Albuterol sulfate is a short-acting selective Beta 2 Adrenergic agonist. It is used to treat and prevent bronchospasm associated with asthma, bronchitis, and other breathing disorders. It is known to show extensive first pass metabolism and it requires frequent dosing which makes it difficult for patients to comply. To avoid these problems, transdermal route of delivery of Salbutamol was explored. Traditional methods of preparation of transdermal drug delivery systems involve the use of solvents and have multiple processing steps. Water scarcity and the emergence of new contaminants are making the world in need of renewable water resources. Slow sand filtration is looked at as an affordable and easy method to filter water with a few weaknesses such as clogging and the inability to remove complex matrix ingredients. To overcompensate these limited weaknesses, graphene-coated sand was studied. Ottawa, concrete, and masonry sand were used, using a reduction method to transform a coating of sugar into elemental carbon in N2 atmosphere at temperatures reaching 750°C. Moxifloxacin is available as a marketed solution (Vigamox(R) which has to be administered 3 or more times a day leading to poor patient compliance and low ocular bioavailability. The overall goal of this research was to use formulation strategies and develop patient compliant dosage forms of moxifloxacin which are sustained-release so as to improve its retention and thereby ocular bioavailability. The first strategy that was explored was to develop sustained release inserts of moxifloxacin hydrochloride using hot-melt extrusion. Current vehicular crashworthy systems and body armor designs are effective in protecting personnel and equipment from disaster due to their energy absorbing characteristics. However, little is understood about how to replicate naturally occurring designs through man-made techniques, and the role of functional grading to further improve the performance of such protective systems. Corn grain alternatives are often not used in finishing rations due to reduced caloric densities and reluctance to deviate from traditional methods. Along with optimum nutrition, cattle need to have proper implant strategies to reap maximum returns on investment. CrossFit is a demanding sport in which athletes perform constantly varied, functional movements at a high intensity, therefore requiring an adequate energy availability to avoid negative health and performance consequences. The purpose of this study was to assess risk of low energy availability (low EA) (phase 1) among recreational, female CrossFit athletes and measure and calculate energy availability using a 7-day dietary to measure energy intake (EI) and exercise energy expenditure (ExEE) (phase 2). In phase 1, using the LEAF-Q (Low Energy Availability in Females Questionnaire), 49% of survey respondents (n=149) were found to be at risk of low EA. …

Total knee arthroplasty (TKA) is a procedure where the arthritic surfaces of the knee is removed and replaced with a combination of metal and polymer implants that recreates the joint line to restore function and quality of life. Implant alignment is important in the success of a TKA. Modern day conventional instrumentation can be cumbersome in the operating room and can be inaccurate when resecting bone and aligning implants. The first review and study explore agility movements and the effect that alternative upper designs in shoes might have on ground reaction force measures of performance. The second review and study evaluate methods of predicting ground reaction forces without the use of a force platform. Cemented and cementless fixation in total knee arthroplasty (TKA) have been successfully used for decades. As the number of younger and more active patients treated with TKA continues to increase, long-term implant survivorship is of increasing importance. One of the most common complications and hence the reason for revision is mechanical loosening (23.1% of all revised TKA). The loosening mechanisms have been proposed for different fixation types. Dose-limiting toxicities of clinically used agents and development of resistance are two significant limitations in cancer chemotherapy. Microtubule targeting agents (MTAs) are a structurally diverse set of compounds that disrupt microtubule dynamics and exert their anticancer effect. Combination chemotherapy with antiangiogenic agents and microtubule targeting agents has shown an advantage against both these drawbacks. Single agents with dual antiangiogenic activity and cytotoxicity would afford a therapy that circumvents pharmacokinetic problems of multiple agents, avoid drug-drug interactions, lower the drug dose, decrease overlapping