Extant research has made clear the link between self-criticism and engagement in non-suicidal self-injury (NSSI). As posited by the Defective Self Model of NSSI, individuals who experience feelings of self-hatred may perceive pain as deserved and engage in NSSI (Burke et al., 2021). Individuals exposed to interpersonal trauma are at higher risk of engaging in NSSI, and also frequently experience trauma-related self criticism, namely shame and self-blame. More work is needed; however, to understand how self-critical post-traumatic cognitions contribute to risk for NSSI engagement for interpersonal trauma survivors. The regulated secretory pathway is important for the secretion of hormones and neuropeptides from large dense core vesicles (LDCVs). Dysregulation of the biogenesis of LDCVs can result in metabolic and neurological diseases. Recent studies have suggested that the peripheral membrane protein HID-1 plays a role in the sorting and storage of granins which contribute to LDCV biogenesis. To further understand how HID-1 influences the regulation of granins, we investigated potential HID-1 interactors. Infertility is a distressing experience for those diagnosed and often exacerbated by engagement with biomedical treatments. This thesis is an exploration of infertility “journeys” posted on YouTube. Through a narrative analysis of 8 YouTube channels, several key themes were identified, specifically embodiment of the infertility label, barriers to treatment, and agency. This work uses the mindful body framework to look at how the individual body, social body, and body politic shape and reinforce these themes in the context of biomedical infertility treatment. The SARS-CoV-2 virus was recently able to cause a global pandemic, largely due to the asymptomatic period that occurs during initial infection. During this period, patients are infectious but do not exhibit symptoms, resulting in the unintentional spread of the virus. This occurs because the virus effectively avoids detection by the immune system during the early stages of infection, and as symptoms are the result of immune system activation, they never manifest. Anti-Asian racism and violence in the U.S. both historically and currently impacts the Asian American community, leading to widespread fear and negative mental health consequences for Asian Americans. This has been widely discussed in the media due to the rise in anti-Asian rhetoric and attacks during the COVID-19 pandemic. The nature of chemical interactions can be difficult to ascertain in the condensed phase; chemistry can be observed, but with the plethora of variables such as solvation, mixes of counter-ions, and complex equilibria, the true intrinsic chemical properties of a given chemical species may be obfuscated. Dysregulation of immune homeostasis is fundamental to the development of autoimmune disorders. Fibroblastic reticular cells (FRCs) within lymph nodes play crucial roles in forming a microenvironment conducive to immune cell interactions and regulating autoreactive T cells driving autoimmune diseases. Studies have shown the potential of FRCs as a form of personalized cell therapy to correct dysregulated immune responses. Inactive sulfated steroids are present in plasma at higher concentrations than unsulfated ones. Steroid sulfatase (STS) is the enzyme that removes the sulfate group from sulfated steroid hormones in local tissues, making them active. High levels of STS are linked to breast and prostate cancers, and low levels of STS cause behavioral disorders and X-linked ichthyosis. STS is present in most tissues, yet its physiological role is uncertain. We sought to characterize STS activity in several rodent model systems in order to learn more about its function. Chinese tallow (Triadica sebifera (L.) Small) is an invasive tree known to have negative impacts on bottomland hardwood forests. Chinese tallow is proficient in reproducing and surviving in bottomland hardwood forests because it is more flood and shade tolerant than most native trees. The long-term effectiveness of Chinese tallow control is an important topic to research as only a few long-term studies of Chinese tallow control have been conducted. This paper investigates the educational experiences of children and adolescents diagnosed with RYR1-related diseases, a spectrum of congenital myopathies affecting muscle function. Despite these conditions typically sparing cognitive abilities, they profoundly impact physical capabilities, presenting challenges in school environments such as participation in physical activities and social interactions. The study utilizes data collected through the RYR1-Related Disease Educational Impact and Satisfaction Survey (RDEISS) and the Behavioral and Emotional Screening System (BESS) from 49 parents of 51 children aged 3-18 years. Ischemic stroke is the second leading cause of death worldwide, and the lack of effective treatment options is an unmet clinical need for stroke survivors. Ischemic stroke-induced oxygen-glucose deprivation (OGD) in the affected brain tissue leads to mitochondrial dysfunction and adenosine triphosphate (ATP) depletion in the brain endothelial cells (BECs) lining the blood-brain barrier (BBB). ATP deprivation in the BECs dysregulates actin cytoskeleton dynamics, resulting in BBB breakdown. Restoration of blood flow (i.e., reperfusion) upon the first-line stroke treatment further leads to BBB breakdown and widens the paracellular spaces between BECs. …

Macromolecules occupy up to 30% of a cell's volume, yet the in vitro study of biological processes is almost exclusively performed in dilute solutions that fail to approximate the crowded interior of a cell. A growing body of literature has identified that macromolecular crowding has significant effects on a number of biochemical processes, including enzymatic activity. However, changes in kinetic parameters with increased macromolecule concentrations have only been determined for enzymes that follow hyperbolic Michaelis-Menten kinetics, despite the fact that over 25% of enzymes deviate from this model. Ascidians are our closest invertebrate relatives and comprise nearly 3,000 species separated into three orders: Aplousobranchia (most speciose), Stolidobranchia, and Phlebobranchia (least speciose). Ascidians can be classified as either solitary or colonial organisms. Species delimitation using morphological characters alone has had varied results. Well known, widely distributed, morphological species have turned out to be catch all species comprised of several cryptic species. Bottomland hardwood forests in the Lower Mississippi Alluvial Valley have been extensively reduced due to agriculture conversion and altered hydrology. Remnant forests have undergone further degradation from sources such as a lack of forest management, invasive species, and reduced regeneration of oaks (Quercus spp.). Recent restoration efforts of bottomland hardwood forests have focused on implementing wildlife forestry practices that prioritize restoring wildlife habitat while retaining profitable timber harvests. Herpes simplex virus type 1 (HSV-1) is a double stranded DNA virus that replicates within the nucleus of host cells. Using accelerated native isolation of proteins on nascent DNA (aniPOND), cellular proteins were found to associate with HSV-1 replication forks. One host factor of interest is proliferating cell nuclear antigen (PCNA), the DNA sliding clamp that tethers DNA polymerases and DNA damage response proteins to replicating cellular DNA. Although HSV-1 encodes its own DNA polymerase processivity factor (UL42), PCNA plays a key role in HSV-1 infection. The research described in this dissertation focuses on the investigation of the structure and dynamics of regions of RNA implicated in two diseases: COVID-19 and amyotrophic lateral sclerosis/frontotemporal dementia (ALS/FTD). Both studies sought to create experimentally corroborated models that were used to characterize the structure and dynamics of the C9orf72 repeat expansion and the s2m region in coronaviruses, respectively and provided the foundation for future work. The connection between suicide and rural residency is not well understood, nor is the relationship between the distribution of prescribers, altitude, and suicide. Emotions are biological responses to stimuli that allow individuals to derive meaning, appraise experiences, and prepare to respond. However, individuals perceive emotions differently based on emotion socialization which not only dictates the way emotions are viewed and managed but also has been directly linked with mental health outcomes. Furthermore, research shows emotion socialization is informed by demographic variables such as gender such that the expectations of emotionality differ; where women are taught to express emotions, men are taught to conceal.
