U. maydis can utilize nitrate as a source of nitrogen from its environment, a process requiring a nitrate transporter to bring the nitrate inside the cell. This is followed by a two-step reduction of nitrate to ammonium. At first, nitrate reductase reduces nitrate into nitrite; in the second reaction, nitrite is reduced to ammonium by nitrite reductase. The goal of the study was to identify the nitrate transporter and nitrite reductase genes in U. maydis and characterize them based on their role in mating and virulence. Our goal with this study was to gain insight into the homeostatic immune cell network in healthy adipose tissue. Specifically, we sought to determine if the immature myeloid cells found in healthy and inflamed adipose were, in fact, tissue-resident myeloid-derived suppressor cells (MDSCs). Using various in vitro and in vivo methods, we have uncovered a population of CD11bHi Ly6CHi Ly6G- MDSCs resident in healthy adipose tissue. Porphyromonas gingivalis, a keystone pathogen for periodontal disease, initiates a complex molecular dialogue with gingival epithelial cells, leading ultimately to disruption of host signaling pathways. Dysbiotic host responses are also thought to contribute to the initiation and progression of oral squamous cell carcinomas. Recently, the homeostatic commensal Streptococcus gordonii has been shown to antagonize P. gingivalis-induced epithelial cell signaling events in host cells, such as proliferation and migration. Biomechanical and physiological parameters related to running performance are usually studied separately. However, evaluating both aspects together could be beneficial in improving athletic performance. The purpose of this study was to observe the change in peak vGRF and asymmetry as speed increases, while observing physiological responses during a O2maxtest. Data from athlete monitoring of 12 cross-country and triathlon athletes were analyzed. The athlete monitoring protocol included three unweighted countermovement jumps and a O2maxtest performed by the athletes. Exploring principles of REA in relation to the HS program model may reveal alignment that will inform high-quality developmentally appropriate practices that are meaningful and enhance learning outcomes. This multiple case study explores the REA in alignment within three Head Start programs in New Mexico, Massachusetts, and Tennessee. In this thesis I argue that the obligation to love based on the Christian tradition requires Christians to disperse their resources in a way that significantly evens out wealth distribution and increases social justice. Christians disagree on the terminology and some tenets of the goal of the Christian life (e.g., deification, beatific vision, communion with God, salvation). However, the requirement to practice love is common to all of these concepts, thus making love normative for Christians. Abstract Noncompliance is a patient’s inability to comply with the recommended treatment for their complete recovery from an ailment, while compliance is the extent to which a patient adheres to the provider’s directive, such as medication and orders given. The purpose of this study was to determine if motivational interviewing when applied to noncompliant pediatric patients from single-parent households, would improve global medical compliance in this population as compared to the standard of care at the end of three months. The objective of this study was to evaluate performance parameters and meat yield of commercial broiler chickens fed diets with different amino acid concentrations, with or without chromium supplementation, and delivered through two different feeding programs over 54 days. The experimental study was completed as a randomized-block design with 4,800, Ross 708 X Ross 708 commercial broiler chickens, picked at random, and evenly divided into six treatment groups (800 birds per group). Background: Approximately 70% of the adult population in the United States has overweight or obesity and increased risk for developing chronic diseases. Dietary self-monitoring adherence using a paper diary is associated with weight loss; however, few studies have examined this association with dietary self-monitoring apps. The selenium oxyanions selenate (Se(VI)) and selenite (Se(IV)) can be utilized by some bacteria and archaea as terminal electron acceptors in anaerobic respiration. Se(VI) and Se(IV) respiration is mediated by a phylogenetically and ecologically diverse array of organisms, suggesting that selenium respiration is ubiquitous in natural environments. Several respiratory Se(VI) reductases have been characterized in bacteria, revealing that Se(VI) respiration has evolved independently several times in this domain. Se(IV) respiration, in contrast, has yet to be characterized. I have purified and characterized the first respiratory Se(IV) reductase from Bacillus selenitireducens MLS10. Pediatric congenital heart disease (CHD) patients have unique risk factors associated with the pathophysiology of abnormal heart function. This vulnerable population is likely at an increased risk of acquiring a pressure injury during hospitalization. There are limited studies that include congenital heart disease patients and more specifically, factors unique to these patients. Surface air temperatures are predicted to increase in the near future, which will likely affect microbial activity and carbon flow in stream ecosystems. I performed an experiment in streamside channels at Coweeta Hydrological Laboratory, NC to assess responses of litter-associated microorganisms to moderate increases in water temperature (5 levels, ambient to +4°C). Post-extubation dysphagia (PED) has been studied in depth in a variety of populations including elective heart surgeries, trauma, critical illness, traumatic brain injury, and those requiring mechanical ventilation, but has not been researched in the overdose population (OD). Over two-thirds of the United States population have overweight or obese (OW/OB) weight statuses due in large part to an obesogenic environment that encourages unhealthful weight related behaviors. The obesogenic environment appears to place a larger burden on women with low incomes as they experience OW/OB disproportionately compared to other groups. The concentrations of metals (Ba, Cd, Co, Cr, Cu, Mn, Ni, Pb, Sr, V, Zn) were investigated in soil and water samples collected over four different sites in the southern part of Lake Nacogdoches over one year from August 2018 through June 2019. Water samples were analyzed using the USEPA method 200.8 part 11.2 for total recoverable metals. Soil samples were analyzed using the USEPA method 3050B for total recoverable metals. Background: Porphyromonas gingivalis adherence to Streptococcus gordonii may be important for P. gingivalis colonization in the oral cavity. Nanoparticles encapsulating synthetic peptide BAR (BAR-NPs) inhibit P. gingivalis adherence more potently than free BAR. However, BAR-NPs would exhibit low retention in an open flow environment. Hypothesis: Targeting BAR-NPs to the streptococcal surface using CafA protein will enhance their efficacy. Methods: CafA-modified NPs encapsulating BAR were synthesized using double emulsion approach. Surface binding and retention, and release kinetics of BAR from CafA-modified NPs was assessed. The objective of this thesis is to translate the framework of geographic imaginaries into an urban context in order to capture a narrative of how residents conceptualize and experience segregation. This framework is rooted in an investigation of local discourses as they exist within a specific social, political, and historical context. Institutionalized segregation and structural racism are the foundations on which the American urban context studied here was built upon. Delineation of the mechanisms underlying tobacco-induced or exacerbated periodontitis is hampered by the lack of an appropriate and reliable animal model.
