Background: Although native to North America, the invasion of the aphid-like grape phylloxera Daktulosphaira vitifoliae across the globe altered the course of grape cultivation. For the past 150 years, viticulture relied on grafting-resistant North American Vitis species as rootstocks, thereby limiting genetic stocks tolerant to other stressors such as pathogens and climate change. Limited understanding of the insect genetics resulted in successive outbreaks across the globe when rootstocks failed. Here we report the 294-Mb genome of D. vitifoliae as a basic tool to understand host plant manipulation, nutritional endosymbiosis, and enhance global viticulture. Results: Using a combination of genome, RNA, …

For the first time, we report successful isolation of exosomes (small membrane-bound extracellular signaling vesicles) from saliva and salivary glands of partially fed or unfed ixodid ticks. Our data showed a novel role of these in vivo exosomes in the inhibition of wound healing via downregulation of C-X-C motif chemokine ligand 12 (CXCL12) and upregulation of interleukin-8 (IL-8). Cryo-electron microscopy (cryo-EM) analysis …

Reaching 1.5 And 2.0 °C Global Surface Temperature Targets Using Stratospheric Aerosol Geoengineering, Cheryl Harrison

Faculty Publications

Abstract. A new set of stratospheric aerosol geoengineering (SAG) model experiments has been performed with Community Earth System Model version 2 (CESM2) with the Whole Atmosphere Community Climate Model (WACCM6) that are based on the Coupled Model Intercomparison Project phase 6 (CMIP6) overshoot scenario (SSP5-34-OS) as a baseline scenario to limit global warming to 1.5 or 2.0 ∘C above 1850–1900 conditions. The overshoot scenario allows us to applying a peak-shaving scenario that reduces the needed duration and amount of SAG application compared to a high forcing scenario. In addition, a feedback algorithm identifies the needed amount of sulfur dioxide injections …

Faculty Publications

Background

The ability of blood-feeding arthropods to successfully acquire and transmit pathogens of medical and veterinary importance has been shown to be interfered with, or enhanced by, the arthropod’s native microbiome. Mosquitoes transmit viruses, protozoan and filarial nematodes, the majority of which contribute to the 17% of infectious disease cases worldwide. Dirofilaria immitis, a mosquito-transmitted filarial nematodes of dogs and cats, is vectored by several mosquito species including Aedes aegypti.

Methods

In this study, we investigated the impact of D. immitis colonization on the microbiome of laboratory reared female Ae. aegypti. Metagenomic analysis of the V3–V4 variable …

BACKGROUND:

Recent clinical and basic research implicated a strong correlation between NAFLD/NASH phenotypes with ectopic manifestations including neuroinflammation and neurodegeneration, but the mediators and critical pathways involved are not well understood. Lipocalin 2 (Lcn2) is one of the important mediators exclusively produced in the liver and circulation during NASH pathology.

METHODS:

Using murine model of NASH, we studied the role of Lcn2 as a potent mediator of neuroinflammation and neurodegeneration in NASH pathology via the liver-brain axis. RESULTS: Results showed that high circulatory Lcn2 activated 24p3R (Lipocalin2 receptor) in the brain and induced the release of high mobility group box …

Rabbiteye blueberry (Vaccinium ashei = V. virgatum) comprises much of the blueberry acreage in the southeastern USA states of Louisiana and Mississippi. Three genotypes of X. fastidiosa were identified from rabbiteye blueberry in Louisiana by multilocus sequence typing. A genotype that was found at two orchards, sequence type (ST) 42, was identical to one previously found in southern highbush blueberry in Georgia and two non-blueberry native species in Texas. Two newly identified genotypes shared most alleles with X. fastidiosa strains considered, like ST 42, to be part of a group that is believed to have resulted from recombination between X. …

Adaptive Responses To Maternal Nutrient Restriction Alter Placental Transport In Ewes, Ashley K. Edwards, Sorin M. Mcknight, Katharine Askelson, Jason R. Mcknight, Kathrin A. Dunlap, M. Carey Satterfield

