Hepatitis C virus (HCV) or human immunodeficiency virus (HIV) infection leads to a phenomenon of inflammaging, in which chronic infection or inflammation induces an immune aged phenotype with T cell dysfunction. Thus, HCV or HIV infection has been deemed as a model to study the mechanisms of T cell infammaging and viral persistence in humans. In this dissertation, T cell homeostasis, DNA damage and repair machineries were investigated in patients with chronic HCV or HIV infection at risk for inflammaging. Childhood adversity is associated with numerous negative outcomes across multiple domains, including mental and physical health, interrelationships, and social functioning. Notably, research suggests that childhood adversity has a dose-response relationship with these outcomes; that is, greater numbers of adverse experiences in childhood are associated with worse outcomes. These outcomes overlap with many risk factors of homelessness. This study sought to address two questions: 1) Does a dose-response relationship exist between childhood adversity and chronic homelessness? 2) Does childhood adversity negatively predict the age at which homelessness first occurs? Suicide is a national public health concern, and college students may be at increased risk. Symptoms of psychopathology (i.e., stress, anxiety, and depression) may contribute to risk, whereas religiosity (i.e., religious attendance, surrendering to God, and feeling forgiven by God) may reduce risk. Students from a rural southeastern university (N=249) completed self-report measures. Serial mediation analyses indicate that attendance and surrender to God are inversely- predictive of suicide risk, both directly and through the indirect pathways of feeling forgiven by God (1st order mediator) and psychopathology (2nd order mediators). In all models, specific indirect effects occurred through feeling forgiven by …

In this thesis, we investigated T cell homeostasis and DNA damage repair machineries in HIV infection. We found that the frequencies of CD4T cells were low, which is associated with cell apoptosis in HIV patients compared to healthy subjects. Importantly, these events were closely correlated to the increase in T cell exhaustion, senescence, DNA damage, and telomere attrition. Mechanistically, while the DNA damage sensors Mer11, Rad50, and NBS1 (MRN) complexes remained intact, the ataxia-telangiectasia mutated (ATM) kinase and its downstream checkpoint kinase 2 (CHK2) were significantly inhibited during HIV infection. Adult women bear a disproportionate burden of overweight and obesity in Sub-Saharan Africa (SSA). Precise information to understand disease distribution and assess determinants is lacking. Therefore, this dissertation aimed to: (i) analyze the prevalence of adult female overweight and obesity combined in lower-level administrative units; (ii) analyze the effect modification of educational attainment and age on the association between household wealth and adult female overweight and obesity; (iii) synthesize qualitative research evidence to describe contextual factors contributing to female overweight and obesity at different life stages. Peptide identification using tandem mass spectrometry depends on matching the observed spectrum with the theoretical spectrum. The raw data from tandem mass spectrometry, however, is often not optimal because it may contain noise or measurement errors. Denoising this data can improve alignment between observed and theoretical spectra and reduce the number of peaks. The method used by Lewis et. al (2018) uses a combined constant and moving threshold to denoise spectra. Recent studies have shown elevated neuroinflammation in a large subset of individuals diagnosed with schizophrenia. A pro-inflammatory cytokine, tumor necrosis factor-alpha (TNFα), has been directly linked to this neuroinflammation. This study examined the effects of two TNFα modulators (PD2024 and PD340) produced by our collaborators at P2D Bioscience, Inc., to alleviate auditory sensorimotor gating deficits and reduce microglial cell activation present in the polyinosinic:polycytidylic (Poly I:C) rodent model of schizophrenia. Auditory sensorimotor gating was assessed using prepulse inhibition and microglial activation was examined and quantified using immunohistochemistry and confocal microscopy, respectively. Refugees are entering the United States in increasing numbers. Identifying factors that promote successful acculturation is an important task for those working to help refugees. As religiosity and social support have previously been linked to better mental health outcomes in refugees, they should be considered when examining acculturation. Using the Duke University Religious Index, the Multidimensional Scale of Perceived Social Support, and the Acculturation Attitudes Scale, this study examined the relations between religiosity, perceived social support, and acculturation strategies. In the wake of infidelity, romantic partners must decide how to move beyond the act of betrayal. Although infidelity can be concealed, sometimes others learn of the infidelity, which may impel a cheater to take steps to repair his or her image. This study examined the use of image repair on social media (Facebook), specifically the tactic of admitting responsibility, in the wake of infidelity. Understanding interactions between nanoparticles and biological systems is fundamental for the development of emerging nano-biotechnology applications. In this thesis, I present an investigation of zinc oxide (ZnO) nanoparticles interactions with biomolecules in two separate studies. The first section of my thesis covers tracking and detection of ZnO nanoparticles using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). ZinkicideTM is a