The redox-active type 1 copper site of amicyanin is composed of a single copper ion that is coordinated by two histidines, a methionine, and a cysteine residue. This redox site has a potential of +265 mV at pH7.5. Over ten angstroms away from the copper site resides a tryptophan residue whose fluorescence is quenched by the copper. The effects of the tryptophan on the electron transfer (ET) properties were investigated by site-directed mutagenesis. Lessons learned about the hydrogen bonding network of amicyanin from the aforementioned study were attempted to be used as a model to increase the stability of another …

Alzheimer's disease (AD) is accompanied by abnormal extracellular deposition of amyloid b (Ab) peptide. This has led to the amyloid cascade hypothesis, causatively relating Ab with AD. While Ab deposits assume a fibrillar cross-b structure, prefibrillar oligomers of Ab have been identified as the main cytotoxic agents in AD. Pyroglutamylated amyloid beta (AbpE) peptides are N-terminally truncated and pyroglutamylated (at Glu3 or Glu11) Ab molecules that display enhanced cytotoxicity and represent up to 50% of total Ab in AD brains. AbpE significantly enhances the toxicity of unmodified Ab by an unknown mechanism. Although in situ Ab populations are heterogeneous, the …

Attitudinal research continues to show a more liberal trend in racial attitudes of Whites towards Blacks across a spectrum of issues (i.e. interracial marriage, residential integration, etc.) leading some to believe that we are moving into a post-racial society. However, a growing body of research has brought into question this utopian post-racial assertion. While Whites express support for racial equality in principle, they vehemently oppose race-targeted policies aimed to address racial inequality. This principle-policy gap has been consistently associated with persistent racial resentment; thus, contradicting the notion that of a post-racial era. Krüppel-like factor 8 (KLF8) has been strongly implicated in breast cancer metastasis. However, the underlying mechanisms remain largely unknown. In this study we report a novel signaling from KLF8 to C-X-C cytokine receptor type 4 (CXCR4) in breast cancer. Overexpression of KLF8 in MCF-10A cells induced CXCR4 expression at both mRNA and protein levels. This induction was well correlated with increased Boyden chamber migration, matrigel invasion and transendothelial migration (TEM) of the cells towards the ligand CXCL12. On the other hand, knockdown of KLF8 in MDA-MB-231 cells reduced CXCR4 expression associated with decreased cell migration, invasion and TEM towards CXCL12. …

Resistance exercise eliciting muscle damage results in an immune response, leading to increases in circulating cytokines, and immune cell mobilization. Classical monocytes respond to muscle damage, however, little is known about the intermediate or nonclassical monocyte response to resistance exercise. Moreover, the impact of polyphenol supplementation in conjunction with resistance exercise on the innate immune response is unknown. The purpose of this study was to examine the immune response following resistance exercise with (PPB) and without (PL) polyphenol supplementation. Kidron Road and Other Stories is a collection of fiction that ranges from the soberly tragic to the magically real. The characters in each selection are molded by their choices, the choices of others, and the cruel whims of fate. I am fascinated by the way fatalism and free will intersect in the human experience. Therefore, my work often explores the paradoxical way lives are molded by past decisions while, at the same time, those decisions seem determined by outside forces.

