Autism Spectrum Disorder (ASD) is a lifelong neurodevelopmental disorder that consists of difficulties with social communication and language, as well as the presence of restricted and repetitive behaviors. These deficits tend to present in early childhood and usually lead to impairments in functioning across various settings. Moreover, these deficits have been shown to negatively impact adaptive behavior and functioning. Thus, early diagnosis and intervention is vital for future success within this population. Background: Porphyromonas gingivalis adherence to Streptococcus gordonii may be important for P. gingivalis colonization in the oral cavity. Nanoparticles encapsulating synthetic peptide BAR (BAR-NPs) inhibit P. gingivalis adherence more potently than free BAR. However, BAR-NPs would exhibit low retention in an open flow environment. Hypothesis: Targeting BAR-NPs to the streptococcal surface using CafA protein will enhance their efficacy. Methods: CafA-modified NPs encapsulating BAR were synthesized using double emulsion approach. Surface binding and retention, and release kinetics of BAR from CafA-modified NPs was assessed. The objective of this thesis is to translate the framework of geographic imaginaries into an urban context in order to capture a narrative of how residents conceptualize and experience segregation. This framework is rooted in an investigation of local discourses as they exist within a specific social, political, and historical context. Institutionalized segregation and structural racism are the foundations on which the American urban context studied here was built upon. This study employs multiple methods, including contextualizing the study area, analyzing discursive content, and visualizing the results. Previous research has focused on prison reentry programs and the impact the program has on reducing rates of recidivism, but there is a lack of research on jail reentry programs and the impact of programs’ individual services have on recidivism. Secondary data was obtained from the Louisville Metro Jail Familiar Faces Action and Community Transition (F2ACT) reentry program, which included basic demographic data of the participants, a record of individual services received by each participant, and the number of times each participant was booked into Louisville Metro Corrections before and after participating in F2ACT. Due to advances in technology, the number of users with hearing devices have increased. These users are often mainstreamed into classrooms with typical hearing peers. However, even with these devices, speech and language impairments may still persist. This study was conducted to analyze school-based speech-language pathologists’ (SLP) comfort, confidence, knowledge, and preparedness in treating students with hearing loss in Kentucky schools. 48 SLPs practicing in all levels of school including elementary, middle, and high, completed an anonymous online survey through the Qualtrics® platform. While a plethora of research studies exist regarding victims’ healing processes post-infidelity, almost no empirical data is available regarding perpetrators’ healing processes post-infidelity. In recent years, researchers have found that self-forgiveness after an interpersonal offense can aid individuals in healing intrapersonally and interpersonally. Self-forgiveness research even suggests a connection between genuine self-forgiveness and relationship satisfaction. The current study set out to explore the possible intrapersonal and interpersonal healing effects of self-forgiveness after participating in infidelity. Lung cancer patients experience high levels of distress, which commonly manifests as depressive symptoms. Importantly, depressive symptoms have demonstrated prognostic significance in cancer contexts, although the biological pathways by which depressive symptoms lead to poorer survival remain unclear and warrant greater attention. In addition to understanding the biological pathways by which depressive symptoms accelerate disease, identification of efficacious and effective psychological treatments for depressive symptoms are needed to improve both quality and quantity of life for cancer patients. The practice and research of mindfulness has increased exponentially in recent decades; however, the study of gratitude maintains modest growth. While the overlap between gratitude and mindfulness may be intuitive, empirical investigation is lacking. The present study sought to investigate the relative and combined influence of mindfulness and gratitude with symptoms of depression in undergraduates. The current study explored these constructs in the context of a mindfulness-based intervention known as Koru. One hundred undergraduate participants provided self-report measures of demographics, mindfulness, gratitude, and symptoms of depression prior to the intervention and again immediately following. Background: Tobacco smoking is the leading environmental risk factor for periodontal diseases. Delineation of the mechanisms underlying tobacco-induced or exacerbated periodontitis is hampered by the lack of an appropriate and reliable animal model. Hypothesis: We hypothesized that Porphyromonas-gingivalis-infected, cigarette smoke-exposed mice would represent reproducible models of acute (ligature model) and chronic (oral gavage model) tobacco-enhanced periodontitis that reflect multiple aspects of the disease noted in human smokers. Sensory Food Aversions occur frequently in children who are likely to appear on Speech-Language Pathologist’s (SLP’s) caseloads. The lack of research regarding intervention for Sensory Food Aversions in schools and the assertion of a gap in school-based services for children with feeding disorders was a significant indicator for the need of the study. The purpose of this study was to investigate the direct and indirect relationships between workplace discrimination and job satisfaction for lawyers. Using the After the JD – Wave 1 data, a national longitudinal survey of law school graduates with a sample size of 4,538, and following a general strain theory approach, structural equation models were conducted to analyze gender-differential effects of perceived discrimination on lawyer’s job satisfaction. The results indicated that men and women lawyers differ in their experience with workplace discrimination and their job satisfaction. The major premise of this dissertation was to transiently display novel immunological ligands on biological membranes as a localized means of modulating innate and adaptive immune responses with applications to bone marrow and pancreatic islet transplantation. In Chapter two, we engineered donor allogeneic bone marrow cells to transiently display a novel form of FasL, SA-FasL, to efficiently purge out alloreactive donor T cells to prevent acute GVHD. This dissertation is theoretically informed by General Strain Theory (GST) (Agnew 1992, 2002), and examines binge drinking within a military veteran population. It begins with an assessment on reform and focuses on shaping knowledge around the issue of binge drinking by military veterans. The current study uses Agnew’s GST to argue for an understanding of how strain and stress can affect individuals who have served in the military and acknowledges how this strain can lead to negative coping mechanisms such as binge drinking. The emergence of the Y en a marre movement in 2011 has reshaped the face of social activism in Francophone sub-Saharan Africa. In less than a decade of its existence, Y’en a marre has become iconic in Senegalese civil society and beyond. This dissertation explores the relationships among short-term financial confidence, academic achievement, six-year graduation success, and pre-college demographic characteristics for first-time, full-time, traditionally aged first-year students. The sample population was from a mid-sized Midwestern four-year degree granting university. These students were enrolled for the first time in Fall 2013 and participated in the survey used for data collection during that term. With single-cell RNA sequencing (scRNA-seq) technology, researchers are able to gain a better understanding of health and disease through the analysis of gene expression data at the cellular-level; however, scRNA-seq data tend to have high proportions of zero values, increased cell-to-cell variability, and overdispersion due to abnormally large expression counts, which create new statistical problems that need to be addressed. Ethanol-induced excessive apoptosis in neural crest cells (NCCs), a multipotent progenitor cell population, is one of the major mechanisms underlying the pathogenesis of Fetal Alcohol Spectrum Disorders (FASD). However, the molecular mechanisms underlying FASD that results from maternal alcohol exposure during pregnancy are poorly understood. Research to date has primarily focused on the amygdala, however, new perspectives suggest that a tiny basal forebrain region known as the bed nucleus of the stria terminalis (BNST) may hold key insights into understanding and treating anxiety disorders. Cigarette smoking is the single largest risk factor for cardiovascular disease (CVD) pathophysiology. Numerous researchers have shown potential associations between aldehydes in tobacco-derived aerosols from mainstream cigarette smoke (MCS) or electronic cigarettes (e-cigs) and their cardiotoxicity by damaging blood vessel endothelium. Idiopathic pulmonary fibrosis (IPF) is a fatal lung disease characterized by extracellular matrix deposition by fibroblasts. Aging and oxidative stress increase the susceptibility to IPF. Redox couples, cysteine/cystine (Cys/CySS) and glutathione/glutathione disulfide (GSH/GSSG), and their redox potentials (Eh) quantify oxidative stress. Fibroblasts from old mice maintain more oxidized extracellular Eh(Cys/CySS) than young mice. Microarray shows down-regulation of Slc7a11 potentially mediates this age-related oxidation. People are living longer and the aging population in the U.S. is showing significant growth, but it does mean they are necessarily living healthier lives. Public health representatives recommended people who want to live a healthy lifestyle as they get older need to regularly participate in sport and physical activity earlier in their life because it improves health and psychological and social well-being. However, approximately 80 percent of U.S. adults/older adults are insufficiently active; they participate for fewer than 150 minutes in moderate-intensity physical activity per week. Salmonella appears in the bloodstream of mice in as little as 15 minutes after oral inoculation and establishes persistent colonies in the spleen and liver. While its pathway to blood is undetermined, this phenomenon is dependent on the activity of Salmonella pathogenicity island 2 (SPI-2) coded type III secretion system (T3SS) and CD18+ phagocytes. We hypothesize that dendritic cells associated with the basal face of the gut epithelium, that are naturally migratory and known to sample for luminal antigens directly transport Salmonella to the bloodstream.
