produce the OncdSyn virus carrying syncytial mutations in both gB(syn3) and gK(syn1). Different approaches have been used to simulate leaf area index (LAI) in soybean (Glycine max L. Merr). Many of these approaches require genotype-specific calibration procedures. Studies modeling LAI dynamics under optimal growth conditions with yields close to the yield potential of soybean have remained scarce. A sink-driven approach was developed and evaluated for LAI simulation in soybean under near-optimal environments. The rate of change in expanding leaf area was simulated using the first derivative of a logistic function accounting for plant population density, air temperature, and water deficit. Tumor necrosis factor alpha (TNFα) is a pleiotropic cytokine with several CNS physiological and pathophysiological actions including sleep, memory, thermal and appetite regulation. Short interfering RNAs (siRNA) targeting TNFα were incubated with cortical cell cultures and microinjected into the primary somatosensory cortex (SSctx) of rats. The TNFα siRNA treatment specifically reduced TNFα mRNA by 45% in vitro without affecting interleukin-6 or gluR1-4 mRNA levels. In vivo the TNFα siRNAα reduced TNFα mRNA, interleukin-6 mRNA and gluR1 mRNA levels compared to treatment with a scrambled control siRNA. The first decision of an injured worker suing his master for a workplace accident was reported in 1837, the year of Queen Victoria's ascension. The second Workman's Compensation Act, a comprehensive social insurance scheme, was passed in 1900, a few months before her death. The Article provides an initial account of the development of employers' liability to their servants for work-related injuries during the Victorian era. It demonstrates that English judges, and especially the Barons of the Exchequer, interpreted the law to resist employers' liability. Hydrogen peroxide and other reactive oxygen species are important signaling molecules in diverse physiological processes. Previously, we discovered superoxide dismutase (SOD) activity in extracellular protein preparations from fiber-bearing cotton (Gossypium hirsutum L.) seeds. We show here, based on immunoreactivity, that the enzyme is a Cu/Zn-SOD (CSD). Immunogold localization shows that CSD localizes to secondary cell walls of developing cotton fibers. Five cotton CSD cDNAs were cloned from cotton fiber and classified into three subfamilies (Group 1: GhCSD1; Group 2: GhCSD2a and GhCSD2b; Group 3: GhCSD3 and GhCSD3s). Five commercial botanical products (Shilianhua [SLH] tablets, Shiu Huo pills, Fenulyn, Bitter Melon, and Glucose Metabolic Support), available in the US market, with reported claims for regulation of metabolism were screened for their effect on body weight gain in high-fat diet-induced obese mice. Pilot results suggested that SLH tablets attenuated body weight gain, whereas Shiu Huo pills and Fenulyn tended to promote weight gain in the mice on the high-fat diet. Epigenetics refers to heritable changes in gene expression that are not attributable to changes in DNA sequence and impacts many areas of applied and basic biology including developmental biology, gene therapy, somatic cell nuclear transfer, somatic cell reprogramming, and stem cell biology. Epigenetic changes are known to contribute to aging in addition to multiple disease states. Epigenetic changes can be influenced by environmental factors that in turn can be inherited by daughter cells during cell division and can also be inherited through the germ line. Since the 1970s, environmental protection goals have gone from general statements of political desire to highly articulated systems of environmental regulation implemented by federal, state, and local governments. Environmental statutes have been enacted giving administrative agencies such as the U.S. Environmental Protection Agency (EPA) the responsibility for translating broad policy goals into specific regulatory requirements. Through its enforcement program, EPA seeks to assure that these general goals are achieved by individual actors. This Article examines a recent trend in EPA's practices, increased reliance on internal agency methods of enforcement. The authors of this article examined how pre-service teachers can use children's and young adult literature about disabilities to enhance understanding of individual differences through a bibliotherapeutic approach. An introduction to bibliotherapy is provided along with related literature from the field. Strategies for using children's and young adult literature to enhance the understanding of issues associated with disabilities are presented along with one teacher candidate's application of the literature in her classroom. The authors have also included a selected bibliography of children's literature on disabilities as well as relevant websites

Bellaes introduction of the fibroblast populated collagen lattice (FPCL) (1) has facilitated the study of collagen-cell interactions. As a result of the numerous modifications of the casting of FPCL's, the in vivo applications of these in vitro findings has been confusing. Here experimental FPCL contraction findings are viewed in regard to three proposed mechanisms responsible for lattice contraction. The cellular mechanisms responsible for generating FPCL contraction are: cell contraction, cell tractional forces related to cell locomotion, and initial cell elongation and spreading.

