Building A Metabolic Bridge Between Glycolysis And Sphingolipid Biosynthesis : Implications In Cancer., Morgan L. Stathem
Building A Metabolic Bridge Between Glycolysis And Sphingolipid Biosynthesis : Implications In Cancer., Morgan L. Stathem
Electronic Theses and Dissertations
Cancer therapeutics has seen an emergence and re-emergence of two metabolic fields in recent years, namely bioactive sphingolipids and glycolytic metabolism. Anaerobic glycolysis and its role in cancer has been recognized in cancer biology over 90 years. In recent decades, the role of sphingolipids in cancer cell metabolism has gained recognition, notably ceramide’s essential role in programmed cell death and the role of the glucosylceramide synthase (GCS) in chemotherapeutic resistance. Despite this knowledge, a direct link between these two fields has yet to be concretely drawn. Herein, we show that in a model of highly glycolytic cells, generation of the …
Inducible Tumor Difference Between Rapid And Slow Rat Nat2 Congenic Fischer 344 Rats Administered Methyl-Nitrosourea., Marcus W. Stepp
Inducible Tumor Difference Between Rapid And Slow Rat Nat2 Congenic Fischer 344 Rats Administered Methyl-Nitrosourea., Marcus W. Stepp
Electronic Theses and Dissertations
Human arylamine N-acetyltransferase 1 (NAT1) is a well-known phase II metabolic enzyme that has been associated with carcinogenesis. Its role in the biotransformation of aromatic and heterocyclic amine carcinogens has been investigated for many years, but more recent investigations focus on a possible endogenous role of human NAT1 in cancer initiation and progression. We conducted in vivo studies using homozygous Fischer 344 rats, congenic at the rat Nat2 locus for high (rapid) and low (slow) activity. Wistar Kyoto inbred rats were used to breed in the slow activity rat Nat2 locus into the Fischer 344 inbred rat, which contains the …
Dispositional Mindfulness And Working Memory In The Context Of Acute Stress., Lauren M. Vines
Dispositional Mindfulness And Working Memory In The Context Of Acute Stress., Lauren M. Vines
Electronic Theses and Dissertations
The neuropsychological domain of working memory and the nearly 3000 year-old Buddhist construct of mindfulness appear to be disparate concepts. However, the measurable decline of working memory capacity (WMC) under stress, in combination with the stress-reducing and attention-focusing effects of mindfulness suggest potential augmentation of working memory through the engagement of mindful practice. A theoretical process through which dispositional mindfulness exerts a moderating effect on WMC reduction following an acute stressor is proposed. To investigate processes within this theoretical framework, a sample of undergraduate college students (N = 67) were assessed across various measures, including level of dispositional mindfulness, affective …
Penalized Regressions For Variable Selection Model, Single Index Model And An Analysis Of Mass Spectrometry Data., Yubing Wan
Electronic Theses and Dissertations
The focus of this dissertation is to develop statistical methods, under the framework of penalized regressions, to handle three different problems. The first research topic is to address missing data problem for variable selection models including elastic net (ENet) method and sparse partial least squares (SPLS). I proposed a multiple imputation (MI) based weighted ENet (MI-WENet) method based on the stacked MI data and a weighting scheme for each observation. Numerical simulations were implemented to examine the performance of the MIWENet method, and compare it with competing alternatives. I then applied the MI-WENet method to examine the predictors for the …

Numerical Investigation Of Hydrodynamic Focusing And Coulter Principle In A Microfluidic Device, Muheng Zhang
Electronic Theses and Dissertations
Particle information such as particle size and amount is significant to many fields such as chemical engineering, biological research, medical analysis, and environment detection. Microfluidic Coulter counter is suitable for this task because it can get fast and accurate analysis. However, the fundamental principle and several design strategies details are still needed to be analyzed and modeled in order to improve its performance. This dissertation focused on several challenges from the numerical simulation aspects. First one is the analysis for the hydrodynamic focusing in microfluidic Coulter Counter. In recent years, many microfluidic Coulter Counter designs have utilized a technique termed …
The Walkable Dividend : The Impacts Of Walkability On Housing And Socio-Economic Composition In Louisville, Ky., Wesley Laurance Meares
The Walkable Dividend : The Impacts Of Walkability On Housing And Socio-Economic Composition In Louisville, Ky., Wesley Laurance Meares
Electronic Theses and Dissertations
Until 2008, there has not been a reliable measure of the social, health, and economic impact of walkable neighborhoods. This changed dramatically when scholars were able to quantify walkability, which measures how accessible daily living activities are by foot. However, most of these studies focus on mega cities, sections of cities or on random parcel data. Absent from the literature is the impacts of walkability on mid-size cities. This dissertation seeks to fill this void by examining the impacts of walkability on neighborhood housing valuation, foreclosures, vacancy rates and socio-economic composition in Louisville, KY. This dissertation employs ordinary least squares …
