This dissertation describes new procedures for synthesizing and purifying Au and Au/Ag nanoplates directly on surfaces as well as controlled binding of proteins to their surface. It also describes fundamental changes in the optical properties of Au and Au/Ag nanoplates upon interaction with different liquids, proteins, or nanoparticles. Finally, this dissertation describes the highly selective and sensitive label-free detection of proteins in an immunoassay format using the optical properties of Au and Au/Ag nanostructures. A chemical seed-mediated growth procedure allowed the synthesis of Au nanoplates directly on surfaces with an initial 23% yield. Purification by either sonication or adhesive tape …

Arsenic In Drinking Water Causes Gene Expression Changes In The Liver Related To Inflammation And Metabolic Dysfunction And Accelerates Atherosclerosis In Apoe-/-Mice, Matthew R. Zajack

Arsenic exposure in drinking-water is a significant worldwide health problem. It causes adverse human health effects, such as cancer, increases the risks for others such as cardiovascular disease, and accelerates atherosclerosis. In this study, we analyze arsenic-induced gene expression changes in the liver of ApoE-knockout mice given 49 ppm arsenic in drinking-water. We hypothesize chronic arsenic exposure accelerates atherosclerosis by disrupting liver homeostasis, causing aberrant gene expression changes. Networks revealed hubs on 3 stress-response MAP kinase pathways, ERK, JNK, and p38. Pathways revealed mitochondrial dysfunction and oxidative phosphorylation enrichment from the gene set. CDCIWHO only "recommends" that rings be removed under gloves because of cross-contamination between health care workers and patients. Microbiological samples of oral streptococci beneath rings and between fingers under non-sterile exam gloves were taken before and after dental procedures (before and after washing with antimicrobial soap) from 50 junior/senior dental students. Subjects wore two rings on one hand, with none on the other (control). Rings were worn on the opposite hand at another session. Handedness, glove tears, and leaks, were also measured. Since the suburban rush and steep rise in household technological devices in the mid-twentieth century, Americans have drawn apart from each other, a shift that has coincided with a rise in both dog ownership and the adoption of handheld mobile devices. This study explored the personal journey of 11 White college administrators who were identified as inclusive leaders at a predominately White institution (PWI), recognized nationally for its work on partnering diversity and excellence. One overall question guided this study: How do White college administrators describe their journey to becoming successful inclusive leaders at a predominately White institution? This question was explored from the perspective of critical race theory (CRT), that is, inclusive leadership for White administrators could be achieved by intentionally examining their construction of Whiteness and their personal racial identity.

This paper details the development of a scale to more accurately assess depression in men. It first summarizes the literature on depression, depression assessment, and current research on men and masculinity. It is argued that current conceptualizations of both depression and masculinity influence prevalence studies, which consistently find that men experience depression half as often as women. It is argued that an assessment measure that accounts for masculine variants of depression (substance use, anger, withdrawal, and emotional restriction) may identify more frequent depression in men than previously expected. According to exit polls, a majority of people who voted for Bush over his opponent, Senator John Kerry, did so because they believed that Bush was the “moral values” candidate. In this dissertation, I assess the moral persona that the President rhetorically constructed during his first term in office. This thesis is a historiographical study of the eugenics and euthanasia programs of Nazi Germany. It traces there development from the end of World War One to the fall of Hitler's Third Reich. We have studied the inhibitory effect of natural and structurally modified polyphenols on Escherichia coli ATP synthase to test (I) if the beneficial dietary effects of polyphenols are related to their inhibitory actions on ATP synthase, (II) if inhibitory effects of polyphenolic compound could be augmented through structural modifications, and (III) if they can act as antimicrobial agent through their actions on ATP synthesis. X-ray crystal structures of polyphenol binding sites suggested that polyphenols bind at a distinct polyphenol binding pocket, at the interface of α,β,γ-subunits. Commercially available organic and conventionally grown vegetables were studied by quantitative determination of selected metals in them and to determine if any differences found are statistically significant. These findings can help the consumers to determine if the vegetable products are within the recommended maximum limits as proposed by the joint FAO/WHO Expert committee on organic foods designation. Eight edible vegetables were purchased from local stores in both the organic and conventionally grown categories. The purpose of this study was to determine the amount of training each sheriff's office requires in North Carolina and if that training includes multiagency exercises designed to mitigate a critical-incident response and identify any concerns from those training events. The study also compared departmental strength (number of sworn officers per agency) with county populations and geographic area of the state the agency is located in with the number of hours required annually by each agency. Siderophores are small, iron chelating molecules produced by many bacteria to help meet the iron requirements of the cell. Multiple metabolic functions require iron as it serves as a cofactor in many enzymes and cellular processes. However, in the presence of oxygen and at physiologic pH, iron forms insoluble ferric complexes that cause the nutrient to be unavailable to bacterial cells. Siderophores alleviate this limitation by chelating the ferric iron, rendering it soluble and available for uptake. The Hermit Thrush is a new breeding bird in the Southern Appalachian high-elevation mountains, having expanded its range southward over the last few decades. Here it is sympatric with the Veery, a congeneric breeding resident. In order to more fully understand why the range expansion took place and to understand more about the local ecology of the newly arrived bird, I measured several habitat variables in 30 Hermit Thrush and 24 Veery territories. Principal Components Analysis and Cluster Analysis brought to light several patterns of habitat preferences for these 2 species. Hermit Thrushes prefer territories with more leaf litter on …

