In the DRM paradigm, experience of semantically related words leads to false memories of related words called critical lures. These false memories serve as evidence that the relationship between items in a DRM list contribute to an overall gist or central concept for the experience. The present studies aim to investigate whether the higher-order semantic relationship that exists between different DRM lists can also influence recognition memory. This dissertation consists of three research projects on cancer screening probability modeling. In these projects, the three key modeling parameters (sensitivity, sojourn time, transition density) for cancer screening were estimated, along with the long-term outcomes (including overdiagnosis as one outcome), the optimal screening time/age, the lead time distribution, and the probability of overdiagnosis at the future screening time were simulated to provide a statistical perspective on the effectiveness of cancer screening programs. Depression remains one of the most prevalent and impactful mental health disorders worldwide. A large number of measures of depressive symptoms are currently available, all designed to assess the same underlying construct. However, the measures tend not to correlate highly with one another. Measurement error cannot fully account for this observation, suggesting that clinical heterogeneity – that is, differences in symptom coverage across measures – might contribute to the lower-than-expected correlation between measures. Leadership in sport organizations continues to be male dominated, even as multiple scholars have argued and proven the importance of women holding sport leadership positions (Burton & Leberman, 2017; Darvin & Lubke, 2021; Schull, 2017). In the first project, we propose a parametric marginal structural ordinal logistic regression model (MS-OLRM) to assess treatment effects on ordinal outcomes. Average treatment effect (ATE) is used to measure the difference of the mean outcomes if all patients would have been treated compared with the outcomes if they would not have been treated. Suicide has proven to be difficult to predict and prevent at the individual level,1–3 so much so that a recent analysis of individual risk assessments found a maximal positive predictive value of 5.5% for suicide death,2 and suicide continues to be a leading cause of death among adolescents age 10-19 years.4 By targeting modifiable risk factors at the population level, policy has the potential to extend our reach to people and subgroups that are missed by individual level interventions. This dissertation presents a series of studies examining mechanisms of deglutition and respiration, and how these vital processes are impacted by opioids. The experiments in Chapter Two investigated the role of the upper esophagus in airway protection through systematic activation of pharyngeal and esophageal mechanoreceptors in a cat electromyography model. Chapter Three compared effects of opioid administration on breathing and swallowing between male and female rats, and found that females are more susceptible to opioid-induced depression of breathing and swallow than males. This dissertation is an examination of spinal cord injury induced neuroplasticity and tests whether noninvasive gene therapy can successfully target neurons in the lumbar spinal cord. It begins with an overview of neural control of locomotion and a brief summary of therapeutics that are used and/or in development for treating spinal anatomically characterize s subset of neurons in the spinal cord, long ascending propriospinal neurons, that are involved in interlimb coordination. Characterization of these neurons allows for subsequent evaluation of their potential involvement in injury induced neuroplasticity. This dissertation explored the relationship between green space usage and individuals’ physical, mental, and social well-being. This empirical research focused on Central Park in Old Louisville to examine how residents use the park and whether it contributes to the well-being of residents. The limited regenerative capacity of adult cardiomyocytes (CMs) is manifested in prevalent morbidity and mortality associated with ischemic heart disease. We previously demonstrated that a combination of four cell cycle factors (4F) promotes efficient cell division in differentiated CMs. The temporal transcriptional reprogramming in the 4F-transduced CMs indicates that epigenetic modifications mediate the cell cycle progression. Through small molecule screening, we evaluated the epigenetic impact on CM’s response in the 4F-transduced CMs. We identified the histone demethylase 5B (KDM5B), which through in vitro and ex vivo genetic knockdown and overexpression studies, exhibited a modulative effect on CM’s cell cycle plasticity …

