Myeloid-derived suppressor cells (MDSC) are a heterogeneous population of immature myeloid cells that promote tumor progression. In this study, we investigated the effect of dectin-1 stimulation by yeast-derived particulate β-glucan in MDSC function and differentiation in cancer. In vivo treatment of mice bearing lewis lung carcinoma and mammary cell carcinoma with particulate β-glucan decreased tumor weight and splenomegaly, and reduced the accumulation of polymorphonuclear-MDSC (PMN-MDSC) but not monocytic-MDSC (M-MDSC) in the spleen and tumor. In addition, particulate β-glucan differentially modulated the function of different MDSC subsets; it enhanced PMN-MDSC respiratory burst and apoptosis, and induced the differentiation of M-MDSC into …

Explanatory models significantly enhance the understanding of etiological influences that place children at risk for anxiety, yet little is known about processes that promote resilient outcomes in children. While contextual factors influence risk and protective processes, cultural constructs have not been incorporated into existing models of anxiety and the role of culture remains obscure. Data reported locally and nationally show that students struggle to understand fractional concepts. The purpose of this study was to examine the effects of an interactive tablet application on student achievement in fractions. Students in grades three through five at the treatment school worked with an interactive tablet application designed to facilitate student construction of fractions knowledge. The interactive tablet application is an open-platform that allowed teachers the autonomy to work with the researcher to load their desired content activities on the topic of fractions to develop instructional sessions for students. Chronic alcohol consumption is a leading cause of liver disease and liver-related death worldwide. Alcoholic liver disease includes, hepatic steatosis, steatohepatitis and ultimately fibrosis and cirrhosis. Emerging evidence has established the important role of the “gut-liver” axis in the development of alcoholic liver disease (ALD). This dissertation integrates classical rhetoric with postmodern understandings of textual fragmentation. “Places in the Polity of Rhetoric” follows two mutually constitutive avenues of inquiry, one of which stresses the importance of understanding textual fragments as rhetorical topoi—that is, as generative “places” that allow writers and speakers to economically evoke larger fields of cultural meaning in the space of a single word, phrase, or image. This dissertation takes a comprehensive approach to the role of dietary carotenoids on redness and the subsequent behaviors in the firemouth cichlid, Thorichthys meeki. I start with a brief introduction into signaling, the importance of carotenoids, and mate choice. The dissertation is then divided into three data chapters which are designed to stand as independent manuscripts. This study made use of the Silene latifolia/Microbotryum lychnidis-dioicae phytopathogen system as the focal system to establish the first reference genome for Microbotryum violaceum sensu lato. In silico analysis was performed on the genome assembly to identify various characteristics of the genome. Using RNA-Sequencing technologies on the Illumina platform, we collected transcriptomic data for both in vitro and in planta life stages of the fungus, providing the most comprehensive look at the gene expression and regulation of this fungus. The development of sight words is essential for individuals with significant disability. There is a plethora of research highlighting the acquisition of sight words for this population but to date, most focus on teacher led interventions (Browder, Algrim-Delzell, Spooner, Mims, & Baker, 2009). This study investigated the use of computer technology vs. one-on-one instruction targeting sight words acquisition for a student with a significant disability. Results showed the participant indicated improved performance using the computer based intervention versus the constant time delay instructor led intervention.

In order for a pathogen to successfully colonize the host, it must be able to acquire essential nutrients and regulate gene expression to respond to environmental fluctuations. One mechanism bacteria have evolved in order to detect these fluctuations and respond is the two component signal transduction systems (TCS). Aggregatibacter actinomycetemcomitans, a dental commensal associated with localized aggressive periodontitis, contains a TCS designated QseBC that was previously shown to regulate expression of several iron acquisition genes, biofilm formation and virulence, but the activating signal was unknown []. The purpose of this study was to investigate the perception of acceptable use of social media among teachers, students, and parents. In doing so the researcher touched on generational factors, communication, instructional practice with social networking and the perceptions of what was acceptable among these 3 groups within a middle school setting.

This study was performed to test the in vitro effects of a Lactobacillus rhamnosus isolate from “amabere amaruranu”, a traditional Kenyan cultured milk, on 3T3-L1 and Caco-2 cell lines. Cultures of fully mature 3T3-L1 adipocytes were treated with bacterial isolate cell extract (CE), filtered spent broth (FSB) from overnight bacterial culture, or with a PBS control. Expression levels of PPAR³1 and 2, C/EBP±, and ATGL proteins in 3T3-L1 cells were upregulated by FSB treatment. CE treatment did not affect protein expression levels. Expression of MTTP and SREBP-1c proteins in Caco-2 cells showed no change with either treatment. Optical density measurements …

Despite his own conservative values, D.H. Lawrence writes sexually liberated female characters. The most subversive female characters in Lawrence’s oeuvre are the Brangwens of The Rainbow. The Brangwens are prototypical models of a form of femininity that connects women to Nature while distancing them from society; his women are cast as monsters, but are strengthened from their link with Nature. They represent what I am calling the Lawrentian-Woman.

Among the many measureable changes induced by anxiety are changes in facial electromyography (EMG), heart rate (HR), and sweat gland activity (EDG). This thesis explores Celsus’s and Origen’s differing understandings of what it means to be “pious” (ὅσιος). Celsus conceived of tradition as the norm for determining piety. On the other hand, Origen maintained that the true norm was found in the Logos and Wisdom of God—i.e., Jesus. This dichotomy of understanding is consistent with the backdrop of the religious revolution happening in the Roman world during the early centuries CE proposed by scholars like Guy Stroumsa.

