Neutrophils and macrophages accumulate at sites of inflammation and cause chronic inflammation leading to various diseases. Therefore, to better understand chronic disease pathways it is important to investigate the properties of macrophage accumulation in inflamed tissues. The I-domain of the macrophage receptor integrin aDb2 plays a vital role in macrophage retention by binding to CEP (carboxyethyl pyrrole), a ligand available at inflammatory sites. This thesis mainly focuses on evaluating the binding site within integrin aDb2 that binds carboxyethyl pyrrole (CEP)-modified proteins. The aim of this qualitative research was to gain an in-depth understanding of how teachers of toddlers (aged 25-36 months) described sound and music learning opportunities and experiences provided to children in their learning environments. In this research, teachers were asked how their personal and professional experiences with sound and music influenced their planning of toddlers’ sound and music learning opportunities. The design of this study was in-depth interviews of six toddler lead teachers in three childcare sites. Data were gathered through photographs, planning documents, and in-depth interviews using a university IRB-approved video conferencing program. Findings showed that all participants …

Infertility impacts numerous individuals during their reproductive journey. Yet, there is little research and information available that examines if having a history of infertility goes on to have further implications after an individual conceives and gives birth. Thus, the current study aimed to examine if infertility impacts the postpartum period, and if there were any particular risk or resiliency factors that contributed to this relationship. In this study, women aged 18 to 50, who had given birth within the last 12 months, participated in an online survey that assessed a variety of physical and mental health constructs. This dissertation was 2 parts of investigations of various acute responses to high-volume combined accentuated eccentric loading and rest redistribution (AEL + RR). Resistance-trained men (n = 12, 25.6 ± 4.4 years, 1.77 ± 0.06 m, 81.7 ± 11.4 kg) completed a back squat (BS) 1 repetition maximum (1RM) and weight releaser familiarization session. Three BS exercise conditions (sets × repetitions × eccentric/concentric loading) comprised AEL + RR 5: 3 × (5 × 2) × 110/60%, AEL + RR 2: 3 × (2 × 5) × 110/60%, and traditional sets (TS): 3 × (1 × 10) × 60/60% 1RM. Urbanization is among the greatest threats to biodiversity on the planet, and is projected to increase in magnitude in coming years. This growing threat makes it important to better understand how urbanization may impact vital ecosystem services, such as pollination. Studies on the impact of urbanization on pollination vary wildly in their findings. This study analyzed multiple aspects of pollination success in common milkweed populations varying in urbanization level, including insect community composition, pollen removal/deposition, and fruit/seed production. Nigerian immigrant women undergo constant navigation of their personal identities when conflicting cultural dynamics sometimes engender a balancing act between their personal beliefs and the ongoing process of acculturation. Their new Southern environment offers its own traditional mores, as well as greater opportunities for economic advancement. This places Nigerian Immigrant Women in a position where they must reconcile their desires for personal independence and empowerment with societal expectations that emphasize more traditional gender roles. Positive parenting practices and secure attachments are consistently linked to healthy child outcomes (Ainsworth & Bowlby, 1991; Waters et al., 2000). Research on cognitive processes that scaffold parental behaviors which contribute to secure attachment is an essential contribution to the literature, particularly given the potential for early intervention with at-risk families. Snake vertebrae are common in the fossil record, whereas cranial remains are generally fragile and rare. Consequently, vertebrae are the most commonly studied fossil element of snakes. However, identification of snake vertebrae can be problematic due to extensive variation. This study utilizes 2-D geometric morphometrics and canonical variates analysis to 1) reveal variation between genera and species and 2) classify vertebrae of modern and fossil eastern North American Agkistrodon and Crotalus. Salmonella is a leading bacterial cause of foodborne illness worldwide. During a previous study investigating the enzymes responsible for regulating cyclic-di-GMP concentrations, a mutant in the cyclic-di-GMP-specific phosphodiesterase STM3615 was identified that displayed a phenotype characterized by decreased survival on agar plates and a shorter bacterium length. I was able to determine that the periplasmic domain of STM3615 was responsible for this phenotype, not the enzymatic phosphodiesterase domain. Based upon a bioinformatic analysis of the protein, I then hypothesized that the periplasmic domain of STM3615 was interacting with a periplasmic protein to give rise to this phenotype. Compared to other instruments for chemical analyses, electrochemical analyzers are relatively simple, inexpensive, easy to miniaturize and require little-to-no maintenance. However, like all commercially available instruments, commercial electrochemical analyzers like potentiostats primarily operate as black boxes with manufacturers providing little or no information about internal circuitry and programming. This practice can limit a researcher’s ability to develop new techniques or adapt an instrument for applications outside its typical use. Underexplored environments such as soil samples continue to be an untapped source of bacterial strains with great potential to produce secondary metabolites for medicinal applications. As a result, these microorganisms represent a broad and yet unknown reservoir of new strains capable of producing these novel compounds. The Gray Fossil Site (GFS) preserves an early Pliocene sinkhole pond that accumulated a rich flora and vertebrate fauna. Fossils of fish are incredibly common at GFS, representing thousands of specimens recovered to date. Comparison with extant species reveals a depauperate freshwater fish fauna consisting of only two species: Lepomis sp. and Micropterus sp. The absence of fishes that are ubiquitous in modern streams suggests no perennial surface hydrologic connection between the Gray Fossil Site and the nearby Holston River. Limited opportunities for dispersal into the pond and deleterious conditions such as eutrophy and high competition may have resulted in …