Objective: This study examined differences between users and non-users of campus food pantry, conducted communication audit, and made recommendations for future food pantry communication. Methods: Cross-sectional twofold study, first analyzing existing intrapersonal, interpersonal, and environmental data to test predictors of campus food pantry usage characteristics, using inferential statistics. Secondly, conducting a communication audit that evaluates the reach of student communication. Results: Significant differences were found between users and non-users of food pantry that support hypotheses in relation to gender, age, race, class, marital status, housing type, housing description, and Pell eligibility. This dissertation is focused on autophagy in OL development and the OL translatome after SCI. The nucleus of solitary tract (NST) and parabrachial nucleus (PBN) represent the first and second central synapses of ascending gustatory information. Neural processing in these nuclei is influenced by descending input from forebrain regions such as the central nucleus of the amygdala (CeA). In mice, we have shown that somatostatin (Sst) expressing neurons of CeA that project to NST and PBN are largely distinct cell populations and optogenetic inhibition of the CeA/Sst-to-NST subpopulation increases the intake of high concentrations of quinine with no apparent effect on sucrose intake. The purpose of this phenomenological study is to examine the influence of power on racial identity and social awareness of minority high schools’ students in Bullitt County, Kentucky. The research will come from the student perspective of those attending high school with students predominantly from another racial background. Using purposeful sampling, 12 students participated in semi-structured interviews, providing insight into three research questions regarding racial identity, social power, and social awareness. Water is stable, benign, and green solvent. Its usage as a solvent in organic synthesis is significantly enhanced by micellar catalysis. However, replacing water as a reaction medium in organic synthesis is not sufficient to meet the sustainability challenges—excessive organic solvents are still required for product isolation and purification. Notably, solvents used in syntheses contribute to more than 80% of waste generation. Networks have the critical ability to represent the complex interconnectedness of social relationships, biological processes, and the spread of diseases and information. Exponential random graph models (ERGM) are one of the popular statistical methods for analyzing network data. ERGM, however, struggle with computational challenges and degeneracy issues, further exacerbated by their inability to handle high-dimensional network data. This study sought to explore the lived experiences of Dysphoric Milk Ejection Reflex (D-MER) during postpartum breastfeeding. A phenomenological method based on Giorgi’s phenomenological underpinnings was used. The setting for this study was virtual interviews in a secure online platform. The sample consisted of 13 participants, self-identifying as experiencing D-MER. Semi-structured interviews, with thematic analysis of transcription were conducted. The study employs qualitative content analysis to analyze select Nigerian music genres to examine how Nigeria is portrayed to international audiences and the economic benefits that can be derived from this form of branding. Sexual trauma is linked to adverse health outcomes (e.g., D’Andre et al., 2011). Given the negative sequelae of sexual trauma, exploration of factors that influence the relation between sexual trauma and health outcomes is crucial. Current literature suggests that psychological flexibility and inflexibility may be factors in promotion of individuals’ health or pathology (e.g., Gloster et al., 2017). Post-transcriptional RNA modifications including N6-methyladenosine (m6A) regulate mRNA stability, splicing, and translation. My research examined m6A in two disease models: breast cancer (BCa) and non-alcoholic fatty liver disease (NAFLD). Acquired resistance to endocrine therapies (ET) develops in approximately 20% of BCa patients with estrogen receptor α positive (ER+) tumors following treatment. The mechanisms by which tumor cells evade ET are not completely understood. Using a cell line model, we investigated the role of an m6A reader protein, HNRNPA2B1 (A2B1) that is upregulated in ET-resistant ER+ BCa cells. This dissertation makes significant contributions to the field of ophthalmology, addressing the segmentation of retinal layers and the diagnosis of diabetic retinopathy (DR). The first contribution is a novel 3D segmentation approach that leverages the patientspecific anatomy of retinal layers. This approach demonstrates superior accuracy in segmenting all retinal layers from a 3D retinal image compared to current state-of-the-art methods. It also offers enhanced speed, enabling potential clinical applications. The proposed segmentation approach holds great potential for supporting surgical planning and guidance in retinal procedures such as retinal detachment repair or macular hole closure.
