Small populations, specifically those that are isolated from others, are more prone to extinction than larger inter-connected populations. The risks that these small isolated populations face include loss of genetic diversity due to founder effects and inbreeding due to population bottlenecks, as well as demographic uncertainty due to fluctuating fecundity and mortality rates and impacts of external environmental factors. Ex situ populations, including those managed as conservation breeding programs with species recovery aims, as well as those that do not have reintroduction goals but are managed for long term population sustainability, suffer from the same extinction risks as small and …

This dissertation focuses on the development of diagnostic tools for mycobacteria using hybridization based technologies including binary deoxyribozyme (BiDz) sensors and microarrays. The genus Mycobacterium, is a diverse group of bacteria containing 150+ species including M. tuberculosis (M.tb) and non-tuberculous mycobacteria (NTM) which exhibit a range of pathogenicity, drug susceptibility and growth characteristics. M. tuberculosis (M.tb) is the causative agent of tuberculosis (TB) and the leading cause of infectious disease related deaths worldwide. Neurofibromatosis type 2 (NF2) is a debilitating disease characterized by the formation of bilateral vestibular schwannomas, which compress the vestibulocochlear nerve and cause deafness. Additional peripheral schwannomas, meningiomas and ependymomas may also form. NF2 is caused by mutations in the NF2 gene, resulting in the loss of function of the merlin tumor suppressor. Merlin functions in multiple signaling pathways and its absence in Schwann cells yields increased cell survival and proliferation, thereby causing schwannoma formation. First line treatment for NF2 is watchful waiting and surgical removal of tumors, potentially resulting in facial paralysis and deafness. To date, there are no …

Animals that co-occur in sympatry with multiple closely-related species use reproductive mate attraction signals not only to assess the quality of a potential conspecific mate (sexual selection), but also to discriminate conspecifics from heterospecifics (species recognition). However, the extent to which sexual selection and species recognition may interact, or even conflict, is poorly known. Neotropical electric fish offer unrivaled opportunities for understanding this problem. They generate simple, stereotyped mate attraction signals that are easy to record and quantify, and that are well-understood from the neurobiological perspective. Peer interaction is an important part of language learning. The results of previous studies showed advantages and disadvantages of peer interaction between native speakers and English learners. Using the educational simulation platform TeachLivE as the interaction platform, this qualitative descriptive case study explored the possibilities of bridging the gap between classroom language practice and real-life second language communication. The study recruited eight participants for a task-based interaction project. Using conversation analysis, the study revealed details in communication between adult English learners and their digital character peers. Sea turtle hatchlings primarily utilize sight to detect differences in elevation and light intensity present along the horizon to navigate from the nest to the water’s edge. The addition of artificial lights can cause visual misdirection, resulting in disorientation (aimlessly wandering in circular paths) or misorientation (moving in distinct paths away from ocean). Extensive research has been done on effects of high levels of artificial light but little on effects of comparatively lower levels of artificial light on hatchling sea turtle orientation. Habitat fragmentation may alter the spatial ecology of organisms inhabiting the fragmented landscape by limiting the area of habitat available and altering microhabitat features. I quantified and compared movement of southern copperheads (Agkistrodon contortrix contortrix) in a fragmented and non-fragmented habitat to determine the effects of habitat fragmentation on the spatial ecology of the southern copperhead. Effective distance moved by each individual was measured by the use of thread bobbins attached via medical tape to the posterior quarter of the snake. Microbial water quality is of significant concern in the two watersheds of the Grand Stand. Polluted runoff, malfunctioning septic tanks, and centralized sanitary sewer failures are common sources of fecal bacteria contamination in coastal areas and pose a threat to human health through recreational use of waterbodies and shellfish consumption. Volunteer water quality monitoring programs are crucial in expanding upon assessments of fecal bacteria contamination by regulatory monitoring. Polyamines are small cationic molecules that play important roles in most vital cellular processes including cell growth and proliferation, regulation of chromatin structure, translation and programmed cell death. Cellular polyamine pools are maintained by a balance between biosynthesis and transport (export and import). Increased polyamine biosynthesis activity and an active transport system are characteristics of many cancer cell lines, and polyamine depletion has been shown to be a viable anticancer strategy. Polyamine levels can be depleted by α-difluoromethylornithine (DFMO), an inhibitor of the key polyamine biosynthesis enzyme ornithine decarboxylase. The objective of this work was to introduce a novel formulation strategy for intranasal delivery of two low permeation candidates: levodopa (L-DOPA; antiparkinsonism drug) and ribavirin (RBVN; an antiviral drug). The central hypothesis states that preparation of binary composite microparticles with Pluronic® triblock copolymers (polyethylene glycol-polypropylene glycol-polyethylene glycol) should allow increased drug transport across nasal mucosal tissue relative to drug alone. Testing the hypothesis necessitated execution of specific aims directed to addressing issues inherent to the proposed approach. The centrality of sacrifice in the Judeo-Christian tradition is undeniable; nevertheless, unidimensional presentations of sacrifice have contributed to an incomplete, and at times distorted understanding. This work does not aim to resolve every ambiguity or misunderstanding, rather, it presents a canonical approach interpretation and brings attention to three underappreciated dimensions of sacrifice: first, the relationship between the external actions of the ritual and the internal dynamic of spirituality and morality; second, the reality that sacrifice refers not only to death, but to a way of living; and third, atonement and reconciliation are not the only reasons to offer sacrifice. The gold standard in 2016 for thoracic aortic grafts is Dacron®, polyethylene terephthalate, due to the durability over time, the low immune response elicited and the propensity for endothelialization of the graft lumen over time. The focus of this research is the ways in which interactions between Indigenous peoples and English settler-colonists were manifested in the landscape at a seventeenth-century site in South Glastonbury, Connecticut. Magnetometry and ground-penetrating radar allowed for the location of anthropogenic and geological features on the landscape, and for the seventeenth-century landscape to be recreated. This reconstruction indicated that Europeans and Indigenous peoples may have been cohabitating the site. Archival research helped to uncover what types of interactions may have been occurring at the site. The hydrodynamic model Delft3D is used to study the impact of Sea Level Rise (SLR) on storm surge and inundation in the coastal region of Bangladesh. To study the present-day inundation scenario, track of two known tropical cyclones (TC) were used: Aila (Category 1; 2009) and Sidr (Category 5; 2007). Model results were validated with the available observations. Eurygaster integriceps Puton (Sunn pest) is the most damaging insect pest of wheat in areas of North Africa, Eastern Europe and Western and Central Asia. Sunn pest injects an enzyme characterized as a prolyl endylprotease (spPEP) into wheat grains that digests the gluten protein (Darkoh et al., 2010). Previous studies have shown that casein peptides generated by Lactobacillus species have inhibitory effects on human (hPEP) and bacterial prolyl endoproteases (bPEP) which makes casein a good natural candidate to inhibit spPEP. The purpose of this study was to (a) determine the locus of control among American college students (b) determine if tobacco use or cessation correlate with any demographic variables to better understand the efficacy of tobacco interventions and help design an intervention most effective in the prominent LOC of college students.

