Numerous studies have detected anomalous precipitation patterns occurring up to 50 km downwind of major cities, providing major evidence that cities inadvertently modify precipitation and atmospheric circulations. Louisville is one such major city with a growing body of evidence of inadvertent precipitation modification. Despite these efforts, the physical mechanisms driving small-scale and unintentional changes in urban precipitation are little understood, being rooted largely in theory rather than in physical observations. This study seeks to build upon previous research by analyzing ground-based precipitation observations recorded by a high-density gauge network located within approximately 40 km of Louisville’s urban center. In the digital inequality literature, the popular notion of a “digital divide” is frequently used to discuss digital inequality; however, this framework is overly simplistic and cannot adequately capture the complex nature of digital inequality. Multiple studies have investigated the occurrence of severe convective-related winds and have increased our understanding of the forces driving severe winds and their spatial and temporal patterns. Data for these studies have come from airport stations maintained by the National Weather Service. Their standardization across the United States makes them ideal for research, but they are limited in their distribution. This study aims to create a similar climatology of severe surface level winds using a mesoscale network (“mesonet”). This study examines the impact of experiences of colorism on the crime trajectory of African Americans. It is qualitative in nature and utilizes semi-structured interviews with incarcerated African American people. Academic underachievement among Black students, particularly Black male students, is a troubling trend that has long attracted scholarly attention. The prevailing consensus is that the shortcomings of some Black male students in academic achievement may be attributable to traumatic experiences arising from environments of violent, inner-city poverty that disproportionately affect Black men. Traumatic experiences have been shown to negatively affect Black men’s self-perception, emotions, self-image, and social and cognitive skills, with consequences in the form of measured shortfalls in retention and graduation rates. This study sought to understand the underlying structural reasons why some rural counties have better or worse than expected health outcomes using a positive deviance (PD) approach. The study is aimed to determine the role of avocado DGAT1 and PDAT1 in seed oil synthesis. Triacylglycerol (TAG) has a nutritional and industrial value and is essential for plant growth. DGAT1 and PDAT1 catalyze the final step of TAG Assembly. We hypothesized that both PaPDAT1 and PaDGAT1, although predominantly expressed in non-seed tissues, could contribute to oil accumulation in seeds. Agrobacterium transformants with PaPDAT1 and PaDGAT1 cloned in pCAMBIA were generated to test this. Conceptualizing, quantifying, and evaluating frailty in human skeletal remains is critical to understanding and interpreting physiological health and stress among past populations. While many researchers focus on frailty in bioarchaeological samples, developing models for mortality risk and individual- and population-specific indices of stress, no current methods directly address frailty in forensic contexts. While different violence typologies exist, the U.S., prioritizes interpersonal violence as its primary target of intervention, often through structural means that foster and reinforce interpersonal violence. Given the diversity and complexity within immunoglobulin (IG) loci, effective mouse models first require characterization of intra-strain differences and construction of high-quality reference assemblies for IG loci in several representative strains. To understand light chain germline diversity across biomedically significant mouse strains, we profiled the expressed IGK and IGL repertoires of 18 commonly used laboratory mouse strains using AIRR-seq. Across strains, we observed germline IGKV sequences shared by three different IGK haplotypes and a more conserved IGLV germline repertoire among common laboratory strains. This dissertation explores the relationship between mitochondrial physiology and development of therapeutics. Mitochondrial dysfunction is associated with both acute and chronic forms of pathophysiology. This work aims to address development efforts at the cell culture and drug-target levels with respect to mitochondria. At the cell culture level, I characterize an approach that has been shown to improve the physiological dependency on mitochondria in tumor-derived cells. I demonstrate that prolonged replacement of glucose with galactose in culture medium induces a global metabolic shift in hepatocellular carcinoma (HepG2) cells to closer reflect a primary hepatocyte phenotype (Chapter 2). This dissertation forwards a rhetorical theory of institutions that centers their genre-based communications, redefines institutions in relation to organizations, and contributes to the field’s understanding of how communities collaboratively rewrite entrenched institutionalized practices from the outside, thus reducing the harm that institutions can enact. Few studies address the lived experiences of transgender and gender nonconforming (TGNC) students in engineering. Grounded in critical trans politics (Spade, 2015), this dissertation contributes to the literature on TGNC students in engineering by examining their experiences negotiating their identities while navigating interrelated systems of oppression in a field dominated by White, heterosexual, cisgender men. Using a critical constructivism framework, I conducted a narrative inquiry to explore the lived experiences of five TGNC students in engineering programs. Participants experienced TGNC oppression at their universities, built LGBTQ+ and TGNC communities, and described more welcoming climates in non-engineering contexts compared to engineering. …

It is now an almost foregone conclusion that classic depictions of vampirism resonate with contemporary queer audiences. A sympathetic response to the monster’s persecution is often the key factor in these arguments, yet little attention is paid to the textual details that prompt such a process of identification. This study posits that the iconography used to establish a connection between monstrosity and non-normative sexuality has its origins in Victorian Gothic fiction, whose descriptions of vampirism were assimilated into the discourse of the fin-de-siècle medical field known as sexology. Retinal fibrosis is detrimental to vision. Retinal pigment epithelial (RPE) cells contribute to several retinal fibrotic diseases. Upon exposure to TGF-β, a key fibrotic cytokine, RPE cells trans-differentiate to myofibroblasts marked by the integration of α-SMA fibers into F-actin stress fibers, which confer strong contractility.