Bdnf Maintains Adult Taste Innervation And Is Required For Taste Nerve Regeneration After Injury., Lingbin Meng
Bdnf Maintains Adult Taste Innervation And Is Required For Taste Nerve Regeneration After Injury., Lingbin Meng
Electronic Theses and Dissertations
Brain derived neurotropic factor (BDNF) is required for the gustatory neuron survival, target innervation, and taste bud maintenance during development. However, whether BDNF has any function in the adult gustatory system in normal conditions or after nerve injury is unclear. To address these issues, I inducibly removed BDNF in all cells expressing BDNF in adult mice. In the experimental animals, Bdnf expression decreased to 5% of control mice in the lingual epithelium and geniculate ganglion (p< 0.01) at both two weeks and ten weeks after tamoxifen administration. I found no effect on taste bud morphology at four weeks following Bdnf gene deletion. However, ten weeks following Bdnf gene deletion, P2X3-positive and TUJ1-positive gustatory innervation to individual taste buds was reduced by nearly half (each with p < 0.01) and both taste bud volume and taste cell number decreased 30% (each with p< 0.01). These experiments demonstrate that BDNF is required for maintenance of normal levels of taste innervation and normal taste bud morphology in adulthood. In addition, taste cells expressing PLCß2 (phospholipase C ß2), a marker for taste cells that respond to sweet, bitter and umami, did not decrease after Bdnf gene deletion in the adult. Thus, the missing taste cells are of another type. This indicates that taste cell loss is not uniform across the various taste cell types, even if nearly all taste cell types receive the P2X3 and TUJ1 innervation. Since BDNF is required for initial innervation of the taste system and supports taste bud innervation and size in adulthood, it could also be required for nerve reinnervation after injury. To determine if Bdnf is still expressed following nerve section, the chorda tympani nerve (taste nerve) was sectioned and Bdnf level was detected with Real Time RT-PCR. Bdnf continued to be expressed at normal levels from two days to two months post-surgery in both geniculate ganglion and tongue epithelium. Therefore, BDNF could be involved with chorda tympani regeneration. To determine if this was the case, the Bdnf gene was deleted in adult inducible transgenic mice (under the control of a Ubiquitin promoter) two weeks before chorda tympani nerve section. Taste bud number was reduced by half in all genotypes at two weeks post-surgery (p < 0.01). For the remaining taste buds, gustatory innervation was nearly gone with only a little innervation from the trigeminal nerve remaining in the taste bud (p < 0.01). Taste bud volume (p < 0.01) and taste cell number (p< 0.01) were reduced by half for both control and experimental genotypes. Eight weeks post-surgery, taste bud number recovered in mice without Bdnf gene deletion, but did not recover in mice following Bdnf gene deletion (p < 0.01). Gustatory nerve innervation returned in 70% of the taste buds in control mice (p< 0.01). For those reinnervated taste buds, both taste bud volume and taste cell number increased to normal levels. However, in mice lacking the Bdnf gene, gustatory fibers only reinnervated 7.8% of the taste buds (p < 0.01); for most uninnervated taste buds, both taste bud volume and taste cell number remained small. These experiments demonstrate that BDNF is crucial for promoting regeneration of gustatory nerve fibers in adulthood. Following gustatory nerve section, considerable adult plasticity has been observed on the contralateral side including enlarged taste buds with more cells (Guagliardo and Hill, 2007). To determine if this anatomical change was associated with alter Bdnf expression. I examined Bdnf level in the geniculate ganglion and tongue epithelium on the contralateral side following chorda tympani nerve section. Results showed Bdnf expression increased two fold at two weeks post-surgery in geniculate ganglion on the contralateral side (p < 0.05), indicating BDNF may involve with the observed plastic changes. To determine if the increase in taste bud size was associated with increased innervation and/or regulated by BDNF, the Bdnf gene was then deleted in inducible knockout mice before nerve surgery, and taste bud size and amount of innervation were measured on the contralateral side. The results showed taste bud volume, taste cell number and a marker for nerve fibers all increased on the contralateral side in mice without Bdnf gene deletion at eight weeks post-surgery. This indicates that larger taste buds could be supported by increased TUJ1 positive fibers from trigeminal nerve. In addition, in mice lacking Bdnf, taste bud volume, taste cell number and innervation did not increase on the contralateral side after surgery, which indicates that Bdnf may contribute to larger taste buds on the contralateral side following nerve section by supporting increased innervation to the larger taste buds.

