Although urbanization is a leading cause of species extinction throughout the world, the impact of urban development on arthropods is little studied and, as a result, poorly understood. I used three distinct approaches to studying arthropod conservation in North America. First, I used landscape genetics techniques to study the impact of urbanization on gene flow among populations of Rabidosa rabida, the rabid wolf spider. While gene flow was not detrimentally reduced, urban development correlated with a reduction in migration rates among populations, and to my knowledge, this is the first study to document isolation by resistance in spiders. Background: Pulmonary mycobacterial diseases describe both tuberculosis (TB) and nontuberculous mycobacteria (NTM). Few data are available measuring the cost burden of mycobacterial diseases on the national level. The purpose of this study was to evaluate the cost burden and measure emerging trends in hospitalization of pulmonary TB and NTM in the US from 2001 through 2012. Methods: This study was a retrospective community based cost analysis of hospitalized patients with a principal diagnosis of pulmonary mycobacterial diseases from 2001 through 2012. Spinal Cord Injury (SCI) in humans is a heterogeneous diagnosis, resulting in variable paralysis and paresthesia based on the mechanism, rostro-caudal location, and severity of injury. Both neurophysiological and anatomical studies have suggested that subclinical residual supraspinal-spinal connectivity exists in a subset of individuals deemed to have motor and sensory complete injuries. Recent reports of volitional movement in chronic, motor complete individuals during epidural spinal stimulation have provided compelling evidence that these residual projections may be capable of mediating volitional movement when the functional state of spinal circuitry is electrically modulated. A reaction-diffusion model and a lattice differential equation are introduced to describe the persistence and spread of a species along a shifting habitat gradient. The species is assumed to grow everywhere in space and its growth rate is assumed to be monotone and positive along the habitat region. We show that the persistence and spreading dynamics of a species are dependent on the speed of the shifting edge of the favorable habitat, c, as well as c*(∞) and c*(−∞), which are formulated in terms of the dispersal kernel and species growth rates in both directions. The median survival for idiopathic pulmonary fibrosis (IPF) patients from diagnosis is a dismal 3 years. This condition is characterized by pulmonary fibroproliferation and excess production and disordered deposition of extracellular matrix (ECM) proteins resulting in obliteration of the original tissue architecture, loss of lung function and eventual death due to respiratory failure. Background: Brain death triggers an inflammatory response that is associated with worse outcomes for recipients of transplanted organs procured from brain dead donors. The intestine has been shown to release inflammatory mediators in other types of shock, but its role is brain death is not well described. Direct peritoneal resuscitation (DPR) improves visceral organ blood flow and has been shown to reduce inflammation after hemorrhagic shock. Better understanding of this inflammatory process within the peripheral organs, the effects of DPR, and the role of the intestine could aid in better resuscitation of potential organ donors. The coexistence of similar species may be related to a variety of resource utilization differences. Dietary resource utilization variation may be the most important difference allowing for the coexistence of sympatric snake species. Many watersnakes (Nerodia spp.) live in sympatry and use similar aquatic habitats feeding on similar prey. While these sympatric watersnakes may have different general foraging patterns, snake diet may be affected by a variety of factors. The review (Chapter 1) overviews our current understanding of the sex bias in autism spectrum disorders (ASD), whereby males are 4 times more likely to develop the condition. The longstanding uncertainty surrounding the etiology of autism can be informed by inclusion of both sexes in future research to better understand the mechanism responsible for this sex bias. Both clinical studies and animal models are useful tools to elucidate the underlying cause(s) of sexual dimorphic morphological, physiological, and behavioral differences in the ASD phenotype. Lassa virus (LASV) is an arenavirus and causative agent of Lassa fever (LF), a viral hemorrhagic fever in West Africa for which there is no vaccine. Lymphocytic choriomeningitis virus (LCMV), is used as a surrogate to mimic LASV-induced liver pathology. LCMV-WE, not LCMV-ARM, causes disease in primates and mice characterized by hepatitis, high viral load, hepatocyte proliferation, and upregulated proliferative triggers (e.g. TNF-α, IL-6 ). We hypothesize LCMV-WE induces pathological hepatocyte proliferation via pro-inflammatory triggers (TNF-α, IL-6) from macrophages, leading to: increased viral replication, modulated cell cycle, and arrested cell cycle. Acute myeloid leukemia (AML) is a highly heterogeneous clonal disorder characterized by an accumulation of malignant immature myeloid progenitors in the bone marrow (BM) that hinder normal hematopoiesis. Patient AML exhibits a dramatic heterogeneity in terms of cytogenetics, disease morphology, and associated prognoses and/or chemotherapeutic sensitivity. Background: Tumor necrosis factor (TNF)-like weak inducer of apoptosis (TWEAK) is a proinflammatory cytokine belonging to the TNF super family. TWEAK produces a variety of cellular responses through the binding to fibroblast growth factor inducible 14 (Fn14), a member of TNF receptor superfamily. Although Fn14 lacks a death domain, TWEAK has been found to induce apoptosis in some cell types by perturbing the activity of certain pathways such as TNF-receptor signaling. High calorie diets have fostered the current pandemic of obesity and comorbid conditions of non-alcoholic fatty liver disease (NAFLD), systemic insulin resistance (IR) and type 2 diabetes (T2D). Hepatic glutathione S-transferases (GSTs) are dysregulated in obesity, NAFLD and diabetes. The multifunctional GST pi isoform (GSTP) catalyzes acrolein metabolism and inhibits JNK (c-Jun NH2-terminal kinase). Spinal cord injury (SCI) results in whole-body dysfunction. While the majority of SCI research is focused on improving locomotor function