As antimicrobial resistance persistently disrupts the treatment of microbial infection, natural product chemistry has played a significant role in identifying novel drugs with novel modes of action critical to getting ahead of resistance. The primary goal of this project is to extract and identify potential novel antimicrobial natural products produced by Arthrobacter sp. Microsporidia are a distant fungal pathogen that have severe clinical consequences for the immunocompromised. Previous work identified a microsporidian pathogen protein termed Microsporidian ADAM or MADAM. This protein has close sequence homology to other ADAM proteins (A Disintegrin and Metalloproteinase) in two microsporidian species, Encephalitozoon intestinalis and E. cuniculi. ADAM proteins have a wide range of functions, including binding to integrins and host signaling. It is known that many pathogens manipulate integrins to invade host cells, and it is predicted that microsporidia are also exploiting this host target. This phenomenological qualitative study examined school leaders' perceptions of using restorative practices within their schools. Participants included twelve school leaders who had experience with utilizing and implementing restorative practices. School leaders included: principals, assistant principals, deans, and administrative assistants. Research indicates that restorative practices are adapted to support specific school goals. Restorative practices are not practiced in isolation, as school leaders often couple restorative justice with social-emotional learning, trauma-informed practices, and other social-emotional supports. Restorative practices are tied closely to CASEL’s social emotional framework.

Glioblastoma is the most commonly diagnosed central nervous system primary malignancy and it is considered a terminal diagnosis with few treatment options available. Glioblastoma tumors frequently develop treatment resistance due in part to their highly heterogenic nature. The heterogeneity of glioblastoma is partially attributed to the presence of glioma stem-like cells (GSC), which are highly invasive and resistant to chemotherapy and irradiation treatments. Signaling of the receptor tyrosine kinase MET is a known regulator of GSC. Glioblastoma patients have an increasingly poor prognosis that corresponds with increasing MET expression. Infant and Early Childhood Mental Health (IECMH) providers engage in uniquely relational work with high-risk families that expose them to the full advantages (i.e., compassion satisfaction [CS]) and disadvantages (e.g., burn-out [BO]) of a helper role. Though the pandemic seemed poised to escalate disadvantage, most early pandemic studies found high CS alongside elevated BO. Unfortunately, as COVID-19 has continued, CS has declined while BO has increased. Given the delayed COVID-19 impact on children, these changes may be particularly acute for IECMH providers. Histological analysis of long bone thin sections is commonly used to infer growth rates and ecology of extinct vertebrates, particularly within Archosauria. However, most comparative neontological studies have used small samples of captive individuals, limiting the scope of variation. To fill this gap, 44 femoral thin sections of wild Alligator mississippiensis were prepared and analyzed. Comparison of slides revealed that larger individuals from cooler climates tend to show more LAGs compared to southerly A. mississippiensis of similar size, however, there is considerable variation. The Turin Pit locality (Monona county, Iowa) has been known to paleontologists since 1908, yet the age of the fauna has been unclear. Early paleontologists considered Turin Pit to date to a pre-Illinoian interglacial (the “Aftonian).” Subsequent researchers suggested it dated to the last glaciation. This study provides a partial list of mammals in the Turin Pit fauna, and together with stratigraphic information, uses the known age ranges of taxa to estimate an age for the assemblage. The purpose of this project was to identify the impact of in-person, hands-only CPR education for community members on their confidence and ability in performing CPR. Bystander-initiated CPR is an important aspect in the survival and outcomes of those that experience a cardiac arrest in the community. The hands-only CPR method was developed to help increase bystander-initiated CPR and improve outcomes for those experiencing a cardiac arrest at home. Abstract The problem investigated in this study was unknown opportunities that exist for educators to meet the academic needs of students who are affected by combat parental deployment and how this challenge of combat deployment disrupts the family’s dynamic, thus challenging their children academically. The Air Force, Army, Navy, Marines, and Coast Guard made up about 1% of the total United States population in 2015. Most of these people were married, 42% had children, and 54% of their children were in kindergarten to 12th grade. Junior officer attrition has increased over the last two decades. The early departure of army officers leaves a dearth of middle- and senior-level leadership, creating risk for future military readiness. West Point leaders theorized that new officers’ poor person–job fit within their specific career fields contributed to attrition. West Point implemented a talent-based system to place new officers into branches that should offer a stronger person–job fit, hoping it would reduce officer attrition. This study examined the relationship between emotional intelligence and conflict management styles in Black women leaders in the United States. The lack of literature surrounding Black women leaders' emotional intelligence or conflict management styles presented an opportunity to offer quantitative data connected to these constructs. A cross-sectional survey design was used to administer three surveys the Situational Test of Emotional Management- B, the Kraybill Conflict Style Inventory, and a modified version the Multidimensional Inventory of Black of Identity, that focused on Black female identity.
