Co-Flowering Community Effects On The Relative Contribution Of Pollen Quantity And Quality Limitation To The Reproductive Success Of Four Clarkia Species, Emma Moore Aug 2023

Co-Flowering Community Effects On The Relative Contribution Of Pollen Quantity And Quality Limitation To The Reproductive Success Of Four Clarkia Species, Emma Moore

Electronic Theses and Dissertations

More than 60% of plant populations experience low pollen availability which limits seed production (pollen limitation). Pollen limitation occurs due to low quantity or quality of pollen delivered to stigmas. Despite its ubiquity in nature, to date we still have limited understanding of the drivers of pollen limitation in plant communities. Plants typically occur in diverse communities where pollinator sharing is common and can influence quality and quantity aspects of pollen limitation. Co-flowering species can attract larger numbers of pollinators or they can compete for pollinators affecting pollen loads on stigmas. Pollinator competition may also lead to higher rates of …

Understanding The Role Of Sabp2-Interacting Protein (Sip) 428: An Nad+-Dependent Deacetylase Enzyme In Abiotic Stress Signaling Of Nicotiana Tabacum, Mariam Onabanjo

Electronic Theses and Dissertations

Abiotic stresses are constantly rising and pose a very high risk to global agricultural productivity and food security. Some plants have evolved several innate pathways for defense against these stresses. Hence, understanding stress signaling pathways can help develop crop plants with higher stress tolerance. The salicylic acid-mediated signaling pathway is important in plants experiencing biotic and abiotic stresses. In previous studies, SABP2-Interacting Protein (SIP-428) has been shown to be a negative regular of plant growth under abiotic stress. This study aimed to investigate the roles of SIP-428 in the ROS signaling of tobacco plants. We investigated transgenic RNAi-silenced lines of …

Gratitude And Sleep Quality In Primary Care: Mediating Effects Of Health Self-Efficacy, Health Behaviors, And Psychopathology, Heather R. Altier

Electronic Theses and Dissertations

Sleep disturbances are prevalent among primary care patients, and psychological dysfunction, including stress, anxiety, and depression, are robust contributors to poor sleep health. Yet, the presence of potential protective characteristics, such as health self-efficacy and engaging in adaptive health behaviors, may mitigate such outcomes. Gratitude (i.e., recognition and appreciation of experiences, relationships, and surroundings), a positive psychological cognitive-emotional characteristic, may serve as a catalyst of these beneficial downstream effects, given its association with improved health functioning and sleep. In a sample of primary care patients (N = 869) from 50 urban and 30 rural practices in Germany, health self-efficacy …

Use Of Transposon Screening For Salicylic Acid-Assisted Desiccation Killing In Salmonella, Shannon D. Elliott

Electronic Theses and Dissertations

Salmonella enterica serovar Typhimurium is one of the most prevalent food-borne pathogens, affecting millions around the world every year, making it a threat to global health. Salmonella possesses the ability to survive the normally lethal condition of desiccation, however, discovery of the genes and mechanisms behind this phenomenon are still ongoing. Using a transposon mutagenesis approach to construct a broad transposon library, this study aimed to uncover genes that may be contributing to changes in Salmonella’s survivability under desiccation, particularly when exposed to the antimicrobial molecule salicylic acid. Building on previous findings showing salicylic acid can alter cell viability …

The Old Family Clock: Exploring High Variation Of Chronotype In The Common House Spider, Parasteatoda Tepidariorum, Caitlin Jones Aug 2023

The Old Family Clock: Exploring High Variation Of Chronotype In The Common House Spider, Parasteatoda Tepidariorum, Caitlin Jones

Electronic Theses and Dissertations

Circadian rhythms are often tightly confined around a mean of 24 hours with little variation in traditional model organisms in the absence of environmental time cues. Some spider species fall well outside this normal range with mean periods of 18 hours and variation around these means of 6 hours or more. This extreme variation in circadian rhythm is not well-understood and is the focus of the current study. Using Parasteatoda tepidariorum as a model organism, we investigated whether variation in eight circadian parameters could be explained by genetic variability, environmental conditions, or an endogenous feature of the circadian system of …

Mapping The Binding Site Within Integrin D2 For Carboxyethylpyrrole (Cep)-Modified Proteins, Afia Prema

Electronic Theses and Dissertations

Neutrophils and macrophages accumulate at sites of inflammation and cause chronic inflammation leading to various diseases. Therefore, to better understand chronic disease pathways it is important to investigate the properties of macrophage accumulation in inflamed tissues. The I-domain of the macrophage receptor integrin aDb2 plays a vital role in macrophage retention by binding to CEP (carboxyethyl pyrrole), a ligand available at inflammatory sites. This thesis mainly focuses on evaluating the binding site within integrin aDb2 that binds carboxyethyl pyrrole (CEP)-modified proteins. So, a recombinant plasmid construct containing the integrin I-domain was developed. Seven non-conserved …

