The objective of this research was to study the utilization of urea by mixed populations of rumen protozoa in vitro in the absence of bacteria. Initial experiments were conducted to determine the validity of chemical determinations used by other researchers in quantitating the nitrogen fractions in the medium following incubation. An experiment was conducted to determine the utilization of urea by rumen bacteria and this served as the basis for evaluating utilization of urea by protozoa. Culture techniques were developed to grow and maintain protozoa under in vitro conditions. Three experiments were conducted in which various nitrogen fractions of fermentation …

The purpose of this study was to isolate the symbiotic endophyte from S. argentea nodules in pure culture and to study the endophyte morphology in situ. In addition to permitting identification of the endophyte, it could be more closely studied for its role in and benefit from the symbiosis. The morphological study in situ would aid in selecting nodule isolates to test for nodule forming ability. The morphological study, therefore, would also aid in identifying the endophyte.

A condition known as “foal heat" scours often develops in young foals during the mare's first estrum, which usually commences at nine days post-partum. Although death due to this affliction is rare, the results such as loss or hair around the anus, unthriftiness of the foals, foul odor in the stable area, and unsight1iness or the foals causes more than minor concern among horsemen. Biever, working in this laboratory, isolated numerous diphtheroidal organisms from cattle and swine sources. These anaerobic diptheroids were morphologically similar to organisms in the genera Corynebacterium and Actinomyces. King and Meyer found that isolates of anaerobic diptheroids from human sources were more closely related to the Corynebacterium based upon metabolic and serologic differentiation. Their work consisted of separating A. bovis from several isolates of anaerobic diptheroids on the basis of the catalase test, litmus milk reactions, and the fermentation of xylose, salicin, and raffinose. A dicarbonyl compound, 2, 3-butanedione, commonly referred to as diacetyl or biacetyl, is considered by many to be-an important flavor constituent of butter and butter cultures. Milk fat has no natural diacetyl content and in the past the diacetyl found in butter resulted from bacterial fermentation of the cream before churning. In recent years commercial lactic cultures have been used, which are added to the fat granules before ''working" of the butter. Bacterial cultures used by the dairy industry generally consist of a mixture of two streptococcus types. A homofermentative type produces primarily lactic acid from the lactose of milk. …

Curd formation in a cultured dairy product is brought about by the addition of a bacterial culture capable of producing lactic acid from lactose. The primary function of this culture is to produce sufficient lactic acid to cause coagulation of the caseinate system of skimmilk. A secondary function of the culture is the production of flavor compounds considered to be desirable in these products. The flavor compounds produced by the normal growth of citric acid fermenting organisms are water soluble, and are largely removed during washing of the Cottage cheese curd. One of the interesting symbiotic relationships in nature is that existing between the ruminants and their microbial population. This arrangement has solved for them the problem of existing on an herbivorous diet. Most mammals can derive little benefit from the potentially available energy in plant materials, which is locked in the complex, insoluble polysaccharide, cellulose. Cellulase is not a mammalian enzyme, but it is part of the chemical machinery of a number of bacteria, some species of which reside in the rumen. The use of commercial fertilizers has become an accepted practice in American agriculture. Among nitrogenous fertilizers, urea possesses characteristics which make it suitable for use in mixed gods and desirable for direct application to soils requiring only nitrogen. Urea is a concentrated fertilizer usually containing about 45% nitrogen. This high proportion of nitrogen is higher than for any other commercial solid nitrogenous fertilizer material. Often urea is cheaper than other forms of nitrogen per pound of plant food when transportation is an appreciable part of the total fertilizer cost.
