We critically worked on the issues of technical and biological heterogeneity. Lung cancer is the leading cause of cancer related death in the United States and worldwide. Five-year survival rates for non-small cell lung carcinoma patients have not improved in decades, and a majority of patients succumb to metastasis. Identification of therapeutic targets for metastatic disease is essential for the development of novel therapies. Collaborative Filtering techniques provide the ability to handle big and sparse data to predict the ratings for unseen items with high accuracy. Matrix factorization is an accurate collaborative filtering method used to predict user preferences. However, it is a black box system that recommends items to users without being able to explain why. This is due to the type of information these systems use to build models. Lipid post-translational modifications enhance a protein’s interaction with membranes and other proteins. In this dissertation, I studied a recently identified Arabidopsis gene, PROTEIN PRENYLTRANSFERASE ALPHA SUBUNIT-LIKE (PPAL), by mapping a second-site mutation that rescues the original mutation phenotype and could be an additional factor involved in sugar homeostasis/sensing. I found that mutant phenotype associated with suppressor line was too weak and variable to consistently score, which resulted in not finding any linked markers. I also generated crosses between ppal-1 and ABA biosynthesis and signal transduction mutants to find any possible connection between these pathways and PPAL. By …

In the United States, high-risk drug use remains a significant social problem. Opioids and stimulants are two drug classes that have contributed to substantial recent increases in drug-related arrests, overdose, and mortality. Kentucky has been particularly devastated by high rates of opioid and stimulant use. Opioid and stimulant effects, while highly rewarding, can result in adverse consequences. Spinal cord injury (SCI) causes severe neurological impairment with widespread motor, autonomic and sensory deficits, leading to a substantial quality of life impairment. The number of individuals with SCI increases by approximately 12,500 annually, and over 80% of individuals with SCI are males. SCI individuals rate sexual function as a top priority quality of life issue, and men with SCI likely suffer from sexual dysfunction, such as erectile and ejaculatory dysfunction, as well as infertility. Acute myeloid leukemia (AML), is a heterogeneous clonal disorder characterized by an accumulation of malignant myeloid progenitors in the bone marrow (BM), hindering normal hematopoiesis. AML exhibits dramatic heterogeneity in terms of cytogenetics, morphology, and chemotherapeutic sensitivity. Partial or complete failure of red blood cell membrane, also known as hemolysis, is a persistent issue with almost all blood contacting devices. Many experimental and theoretical contributions over the last few decades have increased insight into the mechanisms of mechanical hemolysis in both laminar and turbulent flow regimes, with the ultimate goal of developing a comprehensive, mechanistic and universal hemolysis prediction model. The neural rings and ideals as algebraic tools for analyzing the intrinsic structure of neural codes were introduced by C. Curto, V. Itskov, A. Veliz-Cuba, and N. Youngs in 2013. Since then they have been investigated in several papers, including the 2017 paper by S. G\"unt\"urk\"un, J. Jeffries, and J. Sun, in which the notion of polarization of neural ideals was introduced. This dissertation develops a theoretical and empirical approach to the study of professional creative writers and teachers. Specifically, it examines how these writers developed their knowledge of creative writing and writing pedagogy and how that knowledge informs their work as instructors of composition. Similar to the painted creatures that dwell within the illuminated manuscripts of ancient and medieval bestiaries, the beasts in Leonora Carrington’s early work are used metaphorically. She was deeply influenced in her formative years by mythology and animals; tales with heraldic characters, obscure adventures, and symbolic meanings were foundational to her works. World War II and a subsequent internment in a sanitorium initiated Carrington to wrestle with an existential crisis. Her capacity and appetite for sources of inspiration and knowledge was boundless; infinite symbolic references were readily available for her artistic executions. Thermotolerance limit in aquatic organism is set by the ability to sustain aerobic scope to sudden temperature shifts. This study tested the genetic and plastic differences in thermotolerance of Daphnia that can be explained by the differences in the ability to retain mitochondrial integrity at high temperatures. Five genotypes with different biogeographic origins were acclimated to 18ᵒC and 25ᵒC. We developed a rhodamine 123 in-vivo assay to measure mitochondrial membrane potential and observed higher fluorescent in heat damaged tissues as the disruption of the mitochondrial membrane potential. The application of a novel xylose isolation procedure using phenyl boronic acid esterification has been limited to the hydrolyzates of low extractive biomasses like hardwoods and agricultural resides. Hydrolyzates with lower sugar and higher extractive concentrations, like softwoods, have not yielded xylose, or even xylose diester (XDE), the first intermediate product in the new process, under the same conditions. This research applied evaporation concentration, adsorbent separation and membrane filtration to these lower performing biomass hydrolyzates to maximize the yield of the XDE formation step of the new xylose extraction process. Hydrogen molybdenum oxide, known has molybdenum bronze, is a material of interest due to catalyzing electron transfer reactions. Specifically, molybdenum bronze