Impact Of Development On Freshwater Turtle Populations Of Coastal South Carolina: An Examination Of Land Use And Road Density, Andre Dominguez

Previous studies (Haxon 2000; Steen and Gibbs 2004; Marchand and Litvaitis 2004; Aresco 2005) have demonstrated that urbanization and development have disproportionally adverse affects on female turtles. In order to test this paradigm I caught 301 turtles (primarily Trachemys scripta scripta) in ten ponds (5 impacted and 5 unimpacted) across coastal South Carolina. Not only do coastal South Carolina turtles not follow the same male biased pattern of many other studies, there was an overall female bias (154:95). Turtle metrics were between the types of sites existed as well, indicating larger/older population of turtles in unimpacted sites. Water quality parameters …

Few studies have focused on mercury cycling within salt marsh estuaries. Mummichogs (Fundulus heteroclitus) and Atlantic silversides (Menidia menidia) are two species of forage fish that live year round in South Carolina salt marshes. During high tide, mummichogs feed from the water column, sediment, and off of smooth cordgrass while Atlantic silversides prey upon zooplankton. In this study, total and methyl mercury within mummichogs and Atlantic silversides from Dunn Sound, SC were quantified and compared over four seasons throughout 2014. Harmful algae blooms (HABs) and toxins associated with HAB species are a significant threat to the health of marine species and coastal environments. In the South Carolina coastal zone, HABs have proliferated with intensified anthropogenic eutrophication in coastal waters. The raphidophyte species Chattonella subsalsa is a prominent cause of algal blooms resulting in fish kills in South Carolina. Though C. subsalsa is a known ichthyotoxic alga, the fish kill mechanism of this species remains unidentified. The distribution and foraging behavior of predatory salt marsh fishes is shaped by the movements of prey, many of whom seek shelter in shallow and tributary creeks. I hypothesized that the distribution of piscivorous fishes in marsh creek channels would differ with proximity to the mouths of intertidal creeks and with tidal stage. Custom-built trotlines baited with live minnows were deployed during four discrete tidal stages at two microhabitats in main creek channels: (1) adjacent to intertidal creek mouths, and (2) along straight banks in North Inlet estuary, South Carolina. The morphological changes of small tidal creeks, driven by coastal processes, can pose risks to infrastructure and engineered coastlines and often rely on dredging to maintain them. These changes along the beachface can negatively affect the health of associated estuaries where open exchange between the ocean and creek basins is vital. Stable isotope analyses of multiple tissue types have been used to reconstruct life histories of individuals from past populations. This thesis presents the life history reconstructions of a unique population recovered from a 16th to 18th century cathedral crypt located in Vilnius, Lithuania. The sample consists of 23 individuals (21 males, one possible female, and one juvenile). Stable carbon, nitrogen, and oxygen isotope analyses were performed on bone and dentin collagen, and on bone and enamel hydroxyapatite, resulting in eight isotope values per individual, providing both childhood and adult dietary and migration information. The primary question this dissertation aims to answer is how ethics can be meaningfully integrated into health technology assessment (HTA). The contribution of this dissertation is two-fold. The first is to provide an honest and critical evaluation of current HTA practices, including those that involve ethics assessments and that do not. This evaluation will identify gaps and deficiencies in current HTA practices. The second contribution of this dissertation is to propose an approach for meaningful ethics assessment. Hantaviruses, family Bunyaviridae, are present throughout the globe in a variety of mouse, rat, mole, vole, shrew, or bat species. Hantaviruses persist for the lifetime of the animal reservoir, while causing no signs or symptoms of disease. Only the rodent-borne hantaviruses cause disease in humans. In contrast, a “spillover” infection of a hantavirus into a nonreservoir rodent species results in an asymptomatic acute infection. We and others in the field are interested in understanding the biology of these virus-host interactions and mechanisms that underlie these three very different outcomes. Nano-scale particles sized 10—400 nm administered systemically preferentially extravasate from tumor vasculature due to the enhanced permeability and retention effect. Therapeutic success remains elusive, however, because of inhomogeneous particle distribution within tumor tissue. Insufficient tumor vascularization limits particle transport and also results in avascular hypoxic regions with non-proliferating cells, which can regenerate tissue after nanoparticle-delivered cytotoxicity or thermal ablation. Altered energy metabolism is an established hallmark of cancer cells. Fructose-2,6-bisphosphate is an allosteric activator of glycolysis and its concentration in a cell is dictated by the 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatases (PFKFB1-4) family of bifunctional enzymes. The PFKFB family of enzymes are characterized by their different kinase: bisphosphatase activities and were originally determined to be tissue-specific. More recent data however suggest that multiple isoforms are co-expressed in both normal and neoplastic tissues. PFKFB4 is highly expressed in human cancer, strongly induced by hypoxia, and required for the survival of cancer cells. Disordered eating is associated with several negative outcomes and often lead to diagnosable eating disorders. The coping strategies used by those with disordered eating may contribute to such consequences. Past research has found experiential avoidance to be common among those with disordered eating to escape uncomfortable internal experiences. The current study suggested that repetitive thinking and overvaluation of weight and shape are two such mechanisms by which avoidance of negative core beliefs, in particular, is achieved. Similar concepts have been investigated in undergraduate students, but there is paucity of research of attrition in graduate students. It is widely recognized that many cancer therapies are effective only for a subset of patients. However clinical studies are most often powered to detect an overall treatment effect. To address this issue, classification methods are increasingly being used to predict a subset of patients which respond differently to treatment. This study begins with a brief history of classification methods with an emphasis on applications involving melanoma. Nonparametric methods suitable for predicting subsets of patients responding differently to treatment are then reviewed. Polychlorinated biphenyls (PCBs) are persistent environmental toxicants, present in 100% of US adults and dose-dependently associated with nonalcoholic fatty liver disease (NAFLD) in epidemiologic studies. PCBs are predicted to interact with receptors previously implicated in xenobiotic/energy metabolism and NAFLD. These receptors include the aryl hydrocarbon receptor (AhR), pregnane xenobiotic receptor (PXR), constitutive androstane receptor (CAR), peroxisome proliferator-activated receptors (PPARs), liver-X-receptor and farnesoid-X-receptor. The work presented in this dissertation outlines the role of stem cells in the remodeling of adipose tissue under conditions of nutrient excess. Obesity-induced variations in adipose tissue stem cell distribution were uncovered by analysis of the stromal vascular fraction isolated from mice fed a high fat diet for several weeks. Bone marrow cell transplantation was used to determine the derivation of progenitor cells found in adipose tissue; and supplementation of depleted progenitor cell populations achieved via direct cell transplantation, helped to determine the contribution of these progenitor cells to the remodeling process. Current methods are based on using off-the-shelf wires or electrodes to deliver a stimulus voltage to the in-vitro tissue in culture medium. The type IIa sodium-phosphate cotransporter (Npt2a) is expressed in the apical membrane of the renal proximal tubule and is responsible for the reabsorption of the majority of the filtered load of phosphate. Parathyroid hormone (PTH) is secreted by the parathyroid gland in response to a decrease in serum calcium or an increase in serum phosphorus, and acutely induces phosphaturia through the rapid stimulation of endocytosis of Npt2a and its subsequent lysosomal degradation. Chronic PTH stimulation leads to sustained phosphaturia, but the mechanisms for the chronic regulation of Npt2a by PTH remain unclear. We hypothesize that PTH decreases Npt2a mRNA levels …

