This dissertation describes the development of a biotherapeutic agent, EPICERTIN, that fulfills an unmet treatment need for ulcerative colitis (UC): mucosal healing. EPICERTIN is a recombinant cholera toxin B subunit (CTB) variant with a KDEL endoplasmic reticulum (ER) retention motif that is crucial for its wound healing capabilities. Previous studies have demonstrated that EPICERTIN’s mucosal healing activity is due, in part, to its prolonged residence in the ER which initiates an unfolded protein response (UPR) and subsequent TGF-β signaling and barrier repair in colonic epithelial cells. Child welfare organizations throughout the nation grapple with turnover rates estimated to be as high as 65% (e.g., CWLA, 2016; Edwards & Wideman, 2018). Turnover contributes negatively to client outcomes, organizational financial shortages, and to perpetuating stressful conditions for the child welfare workforce. One factor found to contribute to turnover within child welfare is that of secondary traumatic stress (STS). Child welfare workers are known to experience higher rates of STS than any other helping professional (Brady, 2017). Despite advancements in medicine and health sciences, health disparities and declining life expectancy persists among Black women in the United States. For over five decades, there has been substantial research into what in society, or our communities, makes us fearful. Film scores typically feature large instrumental ensembles, sometimes with choir added at pivotal moments. Rarely, however, is the choir entrusted with a greater percentage of the score, and far too scores explore the sonic potential that vocal music has to offer. In fact, in a recent study of 800 films, none contained a score that was completely c. Thus, the body of unaccompanied choral film scores is practically non-existent. Urbanization near estuaries has been shown to affect the growth and survival of juvenile sharks using the system as a nursery. North Inlet and Murrells Inlet, South Carolina, are similarly-sized, tidally-dominated, bar-built estuaries with extensive Sporobolus-lined tidal creeks but differ in degree of human impact. Environmental DNA (eDNA) analysis is an emerging, non-invasive community monitoring tool. This study aimed to determine if eDNA methods can be reliably used in a large brackish, partially mixed estuary by developing and testing three novel eDNA primers, for Sandbar Sharks (Carcharhinus plumbeus), Blacknose Sharks (Carcharhinus acronotus), and Bonnetheads (Sphyrna tiburo). These primers were designed to target 109, 156, and 120 base pair (bp) fragments, respectively, of the highly conserved NAD2 gene in the mitochondrial genome of each species. Presidio Los Adaes (16NA16), located in present day northwest Louisiana, served as the capital of Spanish Texas for much of the 18th century. Maps and historical documents provide architectural plans for the fort and other buildings on the site but differ in the size and location of several buildings. In 2009, a geophysical survey of the site with ground penetrating radar, magnetometry, electrical resistance, and electromagnetic induction showed good preservation of architectural foundations. Visual assessment of the maps and geophysical data, information from archaeological excavations, and spatial statistics suggests that the fort was originally built as specified by the architect, …

Loggerhead shrikes, Lanius ludovicianus, are a declining grassland songbird native to North America. Land-use change has been associated with the decline of grassland bird species. However, the process of urbanization creates novel short-grass habitats reminiscent of shrikes’ natural habitats. Loggerhead shrikes have been observed living in developed areas, particularly in Southeastern coastal states. Understanding the long-term impacts of urban-dwelling on loggerhead shrike populations is vital to identifying whether urban areas offer refuge or act as an ecological sink for shrike populations.

As bycatch continues to impact global shark populations, there is a continuing need for effective bycatch reduction devices. Prior research has shown promise in exploiting sharks' electrosensory ability to this end. We tested the deterrent efficacy of the Select Magnetic and Repellant Treated (SMARTTM) and the newly developed "SMARTER" hooks in experimental longline and hook-and-line trials. Both are magnetized and contain an electropositive metal component made of magnesium (SMART) and a magnesium alloy designed to extend longevity (SMARTER). Subcortical structures of the visual system have been the subject of intense study in recent years, but there remain some important unanswered questions regarding the synaptic relationships linking the nuclei that comprise this important sensory network within the brain. In these studies, we use several modern and traditional approaches, including viral tract tracing, in vitro slice physiology, immunohistochemistry, optogenetics, and electron microscopy to characterize the circuits linking the superior colliculus (SC), parabigeminal nucleus (PBG), and lateral geniculate nucleus (LGN), with particular focus on GABAergic and cholinergic cell types. The continual replacement of taste cells creates interesting questions regarding how the innervating neurons are impacted during this process. Here we ask how innervation within taste buds is affected when taste cell entry is inhibited and reestablished. Inhibition of sonic hedgehog signaling (Shh) is thought to inhibit taste cell turnover. Consistently, fewer new cells were added to individual taste buds after treatment with a Shh-inhibitor compared to vehicle treatment, and taste bud volume decreased after 16 days of treatment. We next examined how taste nerve fiber extension into the gustatory epithelium is affected by preventing taste cell turnover. Ten days …

