This thesis examines a shifting paradigm in the U.S. healthcare system that will re-define the role that the healthcare provider plays in the production of health. This thesis analyzes archival data from archaeological sites with great kivas in the Reserve region of west-central New Mexico dating to A.D. 1000-1350 and examines sociopolitical organization and reform between the dynamic Reserve (A.D. 1000-1100) and Tularosa (1100-1350) Phases. Specifically, studies in this thesis compare great kiva architecture and ceramic types present between sites using methods of descriptive statistics and quantitative analysis, which allowed for interregional variation and change to be identified between those time periods. The results of those analyses are correlated with the archaeological histories of the Mimbres and Chaco societies that bordered the Reserve area in prehistory …

Trans-activation response (TAR) DNA-binding protein 43 (TDP-43) is a natively dimeric 414-residue protein that is encoded by the human TARDBP gene that has important implications in the pathogenesis of the neurodegenerative disorders ALS, FTD, and CTE. TDP-43 has been found hyperphosphorylated and ubiquitinated in the aggregates of the affected neurons of these diseases. The discovery of the presence of TDP-43 positive inclusions in brain matter of patients with CTE has made repetitive brain injury a possible environmental stimulus for aggregation in TDP-43 proteinopathies. Advanced parental age is an important aspect of parental condition that can have both positive and negative effects on offspring fitness, and thus, parental age can be considered a parental effect. The United Methodist Church (UMC) as an institution has strict language against lesbian and gay people and against the ordination of the lesbian and gay people. Prior research has focused on how denominations and congregations discuss issues around sexuality, how attitudes have shifted around issues of sexuality, and how to provide spiritual care for the LGBTQ+ community. This qualitative study interviewed lesbian and gay ordained elders in the UMC to learn about their experiences of serving as clergy people in the UMC even though there are prohibitive statements in church documents. EPR imaging at low frequency is a powerful tool to obtain important biological information in vivo in a non-invasive way. Properties of nitroxide and trityl radical imaging reagents have been studied. Developments in rapid scan imaging techniques are reported that improve efficiency of experiments and user-friendliness of software.
Relaxation and signal-to-noise ratio (S/N) in pulse experiments on trityl radicals were measured at frequencies between 400 MHz and 1.5 GHz. Relaxation time increases as the frequency increases and the radical concentration decreases. Protein disulfide isomerase (PDI) is an essential endoplasmic reticulum (ER) protein that acts as both an oxidoreductase and chaperone. It exhibits substantial flexibility and undergoes cycles of unfolding and refolding in its interaction with cholera toxin (Ctx), which is a unique property of PDI. This unfolding allows PDI to disassemble the Ctx holotoxin, which is required for Ctx activity. Here, we investigated the unfolding and refolding property of PDI and how this affects its interaction with bacterial toxins. PDI showed remarkable redox-linked conformational resilience that allows it to refold after being thermally stressed. Deletion constructs of PDI showed that both …

An additional goal of this project was to characterize the Wnt/β-catenin signaling pathway to unravel the adverse potential of MH on the cardiovascular system (CVS). Our study suggested that MH-mediated cardiovascular injury may be caused due to upregulation of some major components of the Wnt/β-catenin pathway through expression of excessive pro-inflammatory mediators. Moreover MH decreased expression of ErbB3 and NRG-2 suggesting impaired cardiac structure. MH exposure caused a slow heartbeat blood occlusion absence of blood circulation decreased hatching efficiency and increased mortality in medaka embryos in a concentration- and time-dependent manner. Reproductive interference occurs when the mating behaviors of one species negatively impact the fitness of another species. It is of increasing interest in invasive species biology because the introduction of alien species often leads to novel sexual interactions with native taxa, which can contribute to their decline. I examined whether reproductive interference plays a role in the decline of native green treefrogs (Hyla cinerea) following invasions of Cuban treefrogs (Osteopilus septentrionalis) in the southeastern United States. The molecular mechanism mediating the differentiation of neural progenitors to either glia or neurons is guided by the gene glial cells missing (gcm), a master regulator of glial cell differentiation. Knockout of this transcription factor (TF) and its homolog gcm2 in the fruit fly Drosophila Melanogaster results in embryos that develop neurons in place of lateral glia. Conversely, overexpression of gcm in those neural progenitors results in embryos that have substantially fewer neurons and excess glia(Jones, et.al, 1995; Hosoya, et.al, 1995; and Vincent, et.al, 1996). Using a transformative mixed methods approach, I explored the ways in which genres and racial projects are constitutive of each other and contribute to the racialization of Latinx students in higher education and result in disparate outcomes for Latinx students. Latinx research participants revealed the myriad ways in which they are racialized throughout the education system and the numerous ways they encounter whiteness on a daily basis. Specifically, we tackle current challenges in radiology screening process and introduce cutting-edge methods for image-based diagnosis, detection and segmentation. At the time of the Deepwater Horizon oil spill in 2010, very little was known about the inshore bottlenose dolphin (Tursiops truncatus) populations hit hardest by the spill. Without previous population demographic data impacts were challenging to assess. My dissertation was designed to build research capacity moving forward by helping to fill data gaps in the western Florida Panhandle. The first study presented here resulted in the first system-wide assessment of seasonal abundance, survival, and site fidelity patterns of bottlenose dolphins that inhabit the Pensacola Bay system. Accurate digital elevation models (DEMs) in salt marshes are crucial for modeling storm surges and determining the initial DEM elevations for modelling marsh evolution. Additionally, stormwater often contains elevated levels of pathogens which can pollute the receiving water body and impact reuse applications [1-4]. Passive and electrically active coalescence and mixing of pairs of trapped squeezed nanodroplets were studied in this work. PDMS-glass microfluidic devices were designed and fabricated using multilevel photolithography technique. Flow-focusing method was used to generate nanoliter droplets of died glycerol inside oleic acid. The effect of factors such as flow rates and their ratio, interfacial tension, and viscosities on the size and frequency of droplet generation was studied and concluded by demonstrating the capillary number effect. Coastal Wetlands (CW) provide numerous imperative functions and provide an economic base for human societies. Therefore, it is imperative to track and quantify both short and long-term changes in these systems. In this dissertation, CW dynamics related to hydro-meteorological signals were investigated using a series of LANDSAT-derived normalized difference vegetation index (NDVI) data and hydro-meteorological time-series data in Apalachicola Bay, Florida, from 1984 to 2015. NDVI in forested wetlands exhibited more persistence compared to that for scrub and emergent wetlands.
