Periodontitis is an infection-driven inflammatory disease characterized by gingival inflammation and bone loss. The NF-ĸB signaling pathway is pivotal in osteoclastogenesis and infection-induced pro-inflammatory responses. The use of nanoparticles as a vehicle to deliver drug increases stability, loading capacity, and facilitates transmembrane transportation. The hypothesis was that a novel nanoparticle carrying therapeutic NBD inhibitory peptides (NBD-nanoparticles) will inhibit measures of periodontal disease. In this project, we tested the nanoparticles for their ability to directly inhibit osteoclastogenesis and inflammation as an original strategy for periodontitis therapy. This thesis is to chart my discovery that confidence is essential to an actor’s craft. This thesis focuses on my performance as Edmund in King Lear, as well as my production the previous summer playing Hussein in Ayad Akhtar’s Disgraced. I draw from a variety of sources, such as meditation and formal acting exercises, to achieve confidence. Most importantly, I distill my missteps as an actor for a better understanding of what hinders confidence when building a role for performance.

Spatial analysis at the site level offers unique perspectives and has become increasingly more accessible as advances in technology allow rapid 3-dimensional mapping and rigorous statistical analysis. This project focused on the geological stratigraphic level “F”, at Lapa do Picareiro; a shallow cave in the Estremadura region of Portugal. This level, dated to 14,000 years ago calibrated BP, experienced multiple human occupations between the Last Glacial Maximum and the Younger Dryas. This work examines the recovered material from a period of Upper Magdalenian occupations and sheds light on how enclosed space was used in Estremadura Portugal. Distinct distributions of lithic …

Background. The association of chronic ethanol (EtOH) consumption with extracellular matrix (ECM) changes in alcoholic liver disease (ALD) is well established in the context of hepatic fibrosis. However, it is becoming increasingly understood that the hepatic ECM responds dynamically to stress well before fibrosis. The term “transitional tissue remodeling” describes qualitative and quantitative ECM changes in response to injury that do not alter the overall architecture of the organ; these ECM changes may contribute to early disease initiation and/or progression. Verisimilitude, or the appearance of being true, is a concept I turn upside down; relating it to a guise I wear as a contemporary male in a society dictated by learned social behavior and gender norms. Cultural iconography and expected gender norms are tropes I confront within my artwork. Drawings of seemingly everyday objects act as meditations or a fetishized repetition of supposed unobtainable objects and ideals that deal with masculine societal norms. This dissertation aims to explore how the phenomenon of cosplay has been able to produce and sustain a diversity of gender expression due to its emergence from an activity-based community that emphasizes creative play. This creative energy is manifested through cosplay as an active, ritualized practice in which gender diversity is invited to be realized as a distinct possibility, resulting in a display of a full range of masculinities and femininities as well as crossplays and genderbend cosplays. Myeloid derived suppressor cells (MDSCs) are a heterogeneous group of immature myeloid cells that are significantly increased in cancer patients and correlate with higher stage and poor prognosis. MDSCs negatively modulate anti-tumor immunity, suppress T cell activity, promote angiogenesis and increase the risk of metastasis. In this study, we report that monocytic-MDSCs (M-MDSCs) but polymorphonuclear-MDSCs (PMN-MDSCs) over-express 6-phosphofructo-2-kinase/fructose-2, 6-bisphosphatases 3 (PFKFB3), an important regulator of glycolysis. Furthermore in the melanoma model, M-MDSCs but not PMN-MDSCs suppressed T cell function which correlated with PFKFB3 over-expression and increased rate of glycolysis. This dissertation examines the feminist significance of Anya Seton’s historical novels, My Theodosia (1941), Katherine (1954), and The Winthrop Woman (1958). The two main goals of this project are to 1.) identify and explain the reasons why Seton’s historical novels have not received the scholarly attention they are due, and 2.) to call attention to the ways in which My Theodosia, Katherine, and The Winthrop Woman offer important feminist interventions to patriarchal social order. Soil bacteria play important roles in nutrient cycling and other ecosystem functions, and many biotic and abiotic factors can influence bacterial functional diversity. The goal of this field study was to examine differences among bacterial communities in sand dunes of Lake Michigan. I used Biolog Ecoplates™ to compare bacteria functional diversity associated with four different plant species: the native dune-building grass Ammophila breviligulata, invasive species Leymus arenarius and Gypsophila paniculata, and native legume Lathyrus japonicus across 13 sites in Michigan, representing a gradient in abiotic factors such as precipitation and temperature. Background: Although pregnancy is prone to induce more severe periodontal inflammation and tissue damage, its mechanism remains elusive. Periodontal diseases during pregnancy are associated with adverse pregnancy outcomes including preterm birth or low birth weight. Animal models are ideal for investigating immunological mechanisms in periodontal disease. With a newly modified ligature-induced disease model, we managed to develop periodontal disease in pregnant mice and investigate the immunological mechanism of more severe symptoms during pregnancy. Hypothesis: We hypothesis that P. gingivalis infection during pregnancy will lead to a down-regulation of Treg cells, therefore causing increased inflammation and disease severity. Hieronymus Bosch’s (1450-1516) paintings have long fascinated, intrigued, and mystified viewers. In particular, the Garden of Earthly Delights (c.1500) has generated much discussion and speculation. Bosch’s use of medieval symbols and fantastic images, his preoccupation with sinfulness and Hell, and his dismal view of mankind’s future have long been sources of study and speculation. Not only do art historians ponder his work, but so do social historians, philosophers, creative artists and the general viewing public. This study examined whether intake of caffeinated beverages (coffee, tea, and cola) was associated with fecundability (time to pregnancy) in a prospective cohort study. Data from 470 women from the Mount Sinai Study of Women Office Workers (1990-1994) were analyzed. Intake of coffee, tea, cola, and other variables were recorded in daily diaries and calculated as menstrual cycle level means for up to 20 cycles. Pregnancy was assayed using hCG and confirmed by physician diagnosis. The associations of caffeinated beverages with the probability of becoming pregnant during a given cycle were determined using discrete survival analysis, adjusted for potential confounders. …

