Cell-based therapies represent the latest biotechnological revolution in medicine, but significant limitations exist in non-viral intracellular delivery techniques during manufacturing of cell therapies. To address these limitations, a novel acoustofluidic device was developed to deliver ultrasound waves into a flow chamber for rapid molecular delivery to human cells. Our acoustofluidic device delivers biomolecules by inducing expansion and collapse of exogenous gas-filled microbubbles (“cavitation”), which enhances molecular delivery to nearby cells. Experimental studies were conducted to characterize key parameters that influence acoustofluidic-mediated molecular delivery to human cells. This thesis aims to address the role of the queer community in Louisville, Kentucky during the AIDS epidemic. Beginning with the first reported AIDS death in the city in 1983 throughout the 1980s, dialogue focused on those living with AIDS, specifically on education for prevention and aid to those afflicted by the disease. Individuals in the queer community—gay men, lesbians, bisexual men and women, transgender men and women, and others—created resources that were not being provided by the larger city government. Then, in the 1990s, national attention to the AIDS Memorial Quilt encouraged people to participate in rituals of commemoration, …

Given the diversity and complexity within immunoglobulin (IG) loci, effective mouse models first require characterization of intra-strain differences and construction of high-quality reference assemblies for IG loci in several representative strains. To understand light chain germline diversity across biomedically significant mouse strains, we profiled the expressed IGK and IGL repertoires of 18 commonly used laboratory mouse strains using AIRR-seq. Across strains, we observed germline IGKV sequences shared by three different IGK haplotypes and a more conserved IGLV germline repertoire among common laboratory strains. Bacterial vaginosis (BV) is a condition in which healthy lactobacilli are replaced by an overabundance of pathogenic bacteria in the female reproductive tract. Current antibiotic treatments often fail to “cure” infection, resulting in recurrence in more than 50% of women, 6 months post-treatment. Recently, probiotics have demonstrated promise to restore vaginal health; however, as with other active agents, delivery requires once-to-twice daily administration. Poor sanitation behaviors, including open defecation, present risks for diarrhea, which is credited with 600,000 deaths a year in children under age five. Although CLTS is a solution to ending open defecation, more than half of the investment in producing open defecation free (ODF) communities is lost during the maintenance phase. The purpose of this qualitative, case study was to understand stakeholder perceptions of the facilitators and hindrances to becoming ODF certified and maintaining ODF status and sanitary behaviors in a small village community located in Uganda, East Africa. The rates of TBI are significantly higher among individuals experiencing homelessness compared to the general population. Up to half of individuals experiencing homelessness may have a TBI. Accurate prevalence rates of TBI among individuals experiencing homelessness are difficult to obtain due to different methods of sampling participants and differing definitions of TBI; therefore, estimates may be underrepresented. Despite past research that has examined the relationship between TBI and homelessness, there are specific gaps in knowledge such as correlates and risk factors of TBI among individuals experiencing homelessness. Transgender and nonbinary (TNB) people are more likely to experience negative health outcomes than cisgender (i.e., not TNB) people, but less likely to access healthcare services due to the systemic forces of transphobia and cisgenderism. However, in recent years, this highly restrictive diet has gained mainstream popularity for a number of illness management, wellness, and weight loss uses. Atul Gawande, MD, affirms the inevitability of human mortality and the end of life in natural death. He decries current medical delivery’s incessant momentum toward medicalized death and recommends a meaningful conversation between doctor and patient to chart a course of care that supports what is important to the patient. But the tradition of a doctor’s dominance and a patient’s submissiveness disrupts the conversation. New definitions of both doctor and patient, and the intent to respect a patient’s autonomy, will facilitate decision-making and support patients’ values at the end of life, diminishing the momentum of medicalized death.

Influenza (influenza) is a contagious disease, causing thousands of deaths every year. The Advisory Committee on Immunization Practices (ACIP) recommends routine annual influenza vaccination for all people over 6 months of age who do not have contraindications, yet the vaccination coverage for 2018-19 season was 49.2%, around 20% less than the goal set by Healthy People 2030 of 70% coverage. The national foundation of infectious diseases survey showed that 51% of respondents do not think the flu vaccine works, 34% are concerned with the side effects from the vaccine, and 22% are concerned about getting flu from the vaccine. …

The purpose of this dissertation was to: 1) estimate willingness to use PrEP (WTUP) among men who have sex with men (MSM) using meta-analysis; 2) to assess patterns of minority stressors, HIV/AIDS acquisition stress and PrEP stigma among MSM and evaluate the association of such patterns with WTUP, using latent profile analysis; and 3) to elicit MSM’s preferences for a community pharmacy-based PrEP delivery program, using a discrete choice experiment.
First, a meta-analysis using a random-effects model revealed a moderate pooled prevalence proportion for WTUP of 0.58 (95% CI: 0.54-0.61) (or 58 out of 100) among PrEP naïve MSM estimated …

This work combines microbiology, molecular biology, Next-Generation Sequencing and system biology approaches to explore the mechanism of antibiotic persistence: a multi-drug tolerant, non-dividing, and metabolically altered state present in a subpopulation of cells due to phenotypic diversity rather than genetic variation (i.e. mutations). Persister can survive lethal antibiotic state and resuscitate after the treatment period is over. They are considered as the major contributing factor behind recurring infections. They also have a high mutation rate, which increases the chances of bacteria gaining antibiotic resistance. Pneumonia is a major factor affecting populations of free-ranging bighorn sheep (Ovis canadensis) across western North America. Pneumonia can occur in large-scale epizootics, during which greater than half of the population typically dies. After these epizootics, surviving ewes continue to conceive and bear lambs. However, lamb recruitment may remain low due to periodic or annual pneumonia outbreaks causing high lamb mortality rates, sometimes greater than 90%. Our study focused on the Rapid City, South Dakota bighorn sheep (BHS) herd that has recorded pneumonia-induced population decline since 2009. Griffithsin (GRFT) is a carbohydrate binding agent (lectin) that was originally identified in the red alga Griffithsia sp. Q-Griffithsin (Q-GRFT) is an oxidation stable analog of GRFT. GRFT has demonstrated inhibitory activity against HIV-1, Coronaviruses, Hepatitis C, influenza and Ebola viruses. The broad-spectrum activity suggests the potential utility of this lectin in a wide range of viral infections. However, the lectin’s activity in mucosal infections has not been extensively studied. This dissertation is a discursive analysis of the decision in the Dred Scott v Sandford, 1857 case written by Chief Justice Roger Taney. It begins with an overview of the literature on performative speech acts, focusing on the aspects of performatives that relate to Louis Miron and Jonathan Xavier Inda’s thesis that race is a performative speech act. Breaking from their use of race as the analytic, this analysis is situated within a black/nonblack paradigm. Depression/anxiety was defined by actively taking medication and/or diagnosis. The outcomes were ICU admission; ventilation; mortality; and time to mortality. Of 698 hospitalized patients, there were 204 (29%) defined to have either depression or anxiety. There were 109 deaths, and of those, 52 (48%) were diagnosed with depression/anxiety. BACKGROUND: Statins, a class of drugs that treat hyperlipidemia, may have an immunosuppressive effect for patients with community acquired pneumonia (CAP). Retrospective and in vitro studies have suggested an immunomodulatory, antioxidative and anticoagulant effects from statin use in patients with Type 2 Diabetes Mellitus (T2DM) hospitalized for CAP. Prospective studies that have tested any effect of statin therapy on patients with T2DM and CAP have not been found literature. To date, prospective studies showing of the effects statin therapy may have on T2DM patients hospitalized for CAP are not available.