In this work, we study error correcting codes and generalize the concepts with a view toward a novel application in the study of DNA sequences. The author investigates the possibility that an error correcting linear code could be included in the human genome through application and research. We examined several aspects of nectar production in Campsis radicans to better understand how standing crop is affected and how production affects pollinator visitation. In all experiments, nectar and concentration of flowers were measured and total sugar was calculated. Flowers do not produce additional nectar unless nectar is removed, and it is not resorbed. Volume of standing crop and total sugar fluctuates throughout the day, whereas concentration remains constant. Age and time of day significantly affect regeneration of nectar and sugar. The number of removals did not significantly affect the amount of nectar or sugar regenerated; however, concentration declined significantly …

A Re-Evaluation Of The Pleistocene Hellbender, Cryptobranchus Guildayi, And An Overview Of Cryptobranchus Remains From Appalachian Caves., Keila Elaine Bredehoeft

Cryptobranchus guildayi is described as an extinct species of large salamander that is closely related to the hellbender, Cryptobranchus alleganiensis. The validity of this extinct taxon has been questioned, so an expanded osteological sample of modern hellbenders was used for comparative purposes with the C. guildayi fossil material. Based on this analysis, all supposed distinguishing morphological characteristics used to define C. guildayi can be observed in specimens of C. alleganiensis, or are based on misidentifications. Therefore, Cryptobranchus guildayi is considered to be conspecific with C. alleganiensis and taxonomically should be considered a junior synonym of the latter. The …

The genus Aneides (Caudata: Plethodontidae) is an arboreal salamander with a prehensile tail and a distribution that spans North America. It is hypothesized that adaptations for arboreality will be visible in the osteology of the sacral vertebra either by qualitative analysis or linear and morphometric analysis in comparison with other plethodontid salamanders. This study demonstrates that while qualitative and quantitative analyses are successful at making genus-level distinctions between taxa, identification to lower taxonomic levels remains inconclusive. Linear morphometrics and dorsal Procrustes landmarks were the most successful metrics to identify known taxa. Two unidentified fossil salamander sacral vertebrae from Oregon Caves …

In this work we use a graph-theoretic representation of secondary RNA structure found in the database RAG: RNA-As-Graphs. We model the bonding of two RNA secondary structures to form a larger structure with a graph operation called merge. The resulting data from each tree merge operation is summarized and represented by a vector. We use these vectors as input values for a neural network and train the network to recognize a tree as RNA-like or not based on the merge data vector.

Two vertices are said to be identifed if they are combined to form one vertex whose neighborhood is the union of their neighborhoods. A graph is total domination dot-critical if identifying any pair of adjacent vertices decreases the total domination number. On the other hand, a graph is total domination dot-stable if identifying any pair of adjacent vertices leaves the total domination number unchanged. Identifying any pair of vertices cannot increase the total domination number. Further we show it can decrease the total domination number by at most two. Potential dental implant site assessment is initially determined clinically. Subsequent radiographic assessment also provides information about the quantity of available bone. It was hypothesized that the use of radiographic markers in mandibular stents to project the clinically derived placement of implant fixtures would not correspond to the optimal trajectory of the fixture based on acceptable implant placement criteria applied on Cone Beam Computed Tomography images. A retrospective audit of patients with edentulous mandibles scanned with a surgical guide was completed. Determining the etiology of skeletal malocclusion is an important diagnostic step in treatment planning patients with maxillofacial anomalies. Aims: To describe cranial base morphology using cone beam computed tomography (CBCT) images and correlate with the Antero-posterior (AP) jaw discrepancy. Methods: With IRB approval, retrospective analysis was performed on CBCT scans for 40 adult subjects. Cranial flexure and cranial deflection angle were analyzed in relation to three AP variables using linear regression analysis. A hyper basis function network (HyperBF) is a generalized radial basis function network (RBF) where the activation function is a radial function of a weighted distance. The local weighting of the distance accounts for the variation in local scaling and discriminative power along each feature. Such generalization makes HyperBF networks capable of interpolating decision functions with high accuracy. However, such complexity makes HyperBF networks susceptible to overfitting.