A Novel Diffusion Tensor Imaging-Based Computer-Aided Diagnostic System For Early Diagnosis Of Autism., Mahmoud Mostapha
Electronic Theses and Dissertations
Autism spectrum disorders (ASDs) denote a significant growing public health concern. Currently, one in 68 children has been diagnosed with ASDs in the United States, and most children are diagnosed after the age of four, despite the fact that ASDs can be identified as early as age two. The ultimate goal of this thesis is to develop a computer-aided diagnosis (CAD) system for the accurate and early diagnosis of ASDs using diffusion tensor imaging (DTI). This CAD system consists of three main steps. First, the brain tissues are segmented based on three image descriptors: a visual appearance model that has …
The Second Generation Of The Cccm System For In-Vitro Cardiac Tissue Engineering., Mai-Dung Thi Nguyen
The Second Generation Of The Cccm System For In-Vitro Cardiac Tissue Engineering., Mai-Dung Thi Nguyen
Electronic Theses and Dissertations
Cardiovascular disease is the leading cause of death worldwide. When a myocardial infarction occurs, scar tissue compensates the damaged myocardial tissue. This scar tissue increases the stiffness of the heart tissue, reduces the heart’s function, and finally leads to the heart failure (HF) disease. To have the tissue engraftment, in-vitro cardiac tissue should have the same properties as the native mature cardiac tissue. However, current in-vitro cell culture technologies fail to accurately recreate the in-vivo like mechanically physiological environment for in-vitro cardiac tissue culture, and therefore, fail to regenerate the in-vivo like mature cardiac tissue. Hence, a microfluidic cardiac cell …

A Meta-Analytic Review Of The Relationships Between The Therapeutic Alliance, Empathy, And Genuineness In Individual Adult Psychotherapy., Jacob B. Nienhuis
Electronic Theses and Dissertations
This dissertation explored the relationship between the therapeutic alliance and perceptions of therapist empathy and genuineness through a systematic review and meta-analysis. Alliance, empathy, and genuineness are each integral parts of the therapeutic relationship. Prior meta-analyses demonstrated that alliance, empathy, and genuineness each had a moderate relationship to therapy outcome. No previous analysis has explored how therapist empathy and genuineness contribute to the therapeutic alliance. Studies for this analysis were obtained through a multi-part search strategy. Out of 2,141 obtained abstracts, 46 studies contained enough data for inclusion in the meta-analysis. Thirty-six studies reported alliance/empathy relationships, six studies reported alliance/genuineness …
The Impact Of Preconception Maternal Stress On Fecundability And Adverse Pregnancy Outcomes., Shekufe Akhter
The Impact Of Preconception Maternal Stress On Fecundability And Adverse Pregnancy Outcomes., Shekufe Akhter
Electronic Theses and Dissertations
It has been commonly hypothesized (and widely believed) that maternal stress either prior to or during pregnancy can adversely affect fertility and pregnancy outcomes. However, surprisingly few epidemiologic studies have rigorously tested these hypotheses. The current study evaluated the effects of pre-conception self-reported stress on fecundability and spontaneous abortion. The study population was derived from the Mount Sinai Study of Woman Office Workers with 487 women included. Women recorded stress (scale from 1 to 4) and covariate information in a daily diary for 12 cycles or until pregnant. Results indicated a significantly increased risk of spontaneous abortion for women with …
Mirna-21 Has Anti-Inflammatory Effects In The Macrophage Response To Peritonitis., Rebecca Elise Barnett
Mirna-21 Has Anti-Inflammatory Effects In The Macrophage Response To Peritonitis., Rebecca Elise Barnett
Electronic Theses and Dissertations