after injury, cardiovascular (CV) disease continues to be the leading cause of morbidity in the chronic SCI patient population. The combination of injury-induced destruction of autonomic pathways, maladaptive plasticity within lumbosacral circuits, and the progressive decline in physical fitness contribute to the poor CV status of SCI individuals. Women with breast cancer experience significant stress related to diagnosis and treatment, which may negatively affect physiology (e.g., circadian, endocrine, and immune) and contribute to cancer progression. Demographic (e.g., ethnic minority) and neighborhood factors can contribute to cancer-related stress. African American women with breast cancer experience more adverse disease pathology and earlier mortality; however, factors that explain these disparities rarely have been explored. It is hypothesized that psychosocial stressors (i.e., ethnicity, neighborhood stress, and cancerrelated stress) will be related to one another. This thesis examines the way in which the fictional biographies of Virginia Woolf, Orlando and Flush, prefigure central tenets of postmodern fiction. To demonstrate the postmodern elements present in Orlando and Flush, this thesis focuses on how the fictional biographies exhibit three postmodern characteristics: concern for historiography, extensive use of parody, and the denaturalization of cultural assumptions. Born from Woolf’s desire to revolutionize biography by incorporating elements of fiction alongside historical fact, these two novels parallel later works of historiographic metafiction in several key respects. Flavonoids are polyphenolic secondary metabolites found in plants that have bioactive properties including antiviral, antioxidant, and anticancer. Two isomeric flavone were extracted from Gnaphalium elegans and Achyrocline bogotensis, plants used by the people from the Andean region of South America as remedies for cancer. 5,7-dihydroxy-3,6,8-trimethoxy-2-phenyl-4H-chromen-4-one (5, 7–dihydroxy- 3, 6, 8 trimethoxy flavone/ flavone A) and 3,5-dihydroxy-6,7,8-trimethoxy-2-phenyl-4H-chromen-4-one (3, 5–dihydroxy-6, 7, 8–trimethoxy flavone/ flavone B) have shown antineoplastic activity against colon cancer cell lines dependent upon their differentiation status. The purpose of this quantitative study was to determine the perceptions of Northeast Tennessee Educators regarding arts integration. Specifically this study was an examination of the perceptions of district and K-8 school level administrators, K-8 general classroom teachers of math, science, social studies, or literacy, and K-8 arts specialists of dance, drama, music, or visual arts. Nine school districts in Northeast Tennessee agreed to participate in the study. Data were collected through an online survey system, SurveyMonkey.com. This research is focused on the study of the effect of mutating proline 145 to threonine on the substrate and regiospecificity of flavonol specific 3-O-glucosyltransferase (Cp3GT). While the mutant P145T enzyme did not glucosylate anthocyanidins, it did glucosylate flavanones and flavones in addition to retaining activity with flavonols. HPLC was used for product identification and showed mutant P145T glucosylated naringenin at the 7-OH position forming naringenin-7-O-glucoside and flavonols at the 3-OH position. Using the lens of ecofeminism, this study examines the undergirding structures that produce systems such as colonization that oppress and destroy land, people, and other beings. This research argues that with the rise of the middle-class, Victorian England saw the development of a power model in which middle-class men, middle-class women and disenfranchised bodies of children and lower-class women suffered from the demands of bodily domination. Because the bodily health of middle-class men was believed to represent national health, it was imperative that he dominate his body, particularly with regard to sexual urges. One of the most prominent traditions associated with the Southern Appalachians is the art of weaving. Extensive research has focused on the history of Appalachian weaving, but there is little on the current weaving community. Today, the region still serves as an axis for weaving, and many practicing weavers, weaving instructors, and learning institutions can be found in Southern Appalachia. The core of this study is the interviews with ten weavers that reside and practice their work in Appalachia. Two trials were conducted with Santa Gertrudis steers, trial 1 (n = 115) and trial 2 (n = 118), were fed a roughage-based diet during the growing phase and high-grain diet during the finishing phase. Steers were weighed at 14-d intervals and dry matter intake (DMI) measured for 70 d during both phases. Residual feed intake (RFI) is the difference between actual DMI and DMI predicted from linear regression of DMI on mid-test metabolic body weight. Eucalyptus (Eucalyptus spp.) plantations have the potential to boost wood production for pulpwood and biomass feedstocks. This dissertation examines the digital circulation of personal narratives by non-celebrity individuals that become part of larger public and political debates. I posit the “available means of imagination” to describe the ways that narratives – cultural, fictional, and personal – influence our ability to understand the many facets of a given public debate before tracing the interactions among narrative, emotion, and circulation in a series of case studies using new materialist methods. Transgender populations demonstrate significant sexual diversity, with over three-fourths (77%) of transgender individuals identifying as sexual minorities (Grant et al., 2011); however, a majority of research on sex, sexuality, and sexual practice has focused exclusively on cisgender and heterosexual individuals and, perhaps more importantly, addressed these issues a heteronormative and cisnormative perspective (Nichols, 2014). Such frameworks have created significant gaps in research about sex, sexuality, and sexual practice for trans individuals including understanding how gender transition influences sex and sexuality and how trans individuals and their partners engage in sexual practice. Coal ash, a byproduct of coal combustion,is reduced in 47 U.S. states andfrequently contains heavy metals, some of which are known neurotoxins. An estimated 1.5 million children live near sites where coal ash is produced andstored, yet there have been no studies assessing coal ash exposure and children’s neuro behavioral health.