In the creation of autonomous virtual characters, two levels of autonomy are common. They are often called motion synthesis (low-level autonomy) and behavior synthesis (high-level autonomy), where an action (i.e. motion) achieves a short-term goal and a behavior is a sequence of actions that achieves a long-term goal. There exists a rich literature addressing many aspects of this general problem (and it is discussed in the full paper). In this paper we present a novel technique for behavior (high-level) autonomy and utilize existing motion synthesis techniques. The primary goal of the Western Arctic Linkage Experiment (WALE) was to better understand uncertainties of simulated hydrologic and ecosystem dynamics of the western Arctic in the context of 1) uncertainties in the data available to drive the models and 2) different approaches to simulating regional hydrology and ecosystem dynamics. Analyses of datasets on climate available for driving hydrologic and ecosystem models within the western Arctic during the late twentieth century indicate that there are substantial differences among the mean states of datasets for temperature, precipitation, vapor pressure, and radiation variables. Background. The isolation of green fluorescent protein (GFP) and the development of spectral variants over the past decade have begun to reveal the dynamic nature of protein trafficking and organelle motility. In planta analyses of this dynamic process have typically been limited to only two organelles or proteins at a time in only a few cell types. Results. We generated a transgenic Arabidopsis plant that contains four spectrally different fluorescent proteins. Nuclei, plastids, mitochondria and plasma membranes were genetically tagged with cyan, red, yellow and green fluorescent proteins, respectively. We explored the role of aquatic systems in the global N cycle using a spatially distributed, within‐basin, aquatic nitrogen (N) removal model, implemented within the Framework for Aquatic Modeling in the Earth System (FrAMES‐N). The model predicts mean annual total N (TN) removal by small rivers (with drainage areas from 2.6–1000 km2), large rivers, lakes, and reservoirs, using a 30′ latitude × longitude river network to route and process material from continental source areas to the coastal zone. Background: Decreased heart rate variability (HRV) is associated with a higher risk of mortality. Overall, postmenopausal women have lower levels of HRV than premenopausal women, which may be additionally complicated by lifestyle related behaviors such as physical inactivity and obesity. Though cardiorespiratory exercise training increases HRV, little is known regarding the exercise dose necessary to promote improvement.
Methodology/Principle Findings: Our primary aim was to measure HRV in post-menopausal women following 6-moths of exercise training. Mangrove forests are highly productive but globally threatened coastal ecosystems, whose role in the carbon budget of the coastal zone has long been debated. Here we provide a comprehensive synthesis of the available data on carbon fluxes in mangrove ecosystems. A reassessment of global mangrove primary production from the literature results in a conservative estimate of ∼-218 ± 72 Tg C a-1. When using the best available estimates of various carbon sinks (organic carbon export, sediment burial, and mineralization), it appears that >50% of the carbon fixed by mangrove vegetation is unaccounted for. Stable fly, Stomoxys calcitrans (L.) (Diptera: Muscidae), once only a pest of pastured cattle, has become a serious pest of range cattle in the United States. Because of the difficulties associated with stable fly management under range conditions, a pesticide-impregnated cloth target is being developed as a management tool. We conducted studies to determine the influence of weather, time, fabric type, insecticide type, and insecticide concentration on the mortality of stable flies from a susceptible laboratory colony exposed for 30 s to treated cloth targets. We utilized transmission electron microscopy and polymerase chain reaction to identify R. felis in the salivary glands of fed fleas and further define the distribution of R. felis within the arthropod host. We identified Rickettsia-like organisms in salivary glands using electron microscopy. Sequence analysis of portions of the Rickettsia genus-specific 17-kDa antigen gene and R. felis plasmid confirmed the morphological identification of R. felis in cat flea salivary glands. Cellular conditions experienced during energy-limited states - elevated calcium, shifts in cellular adenylate status, compromised mitochondrial membrane potential - are precisely those that trigger, at least in mammals, the mitochondrion to initiate opening of the permeability transition pore, to assemble additional protein release channels, and to release pro-apoptotic factors. These proapototic factors in turn activate initiator and executer caspases. How is activation of mitochondria-based pathways for the signaling of apoptotic and necrotic cell death avoided under conditions of hypoxia, anoxia, diapause, estivation and anhydrobiosis? The suprachiasmatic nucleus (SCN) exhibits circadian rhythmicity in fetal and infant rats, but little is known about the consequences of this rhythmicity for infant behavior. Here, in Experiment 1, we measured sleep and wakefulness in rats during the day and night in postnatal day (P)2, P8, P15 and P21 subjects. As early as P2, day-night differences in sleep-wake activity were detected. Nocturnal wakefulness began to emerge around P15 and was reliably expressed by P21. This chapter joins a discussion of instructional theory that has been ongoing for nearly a century. It departs in some ways from prior discussions: (1) it considers instructional theory as a species of technological theory rather than as a type of scientific theory, a view expressed more fully elsewhere (Gibbons, 2003a), (2) it adopts the viewpoint articulated in earlier chapters of this book that there are multiple distinct bodies of technological theory that pertain to the work of instructional designers, (3) it attempts to articulate a particular view of the nature of two of those bodies of theory by describing …