bactericidal ZnO nanoparticle which has been developed for agriculture. The characterization of Zinkicide in biological media and in solution has been difficult due to its high dispersibility and ultra-small size. This research examined how corporate social advocacy (CSA) and corporate social responsibility (CSR) efforts impacted perceptions of authenticity. Using an experimental survey, participants were randomly exposed to Nike's actions related to the Black Lives Matter (BLM) movement via mock-online news articles. Participants completed a survey that contained Likert-type scale items regarding attitudes (perceived corporate intent, perceived authenticity, brand trust, and brand credibility) and behavioral intentions (word of mouth intentions (WOM), and purchase intention (PI)). Results indicated that positive attitudes significantly increased when Nike implemented an action step after taking a public stance on a controversial social-political issue. This research study examined teachers' understanding of risk factors and characteristics of potential perpetrators of school shootings, as well as the steps to take when they are identified in students. Participants completed a survey that contained Likert-type scale items regarding self efficacy and open-ended questions regarding school violence experiences. A mixed methods analysis revealed three primary conclusions. First, teachers have high levels of self-efficacy regarding the importance of and ability to deal effectively with troubled students. Anticancer drugs like gemcitabine (GEM) are used to treat cancers such as, pancreatic ductal adenocarcinoma (PDAC). However, the use of free gemcitabine yields challenges including cytotoxicity to healthy cells and poor circulation time. By encapsulating GEM in nanoparticles these challenges can be overcome. In this study poly(acrylic acid) (PAA)-GEM nanoparticles are fabricated by coupling GEM onto PAA. The particle formation is driven by the hydrophobic interaction of GEM, which collects in the core of the nanoparticle, forming a PAA shell. A unidimensional Rasch approach was used to explore whether the data collected through the National Survey of Adoptive Parents of 2007 (NSAP) for the well-being items represented a single latent construct and to establish a base model for comparison. A consecutive approach was then used as an exploratory tool to draw out potential multiple dimensions. Finally, multidimensional item response theory (MIRT) was used to confirm the results of the consecutive approach findings while comparing with the unidimensional baseline. Recent research has led to a much better understanding of the evolutionary processes that mold and structure variation within and among populations. How populations diverge at the genome-wide level and how polymorphism is maintained within a species, however, remains unclear. We address these questions with two freshwater color morphs, red and black, of the threespine stickleback fish (Gasterosteus aculeatus) from the northwest United States, in which a shift from red to black nuptial coloration occurred in several locations following glacial retreat. Deficits in Reelin expression and signaling play a pathogenic role in Alzheimer’s disease (AD). Thus, strategies aimed at correcting Reelin deficits may provide a novel therapeutic approach to treating AD. The cysteine-rich, whey protein supplement, Immunocal®, has recently been shown to rescue Reelin expression in a mouse model of Schizophrenia. Forensic practitioners have long sought efficient and reliable means for identifying those samples that are best suited for successful genetic profiling. Traditional serological screening methodologies rely upon enzyme activity and antibody-based serological tests. These tests can be consumptive, laborious and costly while reliance on antibody-based serological testing can be prone to error. Positive results resulting from non-target biological fluids, the potential for cross- reactivity and non-specific binding events yield merely presumptive results. Native Hawaiian people, and especially lesbian, gay, bisexual, transgender, māhū and/or queer (LGBTQM) Native Hawaiians, face health and mental health disparities that are disproportionate when compared with other racial/ethnic minorities in Hawai`i, and when compared to the United States as a whole. Native Hawaiians have the highest mortality rates for numerous biomedical diseases, including higher rates of substance abuse, diabetes, and even asthma, of any ethnic group in the state of Hawai`i (Andrade et al., 2006; Liu & Alameda, 2011). Atmospheric aerosols are ubiquitous throughout the Earth’s atmosphere and can be important with respect to environmental systems and human health. Pollen particles are a class of primary biological aerosol particles (PBAPs) that cost the United States billions of dollars a year in loss of productivity and healthcare costs due to allergy and respiratory effects. Traditional methods of pollen detection rely on collection and subsequent identification by visual microscopy, yet few measurement stations exist in the United States. In the South Bohemian region of the Czech Republic, the landscape is distinguished by a network of long narrow fields bordered by hedgerows clustered in small groups. These unique clusters of hedgerows have been interacting with their environment, effectively mitigating erosion, since they were first established in the High Middle Ages. In this research project I used historical maps to characterize land-use and land-cover (LULC) change relating to hedgerow features in one cadastral territory in the Blanice Watershed. Using georeferenced historical maps from 1837 and 1952, and unreferenced historical maps from 1837 to 1953, I compared the historical LULC to …
