Due to cancer recurrence and the development of drug resistance, metastatic breast cancer is a leading cause of death in women. In the search for a new therapeutic to treat metastatic disease, we discovered CT20p, an amphipathic peptide based on the C-terminus of Bax. Due to inherent properties of its sequence and similarity to antimicrobial peptides, CT20p is a promising cytotoxic agent whose activity is distinct from the parent protein (e.g. does not cause apoptosis). CT20p is not membrane permeable but can be introduced to cells using polymeric nanoparticles, a method that promotes efficient delivery of the peptide into the …

Cardiovascular diseases (CVDs) have been recorded as the number one cause of death worldwide, accounting for 32% of total deaths annually. More than two-thirds of all CVD cases are associated with atherosclerosis, which is the accumulation of fats and other substances causing plaque formation in the interior walls of major arteries. This leads to narrowing of the lumen and hardening of the arteries, ultimately resulting in angina, heart attack and/or stroke. Studies have shown that the pathogenesis of atherosclerosis and associated CVDs is strongly linked to elevated secretion of liver-specific lipoproteins called very-low-density-lipoprotein (VLDL). It is convenient and useful to model the behavior of digital virus outbreaks that occur, globally or locally. Compartmental models will be used to analyze the mannerisms and behaviors of computer malware. This paper will focus on a computer worm, a type of malware, spread within a business network. A mathematical model is proposed consisting of four compartments labeled as Susceptible, Infectious, Treatment, and Antidotal. This research investigates through computational methods whether the physical properties of DNA contribute to its harmonic signature, the uniqueness of that signature if present, and motion of the DNA molecule in water. When DNA is solvated in water at normal 'room temperature', it experiences a natural vibration due to the Brownian motion of the particles in the water colliding with the DNA. Located in Florida's panhandle, the Apalachicola River is the southernmost reach of the Apalachicola-Chattahoochee-Flint (ACF) River basin. Streamflow and sediment drains to Apalachicola Bay in the Northern Gulf of Mexico, directly influencing the ecology of the region, in particular seagrass and oyster production. The objective of this study is to evaluate the response of runoff and sediment loading in the Apalachicola River under projected climate change scenarios and land use / land cover (LULC) change. Fatigue in healthcare providers has been linked to dangerous outcomes for patients, including medical errors, surgical complications, and accidents. Resident physicians, who traditionally work long hours on minimal sleep, are among the most fatigued. In attempt to mitigate the impact of fatigue on resident physician performance and improve patient safety, the Accreditation Council for Graduate Medical Education (ACGME) implemented a fatigue intervention program in 2011 for medical residency programs in the United States. This caused a significant decrease in the number of hours that first-year residents were permitted to work, compared with hours worked by first-year residents in prior years. …

Carboxyphosphate (CP) is an important intermediate involved in reactions catalyzed by acetyl-CoA carboxylase, pyruvate carboxylase, N5-Carboxyaminoimidazole ribonucleotide synthetase, propionyl-CoA carboxylase, urea amidolyase, and carbamoyl phosphate synthetase. Despite its important role, properties for CP have never been reported due to its short estimated half-life (t1/2 70 ms). Thus, the high level ab initio methods, MP2 and CCSD(T), along with the DFT functionals: B3LYP, BB1K, M05-2X, M06-2X, and MPW1K were used to investigate the structure and energetics of CP in both vacuum and the PCM continuum solvation model of water. Parkinson's disease is characterized by the progressive spread of protein misfolding stress, or proteotoxicity, across the brain. During this protracted process, the allocortex of the temporal lobe develops protein inclusions before the neocortex in the frontal and parietal lobes. In the present study we tested the hypothesis that the staged appearance of proteotoxicity in allocortex followed by neocortex is the result of intrinsic vulnerability differences. We microdissected the neocortex and multiple subregions of the allocortex from rat brains and plated the primary neo- and allocortical neurons for parallel in vitro studies. Historically, chlorination has been widely utilized as a primary and secondary disinfectant in municipal water supplies. Although chlorine disinfection is effective in inactivating pathogenic microbes, the use of chlorine creates the unintentional formation of regulated chemicals. The development of a new deposition technique called evaporative vapor deposition (EVD) is reported, allowing deposition and formation of atomically-thin, large area materials on arbitrary substrates. This work focuses on the highly popular monolayer material – graphene oxide (GO). A droplet of a GO solution is formed on a heated polymer substrate, and maintained at steady-state evaporation (all droplet parameters are held constant over time). In the past decade, characterizing spiking neuron models has been extensively researched as an essential issue in computational neuroscience. In this thesis, we examine the estimation problem of two different neuron models. In Chapter 2, We propose a modified Izhikevich model with an adaptive threshold. In our two-stage estimation approach, a linear least squares method and a linear model of the threshold are derived to predict the location of neuronal spikes. Problem: Heart failure is the most frequent cause for hospital readmissions in Medicare recipients, with an estimated