Historically high rates of human impact and recent Supreme Court cases, which have affected the regulation of geographically isolated wetlands in the United States, suggest that Carolina bays may be at high risk of human impacts in the future. Conservation efforts should be established based on applied ecological data and, if regulations by state and federal agencies are unable to conserve Carolina bay wetlands, it may be important to prioritize individual bays for preservation on protected lands. Bats are important to many ecological processes such as pollination, insect (and by proxy, disease) control, and seed dispersal and can be used to monitor ecosystem health. However, they are facing unprecedented extinction risks from habitat degradation as well as pressures from pathogens (e.g., white-nose syndrome) and wind turbines. LiDAR allows ecologists to measure structural variables of forested landscapes with increased precision and accuracy at broader spatial scales than previously possible. This study used airborne LiDAR to classify forest habitat/canopy structure at the Ordway-Swisher Biological Station (OSBS) in north central Florida. The thesis of this dissertation is that in the defining moment in which he is transformed from "uncircumcised of lips" to "god to Pharaoh" (Ex 6:28-7:1), Moses is best understood as Yahweh's idol, undergoing a status change akin to the induction ritual for ancient Mesopotamian idols, the Mīs Pȋ ("Washing/Purifying of the Mouth"). To make this point, I argue that Moses and idols be compared with respect to their status as mediator between divine and human realms. Previous research has clearly shown that urban stormwater runoff quickly transports pathogens, metals, sediment, and chemical pollutants to receiving waterbodies, resulting in the degradation of receiving waters and disruption of ecological networks. Inflammatory Bowel Disease and Crohn's disease represent chronic intestinal inflammatory diseases. It is suspected that bacterial infection is one of the causes of gut inflammation. Studies from others as well as from our laboratory have indicated that peroxidized lipids and their decomposition products are pro-inflammatory. As we consume considerable amounts of dietary oxidized lipids (arising from deep frying of vegetable oils), we hypothesize that dietary peroxidized lipids may also lead to intestinal inflammation. Prior to the 1910s these formats involved a more scientific and objective approach to recording nature. This aesthetic was abandoned in favor for narrative recreations and Hollywood structure after the 1930s. From 1943 to 1946, the U.S. government held over 3,000 German POWs at Camp Trinidad in southern Colorado. In 2013 and 2014, archaeological fieldwork, interviews, and archival research were conducted in order to better understand the daily lives of those incarcerated at the camp. The information gathered about artifacts, environmental features, and personal narratives, reveals insights into the lesser known details of the prisoners' lives. Despite the U.S. military rules and regulations and efforts by American personnel within camp, prisoners created goods they wanted or needed. Clinical intervention to reduce cardiac damage following myocardial ischemia is limited. Polyunsaturated fatty acids have been linked to an overall beneficial effect on cardiovascular health and function. Conjugated linoleic acid (cLA) is an 18:2 unsaturated fatty acid found within the diet in ruminant meat and dairy products. Virtually every ecosystem has been invaded by exotic organisms with potentially drastic consequences for the native fauna or flora. Studying the forms and rates of invading species has been an important topic in spatial ecology. We investigate two two-species competition models with Allee effects in the forms of reaction-diffusion equations and integro-difference equations. We discuss the spatial transitions from a mono-culture equilibrium to a coexistence equilibrium or a different mono-culture equilibrium in these models. Counter to what some people think, urban areas can be biodiversity hotspots. Maintaining this biodiversity can be challenging, since exotic shrubs and vines block sunlight and threaten native plant regeneration. Since 2007, the Louisville Olmsted Parks Conservancy (LOPC) has spent $2 million on invasive plant management in Cherokee Park. Before the project began, long-term transects were established by the LOPC to collect baseline presence/absence data on 11 invasive plant species. In 2014, I revisited these transects and documented presence/absence data on the entire plant community. This dissertation attempts to discover the mechanisms between cardiovascular and respiratory motor control post spinal cord injury (SCI): in normal, non-injured (NI) persons, cardiovascular regulation is dependent upon respiration, but there is nothing that suggests the mechanism for this relationship post-SCI. Thus we hoped to evaluate various aspects of cardiovascular regulation to further illustrate how this relationship is changed or unchanged by SCI. White-footed mice (Peromyscus leucopus) select for areas of greater canopy cover at the macro scale, and for areas with more CWD at the micro-scale. White-footed mice potentially avoid habitats with higher invasive species richness. White-footed mice preferentially foraged under the honeysuckle canopy in response to changes in temperature and humidity. This study suggests that the interaction between P. leucopus and ground layer invasive species is complex, and that the effect of moonlight may be diminished in this urban park. Urban ecosystems demonstrate high levels of anthropogenic land-use change, modification of abiotic inputs, and altered disturbance regimes. These changes result …

Permeable pavements systems and tree boxes are a common type of Green Infrastructure (GI) Stormwater Control Measures (SCMs) that are often used for mitigating the stormwater runoff. In this study two permeable pavement systems and a tree box installed along parking lanes of an urban street in Louisville, KY, were investigated to evaluate their performance on improving stormwater runoff quality. Circulating angiogenic stem cells (CACs) are rare cells found in peripheral blood that have been shown to contribute to endothelial repair and new blood vessel formation. These cells could be biomarkers and/or therapeutic targets for the assessment and prevention of cardiovascular disease (CVD), which is the leading cause of mortality globally and in the United States. This dissertation describes the previously unidentified effects of a plant-produced recombinant cholera toxin B subunit (CTBp) on the gastrointestinal (GI) tract and its ability to protect against inflammation in a mouse model of colonic injury and ulcerative colitis (UC). To comprehensively analyze CTBp’s impacts on the GI tract, we employed global analysis methodologies based on multi-color flow cytometry to analyze immune cell populations in GI and systemic lymphatic compartments, gene expression microarray to decipher transcript-level changes in the colon and small intestine, and 16S rRNA sequencing to characterize fecal microbiota.
