The thesis of this dissertation is that the Descent/Ascent Leitmotif, which includes the language of not only descending and ascending, but also going, coming, and being sent, performs a significant literary and christological function in the Gospel of John. The Evangelist's Descent/Ascent Leitmotif becomes the Gospel's organizing principal, drawing together a constellation of verbs and a number of themes, including Jesus-Son's origin, identity, relationship to God, authority, signs and works, life, and glory.

This thesis examines the connection between town planning and social organization at the small town site of Port Tobacco in south-central Charles County, Maryland from the beginning of the 18th century through to the end of the 19th century. By employing a methodology of both geophysical techniques and archaeological excavations, I was able to locate and map numerous structures and features associated with town planning and examine how these spaces were used. This data was used to show how social order, power, and wealth transformed the town layout from a linear settlement along the river into a grid-like pattern. Specifically, …

Several studies have identified Rho family GTPases (i.e. Rho, Rac, Cdc42) as mediators of diverse critical cellular processes, such as actin cytoskeleton remodeling, gene transcription, cell-cell adhesion, and cell cycle progression. However, more recent data highlight an essential role for Rho GTPases as regulators of neuronal morphology and neuronal survival. In particular, Rac GTPase generally induces neurite outgrowth and promotes neuronal survival while Rho GTPase typically provokes neurite retraction and induces neuronal apoptosis. Animal-assisted interventions (AAIs) are a treatment modality that incorporates a trained animal into a person's healing and learning process in order to benefit the person physically, emotionally and/or socially (Delta Society, 1996). Welcome Home Ranch rockshelter is located in the Palmer Divide region of Colorado. The archaeological evidence on site suggests that this rockshelter was occupied between the Late Archaic period (c. 1000 BC – AD 150) and Early Ceramic period (c. AD 150- 1150). Excavations at Welcome Home Ranch provided a dataset of lithics, ceramics, and features with which to test ideas about prehistoric life during this transitional time frame between the Archaic stage and the Late Prehistoric stage. The historical novel, Cortes Cenquizqui, set in sixteenth century Mexico and Spain, follows the conflicted lives and minds of several characters through an age of freshly crossing culture, language, and power. The narrator, Maria de Quesada of high ranking Spanish and Mexica parents, resents the white world for condemning her work as a female healer or curandera. Yet she acknowledges that she is ill-equipped to leave Mexico City to live in the outlying Indigenous villages. Rural private well-water quality and quantity is a global concern. It is currently a significant concern in Pennsylvania because there are no uniform statewide regulations for well construction. The aim of this study was to determine the prevalence of E. coli and coliforms in private well-water from a small community in Butler County, Pennsylvania. E. coli and coliforms were detected in water samples using EPA standard methods 9222 G. and 9222 B. Moving chronologically from the New Negro Renaissance into the contemporary era, my dissertation examines poetic representations of five blues artists: Gertrude "Ma" Rainey, Bessie Smith, Billie Holiday, Huddie Ledbetter (Leadbelly), and Robert Johnson. Despite extensive scholarship on the blues and these icons, blues tribute poems have remained un(der)studied. Membrane phospholipid synthesis and turnover is a continual process during normal cell growth. The turnover of the glycerophospholipids by B-type phospholipases (PLBs) in Saccharomyces cerevisiae results in the formation glycerophosphodiesters through a deacylation reaction. Here, I address several aspects of the glycerophospholipid deacylation, transport, and reutilization pathway in S. cerevisiae. First, I show the RAS GTPase-activating proteins, Ira1 and Ira2, are required for utilization of the glycerophosphodiester - glycerophosphoinositol (GroPIns) - as a phosphate source. Second, I demonstrate loss of the cell surface associated PLBs, Plb1-3, and/or utilization of GroPIns causes actin cytoskeleton defects and an increased cell size. For …

