Wall shear stress (WSS) is commonly accepted as the primary influence affecting characteristics of anchored endothelial cells when subjected to fluid flow. Orbital shakers are commonly used to study cellular responses due to their ease of use, ability to run several experiments simultaneously, and since they exert physiologically relevant oscillatory shear. These studies require comprehensive resolution of WSS, however the fluid dynamics inside orbiting culture dishes has not yet been well described since the flow is complex and difficult to quantify analytically. College students are at increased risk for weight-gain and obesity compared to their non-student peers. Although many studies have confirmed these findings, few address the underlying causes of weight-related behaviors in college students. Associations between sleep behaviors (sleep duration and sleep quality), stress and weight-related behaviors (physical activity and food choices) have been identified, but studies have not adequately explored these associations among college students. IL-17 producing γδ T cells (γδT17) promote numerous autoimmune diseases such as psoriasis and arthritis, as well as, cancers of the colon, lung and breasts. Yet γδT17 peripheral regulation has yet to be thoroughly explored. In mice deficient in IL-17 signaling, we observed expansion of γδT17 in all major tissues. However, γδT17 expansion was not uniformly distributed systemically and was most prominent in oral draining cervical lymph nodes (LNs) with monoclonal expansion of Vγ6 γδT17. In vitro proliferation assays of these cervical LNs showed endogenous proliferation by γδT17 dependent on cell-to-cell contact with CD103+ DCs. CD86+ and CD80 …

MicroRNAs (miRNAs) are a large number of small endogenous non-coding RNA molecules (18-25 nucleotides in length) which regulate expression of genes post-transcriptionally. While a variety of algorithms exist for determining the targets of miRNAs, they are generally based on sequence information and frequently produce lists consisting of thousands of genes. Canonical correlation analysis (CCA) is a multivariate statistical method that can be used to find linear relationships between two data sets, and here we apply CCA to find the linear combination of differentially expressed miRNAs and their corresponding target genes having maximal negative correlation. This thesis is an examination of the history of environmental change in the defunct Rio Grande river valley in the former Panama Canal Zone. By mining secondary sources and engineering records, this study provides a narrative of the historic river’s role in the formation of the so-called interoceanic corridor through the isthmus of Panama from 1521 to 1950. More importantly, however, as a case study of river histories in Latin American environmental history, this understudied, Pacific-draining river illustrates how nature and non-human nature made Panama’s strategic transit region into a loci for material flows over time. Periodontitis is a chronic inflammatory disease that is characterized by severe tissue destruction of the gingiva and other tooth supporting structures; if left untreated, tooth loss and disintegration of the alveolar bone occurs. This chronic inflammatory state has been linked to other systemic diseases such as cardiovascular disease, diabetes, rheumatoid arthritis, and Alzheimer’s disease. This dissertation is directed toward developing a statistical methodology with applications of the Conway-Maxwell-Poisson (CMP) distribution (Conway, R. W., and Maxwell, W. L., 1962) to count data. The count data for this dissertation exhibit three different characteristics: clustering, zero inflation, and dispersion. Clustering suggests that observations within clusters are correlated, and the zero inflation phenomenon occurs when the data exhibit excessive zero counts. Dispersion implies that the mean is greater/smaller than the variance unlike a Poisson distribution. Glomerulonephritis (GN) is the most common cause of end-stage renal disease worldwide and is characterized by deposition of immune complex, inflammation, proteinuria and podocyte damage. Immunosuppressive therapy is effective in less than 50% of patients, indicating that immunological activity is not the only relevant cause of kidney injury. While methods have been described for neutrophil-mediated immune activation, specific glomerular mechanisms for recruitment of neutrophils and development of GN have not been determined. This dissertation examines the population genetic dynamics of two Florida invasives: the Nile monitor (Varanus niloticus) and Argentine black and white tegu (Salvator merianae). Pseudomonas aeruginosa produces N-(3-oxododecanoyl)-homoserine lactone (C12) as a quorum-sensing molecule that functions to facilitate bacteria-bacteria communication. C12 has also been reported to affect many aspects of human host cell physiology, including evoking cell death in various types of cells. However, the signaling pathway(s) leading to C12-triggerred cell death remains unclear. To clarify cell death signaling induced by C12, we examined mouse embryonic fibroblasts (MEFs) deficient in one or more caspases. Our data indicate that, unlike most apoptotic inducers, C12 evokes a novel form of apoptosis in cells, probably through the direct induction of mitochondrial membrane permeabilization. This study examined the role