is an electrocatalyst toward carbon dioxide reduction. Electrochemical deposition from a peroxymolybdic acid solution is a method for preparing molybdenum bronze films. This work demonstrates reproducible electrodeposition on indium tin oxide substrates and conductive carbon paper. Film thickness depends on concentration, time and pH. After characterization by film thickness, resistance, XRD and XPS, the as deposited films served as the working electrode for electrochemical reduction of carbon dioxide in 0.1 M NaHCO3. Natural caves represent one of the most difficult environments to map with modern 3D technologies. In this study I tested two relatively new methods for 3D mapping in Tipton-Haynes Cave near Johnson City, Tennessee: Structure from Motion Photogrammetry and Computer Vision using Tango, an RGB-D (Red Green Blue and Depth) technology. Many different aspects of these two methods were analyzed with respect to the needs of average cave explorers. Research investigating associations between joint attention and language development have thus far only investigated joint attention by way of visual perceptions while neglecting the potential effects of joint attention engaged through other sensory modalities. Urbanization is increasing at a dramatic rate as the human population increases. While it is well-known that urbanization tends to decrease species diversity (i.e., biotic homogenization), it is not known how urbanization affects the frequency and efficiency of species interactions. Seed dispersal is a plant-animal interaction that depends on disperser feeding rate, disperser diversity, probability of seed dispersal and germination. How these factors are affected by urbanization however is unknown. In this study, we evaluate how urbanization alters these factors. Chronic inflammation is an essential mechanism during the development of cardiovascular and metabolic diseases. The outcome of diseases depends on the balance between the migration and accumulation of macrophages in damaged tissues. Macrophage motility is highly regulated by adhesive receptors, integrins. Namely, intermediate expression of integrin supports macrophage migration, while a high integrin density inhibits it. Our studies are focused on evaluation of the contribution of related integrins αDβ2 and αMβ2 to macrophage migration and development of chronic inflammation.

Branched-chain amino acid (BCAA) supplementation may improve recovery from competitive endurance training by reducing symptoms of central and peripheral fatigue. The purpose of this dissertation was to provide evidence for the use of BCAAs as a training nutrition strategy in order to improve recovery from training and further drive adaptive processes to training by increasing readiness to train. Among major vertebrate groups, anurans are understudied with regards to their visual systems and how they function. This study sampled North American anurans representing diverse evolutionary and life histories and which likely possess visual systems adapted to meet different ecological needs. Using standard molecular techniques, sequences were obtained for four opsins—the protein component of visual pigments—expressed in anuran retinas. Amino acid sequences of the genes RH1, LWS, SWS1, and SWS2 were compared across taxa to identify variable sites, as such variation can shift the spectral sensitivity of visual pigments and thus alter dim-light and color vision. The permanently dark deep-sea, located at oceanic water depths greater than 200 m, represents the largest potential habitat space on Earth. The physicochemical conditions of the planet’s largest biome are tightly coupled to the exchange of matter and energy from terrestrial and sea-floor end-members. In fact, global ocean and climate systems are significantly impacted by deep-sea processes. Background: Left ventricular assist devices (LVAD) are operated at constant speeds (rpm), consequently, pump flow is passively determined by the pressure difference between the LV and aorta. Since the diastolic pressure gradient (~70 mmHg) is much larger than the systolic gradient (~10 mmHg), the majority of pump flow occurs during systole. This limitation results in sub-optimal LV volume unloading, LV washing, and diminished vascular pulsatility that may be associated with increased risk for clinically-significant adverse events, including stroke, bleeding, arteriovenous malformations, and aortic insufficiency. Children in foster care are confirmed victims of maltreatment who may suffer from chronic trauma. These adverse childhood experiences will often increase the risk of physical and psychological adversities such as self-harm (SH), substance abuse, anxiety, post-traumatic stress disorder (PTSD), heart disease, depression, etc. Based on previous and current research studies, youth in foster care are less likely to receive adequate treatment for their self-harm behaviors and mental health needs. The purpose of this research was to determine the effectiveness, barriers and benefits of Supplemental Instruction (SI) on Generation Z students in AGN 331. The theory used as a model for this research was the Pace’s Model of College Impress. Prior research indicated that SI improves test scores, final grades, and persistence in historically difficult courses. Correlations and ANOVA’s were performed on SAS in order to determine a relationship between the non-SI attendees to those who did attend SI. Ellis–van Creveld (EvC) syndrome is a rare autosomal recessive disorder that is characterized by short-limb dwarfism, extra fingers or toes, malformed teeth and nails, and congenital heart defects. Mutations in two genes, EVC and EVC2, which are located on chromosome 4, have been shown to be responsible for EvC syndrome. The protein products of these genes, EVC and EVC2 physically interact and together, they are tethered to the base of the primary cilium of cells.
