The limited regenerative capacity of adult cardiomyocytes (CMs) is manifested in prevalent morbidity and mortality associated with ischemic heart disease. We previously demonstrated that a combination of four cell cycle factors (4F) promotes efficient cell division in differentiated CMs. The temporal transcriptional reprogramming in the 4F-transduced CMs indicates that epigenetic modifications mediate the cell cycle progression. Through small molecule screening, we evaluated the epigenetic impact on CM’s response in the 4F-transduced CMs. We identified the histone demethylase 5B (KDM5B), which through in vitro and ex vivo genetic knockdown and overexpression studies, exhibited a modulative effect on CM’s cell cycle plasticity …

While there is a body of research about student conduct policies and procedures, there is a lack of research about how different role groups work together to implement these procedures and have a positive working interpersonal relationship. This sequential mixed methods case study design takes place in a large urban school district. In this study, I investigate the perceptions that educators have on the implementation of student conduct procedures in their school. A thorough understanding of receptor regulation is imperative to predict expression in varying contexts of disease or treatment. Lymphocyte surface receptors are often used as biomarkers and drug targets, making them particularly important for study. For receptors of debated functionality, such as the Fc receptor for IgM (FcMR), understanding regulation can also help to predict expression in vivo to supplement hypotheses of biological roles. This study investigated perceptions of knowledge and preparation regarding administrative responsibilities among in-service and preservice music teachers. Samples were selected randomly through NAfME and NASM databases. Data collection and analysis answered research questions asking how knowledgeable these groups felt about administrative topics, how they felt their undergraduate coursework prepared them for administrative responsibilities, and if there were any significant differences in preparedness among different subject areas taught by in-service directors. The findings report in-service teachers felt more knowledgeable of all administrative topics. Objective: This study examined differences between users and non-users of campus food pantry, conducted communication audit, and made recommendations for future food pantry communication. Methods: Cross-sectional twofold study, first analyzing existing intrapersonal, interpersonal, and environmental data to test predictors of campus food pantry usage characteristics, using inferential statistics. Secondly, conducting a communication audit that evaluates the reach of student communication. Results: Significant differences were found between users and non-users of food pantry that support hypotheses in relation to gender, age, race, class, marital status, housing type, housing description, and Pell eligibility. Findings could not support hypothesized differences in first-generation status. …

Loss of myelin causes severe neurological disorders and functional deficits in white matter injuries (WMI) such as traumatic spinal cord injury (SCI). This dissertation is focused on autophagy in OL development and the OL translatome after SCI. Chapter I describes the history of myelin, OL development, and their involvement in neurodegenerative diseases and SCI. The proteostasis network, in particular autophagy, and its contributions to white matter pathology is discussed. The nucleus of solitary tract (NST) and parabrachial nucleus (PBN) represent the first and second central synapses of ascending gustatory information. Neural processing in these nuclei is influenced by descending input from forebrain regions such as the central nucleus of the amygdala (CeA). In mice, we have shown that somatostatin (Sst) expressing neurons of CeA that project to NST and PBN are largely distinct cell populations and optogenetic inhibition of the CeA/Sst-to-NST subpopulation increases the intake of high concentrations of quinine with no apparent effect on sucrose intake. Fibrinogen (Fbg) is a coagulation protein critical for clot formation. Coagulation Factor XIII (FXIII) is a calcium-dependent transglutaminase that crosslinks reactive glutamines (Q) and lysines (K) between fibrin and other anti-fibrinolytic proteins. In the presence of Ca2+, FXIII could be activated non-proteolytically (FXIII-A°), or proteolytically by thrombin (FXIII-A*). Significant increases in clot stability and red blood cell retention are linked to FXIII activity in the fibrinogen αC region (Fbg Aα 221 – 610). This region contains several FXIII-reactive glutamines and lysines, as well as a binding site for FXIII-A* (Fbg αC 389 – 402) that includes a key …