This project investigated the role of miRNA-21 in the macrophage response to peritonitis; with the goal of understanding whether the associated mechanism has potential benefits in the clinical treatment of peritonitis. A novel therapeutic intervention in the treatment of peritonitis could modulate the host immune response to decrease an exaggerated pro-inflammatory innate response and then prevent or ameliorate the development of sepsis and organ failure. We found the following: 1. MiRNA-21 expression increases in peritoneal macrophages after lipopolysaccharide (LPS) stimulation. 2. Over expression of miRNA-21 decreases TNF-a secretion from macrophages after LPS stimulation. Suppression of miRNA-21 expression results in increased …

Nanoporous Bi2te3 Thermoelectric Based Knudsen Gas Pump., Abderrazzak Faiz
Electronic Theses and Dissertations
This dissertation reports on the fabrication of a bi-directional Knudsen gas pump using a multifunctional nanoporous thermoelectric material. The integration of the channels into the thermoelectric material has the advantages of reducing heat losses and power consumption, improving the pump efficiency, and reducing the overall pump size. The Knudsen pump operates on the thermal transpiration principle, the phenomenon that gas molecules drift from the cold end to the hot end of a narrow channel. The gas pumping is realized in the transition or free molecular regime. The Knudsen pump is needed in a great number of applications such as gas …

Evaluation Of The Safety And Pharmacokinetic Profile Of The Broad Spectrum Antiviral Lectin Griffithsin., Christopher Lynn Barton
Electronic Theses and Dissertations
Carbohydrate binding agents that target viral envelope glycans are being studied for their potential use as microbicides and antiviral therapeutics. Griffithsin is a lectin originally identified in a red alga Griffithisia sp. Multiple studies have shown that GRFT inhibits HIV-1, Coronaviruses, Hepatitis C, influenza and Ebola virus replication in vitro. This antiviral activity suggests potential uses in chemoprophylaxis and disease treatment. However, safety of GRFT administration has not been extensively studied. In vivo testing--chronic subcutaneous treatment as well as single dose subcutaneous, oral, and intravenous administrations of Griffithsin in Sprague Dawley Rats (Rattus Norvegicus)--was used to assess Griffithsin’s pharmacokinetic properties …

Electronic Theses and Dissertations
This thesis examined whether water quality diminished as a result of increasing pollution downstream along the Middle Fork of Beargrass Creek, Louisville, Kentucky in response to urbanization. Samples of key water quality indicators/pollutants were collected for 12 weeks (November - January) along the Middle Fork of Beargrass Creek at three different stream gauges. A spatial and statistical analysis was performed to verify any significant correlations between the water quality indicators and pollutants, and whether basin wide landcover/sewer outlet numbers led to an increase in these indicators downstream. Following ANOVA and Kruskal-Wallis statistical testing it was established that turbidity significantly increased …
Bone Strength And Architecture : Pharmacological Targeting Of Camkk2 As A Method For Enhancing Bone Quality., Zachary James Pritchard
Bone Strength And Architecture : Pharmacological Targeting Of Camkk2 As A Method For Enhancing Bone Quality., Zachary James Pritchard
Electronic Theses and Dissertations
Mice lacking or pharmacologically inhibited for calcium/calmodulin-dependent protein kinase kinase 2 (CaMKK2) have enhanced bone mass and microarchitecture. This enhanced bone mass and architecture is due to changes in osteoblast and osteoclast numbers as well as activity. Whether the enhanced bone mass translated to increased bone quality and strength was further explored by developing a method to directly test the trabecular strength of the distal femur. Micro computed tomography (CT)-based measurement of the length of the epiphysis of the distal femur, aided in its removal, allowing the exposure of the trabecular bone volume used to determine the microarchitecture parameters. Following …
How Do Individual Factors Influence Moral Decision Making In Entrepreneurship? : The Role Of Self-Construal, Temporal Construal And Moral Identity., Shanshan Qian
Electronic Theses and Dissertations
