Few previous studies have compared microbial communities in subterranean and surface environments. Chemical analyses used to characterize the surface and cave microbial environments indicated limited exchange between surface and subsurface waters. Here, bacterial communities from four distinct cave sediments collected in central Kentucky and adjacent surface streams were compared using a combination of culture and molecular methods. Predictions were that: 1) Cave communities will have unique community structures when compared to those of nearby surface sediments, and 2) Genetic diversity of bacterial communities in cave sediments will be higher than those in nearby surface sediments. The Adoption and Safe Families Act created specific outcomes for permanency for children in foster care. The purpose of these outcomes is to decrease the number of children in long term foster care. Seven years have passed since ASFA was signed into law. Audits completed in each state by federal reviewers have found that no state has meet ASFA's primary outcome of permanency and stability of placement for foster children. Relationships Among Demographic Variables, Organizational Culture, Interpersonal Self-Efficacy And Perceived Job Performance., Maria D. Vasquez-Colina

The goal of this study was to investigate the relationships among demographics variables (age, sex, and ethnicity), organizational culture, interpersonal self-efficacy, and perceived job performance of nonprofit human service workers. The 13 participating organizations provided services such as adoption assistance, adult daycare services, child care resource and referral help, children's daycare services, family counseling, children and youth mentoring, residential care for elderly, residential care for persons with disabilities, and substance abuse treatment programs. Only 607 full-time workers filled in the questionnaire. The response rate was 54%. Findings in the present study found that self-efficacy is a major predictor of performance. …

This thesis begins by reviewing human N-acetyltransferases (Chapter I), then outlines experiments involving human hepatocytes and rat N-acetyltransferases (Chapters II-V). These experiments facilitated the development of a dissertation project designed to study N-acetyltransferase tissue-specific expression in inbred rat strains (Chapter VI). Real-time PCR experiments concluded that genetic and/or environmental factors influence N-acetyltransferase expression in cryopreserved human hepatocytes. Therefore, an animal model must be used to study N-acetyltransferase expression. Initial studies of recombinanatly expressed rat N-acetyltransferases (Nats) revealed a correlation between rat and human N-acetyltransferases in nucleotide and amino acide sequence, catalytic activity, substrate selectivity and thermostability, suggesting the rat is …

Reperfusion of the ischemic lung causes pulmonary arteriolar vasoconstriction and reduces alveolar perfusion. Lung ischemia-reperfusion (IR) injury leads to platelet and leukocyte activation which could contribute to decreased alveolar perfusion during reperfusion by platelet-arteriolar wall interactions (rolling and adhesion) and subsequent microvascular constriction. During the oxidative stress of ischemia and reperfusion, formation of reactive nitrogen species (RNS) may promote platelet adhesion to the vascular wall with subsequent formation of microthrombi and release of vasoactive substances. The purpose of this study was to determine if obesity impacts craniofacial dimensions in adolescent and teenage subjects. Twenty-one cephalometric measurements were selected as a basis for comparison between normal weight, overweight and obese children. The subjects were weighed, measured, and placed into one of the three groups based on their Body Mass Index (BMI). Lateral cephalometric radiographs were taken, traced and digitized on all patients. The various measurements were analyzed to determine if there were any statistically significant differences between any of the measurements when compared on the basis of BMI. Although the juvenile justice system was conceived and developed to provide individualized, benevolent treatment to young offenders, the system has gradually shifted to resemble a more punitive, adult criminal justice model. Steller sea lions, the largest Otariid, fast during their breeding season; during this time they refrain from ingesting food for a period of 12-43 days. Fasting, while undertaking an extremely energetically demanding activity (breeding and pupping), requires specific physiological adaptations. This study examined the physiological response to fasting of two age classes, juveniles and sub-adults, during the breeding and non-breeding seasons to determine how these animals utilize lipids and the pattern of fatty acid mobilization from lipid stores during fasting. Novel Protein-Protein Interactions Regulate The Proteolytic Activity Of The Pro- Apoptotic Serine Protease, Omi/Htra2, Supriya Singh

Omi/HtrA2 is a mitochondrial serine protease with high homology to the bacterial HtrA proteins. Omi promotes caspase-dependent apoptosis by binding and degrading IAPs-inhibitor of apoptosis proteins. Omi can also induce caspase-independent apoptosis but the actual mechanism is still unknown. IAP's are not the only substrates cleaved by Omi. There are at least two more known substrates of Omi, the HAX-1 and the ped/pea-15 proteins. HS1-associated protein X-1 (HAX-1) is a mitochondrial protein, degraded by Omi after induction of caspase-dependent apoptosis. Ped/pea-15 is also an anti-apoptotic protein and is cleaved by Omi after induction of caspase-independent apoptosis. The proteolytic activity of …