This creative thesis is a collection of fictional stories, connected by a common geographical and cultural thread: Kuwait. The stories unravel across a span of four thousand years. The poetic-prose style of the opening vignette is a nod to the earliest form of writing that makes up the bulk of Kuwait’s literature: poetry. The abstractedness of the two opening pieces reflects the lack of a deep-rooted national identity, and a brevity in Kuwait’s historical record. Therapies targeting estrogen receptor α (ERα) including selective estrogen receptor modulators (SERMs), e.g., tamoxifen (TAM); selective estrogen receptor downregulators (SERDs), e.g., fulvestrant (ICI 182,780); and aromatase inhibitors (AI), e.g., letrozole, are successfully used in treating breast cancer patients whose initial tumor expresses ERα. Unfortunately, the effectiveness of endocrine therapies is limited as ~ 40% of breast cancer patients will eventually acquire resistance to them. This dissertation describes and investigates a debate about differences in the visual system’s anisotropic sensitivity to orientation as well as where and how the brain encodes visual orientation information. Three different visual perceptual patterns are discussed: the Class 1 and 2 oblique effects and the horizontal effect. The oblique effect is the ability to perceive cardinal orientations more easily than oblique orientations, and is found using narrowband stimuli. In the horizontal effect, oblique orientations are perceived most easily, horizontal orientations are perceived worst, and vertical orientations fall in-between, and is found using broadband stimuli. This work examines the construction and impact of the master narrative breast cancer which is supported and reified by the contemporary breast cancer awareness movement. I contend that historical problematic constructions of the female body were interwoven with the beginning of the movement around the turn of the twentieth century. As a partial result, the female body with breast cancer is abject, and therefore subject to policing and discipline. The master narrative of breast cancer, through pinkwashing, enacts this policing and discipline. Lunasin is a 44 amino acid peptide that has been shown to have cancer chemopreventative and chemotherapeutic properties. This study investigated the potential utility of Lunasin as a chemotherapeutic in melanomas. Studies showed that Lunasin had little activity against established melanoma cell lines using adherent culture methods; however, Lunasin’s in vitro activity was significantly higher in non-adherent colony-forming assays and oncosphere formation. These results led to the investigation of whether or not Lunasin has selective effects on cancer initiating cells (CIC) that are known to be present in melanomas. The Epidermal Growth Factor Receptor (EGFR) is a 170-kilodalton transmembrane protein that belongs to the ErbB family of receptor tyrosine kinases. Upon ligand-mediated activation, the EGFR is responsible for cell growth, proliferation, and tissue homeostasis; however, the EGFR is overexpressed in many human malignancies, including MDA-MB-468 cells, a metastatic breast epithelial cell line. Studies within this cell line, and other cell lines characterized with high EGFR levels, have shown that EGF stimulation results in the induction of apoptosis. This dissertation is focused on dissecting the functional role of two anatomically-defined pathways in the adult rat spinal cord. A TetOn dual virus system was used to selectively and reversibly induce enhanced tetanus neurotoxin expression in L2 neurons that project to L5 (L2-L5) or C6 (long ascending propriospinal neurons, LAPNs). Results focus on the changes observed during overground locomotion. The dissertation is divided into four chapters. Governing social-ecological systems, such as the urban water commons, is a multi-scale and multi-sector (polycentric) human-environment process. This dissertation interrogates this process by situating itself within the Ecology of Games Framework by Norton Long (and updated by Mark Lubell) and the literature on polycentric governance by the Bloomington School of Political Economy. Anaerobic digestion (AD) of municipal sludge is a widely used stabilization process at municipal wastewater treatment plants. It is highly effective in reducing sludge volume and produces methane gas which is used as fuel in the wastewater plant. Following digestion sludge is sent to a centrifuge to separate the solids from the liquids. Louisville MSD adds flocculent to material in the centrifuge to better coagulate the solids, thereby aiding in separation of the solids from the liquids. Increased demands on water supplies caused by population expansion, saltwater intrusion, and drought have led to water shortages which may be addressed by use of recycled water as recycled water products. Study I investigated recycled water production in Florida and California during 2009 to detect gaps in distribution and identify areas for expansion. Gaps were detected along the panhandle and Miami, Florida, as well as the northern and southwestern regions in California. Lilium grayi (Gray’s Lily), a southern Appalachian endemic species, is threatened by a Lilium-specific fungal pathogen, Pseudocercosporella inconspicua. The disease is characterized by tan lesions that can cause early senescence, while also lowering seed production and viability. This project tested for P. inconspicua conidia and accessed health at nine locations. The disease was present and ubiquitous across the range of L. grayi. Organisms living in aquatic environments are subject to a number of stressors from natural (temperature, predation, food availability) and anthropogenic origins (pesticides, metals, etc). Natural stressors may amplify the effects of contaminants and increase an organism’s sensitivity to them. Understanding the impact of these combined factors is therefore essential for the practical management of contaminants. Lapa do Picareiro, a cave located in Portuguese Estremadura, contains continuous deposits dated to the Late Pleistocene. As one of the highest elevation Upper Paleolithic sites currently known in Portugal, questions are raised about the function of the site during this time. This thesis presents a comprehensive analysis of the lithic assemblage recovered from the Late Magdalenian layer of F/G (13,442 to 14,011 cal BP). The data generated from this study, when looked at in conjunction with the previously analyzed faunal assemblage, helps to provide a broader understanding of the site’s function during the Late Magdalenian.