Some entrepreneurs are often perceived to do almost anything to succeed and pursue self-interests while breaking moral and ethical standards. This is particularly severe when the ventures are at the early stage because entrepreneurs face scarce resources, high uncertainty and a competitive environment. It is noted that entrepreneurs' behavior of conforming business ethics and morality is profound for entrepreneurs’ firms viability. Thus, this dissertation employs self-construal theory and construal level theory and identifies how entrepreneurs’ cognitive development influences entrepreneurs to make moral decisions. In addition, I address the role of entrepreneurs’ moral identity in the focal relationships. Data were collected …
Investigation Of Stabilization Mechanisms For Colloidal Suspension Using Nanoparticles., Qingwen He
Investigation Of Stabilization Mechanisms For Colloidal Suspension Using Nanoparticles., Qingwen He
Electronic Theses and Dissertations
Over the past decade, charged nanoparticles have been found to enhance the stability of colloidal suspensions. One promising explanation of this stabilization mechanism is “Nanoparticle Haloing”- the formation of a non-adsorbing nanoparticle layer surrounding neutral colloids that would induce an effective electrostatic repulsion between them. The objective of this work was to investigate the effect of additional charged nanoparticles on the interaction between neural colloids in nanoparticle-regulated complex fluids. Firstly, colloidal probe Atomic Force Microscopy (CP-AFM) is used to directly measure the interaction force in such system with nanoparticle volume fractions varying from 10-6 to 10-2. It is found that …
Genetic And Mechanistic Analysis Of Rat Mammary Cancer Susceptibility., Jennifer Sanders
Genetic And Mechanistic Analysis Of Rat Mammary Cancer Susceptibility., Jennifer Sanders
Electronic Theses and Dissertations
Breast cancer is a complex disease, which is influenced by genetic, epigenetic and environmental components. Genetic susceptibility to breast cancer is made up of high, moderate and low penetrance alleles. High and moderate penetrance alleles are rare and constitute only a small percentage of the genetic susceptibility. Most variation in genetic susceptibility is controlled by low- penetrance, common polymorphisms. Comparative genetics uses model organisms to study human disease. Rat strains exhibit different susceptibility phenotypes to chemical induced carcinogenesis. The Wistar-Furth (WF) rat strain is susceptible to chemically induced mammary carcinogenesis, while the Wistar-Kyoto (WKy) and Copenhagen (COP) rat strains are …
Classification System Of The Physical Streambed Habitat., Abolfazl Shafaie
Classification System Of The Physical Streambed Habitat., Abolfazl Shafaie
Electronic Theses and Dissertations
A streambed is the channel bottom of a stream, river, or creek; the physical confine of the normal water flow. It provides necessities for fish and microinvertebrate including shelter, food, and breeding spots. Therefore, it is an essential part of aquatic animal habitats and often seen as an index of stream health. There is no generally accepted method to quantify streambed physical components and depending upon state and organization a variety of methods are used. The procedure provided in the rapid bioassessment protocol (RBP) (Kaufmann et al., 1999; Sylte and Fischenich, 2002) is probably the most widely used method to …
Biophysical And Computational Investigations Into G-Quadruplex Structural Polymorphism And Interaction With Small Molecules., Huy Tuan Le
Electronic Theses and Dissertations
In the cell, guanine-rich nucleic acids can self-assemble into unique four stranded tertiary structures known as G-quadruplexes. G-quadruplex formation in the telomere leads inhibits telomerase, an enzyme activated in cancer cells to maintain the telomere and allowing for cancer cells to achieve immortality. G-quadruplex formation in the promoters and 5’-untranslated regions regulates the expression of many oncogenes. Furthermore, G-quadruplex formation during cellular replication promotes genomic instability, a characteristic which enables tumor development. Because of their implication in cancer, G-quadruplex structures have emerged as attractive drug targets for anti-tumor therapeutics. In the current dissertation work, we present three experimental approaches to …
