Migraine headaches are ranked as one of the highest disabling conditions in the United States. Women who are in the perimenopausal transition phase have a higher prevalence of disabling migraines that can result in a decreased quality of life, loss of income, decreased productivity, and a higher risk for emotional difficulties. The gap in practice is the lack of education on complementary alternative medicine (CAM) and the use of a migraine diary to recognize individual symptoms as a substitute or cotherapy with pharmacologic treatments. Lewy body disorders are a family of neurodegenerative conditions characterized by proteostatic and redox disequilibrium, leading to deposition of the abundant protein α-synuclein in hallmark inclusions and oxidative damage to DNA. Sublethal oxidative DNA damage in neurons is repaired predominantly by the base excision repair pathway, which involves the coordinated activity of several enzymes, including apurinic/apyrimidinic endonuclease 1 (APE1). Despite emerging connections between α-synuclein, oxidative stress, and DNA damage/repair, along with evidence demonstrating the multifunctional nature of APE1, the link between APE1 and α-synuclein has not yet been explored.

Alzheimer's disease (AD) is a progressive neurodegenerative disease that impairs memory and cognitive judgment, mostly in older people. It is the leading cause of dementia in late adult life. The hallmarks of AD include neuronal loss, neuroinflammation, a build-up of extracellular amyloid beta (Aβ) plaques, intracellular neurofibrillary tangles, and extracellular neuritic plaques. Current treatment, although useful, is not adequate. Identification of therapeutic agents with the potential to alter the progress of AD is a high priority. In Lewy body disorders, α-synuclein aggregates are believed to destabilize the microtubular framework. However, microtubules are a challenging therapeutic target, as excessive rigidity or flexibility afforded by microtubule modulators may be neurotoxic. Thus, the objective of the present study was to evaluate dual-acting microtubule- stabilizing/destabilizing agents in cellular and animal models of early-stage Lewy body disease.

In Aim 1 we tested the dual-acting tricylic pyrimido[4,5-b]indole analogs AG161-47 and AG161-41 in an in vitro preformed fibril induced model of Lewy body disease. We found that both compounds reduced preformed fibril-seeded α-synucleinopathy without toxicity in primary hippocampal cultures. AG161-47 subtly impacted microtubule …

Tumor-associated macrophages (TAMs) play a substantial role in sculpting the tumor microenvironment (TME) into an immunosuppressive bastion that undermines immune surveillance and enables tumor progression. They therefore represent a promising therapeutic target to make the TME more conducive for anti-tumor immunity and greatly improve treatment outcomes in cancers that are highly resistant to conventional forms of therapy. Several medications have been developed to modulate the activities of TAMs, showing tremendous potential, but these medications lack specificity for TAMs making their use susceptible to adverse side effects that could outweigh prospective benefits. This dissertation explores the intersection of biophilia and counseling, aiming to illustrate the potential benefits of integrating nature-based interventions into therapeutic practice. Biophilia, the innate human tendency to connect with nature, has garnered attention in various fields, yet its implications for counselors remain unexplored. This qualitative study employs a hermeneutic phenomenological approach to investigate the lived experiences of counselors who have incorporated biophilic elements into their practice and the perceptions of clients who have engaged with these interventions.

Through semi-structured interviews with licensed counselors, themes emerged regarding the impact and use of biophilic interventions on the therapeutic process. Findings suggest …

Background: Disasters are dangerous for schools where many children are located in one area. Children were significantly impacted by a biological incident during the 2009 H1N1 influenza pandemic, and more recently the COVID-19 pandemic. Research is needed to determine the values, perceptions, and beliefs of school disaster preparedness through a cultural lens.

Methods: A focused ethnography was used to interview school administrators and nurses from April-December 2022 to explore and understand the unique culture of school disaster preparedness. Data was analyzed according to Leininger’s Four Phases of Data Analysis.