Host cells respond to bacterial lipid A through Toll-like receptor 4 (TLR4). Activation of TLR4 by lipid A triggers a response that involves two main adaptor proteins, MyD88 and TRIF. MyD88-dependent gene expression is associated with proinflammatory protein production, while TRIF-dependent gene expression is essential for optimal activation of adaptive immunity by antigen-presenting cells. Detoxified, monophosphoryl lipid A agonists (MPLA or synthetic MLA) were previously suggested to elicit TRIF-biased TLR4 signaling; that is, induction of weaker MyD88-associated gene expression but relatively intact TRIF-dependent gene expression when compared to fully active diphosphoryl lipid A (lipid A). Invasive species are one of the leading causes of biodiversity loss and their presence can significantly impact the structure and function of native ecosystems. In this dissertation, the impacts of the invasive honeysuckle shrub, Lonicera maackii, on exotic earthworm populations, leaf litter decay responses, and inorganic and organic nutrient pools in an urban woodland park are assessed. Chapter 1 provides a brief review of honeysuckle's effects on forest ecosystems. Detrusor-sphincter dyssynergia after chronic spinal cord injury (SCI) leads to over-distention, high bladder pressures and vesico-ureteric reflux into the kidney, resulting in repeated infections and pyelonephritis. Given the importance/limited knowledge of SCI effects on the upper urinary tract, this study assessed expression of two proteins in the kidneys of contused (T9) male rats relative to controls that are indicative of tissue stress/damage (TGFβ, CD11b). The reaction of an aminooxy moiety (RONH2) with an aldehyde or ketone carbonyl, an oximation reaction, results in the formation of robust oxime ether linkages. Oximation is a form of click chemistry and is chemoselective, occurs in a variety of solvents including water, and produces high yields with little to no purification. We were inspired to exploit the advantages of oximation reactions by attaching aminooxy-functionalized molecules to solid supports, thus allowing us to employ aminooxy chemistry in ways that cannot be achieved using solution phase. Running economy (RE) is the amount of oxygen consumed while running at a submaximal intensity. Along with aerobic capacity (VO2max), RE is an important predictor of running performance. Little research has investigated changes in RE with restricted breathing training [i.e. controlled breath-holding (CFB)] during exercise. RE may improve ~6% amongst a novice swimming cohort through CFB training, but this has not been established in elite swimmers. In tumor cells, glutamine metabolism provides anaplerotic carbon for the TCA cycle, fatty acid synthesis, and precursors for the production of nucleotides and glutathione. This metabolic alteration is primarily driven by disruptions in oncogenic or tumor suppressor function and results in glutamine dependency for tumor cell survival. Troglitazone, a PPARγ agonist, has been reported to alter glutamine metabolism. This thesis project addresses whether troglitazone treatment could disrupt glutamine metabolism in glutamine-dependent tumor cells. Results obtained with troglitazone treatment include: dose-dependent inhibition of cell proliferation, glutamine uptake, glutaminolytic protein expression, steady-state ATP levels, and glutamine carbon flux into the TCA cycle. …

An inventory of vascular plants was conducted in Steele Creek Park in Sullivan County, Tennessee from March 2013 to September 2014. The park covers an area of 892 ha and lies within the Ridge and Valley Province. The inventory of vascular plants documented 547 species of 323 genera and 101 families. Two hundred sixteen taxa were newly reported for Sullivan County. Tennessee Special Concern Species included Cardamine rotundifolia, Castanea dentata, Lonicera dioica, Allium tricoccum, Cypripedium acaule, and Panax quinquefolius. Plants are in a perpetual evolutionary arms race with a wide range of pathogens. Their sessile nature has led plants to evolve defense mechanisms that can quickly recognize a unique stressor and deploy a resistance tailored for a specific attack. The salicylic acid (SA) mediated defense pathway has been shown to be one of the major defense tactics plants can initiate to defend themselves against microbial pathogens. Following a pathogen attack high levels of methyl salicylate (MeSA) are produced that can be converted to SA by the enzyme salicylic acid binding protein 2 (SABP2).