annual cost of $12 billion. Heart failure hospitalizations are also an independent risk factor for increased mortality. Self-care, thought to be enhanced by perceived social support, is key to managing this syndrome, and up to 50% of readmissions are considered the result of inadequate self-care. The US Military requires specific fitness testing, known as the Physical Readiness Test (PRT), for its members to determine their overall fitness levels. The test currently being used has been shown to have bias towards heavier mass individuals of up to 20%. Backround. A shift from a weight-focus to a health focus -Health At Every Size (HAES)-has been suggested. Yet, little research has compared the impact of this approach to a traditional weight loss program on diet, while physical activity, anthropometric and health indicators have been mixed. Objective. A statewide gear comparison was performed in South Dakota during 2013 and 2014 between current South Dakota Department of Game, Fish and Parks (SDGFP) sampling gears (i.e., gill nets and modified fyke nets) and their equivalents described in Standard Methods for Sampling North American Freshwater Fishes (Standard). The main limitation of meals from canola and other Brassica spp. is the presence of glucosinolates (GLS), which are anti-nutritional and can even be toxic at high ingestion levels. Furthermore, large amounts of GLS can reduce palatability for livestock and thus reduce intake and growth rates. For this reason canola was bred to contain lower levels of GLS (<30 >μmol/g) and erucic acid (< 2%). However, feed inclusion rates are still limited to ~30%, and this reduces the value of canola meal. The goal of this thesis was to optimize a pretreatment and fungal conversion process to enhance the nutritional value of canola meal. Various combinations of physical/chemical pretreatments, fungal cultures, and incubation methods were investigated to metabolize GLS into cell mass, CO2, or other non-toxic components. These treatments also served to hydrolyze canola meal fiber into carbohydrates which were then metabolized by the fungi into single cell protein. Solid-state incubation conditions were initially tested, since filamentous fungi are well adapted to grow at lower moisture levels, and this would potentially reduce contamination issues with bacteria. Flask trials were performed using 50% moisture, hexane extracted (HE) or cold pressed (CP) canola meal, with incubation for 168 h at 30ºC. On HE canola meal Trichoderma reesei (NRRL-3653) achieved the greatest increase in protein content (23%), while having the lowest residual levels of sugar (8% w/w) and GLS (0.4 μM/g). Porcine reproductive and respiratory syndrome virus (PRRSV) is an enveloped, single stranded, positive sense RNA virus and a member of Arteriviridae. Its genome encodes 10 open reading frames for at least 7 structural proteins and 14 non-structural proteins. Membrane (M), Nuclepcapsid (N), and Glycoprotein-5 (GP5) are the major structural proteins of PRRSV, while Envelope (E), Glycoprotein-2 (GP2), Glycoprotein-3 (GP3), and Glycoprotein-4 (GP4) are the minor structural proteins of PRRSV. Four different grazing systems: two rotational, a continuous, and a high-intensity, short-duration (mob) system, replicated twice, were evaluated from an economic perspective. Mob grazing is defined as a system having very high stocking rates for a small amount of time. Livestock are forced to eat or trample the vegetation. Stocking rates and average daily gains (ADG) were collected from the UNL Barta Brothers ranch near Rose, Nebraska. The study started in 2011 and lasted until 2014. We conducted a 2-year (2012–2013) study of survival and cause–specific mortality using individually marked adult cow and neonate elk (Cervus elaphus) occupying the southwestern region of the Black Hills. We used known-fate analysis in Program MARK for survival analysis of adult cows and calves. We estimated survival and cause-specific mortality of 49 adult female elk over the 2 years of the study. Annual adult cow survival was 0.85 (95% CI = 0.72–0.87). We documented 12 mortalities with harvest (58.3%) and predation (16.6%) accounting for the majority of known mortalities. We captured and fit 71 neonates < 10 days of age with expandable Very High Frequency (VHF) radiocollars during summer 2012 (n = 37) and 2013 (n = 34). Annual (12 month) survival of elk calves was 0.75 (95% CI = 0.61–0.84) while summer (20 weeks; 15 May–25 September) survival was 0.79 (95% CI = 0.68–0.88). Predation accounted for 87.5% of mortalities; remaining mortalities were from starvation (6.3%) and unknown (6. 3%) causes. We used Global Positioning System (GPS) collars to collect 167,707 locations to determine home range, movement ecology, and macroscale resource selection of 48 adult cow elk. We documented elk using a variety of migration strategies (obligate migrator, resident, conditional migrator, disperser); the majority of the population (58%) was migratory. Spring migration distance travelled ranged from 2.45 km – 74.44 km (n = 42); fall migration distances ranged from 6.41km – 153.95 km (n = 46). We used 99% Brownian Bridge Movement Models to create seasonal and overall home ranges of adult cow elk. Mean overall home range size for conditional migrators was 249.28 km2 (SE = 28.60, n = 7, range = 233.75), for obligate migrant elk it was 227.18 km2 (SE = 13.94, n = 29, range = 346.83), and for resident elk it was 175.65 km2 (SE = 22.75, n = 11, range = 216.04). We used discrete choice models to determine resource selection at the macro-habitat scale of collared adult elk using ArcMap 10.1 data. Adult cow elk selected for open grassland/herbaceous areas and early successional forest areas close to forested edges at higher elevations. Our study showed that elk populations have the ability to thrive within an ecosystem with healthy predator populations.
