Unraveling The Unknown: Characterization Of A Novel Antimicrobial Compound Found In Klebsiella Pneumoniae, Jasmal Cruz Perez

Klebsiella pneumoniae, a pathogenic bacterium resistant to numerous antimicrobials, is a worldwide health issue due to its ability to cause healthcare-associated diseases such as pneumonia and urinary tract infections. This study investigated an antimicrobial compound secreted by K. pneumoniae. The compound was found to possess inhibitory activity against Enterobacteriaceae members, including other Klebsiella pneumoniae strains, Citrobacter freundii, and Enterobacter cloacae. The inhibitory activity is mediated via a secreted protein that is active across a wide range of pH and temperature conditions. Higher concentrations of the compound enhanced antimicrobial activity. The antimicrobial protein suppressed biofilm formation during both …

Physics-informed deep learning is a methodology in artificial intelligence aimed at combating the large training data requirement and the barrier of domain awareness that deep learning architectures commonly face in applications. Stochastic modeling integrated into the predictive models provides that domain knowledge. Variations of the Intelligent Driving Model impact the learning behaviors of the joint-training architecture. This thesis examines the effect of substituting the standard linear Intelligent Driving Model with a modified nonlinear version, as applied to real human driving behavior on the I-80 interstate. The artist, Cait Maltbie, discusses their Master of Fine Arts exhibition, Inner Bonds, which was installed at Slocumb Galleries in Johnson City, TN, from March 24, 2025, through April 4, 2025, with a public reception held on March 27, 2025. The exhibition consisted of multi-disciplinary work: zines, screenprints, and a sculptural installation mimicking the scenery and emotional environment of the imaginary Inner Bonds world. Alcaligenes faecalis, a generally nonpathogenic, Gram-negative bacterium, has been observed by our laboratory to possess antagonistic activity against antimicrobial-resistant genera Staphylococcus, Candida, and Bacillus. This study focused on exploring the inhibition mechanism of A. faecalis against the three genera, with an emphasis on S. aureus. A library of A. faecalis transposon mutants was constructed and identified fourteen possible genetic factors linked to this inhibitory activity. Characterization of this inhibition indicated that A. faecalis suppressed S. aureus planktonic and biofilm growth via a contact-dependent mechanism, with pronounced effects during biofilm attachment phase. Cellulose, the most abundant biomass on Earth, offers a promising pathway for alternative energy through conversion into alkane hydrocarbons. Traditional methods rely on high temperatures and high-pressure hydrogen, often derived from petroleum. In this study, we investigated the conversion of Microcrystalline Cellulose using hydrogen metal oxides (bronzes) under milder conditions. Following ASTM E3417-24 hydrolysis protocols, we employed hydrogen bronzes with toluene for product extraction. This approach enabled conversion at lower temperatures without pressurized hydrogen. Products were analyzed using Gas Chromatography Mass Spectrometer for identification and Gas Chromatography Flame Ionization Detector for quantification. In this experiment, the goal was to design, build, and test an Electroencephalogram (EEG) circuit that would then be paired with a touchscreen display as a portable unit for use in Schizophrenic research at East Tennessee State University at the Quillen College of Medicine. Instructors often prioritize traditional instruction in the classroom, despite evidence supporting the use of active learning methods. Exploratory learning, presenting novel problems to students before presenting information about the procedure and underlying concepts, can improve students’ conceptual understanding of instructional material. In exploratory learning, collaborative learning is often an important component of the instructional design. However, few researchers have directly compared collaborative and individual exploration to determine if there are benefits of adding collaboration to exploration. This study conducts a raw material sourcing analysis of Clovis lithic artifacts from Copelin Valley, Hart County, Kentucky, a key Clovis occupation site. Over a decade of documenting private collections and recent archaeological investigations confirm the site's significance within the Paleoindian landscape. Using reflectance spectroscopy, a non-destructive technique, this research assesses chert source probabilities for over 100 Clovis points and tools. By integrating spectral data with geological sourcing frameworks, this study examines raw material procurement strategies and their implications for Clovis mobility and settlement. Retinitis Pigmentosa (RP) is an inherited retinal disease characterized by rod and then cone photoreceptor degeneration. ~25% of cases are caused by a genetic mutation that results in autosomal dominant RP (adRP). There has been a lack of exploration of the progression of degeneration in adRP models. In Chapter Two, I examined visual acuity (VA) changes, retinal structure, and function from early to late-stage disease in a murine P23H Rho ‘knock-in’ mouse model (RhoP23H/+), the most common form of adRP in North America. Research has shown that engineering students from underrepresented minority (URM) populations experience significant retention gaps compared to their majority peers (Dickerson et al., 2014). One factor that can significantly promote academic success, retention, persistence, and psychological adjustment is a sense of belonging (Hausmann et al., 2007; Hoffman et al., 2002). This thesis is an ethnographic case study and archeological-environmental assessment of the North Shore Cemeteries to derive condition commentary and care recommendations for the landscape, cultural resources, descendant community, and Great Smoky Mountains National Park. Framed by theories of space-place culture, dispossession, and public lands co-management, this multi-methods, interdisciplinary thesis combines historical context, environmental conditions, archeological documentation, participant observations, and ethnographic interviewing of NPS and community affiliates to derive applied strategies for cemetery repair, operational efficacy, and this specific, public lands “co-management” relationship. This dissertation explores the integration of multimodal data streams and artificial intelligence pipelines to understand human affect in neurotypical and children with Autism Spectrum Disorder (ASD). This dissertation captures human affect in the context of human-robot interaction. For this, multiple studies have been presented with both children with ASD and neurotypical adults. This dissertation makes four contributions: 1) The first study introduces autonomy during perspective-taking teaching sessions by making verbal content generation through large language models (LLMs). During acute myocardial infarction (MI), the composition of the extracellular matrix (ECM) undergoes significant changes. While collagen accumulation is one of the most well-documented ECM alterations, hyaluronan (HA) is equally abundant. However, the cellular sources, enzymatic regulation, and functional significance of HA accumulation remain largely unexplored. Here, we hypothesized that activated cardiac fibroblasts produce HA via hyaluronan synthase 2 (HAS2), facilitating scar formation and stability. Our findings demonstrate that HA accumulates post-MI and persists for at least four weeks. Through HA sizing techniques, we determined the molecular weight of this persistent HA. This project explores undergraduate students’ digital literacy practices and algorithmic beliefs and awareness on algorithmically driven social media platforms like the TikTok For You Page (FYP). TikTok is a very successful social media platform with over 135 million users in the U.S. alone, nearly half of the U.S. population (Statista, 2025a). Central to TikTok’s popularity and success is its unique technological infrastructure, specifically the underlying algorithm of the FYP that shapes user experience. Women and gender non-conforming students remain underrepresented in Science, Technology, Engineering, and Math (STEM) majors within higher education, and gender disparities persist in advancement to high-paying STEM careers (Maloy et al., 2021; NSF, 2015; Riegle-Crumb et al., 2018). This three-article style dissertation uses qualitative interviews with 29 women and gender non-conforming introductory STEM students and 11 STEM faculty to investigate how these students navigate through their introductory STEM courses. Women and gender non-conforming students in this study faced academic, familial, and gendered pressures to succeed in introductory STEM courses. This dissertation examines the development and impact of the Jefferson County Public Schools (JCPS) Racial Equity Policy through a historical analysis and the perspectives of African American community leaders. The study explores how systemic inequities, rooted in a legacy of racial discrimination, continue to shape the educational experiences of Black students within JCPS. Drawing on archival research and interviews with key community figures, it evaluates the policy’s intended role in dismantling structural barriers and the community’s cautious optimism regarding its effectiveness. Longitudinal data in real-world settings are frequently found to be heterogeneous and exhibit intricate spatio-temporal dependence structures. Analyzing such complex data to obtain reliable estimation while quantifying uncertainty necessitates using sophisticated Bayesian methodology. In this work, we present novel Bayesian methods developed to address these challenges. We often observe heterogeneity in longitudinal data, where the mean and variance for certain profiles meaningfully differs from the rest. Some profiles may also exhibit outliers at a limited number of measurements. Using a standard mixed effects model, which assumes homogeneity, can lead to overestimating the residual variance and inefficient estimation. In this work, …

