The Northern Great Plains (NGP) mixed-grass prairie developed under fire, grazing, and periodic droughts, which created a heterogeneous landscape. Since European settlement, heterogeneous landscapes have declined due to fire suppression and the maximization of livestock production, leading to the need for an alternative management practice. Our overall project objective was to determine if heavy winter grazing could be used as an alternative to patch-burn grazing for promoting heterogeneity and improving ecosystem services in the NGP. Many in-field and edge-of-field management practices have been used to reduce nutrient loads from agricultural fields. The denitrification woodchip bioreactor (WB) is one edgeof- field management practice that has proven to be effective in removing nitrate from subsurface drainage water. The success in nitrate removal achieved with WBs has raised interest in expanding their capabilities for removing other agricultural pollutants, including phosphorus, by using other types of media like phosphorus-sorbing filters or combining these filters with woodchips to remove both nitrate and phosphorus as dual-nutrient removal systems. Managing fisheries requires balancing predators and prey. Prey stockings are often used when food sources may be limiting to support fish communities with multiple predators. Stocking of prey often results in increased predator size and could potentially lead to the consumption of nuisance species such as Black Bullheads Ameiurus melas. Multidrug resistance protein 1 (MRP1/ABCC1) actively transports a variety of drugs, toxic molecules and important physiological substrates across the plasma membrane. It can confer broad-spectrum drug resistance and can decrease the bioavailability of many important drugs such as anti-cancer agents, antibiotics, antivirals, antidepressants and antiinflammatory drugs. Calcein-AM, a fluorescent reporter commonly used for studying compound interactions with MRP1 was recently used in the development of a high content imaging-based assay by our group. This assay was robust and had better sensitivity than fluorescent plate readouts. The assay identified 12 MRP1 inhibitors after screening an anticancer library of 386 compounds. Due …

In South Dakota, grasshoppers (Orthoptera: Acrididae) are sporadic pests that can cause economic injury to rangeland and crops during outbreaks. It is important to know which grasshopper species are present as not all have the same potential to cause damage. USDA-APHIS conducts annual grasshopper surveys in western South Dakota rangelands, but the last published survey was in 1925. Of the potential biological control agents existing, grasshopper mites feed on grasshopper eggs and the larvae are ectoparasites of nymph and adult grasshoppers. Identifying precise transcription factor binding sites (TFBS) or regulatory DNA motif (motif) plays a fundamental role in researching transcriptional regulatory mechanism in cells and helping construct regulatory networks for biological investigation. Chromatin immunoprecipitation combined with sequencing (ChIP-seq) and lambda exonuclease digestion followed by high-throughput sequencing (ChIP-exo) enables researchers to identify TFBS on a genome-scale with improved resolution. A new species of Acrisis Foerster was reared and is described from the native prairie legume Glycyrrhiza lepidota (Nuttall) Pursh, a plant studied for its potential for conservation and remediation. When combining imaging and therapeutic motifs, known as theranostics, amphiphilic diblock copolymers have a strong precedent of success. These polymers can self-assemble into a variety of morphologies that are known to concentrate in areas of inflammation such as tumors. However, prediction of size and morphology from polymer structure still remains an open challenge. Phenotypic plasticity is an adaptation to unpredictable environments whereby an organism of a single genotype may express more than one phenotype under differing environmental conditions. Phenotypic plasticity can manifest as polyphenisms, which is an extreme form of phenotypic plasticity that produces two or more discrete, alternative phenotypes. The expression of alternative phenotypes is controlled by biotic and abiotic environmental factors, which variably affect the strength and direction of phenotypic outcomes. The need to improve behavior in order to improve academic achievement became evident after desegregating data and listening to community and staff members voice concerns. Using three intervention strategies, group counseling, teaching classroom management training, and pro-social activities, the program sought to decrease the number of discipline referrals written for a targeted group of students and students school-wide. Juvenile dragonflies (nymphs) may possess the ability to pass their microbiome to the adult life stage through metamorphosis. If this is so, the environment in which the nymph develops may have an effect on the adult microbiome. In this study, the gut microbiomes of 13 species of dragonfly were compared across life stages and when collected from environments at different levels of urbanization. The analysis of human remains in archaeological contexts is often complicated by the presence of highly fragmented and commingled remains. The standard methods used to help quantify the number of individuals and elements in these contexts are based upon the segmentation of whole bones. The methods provide standardization and are flexible enough to allow for the idiosyncratic nature of each context. However, this results in a lack of transparency, which is necessary to reanalyze the same sample or to compare “like” contexts, as the data collected will vary.