Global climate change and anthropogenic activity have facilitated the movement and invasive potential of nonnative plants in native environments. These invasions can have negative effects on ecosystem diversity and function. The nonnative and invasive plant, Chinese Tallow (Triadica sebifera), has already invaded much of the south eastern US where it is outcompeting native tree species and changing ecosystem diversity in a variety of habitats. Light information is transmitted to the brain through approximately 30 ganglion cells (GCs), the retinal output neurons. Unusual monoenoic fatty acids (UMFA’s) and specialized metabolites called anacardic acids (AnAc) are produced in glandular trichomes of Pelargonium ´ hortorum (geranium). The UMFA’s, 16:1∆11 and 18:1∆13 are precursors for the synthesis of unsaturated AnAc 22:1n5and 24:1n5 that contribute to pest resistance in geraniums. UMFAs and their derived AnAc metabolites not only provide a useful biological marker that differentiates the biosynthetic pathway for unusual mononenes from the common fatty acids (i.e. stearic, palmitic, oleic, linoleic and linolenic) but also have industrial, medical and agricultural applications. Vitamin A metabolism, which produces the signaling molecule Retinoic Acid (RA), has been demonstrated to be important for growth and branching morphogenesis of mammalian embryonic salivary gland epithelium. However, it is not known whether RA functions directly within epithelial cells or in associated tissues that influence morphogenesis of salivary epithelium. Moreover, downstream targets of RA transcriptional regulation have not been identified. Here we show that canonical RA signaling occurs in multiple tissues of embryonic mouse salivary glands, including epithelium, associated parasympathetic ganglion neurons, and non‑neuronal mesenchyme. Trauma, major surgery and systemic infection can lead to a subsequent immunosuppressed state which can predispose patients to nosocomial infection and death. Approximately a sixth of patients will have impaired monocyte function, as determined by decreased HLA-DR expression or decreased TNF-α production in response to ex-vivo lipopolysaccharide (LPS) stimulation. This project aimed to develop an in-vitro model of impaired monocyte function using isolated monocytes from healthy volunteers, to study the effect of monocyte impairment on IκK signaling, and determine the role of IκK in the impaired monocyte response. These studies have provided the following results:

Since Durkheim’s characterization of the sacred and profane as “antagonistic rivals,” the strict dichotomy has been framed in such a way that “being religious” evokes images of a life filled with profound meaning and value, while “being secular” evokes images of a meaningless, self-centered, superficial life, often characterized by materialistic consumerism and the cold, heartless environment of corporate greed. Consequently, to identify as “neither religious nor spiritual” runs the risk of being stigmatized as superficial, untrustworthy, and immoral. Conflicts and confusions encountered in the process of negotiating a nonreligious/nonspiritual identity, caused by the ambiguous nature of religious language, were explored …