Faculty Publications

Introduction: Maternal nutrient partitioning, uteroplacental blood flow, transporter activity, and fetoplacental metabolism mediate nutrient delivery to the fetus. Inadequate availability or delivery of nutrients results in intrauterine growth restriction (IUGR), a leading cause of neonatal morbidity and mortality. Maternal nutrient restriction can result in IUGR, but only in an unforeseeable subset of individuals. Methods: To elucidate potential mechanisms regulating fetal nutrient availability, singleton sheep pregnancies were generated by embryo transfer. Pregnant ewes received either a 50% NRC (NR; n = 24) or 100% NRC (n = 7) diet from gestational Day 35 until necropsy on Day 125. Maternal weight did …

A distinctive pattern is the so-called “circum-Amazonian distribution,” which is observed in species that do not occur in Amazonia but rather along a belt of forested habitats spanning south and east of Amazonia, the Andean foothills, and often into the Venezuelan Coastal Range and the Tepuis. Although this pattern is widespread across animals and plants, its underlying biogeographic mechanisms remain poorly understood. The Variable Antshrike (Thamnophilus caerulescens) is a sexually dimorphic suboscine passerine that exhibits extreme plumage variation and occurs along the southern …

Decision makers need an accurate understanding of aquifer storage trends to effectively manage groundwater resources. Groundwater is difficult to monitor and quantify since the data collected from monitoring wells are often available only at irregular and infrequent intervals. We present an open-source web application (app) to visualize groundwater data over time and automatically calculate changes in aquifer storage volume to help managers assess aquifer sustainability. This app uses a novel multi-linear regression (MLR) algorithm to impute missing data for infrequently sampled wells, using correlated data from other wells in the same aquifer. The app uses this MLR-imputed data to spatially …

Species have been commonly hypothesized to have high population densities in geographic areas which correspond to either the centre of the species geographic range or climatic niche (abundant–centre hypothesis). However, there is mixed empirical support for this relationship, and little theoretical underpinning. We simulate a species spreading across a set of replicated artificial landscapes to examine the expected level of support for abundant–centre relationships in geographic and niche space. Species niche constraints were modeled as a single axis which was related directly to population growth rates. We found strong evidence for abundant–centre relationships when populations follow deterministic growth, dispersal is …

Cooperative Policing Behaviour Regulates Reproductive Division Of Labour In A Termite, Qian Sun, Jordan D. Hampton, Austin Merchant, Kenneth F. Haynes, Xuguo Zhou

Reproductive conflicts are common in insect societies where helping castes retain reproductive potential. One of the mechanisms regulating these conflicts is policing, a coercive behaviour that reduces direct reproduction by other individuals. In eusocial Hymenoptera (ants, bees and wasps), workers or the queen act aggressively towards fertile workers, or destroy their eggs. In many termite species (order Blattodea), upon the death of the primary queen and king, workers and nymphs can differentiate into neotenic reproductives and inherit the breeding position. During this process, competition among neotenics is inevitable, but how this conflict is resolved remains unclear. Here, we report a …

Leishmania major is the causative agent of cutaneous leishmaniasis (CL). No human vaccine is available for CL, and current drug regimens present several drawbacks, such as emerging resistance, severe toxicity, medium effectiveness, and/or high cost. Thus, the need for better treatment options against CL is a priority. In the present study, we validate the enzyme methionine aminopeptidase 1 of L. major (MetAP1Lm), a metalloprotease that catalyzes the removal of N-terminal methionine from peptides and proteins, as a chemotherapeutic target against CL infection. The in vitro antileishmanial activities of eight novel MetAP1 inhibitors (OJT001 to OJT008) were investigated. Three compounds, OJT006, …

Over evolutionary time, pathogen challenge shapes the immunepheno type of the host to better respond to an incipient threat. The extent and direction of this selection pressure depend on the local pathogen composition, which is in turn determined by biotic and abiotic features of the environment. However, little is known about adaptation to local pathogen threats in wild animals. The Gentoo penguin (Pygoscelis papua) is a species complex that lends itself to the study of immune adaptation because of its circumpolar distribution over a large latitudinal range, with little or no admixture between ''different clades. Inthis study, we examine the …

Tropical cyclones constitute major disturbances producing direct effects (damage, mortality) in many coastal communities worldwide. Subsequent reassembly involves changes in composition and abundance for which the underlying mechanisms (deterministic and stochastic processes) are still not clear, especially for mobile organisms. We examined tropical cyclone-induced changes in composition and reassembly of entire insect communities in 16 Louisiana coastal salt marshes before and after Hurricane …