Electronic Theses and Dissertations

Urbanization is among the greatest threats to biodiversity on the planet, and is projected to increase in magnitude in coming years. This growing threat makes it important to better understand how urbanization may impact vital ecosystem services, such as pollination. Studies on the impact of urbanization on pollination vary wildly in their findings. This study analyzed multiple aspects of pollination success in common milkweed populations varying in urbanization level, including insect community composition, pollen removal/deposition, and fruit/seed production. By analyzing multiple pollination aspects we can determine the overall mechanism in which urbanization instills on the entire pollination process. Overall, urbanization …

Behavioral Indicators Of Reflective Functioning In Mother-Child Dyadic Interactions, Rachel Clingensmith Aug 2023

Behavioral Indicators Of Reflective Functioning In Mother-Child Dyadic Interactions, Rachel Clingensmith

Electronic Theses and Dissertations

Positive parenting practices and secure attachments are consistently linked to healthy child outcomes (Ainsworth & Bowlby, 1991; Waters et al., 2000). Research on cognitive processes that scaffold parental behaviors which contribute to secure attachment is an essential contribution to the literature, particularly given the potential for early intervention with at-risk families. Parental Reflective Functioning (PRF) is a construct of increasing interest which has been linked to secure attachments and positive child outcomes, with one commonly used self-report measure of PRF being the Parental Reflective Functioning Questionnaire (PRFQ; Camoirano, 2017; Clingensmith, 2021; Luyten et al., 2017). As such, the purpose of …

From the beginning of May 2022, cases of mpox have been recorded from non-endemic nations, and the illness has continued to be reported in other endemic nations. Majority of confirmed cases have been recorded in Europe and North America. In this thesis, we compare the spread of the outbreak across the top ten countries using a combination …

Investigation Of The Protein Components In A Periplasmic Mechanism Regulating Bacterial Morphology, Alexandra Pulliam Aug 2023

Investigation Of The Protein Components In A Periplasmic Mechanism Regulating Bacterial Morphology, Alexandra Pulliam

Electronic Theses and Dissertations

Salmonella is a leading bacterial cause of foodborne illness worldwide. During a previous study investigating the enzymes responsible for regulating cyclic-di-GMP concentrations, a mutant in the cyclic-di-GMP-specific phosphodiesterase STM3615 was identified that displayed a phenotype characterized by decreased survival on agar plates and a shorter bacterium length. I was able to determine that the periplasmic domain of STM3615 was responsible for this phenotype, not the enzymatic phosphodiesterase domain. Based upon a bioinformatic analysis of the protein, I then hypothesized that the periplasmic domain of STM3615 was interacting with a periplasmic protein to give rise to this phenotype. To identify this …

An Early Pliocene Fish Assemblage From The Southern Appalachians: Ichthyofauna Of The Gray Fossil Site, Shay Maden Aug 2023

An Early Pliocene Fish Assemblage From The Southern Appalachians: Ichthyofauna Of The Gray Fossil Site, Shay Maden

Electronic Theses and Dissertations

The Gray Fossil Site (GFS) preserves an early Pliocene sinkhole pond that accumulated a rich flora and vertebrate fauna. Fossils of fish are incredibly common at GFS, representing thousands of specimens recovered to date. Comparison with extant species reveals a depauperate freshwater fish fauna consisting of only two species: Lepomis sp. and Micropterus sp. The absence of fishes that are ubiquitous in modern streams suggests no perennial surface hydrologic connection between the Gray Fossil Site and the nearby Holston River. Limited opportunities for dispersal into the pond and deleterious conditions such as eutrophy and high competition may have resulted in …

Microsporidian Spores And The Integrin Binding Loop Of The Madam Protein Are Important For Integrin Signaling And Attachment To Host Cells, Cindy L. Barrett Aug 2023

Microsporidian Spores And The Integrin Binding Loop Of The Madam Protein Are Important For Integrin Signaling And Attachment To Host Cells, Cindy L. Barrett

Electronic Theses and Dissertations

Microsporidia are a distant fungal pathogen that have severe clinical consequences for the immunocompromised. Previous work identified a microsporidian pathogen protein termed Microsporidian ADAM or MADAM. This protein has close sequence homology to other ADAM proteins (A Disintegrin and Metalloproteinase) in two microsporidian species, Encephalitozoon intestinalis and E. cuniculi. ADAM proteins have a wide range of functions, including binding to integrins and host signaling. It is known that many pathogens manipulate integrins to invade host cells, and it is predicted that microsporidia are also exploiting this host target. Previous work with the MADAM protein demonstrated that this protein has a …

Effects Of Environmental Pollutants On Liver Disease: Role Of The Gut-Liver Axis., Ngozi Victoria Adiele Aug 2023

Effects Of Environmental Pollutants On Liver Disease: Role Of The Gut-Liver Axis., Ngozi Victoria Adiele