of instructional assistant principals in distributed instructional leadership in three middle schools in a large urban school district. Using the leadership functions in Hallinger’s (2011) framework of instructional leadership, the distribution of instructional leadership functions were examined. Interviews, document analysis, and an observation determined that the assistant principal in only one out of three cases was engaging in distributed instructional leadership. One was named an instructional assistant principal and functioned as an operations manager and the third was called a “lead” assistant principal and also spent most of his time on operational/management functions. Previous studies of child molestation have largely used a sample of either incarcerated or convicted sex offenders. Previous studies have also failed to provide substantial analyses regarding how physical contacts between child victims and adult perpetrators were initiated and terminated at the crime scene; few studies have focused on “what exactly happened” while physical touching was taking place through an analysis of behavioral sequences of both child victims and adult offenders. Alcohol consumption can cause alcoholic liver disease (ALD), which remains a major cause of morbidity and mortality in the United States. Chronic alcohol consumption causes a pro-oxidant environment in the liver and increases hepatic lipid peroxidation. Acrolein is the most reactive and toxic aldehyde generated through lipid peroxidation. Acrolein forms protein adducts and triggers endoplasmic reticulum (ER) stress and hepatocyte apoptosis, which are recognized etiologic factors in ALD. The isometric mid-thigh pull (IMTP) has been cited often in the scientific literature; however, there is still a lack of agreement as to the ideal body position used during this test, and how body position impacts the relationship between IMTP performance and dynamic performance. MicroRNA (miR) dysregulation alters cancer-associated gene expression, which contributes to cancer pathogenesis. For example, miR-186 over expression lead to enhanced proliferation and migration in pancreatic cancer cell models. However, the role of miR-186 in prostate cancer (PCa) remains controversial. Previously, miR-186-5p was up-regulated in PCa patient serum (stage III/IV) compared to controls. Furthermore, miR-186-5p was up-regulated in metastatic PCa (PC-3, MDA PCa 2b, LNCaP) relative to normal prostate epithelial cells (RWPE1). We hypothesized miR-186 inhibition will reduce aggressive PCa using metastatic cell models. Myocardial voltage-gated potassium (Kv) channels regulate the resting membrane potential and the repolarization phase of the action potential. Members of the Kv1 and Kv4 family associate with ancillary subunits, such as the Kvβ proteins, that modify channel kinetics, gating and trafficking. Previous investigation into the function of cardiac β subunits demonstrated that Kvβ1 regulates Ito and IK,slow currents in the heart, but the role of Kvβ2 in the myocardium remains unknown. Since the 1970s, feminist art historians have extensively treated Mary Cassatt and Berthe Morisot. In particular, focusing on class-bound womanhood and domesticity, Griselda Pollock, Linda Nochlin, and Anne Higonnet have provided compelling psychoanalytic, Marxist, and semiotic analyses, seemingly exhausting all potentials for any further historical exploration of these artists. This project draws upon the work of Michel Foucault, Judith Butler, and Donna Haraway to critically analyze the political power and utility of origin stories as they are used within discourses of identity. I specifically examine the dominant cultural and counter-origin stories of transgender and fat bodies and argue that the counter origin stories constructed by both trans and fat studies/activism continue to engage with norms that regulate identity. MicroRNA-21 (miR-21) is an evolutionarily conserved microRNA, abundant in most cardiovascular tissues. It has been implicated in the pathogenesis of several cardiovascular diseases including restenosis, myocardial infarction, and heart failure. However, little is known about the contribution of miR-21 in atherosclerosis. My data show that expression of miR-21 is increased by >1.5-fold in murine atherosclerotic lesions and by 1.5-2.0-fold in the macrophages of Western diet (WD)-fed LDLR-KO mice (for 12-20 weeks). In vitro, LDL, oxidized LDL, acetylated LDL and LPS induced miR-21 by 2-4-fold and down-regulated its target protein, PDCD4, in bone marrow-derived macrophages. Methods: Our study population included 33,342 children, from Smile KY! annual screenings from 2002 to 20014. Data collection was through questionnaire forms. Our primary risk factor was “dental health instructions received” (yes/no) and our outcome of interest was “parental assessment of your child’s dental health” (good/fair/poor). Purpose: Left ventricular assist device (LVAD) therapy can be live-saving for advanced heart failure patients. Conventional anastomosis (surgical connection) of LVAD outflow grafts to the aorta requires aortic clamping and hand-suturing. Aortic clamping increases the risk for neurological complications. Hand-suturing may be time-consuming and requires significant surgical dexterity. There is currently no commercially available device for sutureless anastomosis of large vascular grafts (diameter > 5mm).
