Therapeutic proteins represent an essential piece of a health management plan for diseases such as diabetes, cancer, hemophilia, Crohn's Disease, and myocardial infarction. These proteins, however, must be maintained in their correct, biologically active conformation throughout processing, transportation, and delivery. This requirement poses serious engineering challenges because of a protein's susceptibility to thermodynamic instabilities resulting from the weak bonds driving the tertiary structure of the molecule. A particularly problematic type of protein degradation is aggregation. Administration of aggregated proteins, a particularly problematic degradation form, can have dire consequences, including blocking a patient's responsiveness to therapy, inducing immunogenicity, and even anaphylactic …

The postmodern criticism of humanist agency initiated by Dilip Gaonkar nearly twenty years ago set in motion a discipline wide discussion concerning the conceptualization rhetorical agency. Rhetorical agency is difficult but vital to conceptualize because the term bears directly on the discipline's theorizing about the speaker or rhetor, the effect of the speaker or rhetor's rhetoric on an audience, and the extent to which the speaker or rhetor's agency is constrained by ideology and discourse. Studies indicate that ATP is one of the primary neurotransmitters in taste transduction. ATP release occurs from taste cells via specific hemichannels such as pannexin/connexin hemichannels (Huang et al., 2007; Romanov et al., 2007). We hypothesize that Type II (receptor) and possibly Type III (presynaptic) cells release ATP at sites containing pannexin/connexin hemichannels. In this study, we examine the presence of connexin–32–LIR (Like Immunoreactivity) and connexin–43–LIR in rodent taste buds through immunocytochemical analysis and DAB (Di–amino–benzidine) immunoelectron microscopy. We observed that connexin–32–LIR co–localizes with P2X2–LIR in nerve fibers and in a small subset of NCAM–LIR cells. Connexin–32–LIR does not co–localize …

This dissertation endeavors to build a much-needed bridge between the fields of communication and education. Using critical pedagogy and critical communication pedagogy as theoretical frameworks, this project advances an understanding of classroom communication as constitutive of power relations, and teachers and students as agents who can work together to foster learning potential and social justice. Smoking is the leading preventable cause of death, and smoking during pregnancy can lead to significant health complications for both the mother and developing child. In the Appalachian region of East Tennessee, pregnancy smoking rates are as high as 40% in some counties which is almost 4 times the national average. Weight and body image concerns have previously been identified as variables contributing to maintenance of smoking behavior. The current study was designed to examine the relationship between prepregnancy weight and body image concerns and smoking status at third trimester. Regarding the myriad disputed documents encountered within the science of forensic document examination, questioned handwriting is the most prevalent. This includes the simulation or alteration of and or additions to handwriting and signatures. The current study examined the changes that may occur in writing when given a limited amount of space. Several participants completed a survey wherein writing samples were taken under varying space allowances. A consensus model combines statistical methods with signal processing to create a better picture of the family of related signals. In this thesis, we will consider 32 signals produced by a single electroencephalogram (EEG) recording session. The consensus model will be produced by using the S-Transform of the individual signals and then normalized to unit energy. A bootstrapping process is used to produce a consensus spectrum. Schizophrenia affects about 1% of the population. A hallmark of the disorder is increased dopamine D2 receptor sensitivity in the brain. Studies have shown that schizophrenics smoke cigarettes at approximately 4 times the rate of the general population. It has been suggested that nicotine use is a form of self-medication for symptoms in schizophrenia. Smoking behaviors typically begin in adolescence. We assessed effects of nicotine on behavior and brain plasticity in an adolescent rodent model of schizophrenia with the goal of identifying targets for smoking cessation. Alcohol is the most abused drug in the United States and most frequently performed assay in forensic laboratories. Alcohol is routinely present in biological specimens from fatal residential fires and forensic toxicologists must interpret if these individuals are impaired by determination of their blood alcohol concentrations on post-incineration blood collected at autopsy. There is no known data available to confirm or refute blood alcohol concentrations and impairment in fire-related deaths. Quantum dots are inorganic artificial semiconductor nanocrystal whose electrons influence their physical and chemical properties. This thesis tests various hypotheses from a variety of research traditions that predict the likelihood for potential sex differences in "talking": a newly-emergent phase of romantic courtship. Data for this study was derived from a purposive sample of 566 students enrolled during the Fall 2011 semester generated using a self-administered survey available on the East Tennessee State University SONA system. Statistical analyses using chi-square, analysis of variance (ANOVA), and binary logistic regression reveal statistically significant differences for males and females on beliefs about sexual exclusivity and sexual activity during the "talking" phase. The purpose of this quantitative study was to determine if there were significant differences in student success in face-to-face and online biology courses as categorized by gender, major, and age; and as measured by lecture grades, lab grades, and final course grades. The data used for analyses included data from 170 face-to-face sections and 127 online sections from a biology course during the fall and spring semesters beginning fall 2008 through spring 2011.

The primary sources consulted include newspapers, military medical reports, photographs, memoirs, speeches, and archival collections. Antibiotics have been commonly used in medical practice for over 40 years. However, the misuse and overuse of current antibiotics is thought to be the primary cause for the increase in antibiotic resistance.
Many current antibiotics target the bacterial ribosome. Antibiotics such as aminoglycosides and macrolides specifically target the 30S or 50S subunits to inhibit bacterial growth. During the assembly of the bacterial ribosome, ribosomal RNA of the 30S and 50S ribosomal subunits is processed by bacterial ribonucleases (RNases). The manner in which this pathogen is able to interact with its host is difficult to define, as is the case with most microbes. Through the use of alternate sigma factors and other regulatory processes, S. Typhimurium is able to invade host cells to establish systemic infections, and survive the assaults of the host immune system. While most strains of S. The purpose of this study was to develop a mechanistically reliable model capable of identifying xenoestrogens. Our hypothesis was that there are identifiable structural characteristics among a diverse set of estrogen receptor ligands that differentiate estrogenic and nonestrogenic compounds. Several technical and economic obstacles currently hamper the industrial development of ethanol from biomass. One of the key bottlenecks is the slow kinetics of the enzymatic hydrolysis of cellulose, and the subsequent rate reduction as the reaction proceeds. As a result, this research focused on understanding underlying causes for the slow kinetics, rate reduction, and low yield during cellulose hydrolysis. We formulate an integro-difference model to predict the growth and spatial spread of a perennial plant population with an age-structured seed bank. We allow the seeds in the bank to be of any age, producing an infinite system of equations. The production of new seed can be density-dependent and so the function describing this growth is allowed to be non-monotone. The functions describing the seed bank are linear. We develop properties about the non-spatial model, including the existence of a positive steady-state and conditions under which solutions converge to this steady-state.