Glioblastoma (GBM) brain tumors are highly aggressive gliomas due to genetic and cellular heterogeneity. Current GBM treatment consists of surgical resection of the tumor combined with radio- or chemo-therapies. While these treatments have increased the life expectancy for GBM patients up to 20 months, they have had little effect on the 5-year survival rate. The complex cellular and genetic composition of the tumor makes current treatments less effective long term. Organizations have used beekeeping to address poverty and other development goals with mixed outcomes. Beekeeping presents a viable option to promote AIG. The Central Appalachian region of the United States disproportionately experiences higher rates of cardiovascular diseases (CVD) and associated risk factors. Primary risk factors for CVD include hypertension, hypercholesterolemia, and tobacco use, all of which have the potential to be mitigated through lifestyle behavior changes. Dietary and physical activity practices are the two main driving forces for the development of hypertension and hypercholesterolemia, while tobacco use is a behavioral choice on its own. The hyoid apparatus in laryngeally echolocating bats forms a mechanical connection between the larynx and auditory bullae and has been hypothesized to transfer the outgoing echolocation call to the middle ear during echolocation call emission. We used µCT data to build models of the hyoid apparatus and middle ear from six species of bats and used finite element modeling (FEM) to measure the vibroacoustic response of the tympanic membrane due to hyoid-borne sound generated during echolocation. We found that hyoid-borne sound in all six species stimulated the eardrum within a range likely heard by bats. Although there were minor differences …

Here, a morphological description of the extinct meline badger Arctomeles dimolodontus from the Gray Fossil Site (GFS), Tennessee is provided with comparison to extant Meles meles. Originally described solely on upper teeth of a single individual, the recovery of several crania and jaws provides a sample of at least five individuals, affording the largest known fossil sample in North America of the enigmatic Melinae. Examination of A. dimolodontus skull material reveals extreme intraspecific variation in previously identified diagnostic characters, highlighting the need for caution when identifying fossil taxa based on similarly fragmentary remains. Tick populations have been immigrating into northeast Tennessee from east Tennessee, Kentucky, Virginia, and North Carolina. Counties in states bordering northeast Tennessee harbor tick species associated with human illness. Human diseases transmitted by ticks include ehrlichiosis, spotted fever rickettsial group diseases, tularemia, anaplasmosis, babesiosis, Lyme disease, alpha-gal syndrome, Heartland virus, Powassan virus, and southern tick-associated rash illness (STARI). These diseases cause morbidity and mortality in human populations and may pose a high risk to individuals, wildlife, and livestock. The Cherokee National Forest covering the east Tennessee border provides a permissible environment for ticks to immigrate and thrive. Residents of northeast …

Cannabis is widely consumed by humans for pharmacological effects that are mediated by THC, though there is little evidence that THC is a primary reinforcer in non-human animal models. We hypothesized that THC may have potent reinforcement enhancing effects, comparable to other drugs (e.g., nicotine and caffeine) which are also widely consumed by humans, but difficult to establish as primary reinforcers in non-humans. Ralph Waldo Emerson’s bee poem, “The Humble-Bee,” expresses the nineteenth-century transcendentalist philosophy of finding wholeness and oneness in nature while Sylvia Plath’s twentieth-century bee poems function as a response to Plath’s feelings of alienation and repression, indicating that transcendental peace is lost in the postmodern era. Emerson’s poem indicates the spiritual fulfilment found through observing bees and highlights the harmony between humans and nature, but women of the nineteenth century find difficulty achieving this same level of freedom; Emily Dickinson reclaims the language of transcendentalism in her bee poetry to explore a world otherwise denied to her. Since the early days of the discovery of the Gray Fossil Site (GFS), meticulous efforts to preserve the spatial data were a priority. Direct surveying of fossils prior to recovery, as well as grid mapping the site, provided relative spatial data within a square meter. Such efforts meant that even fragments and microfossils, recovered during the screening operations and eventual concentrate picking, maintained some spatial data. Available spatial data are used to identify smaller deposits within the greater system, as well as non-random distribution patterns among a number of GFS taxa. A rising problem in agriculture is the increase of antibiotic-resistant Salmonella cases associated with chicken eggs, which transmit infection to humans. To counter this, new approaches to combat Salmonella in chickens and desiccated on eggshells are vital in the prevention of human foodborne illness. Disrupting signaling pathways with naturally occurring compounds provides a potential novel avenue for prevention of Salmonella infections, as this would disrupt sensing of these environments and inhibit subsequent optimal gene expression. Food is a universal human necessity, yet food often serves more than a biological purpose as it informs individual and communal identities, and even facilitates memory. This thesis explores personal memory, the development of identity, and an almost reverential connection to nature in several food poems by Li-Young Lee in Rose (1986) and Behind My Eyes (2008). Born in 1957, Lee has been writing poetry since he was young, studying under Gerald Stern in the late 1970s, and he is known for writing sublime, transcendent yet incredibly accessible and expressive poetry. Mankind Is Machine: A Monstrous Posthuman Reading Of Philip K. Dick’S Selected Works, Gabriel Davis

The works of Philip K. Dick act as an ideal template for readers to explore what it means to be human in a technologically dominated world. Dick’s emphasis on the usage of androids and artificial intelligence as literary monsters allows for a posthuman reading of the traditional literary monster, notably in how their uncanny nature and behavior helps reveal the synthetic tendencies of humanity. In Do Androids Dream of Electric Sheep?, “Imposter,” and “I Hope I Shall Arrive Soon,” each narrative incorporates artificial intelligence and androids acting as others to reveal the machine-like qualities of Dick’s human characters.
