Macrophages maintain tissue homeostasis by identifying and eliminating threats within their tissue microenvironment. Pattern recognition receptors allow macrophages to recognize and internalize specific ligands while macropinocytosis allows the internalization of all extracellular solutes from their environment. Without pattern recognition receptors, pathogens may grow unchecked if they cannot be detected, and without macropinocytosis, macrophages struggle to detect and move around their environment. This thesis presents the results of CRISPR/Cas9 whole-genome screens used to identify the regulators of both endocytosis (Chapter 2) and macropinocytosis (Chapter 3). Termites in the genus Reticulitermes (Blattodea: Rhinotermitidae) are distributed across the eastern United States, including the southern Appalachian Mountains, a region incredibly rich in biodiversity. The eastern subterranean termite, Reticulitermes flavipes, has been uninentionally introduced to South America and Europe, and is predicted to further expand its geographic range. My goal was to determine how eco-evolutionary processes, operating at both long and short timescales, may have contributed to R. flavipes becoming an invasive species. Ground nesting bird species are on a considerable decline and research efforts are being made to increase these populations throughout the Great Plains. Ground nesting bird communities found in the Northern Great Plains are driven greatly by varying amounts of cover and area size. Past research implemented patch-burn grazing to increase structural heterogeneity and to increase grassland bird habitat in the tallgrass prairie. While bird populations were very responsive to this management in the Northern Great Plains fire management is viewed negatively, especially for Midwest cattle ranchers. There has been a recent push for fortification of fruit juices due to rising consumer health concerns such as obesity and high sugar consumption. Bacterial endophytes have the capability to enhance plant growth by producing plant growth hormones, solubilizing phosphates, suppressing pathogenic fungi, and reducing plant stress hormones. These capabilities make them desirable limiting the amount of nutrients and pesticides that are applied to crops. Through these assays and isolations, it is possible to identify novel bacterial species. In-vitro testing had shown 9 of the 20 isolates possess the ability to produce indole-3-acetic acid (IAA) with Pantoea agglomerans BC09 producing a concentration of 30.2 ng/μl over 4 days. Ultrasound technology provides cattle breeders a relatively quick, non-invasive, and economical way to gather carcass data on live animals. Ultrasound provides the means to accurately predict body composition and develop estimated breeding values; however, national cattle genetic evaluations assume homogenous additive genetic and residual variances. Fusarium head blight (FHB) is a severe disease of wheat (Triticum aestivum L.). It not only reduces the quantity of the harvested grains but also decreases the grain quality due to mycotoxins contamination, especially Deoxynivalenol (DON). Qfhb1 (or simply called Fhb1) is the most important quantitative trait locus (QTL) for FHB resistance. Our lab has previously identified wheat gene WFhb1-1 (aka. Background: Childhood obesity is continuing to rise, leading to long-term health consequences. Research shows that rural populations have higher rates of childhood obesity. There is a lack of research on how the home environment may affect this health disparity. The high fiber content of corn dried distillers’ grains with solubles (DDGS) can limit its utilization in swine diets. Pretreatment with heat and enzymatic predigestion of whole stillage (WS; slurry material that is dried into DDGS) can alleviate negative effects of dietary fiber and can improve digestive and fermentation characteristics of the feedstuff. Student retention has historically been an important discussion in higher education. While the importance of student retention remains, a new generation of students have filled colleges and universities. This study examines how interactions with faculty influence Generation Z college students’ overall satisfaction and student retention. The quantitative study used the NSSE survey instrument at a public land-grant institution in the Midwest. The results of a Mann-Whitney U analysis indicated a significant difference in student satisfaction reported between students who returned to the same institution the following year and students who did not return. Anabaena sp. PCC 7120 is a filamentous, nitrogen-fixing cyanobacterium that uses spatial separation to perform photosynthesis and nitrogen fixation simultaneously. Under fixed nitrogen limiting conditions, Anabaena 7120 forms specialized cells, called heterocysts, to fix nitrogen. In this research, I sought to add to the knowledge surrounding the process of heterocystous nitrogen fixation in Anabaena 7120. Several decades of above average precipitation in South Dakota has increased the area of saline and sodic soils, which reduce crop yields and inhibit sensitive plant growth. Saline and sodic soils are difficult to remediate using traditional agricultural crops. Establishing salt tolerant perennial species may restore productivity to salt affected areas. The thesis presents new results for the reflection and transmission coefficients for Cherenkov photons in a strong magnetic field at the interface between the fused quartz of and the surrounding nitrogen gas within the iTOP particle detector, a sub-system of the Belle II experiment. The iTOP is a component of the particle identification system at Belle II which distinguishes between kaons and pions in final states after collision. This thesis connects late nineteenth-century southern Democrat and former Confederate L.Q.C. Lamar to the status politics of the Northeastern “small imperialist elite.” Moreover, this work traces Senator, Secretary of the Interior, and Associate Justice Lamar’s legacies in American political thought, such as a regional memorialization and the example of his national influence through the policies of President Theodore Roosevelt specifically. Though he inspired generations of politicians with varying views, Lamar himself maintained a strict constructionist political philosophy and worked for home rule throughout his career. The first essay attempts to explain the heterogeneity in the stock market’s reaction to a firm’s lobbying for good efforts. It is hypothesized that higher (a) CEO equity-to-pay ratio, (b) marketing influence in the TMT, (c) marketing capabilities, (d) corporate social performance, and (e) institutional ownership, will be associated with less negative abnormal stock returns. Results indicate that firms with a greater amount of marketing influence in the top management team experience greater negative abnormal returns. The goal of the present study was to develop a nanoemulsion (NE) system for efficient and prolonged delivery of Fingolimod Hydrochloride (FTY720; FT), to the posterior segment of the eye through the topical route. The overarching goal of this research is to explore the biological mechanisms of aging and formulate novel therapeutic approaches to mitigate age-related impairments. The presence and pervasiveness of ~24 hour (circadian) rhythms in molecular and behavioral dynamics suggests that the mechanisms regulating these rhythms are integral to biological aging and possibly amenable to therapeutic interventions. Based on evidence presented in this dissertation, we conclude that the endocannabinoid system is a mediator of both central and peripheral circadian physiology. Additionally, the hormetic pharmacological properties of the synthetic cannabinoid CP55940 suggest that bidirectional modulation of circadian rhythms via cannabinoids is possible. Chapter …

Natamycin (NT) is a front-line drug in the management of ocular fungal infections (OFI). An ophthalmic marketed NT suspension (Natacyn®) is currently the only FDA approved medication prescribed in the pharmacotherapy of OFI. Current NT pharmacotherapy requires frequent topical administration (every hour or 2-hours over 6-8 times in a day) due to it being instilled as eye-drops. This leads to higher precorneal losses and subsequent poor permeation and bioavailability.