The purpose of this qualitative case study is to understand how preservice teachers develop their criticality within a semester long undergraduate course focused on children’s literature. The “Rumble in the Jungle” is often remembered as Muhammad Ali’s triumphant return to boxing glory, yet its deeper historical and cultural context remains understudied. This thesis reframes the 1974 heavyweight championship in Zaire as a global spectacle shaped by Pan-Africanism and Paul Gilroy’s theory of the Black Atlantic. More than a fight, it was a performance of Black identity, unity, and cultural return. This thesis, with a contextual focus on Afghanistan, looks into the failure of the concurrent implementation of Islam and democracy in that country. I explored two genetic mechanisms—parent-of-origin (PoO) effects and genome-wide polygenic variation, as measured by polygenic scores (PGSs)—to elucidate possible influences on cognitive variability in WS. This qualitative study investigates the root causes of teacher attrition, retention, and turnover in Eastern Kentucky through the perceptions and lived experiences of highquality veteran educators—those who have persisted in the profession for five or more years. Grounded in participatory action research, asset-based community development, and future-creating methodologies, the study centers the voices of experienced teachers not only as subjects of inquiry but as co-constructors of knowledge and potential solutions. Drawing on empathy interviews with educators across Eastern Kentucky districts, the research identifies core tensions contributing to attrition, including policy misalignment, lack of autonomy, and emotional exhaustion. Inspired by Gnostic myth, biosemiotic forms and Basilides of Alexandria's Seven Sermons to The Dead, Gastropoda tells a creation myth of the opposite yet complementary processes of birth and consumption. The subject of the myth, an unnamed Gastropod, is thrust from the formless ocean to a land more differentiated; full of boundaries and obstacles which obscure the source from wence it came. Immediately following myocardial injury, a robust reparative response is orchestrated through intercellular communication. While various cell populations play critical roles in this process, cardiac fibroblasts have long been recognized as essential mediators of collagen synthesis and scar maturation; however, recent investigations have uncovered significant heterogeneity within the cardiac fibroblast population, revealing distinct subpopulations with roles that extend beyond their traditional functions. Notably, after tissue injury, fibroblasts exposed to damage signals can transition to an inflammatory phenotype, utilizing paracrine and autocrine signaling to coordinate immune cell recruitment and activation.