toxicities, and delay or prevent tumor cell resistance. Depression is a mental health disorder affecting greater than 350 million people worldwide with roughly 7% of the United States population diagnosed as of 2017. The selective serotonin reuptake inhibitors (SSRIs) have been the mainstay of pharmacotherapies for depression for the last 40 years. The SSRIs target the serotonin transporter (SERT), a monoamine transporter (MAT) responsible for terminating serotonergic neurotransmission. The SSRIs are not perfect therapeutics and suffer from delayed response times, inconsistent efficacy among patients, and often produce intolerable side effects. Tamoxifen is a first-generation selective estrogen receptor modulator, which reduces the risk of both invasive and non-invasive breast cancer (BC) as well as tumor recurrence in several clinical trials. However, chronic use of tamoxifen can increase uterine cancer risk and induce tamoxifen resistance. In past studies, melatonin alone reversed tamoxifen resistance induced by light exposure at night in rodents. This study demonstrates that melatonin or melatonin-based ligands (e.g., melatonin-tamoxifen drug conjugates) may be novel BC therapies that are efficacious and free of side effects. The ability to perform accurate, repeatable, and defensible elemental and molecular speciated analysis is immensely significant for measurements that support human health, environmental science, and industry. This is especially true since trivalent chromium [Cr(III)] is necessary for proper nutrition, while hexavalent chromium [Cr(VI)] is extremely toxic, genotoxic, and carcinogenic. The main challenges associated with speciated analysis are related to reactive species that are continuously transformed or converted to other species during sample processing. In addition, damage caused to structures such as bridges, pipelines, and boats, corrosion also has a profound effect on the biomedical community. Implanted metallic devices (i.e., vascular stents and artificial joints) are prone to pitting corrosion caused by aggressive ions present in extracellular fluid. Abandoned mine drainage (AMD) has resulted in worldwide devastation. Passive remediation systems (PRS) are a cost-effective option to treat AMD. While the geochemistry (abiotic) of these systems is well understood, little work has been done to characterize the microbial communities (biotic) that naturally form within these systems. The studies outlined in this dissertation encompass a broad focus, relating to the pathogenesis of neurological disorders such as Alzheimer’s disease, fragile X syndrome, and amyotrophic lateral sclerosis, as well as a novel infectious disease, COVID-19. However, all four studies detailed within this dissertation contain similar elements, namely, the coordinated role of RNA structure and function in essential molecular mechanisms. The Arizona toad (Anaxyrus microscaphus) and Jones’ waxy dogbane (Cycladenia humilis var. jonesii) are habitat specialists with historical ranges in the desert southwest and specifically, Zion National Park (ZION). The machine learning method, MaxEnt, constructed species distribution models (SDMs) in ZION for the two study species at 30 m and 900 m spatial resolutions using climate, topographic, and remotely sensed data. Additionally, 900 m forecasting models were constructed to observe the shifts in suitable habitat for the years 2050 and 2070, based off two representative concentration pathway scenarios. The objective of this study was to evaluate if holding time had an effect on mare whole blood progesterone levels. This research was completed using 12 mares, with blood being collected on multiple cycles to get a total of 28 separate blood collections. At each collection 5 vials of blood were taken for different time treatments of 0, 12, 24, and 48 hours. Cuban Treefrogs (Osteopilus septentrionalis) are established in Florida and Louisiana and have invasive potential further westward, possibly impacting native anurans at multiple life stages. In anurans, competition at the larval stage can decrease adult fitness through slower development and smaller size at metamorphosis, ultimately decreasing recruitment rates. Regional vegetation patterns of Texas beach plant communities were analyzed using cluster analysis, ANOSIM, SIMPER, NMDS, and ISA for fives zones representing the foredune complex of twenty Gulf Coast beaches. ANOVA revealed that zones differ in terms of percent bare sand, percent vegetative cover, and species richness. Cluster Analysis, ANOSIM, and SIMPER results indicate that Texas beaches can be divided into northern and southern regions based on differences in species composition of plant communities.