The purpose of this phenomenological study is to examine the influence of power on racial identity and social awareness of minority high schools’ students in Bullitt County, Kentucky. The research will come from the student perspective of those attending high school with students predominantly from another racial background. Using purposeful sampling, 12 students participated in semi-structured interviews, providing insight into three research questions regarding racial identity, social power, and social awareness. There have existed techniques for manipulating the perception of images since the creation of the camera. The techniques examined in this paper are editing, staging, and the use of captions. In recent years, perception has become a powerful tool for political parties to manipulate. This manipulation of perception has led to an increasing distrust in the images the media presents to the public. The purpose of this qualitative study was to investigate how the perceptions about family engagement structures influence the sense of belonging for Black families at their child’s school. Qualitative participatory action research (PAR) design was used for this study and purposeful homogenous sampling (Creswell, 2012) was used to select participants. The participants for this study were Black parents/guardians of students in grades K-12 in Jefferson County, Kentucky. This dissertation develops an ambient rhetoric: a living practice that is more disclosive than communicative, more entangled than intersectional, and more subtle than explicit and evidential. Rather than exploring the symbology, iconography, texture, and artistic integrity of Khajuraho monuments, I claim that Khajuraho monuments aim at convincing visitors with their persuadability and affectability inherent in their material bases, architectural panels, fractal design, allegorical cast, aura of attunement, and the environment in which the monument complex is nestled. In this dissertation, I examined the behavior of foraging twig ants, explored the sensory profile of their homing behavior, and investigated the role of habitat use in shaping the sensory neurobiology of species within the genus Pseudomyrmex (Hymenoptera: Formicidae). In chapter one, I reviewed the ecology of Pseudomyrmex, and introduced the focal species, P. boopis, a neotropical understory-dwelling twig ant. In chapter two, I described the natural history of P. boopis on Barro Colorado Island, Panama—specifically, their nesting habits, activity rhythm, and foraging behavior on understory surfaces. Water is stable, benign, and green solvent. Its usage as a solvent in organic synthesis is significantly enhanced by micellar catalysis. However, replacing water as a reaction medium in organic synthesis is not sufficient to meet the sustainability challenges—excessive organic solvents are still required for product isolation and purification. Notably, solvents used in syntheses contribute to more than 80% of waste generation. Networks have the critical ability to represent the complex interconnectedness of social relationships, biological processes, and the spread of diseases and information. Exponential random graph models (ERGM) are one of the popular statistical methods for analyzing network data. ERGM, however, struggle with computational challenges and degeneracy issues, further exacerbated by their inability to handle high-dimensional network data. Bayesian techniques provide a promising avenue to overcome these two problems. Large knowledge gaps concerning the effect of ocean surface waves on near-surface vertical distributions of temperature and humidity in the marine atmospheric surface layer exist due to practical limitations and sensor fidelity challenges of direct measurements. Wave effects on these distributions between the wave trough and crest are least studied within the literature. These scalar distributions influence atmospheric refractivity, which can lead to anomalous propagation of electromagnetic energy. Measurements of temperature and humidity are classically made using rocket- or radiosondes and fixed weather stations, and can utilize tethered profiling systems. The Ordovician Utica shale play is rapidly developing throughout the Appalachian Basin. The play is a mixed carbonate-siliciclastic system that grades from primarily carbonates at its base in the Lexington/Trenton limestones into primarily shale within the Utica shale. As of January 2021, it is the third highest-producing dry shale gas play in the contiguous U.S., although it is known to produce wet gas and oil in various parts of the basin. This study sought to explore the lived experiences of Dysphoric Milk Ejection Reflex (D-MER) during postpartum breastfeeding. A phenomenological method based on Giorgi’s phenomenological underpinnings was used. The setting for this study was virtual interviews in a secure online platform. The sample consisted of 13 participants, self-identifying as experiencing D-MER. Semi-structured interviews, with thematic analysis of transcription were conducted. Four major themes were identified: 1) Breastfeeding concessions and toleration; 2) Something does not feel “normal”; 3) I remember a feeling like this before 4) This is real, and recognition is validating. This thesis explores Afrofuturism and positions Afrofuturist drama as a new, 21st -century genre of theatre. My thesis defines Afrofuturism and characteristics of Afrofuturist plays by Black women. My examination of Afrofuturist pieces and performance strategies is for Black female performers working in Afrofuturism. Post-transcriptional RNA modifications including N6-methyladenosine (m6A) regulate mRNA stability, splicing, and translation. My research examined m6A in two disease models: breast cancer (BCa) and non-alcoholic fatty liver disease (NAFLD). Acquired resistance to endocrine therapies (ET) develops in approximately 20% of BCa patients with estrogen receptor α positive (ER+) tumors following treatment. The mechanisms by which tumor cells evade ET are not completely understood. Using a cell line model, we investigated the role of an m6A reader protein, HNRNPA2B1 (A2B1) that is upregulated in ET-resistant ER+ BCa cells. This dissertation makes significant contributions to the field of ophthalmology, addressing the segmentation of retinal layers and the diagnosis of diabetic retinopathy (DR). The first contribution is a novel 3D segmentation approach that leverages the patientspecific anatomy of retinal layers. This approach demonstrates superior accuracy in segmenting all retinal layers from a 3D retinal image compared to current state-of-the-art methods. It also offers enhanced speed, enabling potential clinical applications. The proposed segmentation approach holds great potential for supporting surgical planning and guidance in retinal procedures such as retinal detachment repair or macular hole closure. Dryolestoids are extinct cladotherians mammals from the Jurassic and Cretaceous periods. I describe a collection of dryolestoid specimens from the Late Cretaceous localities of Cerro Tortuga (Allen Formation), Anfiteatro 1, and Shining (both La Colonia Formation) from Patagonia, Argentina. Using comparative morphology, I identify a new species of meridiolestidan dryolestoid based on eleven specimens across both formations. The new species’ recovery from La Colonia Formation represents the first dryolestoid connection between the two approximately contemporaneous formations. The species’ morphology may represent an ecological shift within Meridiolestida from insectivory to herbivory, showing a transition in characters between the plesiomorphic sharp-toothed meridiolestidans …

This study is an autoethnography written in the form of layered accounts with a theoretical framework of queer theory. It was based on my journey as an LGBTQ+ individual and educational leader. The purpose of this study was to utilize my experiences as an LGBTQ+ educational leader to investigate the conditions that supported and limited my self-efficacy as an LGBTQ+ educational leader.