Symbiodinium photosynthesis fuels the growth of an array of marine invertebrates, including scleractinian corals and octocorals. Studies examining the symbiosis between Caribbean octocorals and Symbiodinium are sparse, even though octocorals blanket the landscape of Caribbean coral reefs. Here, I compared the photosynthetic characteristics of Symbiodinium in four comCaribbean octocoral species (Pseudoplexaura porosa, Pseudoplexaura wagenaari, Eunicea tourneforti, and Pterogorgia anceps) in outdoor aquaria at ambient temperature. The four octocoral species exhibited similar photochemical efficiencies despite significant differences in Symbiodinium density, chlorophyll a per cell, light absorption by chlorophyll a, and rates of photosynthetic oxygen evolution. Approximately 60% of Major Depressive Disorder (MDD) patients do not respond to FDA-approved antidepressants. Introducing effective pharmacotherapies for this treatment-resistant population is hindered by the lack of pre-clinical screening assays that accurately model the clinical features of TRD. The purpose of this research was to screen representatives of different classes FDA-approved antidepressants and one novel antidepressant in two genetic lines of domestic fowl chicks that have previously been identified as stress-vulnerable and stress-resilient in the chick anxiety depression model. Migration is a widespread and complex phenomenon in nature that has fascinated humans for centuries. Connectivity among populations influences their demographics, genetic structure and response to environmental change. Here, I used the loggerhead turtle (Caretta caretta, L.) as a study organism to address questions related to migratory connectivity and carry-over effects using satellite telemetry, stable isotope analysis and GIS interpolation methods. Telemetry identified foraging areas previously overlooked for loggerheads nesting in Florida. In the state of Florida, Old World climbing fern (Lygodium microphyllum) is an invasive plant listed by FLEPPC as a category one invader with significant ecological impacts that threaten native plant diversity. Purpose: To examine the effects of 28 days of β-hydroxy-β-methylbutyrate free acid (HMB) and high-intensity interval training (HIIT) on maximal oxygen consumption (VO2peak), ventilatory threshold (VT), respiratory compensation point (RCP) and time to exhaustion (Tmax) in collegeaged men and women. Methods: Healthy men and women (n=34, age and VO2peak= 22.7+3.1yr and 39.3+5.0 mL. kg-1.min-1, respectively) participated in this study. All participants completed a series of tests prior and subsequent to treatment. A maximal oxygen consumption test was performed on a cycle ergometer to assess VO2peak, Tmax, VT, and RCP. Amyotrophic Lateral Sclerosis (ALS) is an always lethal motor neuron disease with unknown pathogenesis. Inhibitors of the molecular chaperone heat shock protein 90 (Hsp90) have limited neuroprotection in some models of motor neuron degeneration. However the direct effect of Hsp90 inhibition on motor neurons is unknown. Here we show that Hsp90 inhibition induced motor neuron death through activation of the P2X7 receptor. Motor neuron death required phosphatase and tensein homolog (PTEN)-mediated inhibition of the PI3K/AKT pathway leading to Fas receptor activation and caspase dependent death. Excess fertilizer can be lost either via surface runoff or by leaching through the soil mass eventually reaching water bodies and leading to eutrophication. The present study compared the effectiveness of different mentoring programs at reducing feelings of stereotype threat experienced by women in science, technology, engineering, and math (STEM) fields. Stereotype threat refers to the extra pressure a person feels to disprove a negative stereotype that applies to him or her. This dissertation studies the question of how to conceive of revolution without necessarily drawing any concepts from the state. When concepts, figures, or objects drawn from the state are assumed to exist following a revolution, the ability of revolution to radically change the state is constrained. This problem limits earlier theories of revolution, such as social contract theory and Marxism, both of which present certain parts of the state as unchangeable. In the past half-century, two raptors with similar life histories, bald eagles (Haliaeetus leucocephalus) and golden eagles (Aquila chrysaetos canadensis) were the subjects of either broad-scale reintroduction or translocation projects in North America. These two different conservation approaches provided the framework for a retrospective natural experiment. The goal of my research was to determine the effects of these two conservation management approaches on the genetic population structure of each species. In 1994, Cynthia Selfe and Richard Selfe claimed that interfaces are maps of cultural information and are therefore ideological (485). For Selfe and Selfe and other scholars, these interfaces carried a colonial ideology that resulted in Western dominance over other cultures. Since this early scholarship, our perspectives on interface have shifted with changing technology; interfaces can no longer be treated as having persistent and predictable characteristics like texts.