Oncolytic adenoviruses (Ads) have great therapeutic potential for lung cancer. Cancer selective E1b-deleted Ads are safe, however their efficacy remains limited clinically. To improve E1b-deleted Ads, Adhz60 was selected for greater anti-cancer efficacy by a process called bioselection, producing AdUV. AdUV preferentially lysed A549 and H1299 lung cancer cells more effectively than both the cancer selective Adhz60 and non-selective Ad5. AdUV induced greater LC3-II expression, relative to LC3-I, indicating that AdUV (30.9-fold) induced autophagy more effectively than Ad5 (12.2-fold) and Adhz60 (7.8-fold) in A549 cells. Mice treated with AdUV had significantly smaller tumors (p-value < 0.001) and prolonged survival (p-value = 0.0005) than mice treated with the negative control AdGFP. These results suggest that AdUV efficiently lyses lung cancer cells in vitro and in vivo.

The goal of this project is to characterize a new mouse model of alcohol-enhanced acute lung injury (ALI) and to determine the role of plasminogen activator inhibitor-1 (PAI-1) in this model. Male mice (WT and PAI-1-/-) were exposed to ethanol-containing Lieber-DeCarli diet or pair-fed control diet for 6 weeks; some animals were administered intraperitoneal lipopolysaccharide (LPS) prior to sacrifice. Chronic alcohol feeding enhanced induction of the chemokines MIP-2 and KC (murine IL-8 homologues) after LPS injection in wild type animals. This enhanced chemokine expression did not correlate with enhanced pulmonary neutrophil infiltration, however animals exposed to chronic ethanol showed sustained …

Receptor Tyrosine Kinases (RTK’s) like EGFR, Insulin Receptor, IGF1R and others mediate growth, proliferation and balance between apoptosis and survival. Dysregulation of RTK activity has been identified in a wide range of cancers and contributes to initiation, progression and metastasis of tumors. Ubiquilins (UBQLN) are fairly novel proteins and are studied mostly in the field of neurodegenerative disorders. Our laboratory recently discovered a link between UBQLN1 function and tumorigenesis. Hepatocellular Carcinoma (HCC) is a challenging condition to treat in the clinic due to its distinct tumor phenotype, aggressive growth, and chemotherapy resistance. Cancer Stem Cells (CSCs) are a poorly differentiated subpopulation of cells within the tumor microenvironment, and role of CSCs in tumor initiation and chemotherapy resistance is not well understood in HCC. In this study, we have successfully identified HCC cells with CSC-like properties that showed resistance to Doxorubicin. Using the established serum free culture techniques, we have enriched CSC-like cells among hepatocellular cancer cells. A double-blind placebo-controlled trial determined the long term effects of Protandim® supplementation on 5-km running performance, as well as on acute and long-term oxidative damage assessed by serum lipid peroxides (Thiobarbituric acid-reacting substances: TBARS). Thirty-eight subjects were placed into one of two groups: an experimental (675 mg/day Protandim®) or control group (675 mg/day corn starch). 5-km runs were performed at baseline and 88 (SD 4) days post-supplementation with samples taken immediately pre- and 10-min post 5-km run, as well as at rest ∼30 and ∼60 days during the supplementation period. In rodent models, suramin improves recovery following acute kidney injury (AKI). We hypothesized that suramin would be useful in protection against cisplatin-induced AKI. This hypothesis was tested by pre-treating C57BL/6j mice with suramin prior to cisplatin. Our data indicates that suramin protects the kidney from injury by decreasing cisplatininduced decreases in kidney function. Renal histology indicated that suramin significantly protected from cisplatin-induced AKI. Data indicate that suramin pretreatment attenuated mRNA expression of pro-inflammatory cytokines and chemokines following cisplatin treatment, while also decreasing markers of cell stress and cell death. Upon ligand-mediated activation, the EGFR responsible for cell growth, proliferation, and tissue homeostasis in epithelial cells; however, the EGFR is overexpressed in many human malignancies including MDA-MB-468 cells, a metastatic breast epithelial cell line. Background: Hyperhomocysteinemia (HHcy) is heavily implicated in diabetes and cardiovascular disease. HHcy is known for causing inflammation and vascular remodeling, particularly through production of reactive oxygen species (ROS) and matrix metalloproteinase-9 (MMP-9). Although its impact on skeletal muscle is rather unclear, HHcy is known to cause skeletal muscle weakness and functional impairment. The impact of HHcy on inflammation makes it seem likely that HHcy causes skeletal muscle fibrosis through induction of inflammatory factors and destructive macrophages. This study explores the mediating effect of one’s perceived relationship with God on the relationship between prayer and mental health in a cultural context. As an interpersonal activity, prayer may operate along relational constructs. Research supports a theoretical mechanism of prayer that aligns with the benefits of supportive interpersonal relationships. It is possible, then, that as relational processes change across dimensions of individualism and collectivism, prayer may mirror these same changes. c-kit+ cardiac progenitor cells (CPCs) have recently gained much attention due to the therapeutic effects they exert on cardiac function following their administration into the infarcted heart as evidenced by animal studies and by a recent clinical trial (SCIPIO). The relationship between the processing of orientations by the human visual system has been related to the orientation content of the natural environment; horizontal orientations, while predominant in natural environments, are perceived less well than vertical and oblique orientations are perceived best, though they are least prevalent in the natural world. This ‘horizontal effect’ has further extended the well-studied relationship between visual encoding and natural scene statistics as the differential perception of orientations in broadband scenes inversely matches their differential representation in the natural environment.