Il-12 Induced Modulation Of Tumor Derived Myeloid Cell Inflammatory Responsiveness., Courtney Jetun Mitchell
Il-12 Induced Modulation Of Tumor Derived Myeloid Cell Inflammatory Responsiveness., Courtney Jetun Mitchell
Electronic Theses and Dissertations
Tumor infiltrating and tumor associated myeloid cells (TIMs and TAMs) elaborate an array of factors that promote tumor growth and metastasis. IL-12, a potent inflammatory cytokine has been shown to induce regression of many cancers. We hypothesize that IL-12 augments the ability of TIMs and TAMs to respond to inflammatory stimuli providing a window in which these stimuli are more likely to promote tumor destruction. Related to this hypothesis, we asked two questions: (1) Does IL-12 directly change signaling events associated with inflammatory signal transduction? (2) Is IFN? required for the entirety of IL-12 induced enhancement of the response of …

Interleukin-6 Production And Epithelial Cell Signaling By Telomerase Immortalized Gingival Keratinocytes In Porphyromonas Gingivalis Infection., Himabindu Vuddaraju
Electronic Theses and Dissertations
P. gingivalis is a major periodontal disease causing pathogen. Gingival epithelial cells being the first line of defense, interact and internalize the bacteria evoking a cell signaling response that finally leads to the release of cytokines. IL-6 is a potent pro- inflammatory cytokine, which is one of the responsible factors for bone resorption, a clinical manifestation of periodontitis. The experimental model study hypothesizes that P. gingivalis infection increases the production of IL-6 in telomerase immortalized gingival keratinocytes (TIGKs) through up-regulation of c-Jun (AP-1 reporter). We observed the IL-6 production post-infection with P. gingivalis WT ATCC33277 and mutants ?0482, ?Ndk and …

Electronic Theses and Dissertations
Although the diagnostic criteria for mild cognitive impairment have evolved considerably since their inception, they remain varied and able to be interpreted and implemented in different ways depending on the judgment of the clinician. Because of this issue, a wide range of incidence, prevalence, and conversion rates are found in the research literature. Using data collected from 400 patients with amnestic mild cognitive impairment, limitations inherent in current mild cognitive impairment diagnostic criteria were addressed. First, using Alzheimer’s Disease Neuroimaging Initiative diagnostic criteria, an equation was constructed to predict conversion from mild cognitive impairment to dementia by analyzing the predictive …

Electronic Theses and Dissertations
The treatment of cancer with chemotherapeutic agents has made great strides in the last few decades but still introduces major systemic side effects. The potent drugs needed to kill cancer cells often cause irreparable damage to otherwise healthy organs leading to further morbidity and mortality. A therapy with intrinsic selective properties and/or an inducible activation has the potential to change the way cancer can be treated. Gold nanoparticles (GNPs) are biocompatible and chemically versatile tools that can be readily functionalized to serve as molecular vehicles. The ability of these particles to strongly absorb light with wavelengths in the therapeutic window …

The Effect Of Romantic Jealousy On Self-Control: An Examination Of Trait Constructs And Sex Differences Based On Survey And Experimental Data, Lyndsay A. Nelson
Electronic Theses and Dissertations
A large body of research has demonstrated that the experience of romantic jealousy is often associated with a variety of negative outcomes. However, evolutionary psychologists have provided evidence that jealousy is an adaptive emotion that can aid with mate retention. Together these lines of research suggest that jealousy may at times work to protect and enhance one’s relationship, whereas in other cases it could lead to harmful consequences. Considering the varying outcomes of jealousy, it is critical that research explore more specifically how this complex state operates and how it affects individuals’ functioning. In the present research I conducted 2 …