Arsenic contamination of drinking water is a real public health problem in certain areas of South-East Asia where chronic exposure has been correlated to higher rates of lung, skin, bladder, kidney, and liver cancer. Although arsenic carcinogenicity is well established, the mechanism by which it induces cancer is poorly understood. An estimated 180 million people are infected globally (WHO). There is no vaccine available to prevent hepatitis C. While direct measurements of energetic demands are nearly impossible to collect on large cetaceans, comprehensive bioenergetic models can give insights on such parameters by combining physiological and ecological knowledge. This model was developed to estimate necessary food intake of gray whales, Eschrichtius robustus, of the Eastern North Pacific stock. Field Metabolic Rates (FMR) for gray whales were first estimated based on various assumptions (e.g. volumetric representation of gray whales, extent of their feeding season, and blubber depth distribution) using morphometric data, energetic costs, and food assimilation according to age and gender specific requirements. Marine turtle conservation is most successful when it is based on sound data incorporating life history, historical population stability, and gene flow among populations. This research attempts to provide that information through two studies. In chapter I, I identify historical patterns of gene flow, population sizes, and contraction/expansion during major climatic shifts. In chapter II, I reveal a life history characteristic of loggerhead turtles previously undocumented. I identify a pattern of juvenile recruitment to foraging grounds proximal to their natal nesting beach. Discrimination and inequality encountered by gays and lesbians in the United States is profuse. A cornerstone of the gay rights movement, equality in the workplace has been a pivotal struggle for gays and lesbians. This study examined the attitudes and opinions of registered nurses (RNs) regarding homosexuals in general and the protection of homosexuals in the workplace through a nondiscrimination policy. Populations of Crassostrea virginica within Mosquito Lagoon, Florida have recently undergone significant die-offs, which are a subject of major concern. Restoration efforts within Mosquito Lagoon are focusing on reconstructing the three-dimensional reef habitats. Before effective protocols can be established, however, important questions about the sources of juvenile and adult oyster mortality must be answered. Conflict resolution programs are discussed as are causes of violence in school age children. Hemozoin formation, the consequential end product of the proteolysis of hemoglobin by Plasmodium falciparum, is essentially a biomineralization process whereby toxic free heme is aggregated into an inert crystalline solid, also known as malaria pigment or ß-hematin. The histidine-rich protein II (HPR II), isolated from the digestive vacuole of the parasite, has been implicated in the mediation of this biomineral through the protein's ability to bind heme, aggregate the biomineral, and be effectively inhibited by known antimalarials, including chloroquine. Selenium is a required micronutrient in mammalian cells. It is incorporated in the form of selenocysteine into selenoenzymes such as glutathione peroxidase and thioredoxin reductase, and is absolutely required for activity. Thioredoxin reductase is necessary for reduction of oxidized thioredoxin and therefore plays a major role in maintaining the redox status of the cell. Glutathione peroxidase is responsible for reducing peroxides into their corresponding alcohols and water. Together, these selenoenzymes constitute a significant part of the cell's arsenal to defend itself against oxidative stress. Security managers have a variety of tools at their disposal -- firewalls, Intrusion Detection Systems (IDSs), encryption, authentication, and other hardware and software solutions to combat computer crime. Many IDS variants exist which allow security managers and engineers to identify attack network packets primarily through the use of signature detection; i.e., the IDS recognizes attack packets due to their well-known "fingerprints" or signatures as those packets cross the network's gateway threshold. This study focused on hospital emergency departments in Florida and the activities, policies, and procedures involved in preparing for a bioterrorist attack. This study addresses whether or not male and female intimate partner homicide offenders differ in a variety of characteristics using data from the Chicago Women's Health Risk Study, 1995-1998. Frequencies of male and female intimate partner homicide offender's risk factors were compared to look at how they differ. The areas that were explored were demographics, prior abuse, and the criminal justices response. A number of gender differences were found. Directions for future research pertaining to intimate partner homicide offenders are discussed.

Embankments are key elements in the infrastructural development of structures such as dams, bridges, and roads. Residual soils are generally used as fills in the construction of embankments in areas were residual soils such as laterite is the dominant soil types. Laterite soils have the characteristics of losing its shear strength with time and in fully saturated conditions and its properties varies from region to region.