Running economy (RE) has been identified as a critical component of endurance performance. Alterations in footwear have shown to significantly impact RE. Quantifying reliability of within-day RE measures may determine differences that could be expected when testing footwear. PURPOSE: Assess within-day reliability of RE testing on a calibrated metabolic system. On a routine trip to the grocery store, or while scrolling online, you might encounter the provocative and puzzling discourse of Intactivism, a viral, single-issue social movement that opposes infant male circumcision, and wonder “Huh? Why is this here, now?” This project situates Intactivism in a history of circumcision and anti-circumcision that spans from ancient Sumerian tents to the front pages of Reddit. It takes an immersive qualitative clinical ethnographic approach, nested in a broader cultural psychology that is non-reductive and pluralistic, to articulate an Intactivist ethos in its public and private dimensions. The Colorado River Basin (CRB) is one of the most critical water sources for the western United States (U.S.) and Mexico. For over a century, numerous policies and regulations have been implemented in attempts to manage the Colorado River Basin to address the concern of overuse, collectively referred to as the “Law of the River.” The USGS hydrologist E.C. LaRue warned that the optimistic estimates used in negotiating the Colorado River Compact did not align with the actual water availability in the river. Historical and current extraction operations and infrastructure, including natural gas pipelines, conventional and unconventional oil and natural gas drilling, longwall coal mining, and strip coal mining, occur throughout the Mingo Creek watershed of Washington County, Pennsylvania, including Mingo Creek County Park. Mingo Creek and several associated tributaries flow through Mingo Creek County Park, eventually discharging into the Monongahela River. Glycerophosphocholine (GPC) is an abundant metabolite in eukaryotes. In baker’s yeast, Saccharomyces cerevisiae, GPC can be produced via deacylation of the major membrane glycerophospholipid phosphatidylcholine (PC). My lab has identified a novel pathway to re-synthesize PC from GPC by acylating it at the sn-1 and sn-2 positions; this pathway has been termed the PC Deacylation-Reacylation Pathway (PC-DRP). We previously characterized the acyltransferase Gpc1 and its ability to synthesize one-tailed lyso-PC from GPC, as well as that noting that GPC1 is upregulated in inositol-free conditions and results in more saturated PC. Characterization Of A Novel Metalloid Reductase Outer Membrane Porin Rara From Sulfurospirillum Barnesii Ses-3, Narthana Jeganathar Kanmanii

Sulfurospirillum barnesii SES-3 is a gram-negative epsilon proteobacterium that grows anaerobically by coupling the oxidation of lactate to the reduction of arsenate. Previous work on metal and metalloid metabolism in S. barnesii had identified a redox active protein, designated RarA that when provided an artificial electron donor (i.e., reduced methyl viologen, FMN, or NADH), could reduce a variety of substrates, including arsenate, selenate, selenite, nitrite, and even phosphate. The enzyme’s inferred amino acid sequence showed weak similarity (30%) to outer membrane porins with homologs found in only certain species of Sulfurospirillum. Male reproductive protein-coding genes are among the most rapidly evolving genes across many animal taxa, commonly attributed to sexual selection; however, pathogen defense mechanisms might also be contributing factors. This study aimed to both computationally and experimentally investigate the selective pressures responsible for the rapid evolution of seminal proteins. The research work presented in this dissertation focuses on the synthesis, characterization, and property assessment of new and existing quaternary diamond-like semiconductor (DLS) materials for the potential application in infrared (IR) nonlinear optics (NLO). Chapter 2 discusses the previously discovered temperature-dependent polymorphs of Cu2ZnGeS4 (CZGS); however, the IR NLO properties, neutron powder diffraction data, and single crystal X-ray diffraction structures were previously undetermined. With over 6 million death counts, cancer is the second leading cause of death in the United States. Lung cancer was the leading cause of cancer death, accounting for 23% (136,084 death count) of all cancer deaths. Other than that, colon and rectal (9%, 51,869 death count), pancreas (8%, 46,774 death count), female breast (7%, 42,275 death count), prostate (5%, 32,707 death count), and liver and intrahepatic bile duct (5%, 28,227 death count). The capacity to identify bacteria quickly and accurately is critical for applications such as medical diagnosis, environmental monitoring, and food safety. For instance, sepsis diagnosis requires blood cultures that take as long as 1-5 days, preventing timely intervention and increasing mortality rate. Here, we propose the joint use of microfluidics and a machine learning algorithm for rapid bacterial cell capture and quantification. The combination of high-throughput droplet microfluidics and a support vector machine (SVM) enables analysis and quantification of bacterial samples within as short as 4 hours. The first principles of bioethics, autonomy, beneficence, nonmaleficence, and justice have contributed immensely to healthcare ethics. The procedural approach streamlines the decision-making process, protects vulnerable subjects, and fosters stability and trust in the patient-caregiver relationship. This dissertation interrogates the use of poetic fragmentation as a tool of ecological representation and coins the term “ecotonal poetics.” Ecotonal poesis, this dissertation explains, is the stretching of lyric across the field of the page, using space, sound, language, and parataxis in its attempt to not only represent but create ecotonal ecologies through text, sound, and visual cues. This dissertation puts forth four case studies of contemporary American women poets turning the space of the page into an ecotone where text, white space, author, reader, body, and environment meet. The re-establishment of historic fire return intervals is vital to restoring forest health, species diversity, natural succession regimes, and reduction of some invasive species. Studies on the effect of fire frequencies on southern pine understory plant communities have revealed increased plant diversity and richness in response to fire, particularly in longleaf pine. The National Forests and Grasslands of Texas (NFGT), The Nature Conservancy (TNC), and the privately-owned Winston 8 Ranch are intensively managed using prescribed burns at different frequencies for a variety of management objectives.

A 2x2 design was undertaken to determine the effects of vignette character gender and autism disclosure status on social distance scores, reading comprehension performance, and false memory induction. The introduction focuses on the theoretical threads taken from the autism, false memory, and gender literatures and how they combine to create the underpinning of the current design. A second more controlled study was compelled by initial false impressions of the deleterious effects of fast completion times.