Electronic Theses and Dissertations

Polychlorinated biphenyls (PCBs) and polystyrene (PS) microplastics are environmental pollutants associated with various diseases including toxicant associated fatty liver disease (TAFLD). Previously, exposure to Aroclor 1260 (a PCB mixture) has been shown to exacerbate TAFLD, partly through Aroclor 1260 induced gut dysbiosis. Studies has also demonstrated that PS exposure alters gut microbiome composition and induced obesity. However, the effects of Aroclor 1260 on the gut and the effects of PS on the liver are still understudied. Therefore, we hypothesize that environmental pollutants will disrupt the gut-liver axis through changes in gut and its microbiome as well as changes in liver …

Interactive Computer-Based Simulations As Exploratory Learning Activities., Derek Mcclellan Aug 2023

Interactive Computer-Based Simulations As Exploratory Learning Activities., Derek Mcclellan

Electronic Theses and Dissertations

Typical college lectures follow a direct instruction framework, where instructors deliver a lecture, followed by an activity. Exploratory learning flips this routine by providing students with an activity prior to instruction. Research suggests that this inversion benefits students’ conceptual understanding and ability to transfer their knowledge. The majority of exploratory learning tasks in the literature are problem-solving activities. The current work investigates the use of computer-based simulations during exploratory learning, and whether manipulating the cognitive load of the activity impacts learning. In Experiment 1, undergraduate students (N=66) were randomly assigned to explore a simulation-based circuit construction activity prior …

Why Are Measures Of Depression Imperfectly Correlated? An Empirical Investigation., Eric Joseph Smith Aug 2023

Why Are Measures Of Depression Imperfectly Correlated? An Empirical Investigation., Eric Joseph Smith

Electronic Theses and Dissertations

Depression remains one of the most prevalent and impactful mental health disorders worldwide. A large number of measures of depressive symptoms are currently available, all designed to assess the same underlying construct. However, the measures tend not to correlate highly with one another. Measurement error cannot fully account for this observation, suggesting that clinical heterogeneity – that is, differences in symptom coverage across measures – might contribute to the lower-than-expected correlation between measures. We conducted two concurrent studies to examine this hypothesis. In Study 1, we developed a coding scheme to categorize items into one of 10 symptom categories. We …

We investigated the efficacy of these combinations in treating in vitro and in vivo tissue culture models …

Kdm5b As A Potential Epigenetic Regulator Of Cardiomyocyte Regeneration And Cell Cycle Activation., Dana Hammouri Aug 2023

Kdm5b As A Potential Epigenetic Regulator Of Cardiomyocyte Regeneration And Cell Cycle Activation., Dana Hammouri

Electronic Theses and Dissertations

The limited regenerative capacity of adult cardiomyocytes (CMs) is manifested in prevalent morbidity and mortality associated with ischemic heart disease. We previously demonstrated that a combination of four cell cycle factors (4F) promotes efficient cell division in differentiated CMs. The temporal transcriptional reprogramming in the 4F-transduced CMs indicates that epigenetic modifications mediate the cell cycle progression. Through small molecule screening, we evaluated the epigenetic impact on CM’s response in the 4F-transduced CMs. We identified the histone demethylase 5B (KDM5B), which through in vitro and ex vivo genetic knockdown and overexpression studies, exhibited a modulative effect on CM’s cell cycle plasticity …

Illuminating Underappreciated Mechanisms Of Receptor Regulation In Human Lymphocytes., Cassandra Woolley

Electronic Theses and Dissertations

A thorough understanding of receptor regulation is imperative to predict expression in varying contexts of disease or treatment. Lymphocyte surface receptors are often used as biomarkers and drug targets, making them particularly important for study. For receptors of debated functionality, such as the Fc receptor for IgM (FcMR), understanding regulation can also help to predict expression in vivo to supplement hypotheses of biological roles. Various mechanisms exist for altering receptor surface expression, including direct feedback mechanisms such as ligand-induced endocytosis and broader mechanisms such as transcriptional and translational control. In this dissertation, we explore selected underappreciated mechanisms of lymphocyte receptor …

Connectivity Of Amygdala Somatostatin-Expressing Neurons And Their Role In Taste-Guided Behavior., Jane Jeruto Bartonjo

Electronic Theses and Dissertations

The nucleus of solitary tract (NST) and parabrachial nucleus (PBN) represent the first and second central synapses of ascending gustatory information. Neural processing in these nuclei is influenced by descending input from forebrain regions such as the central nucleus of the amygdala (CeA). In mice, we have shown that somatostatin (Sst) expressing neurons of CeA that project to NST and PBN are largely distinct cell populations and optogenetic inhibition of the CeA/Sst-to-NST subpopulation increases the intake of high concentrations of quinine with no apparent effect on sucrose intake. Synaptic connectivity of these cells is needed to understand the possible mechanisms …
