The human intestine encompasses a vast community of microorganisms known as the gut microbiota that play a crucial role in maintaining health. Common perturbations such as changes in the normal diet, antibiotic treatment, and changes in environmental conditions can alter the gut microbiome. This can create dysbiosis in the gut leading to disease conditions. Therefore, it becomes important to determine the forces that influence the gut microbial ecology. Kernel size is a vital milling characteristic for Avena sativa L. (oats), with larger kernel size being desirable for milling facilities. This project evaluated the effect of genotype, environment, and their genotype by environment interaction (G by E) on kernel width, length, and area for a set of 27 oat cultivars grown at multiple locations in South Dakota over three years. Findings indicated significant differences among oat varieties, and that environment had a substantial effect on kernel size. However, we did not see a strong influence of genotype by environment interaction on kernel size. The lack of impact for genotype …

Bobcat (Lynx rufus) populations experienced declines in the Midwest during the 20th century due to land conversion for agriculture and over exploitation and were practically nonexistent in areas by the 1970-80s. Populations have been recovering following changes in land-use practices and habitat improvement. Eastern South Dakota was closed to bobcat harvest in 1977 but reopened in 2012 to select counties. Bobcats are elusive, have large home ranges, and occur at low densities, making monitoring their populations difficult. A century ago, the North American grasslands and prairie ecosystems were dominated by bison. At least 30 million bison roamed the Great Plains when the first explorers arrived. By 1900, there were little over a thousand bison remained in the United States and Canada. Recovery efforts has been made since the 20th century to reestablish the herds and increase the bison population. Nest survival of upland nesting waterfowl (e.g., mallard [Anas platyrhynchos], blue-winged teal [Anas discors]) has been studied extensively in the Prairie Pothole Region (PPR) of North America. Conversely, over-water nesting diving ducks’ nest survival is poorly understood, likely because over-water nests are more challenging to find, may be less concentrated, and searching for these nests is physically demanding. Nest depredation is considered a leading cause of duck nest failure and is considered a limiting factor on upland nesting waterfowl production in the PPR. Purpose/Objectives: The objectives of the study were two-fold: 1) Determine the motivators and barriers to local food and local traditional food procurement among Food Service Directors (FSDs) of South Dakota (SD) schools serving Native American (NA) students, and 2) determine what traditional foods are desired for local procurement by the same population.
Methods: A cross-sectional survey design was used. An online survey was emailed to SD FSDs at 42 schools with high enrollments NA students. The gut development of young calves is crucial for the producer and the calf's future. Many factors can affect the development of a young calf's gastrointestinal system, and there has been little research into the fecal microbiome development of calves from 0 to 12 weeks of age. To gain further insight into this process, development of the fecal microbiome in 12 dairy calves was investigated. Bauxite, the principal ore of aluminum is a potential low-cost sorbent to treat heavy metal-contaminated water. This work studied the ability of raw bauxite to adsorb Pb2+, Cu2+ , Ni2+, and Co2+ ions in natural waters and the dependency of the sorption efficiency on the initial metal ion concentration in water, contact time, the ‘type’ of bauxite, and the effect of coexisting metals. Gram-negative unicellular myxobacteria, along with their multicellular lifestyle andbiologically active specialized metabolites, are known for the predatory interactions with Gram-negative/ Gram-positive bacteria and fungi. Although myxobacterial predation range have been exploited extensively, little is known about the prey associated molecules contributing to myxobacterial predator-prey dynamics. By employing transcriptomics and untargeted metabolomics approaches, we demonstrate two structurally distinct classes of signaling molecules from Gram-negative bacterial prey elicit significant omics responses from myxobacteria, Myxococcus xanthus and Cystobacter ferrugineus. The use of Δ9-Tetrahydrocannabinol (THC) and Δ9-Tetrahydrocannabinol-Valine-Hemisuccinate (THC-VHS; NB1111) has recently been investigated in the management of intraocular pressure (IOP) lowering activity. The current studies were undertaken to develop a THC-VHS loaded nanoemulsion (NE; THC-VHS-NE) with and without Carbopol® 940 as mucoadhesive agent (NEC; THC-VHS-NEC) that could increase the drug load and the duration of activity. Primaquine (PQ)-loaded solid lipid nanoparticles (SLN), nanostructured lipid carriers (NLC), and nano-emulsion (NE) were developed to investigate the influence of lipid excipients, surfactants, and drug loading on the physicochemical properties of lipid formulations. To investigate the drug release mechanisms from the lipid-based formulations, empirical release kinetic models such as zero-order, Higuchi, Korsmeyer-Peppas, and Hixson-Crowell were used. In an ex vivo hemolysis toxicity study, the efficacy of SLN, NLC, and NE to protect erythrocytes against PQ-induced cell lysis was investigated. The results show that all lipid formulations reduced the erythrocyte hemolysis by approximately 4.5-fold compared to the drug solution, suggesting that …

Chronic hyperglycemia is one of the main characteristics of type 2 diabetes mellitus (T2DM). With the increased prevalence of T2DM, the risk of cardiovascular complications, including heart attack or stroke, is increased. Complications linked to the vasculature are increased vascular calcification (VC) or mineral deposition within the medial layer of conducting arteries. Under calcification conditions, vascular smooth muscle cells (VSMCs) undergo an osteogenic switch to an osteoblast-like cell and begin producing bone proteins. Chronic hyperglycemia can also result in increased extracellular matrix (ECM) synthesis, accumulation, and crosslinking. This form of feeding makes these bivalves especially sensitive to conditions in their environment. Gut microbial communities (microbiomes) have been recognized as important to both host organism and ecosystem health; however, how microbiomes are organized and influenced is still unclear. Admicellar polymerization was developed in the 1980s as an economical method for modifying various surfaces. In admicellar polymerization, a low monomer to initiator (M/I) ratio was traditionally required to overcome this limitation and achieve high polymer conversion when oxygen was present in the headspace and dissolved in the reaction medium. This comparative analysis, focusing on the 150 highest-frequency phrasal verbs in the phrasal verb pedagogical list (Garnier and Schmitt, 2015), highlights the limited occurrence across English language textbooks for Japanese junior high school. The spoken and written data of Japanese learners of English (i.e., NICT-JLE and NICE) shows considerable differences in the usage patterns of phrasal verbs compared to native English speakers. Diabetic individuals have an increased risk for developing cardiovascular disease due to stiffening of the left ventricle, which is thought to occur, in part, by increased AGE/RAGE signaling. Advanced glycation end products (AGEs) accumulate within the body over time, and under hyperglycemic conditions, the formation and accumulation of AGEs is accelerated. AGEs exert their effect by binding to their receptor (RAGE) and can induce myofibroblast differentiation, oxidative stress, cell-extracellular matrix contraction, and cell migration. This study examines the effects of non-native phonotactic constraints on the perception of nasal-initial consonant clusters (NICC), specifically whether native English speakers can perceive NICC as accurately as native Swahili speakers. The ABX task was conducted over Zoom to test L1 English speakers (N=12) and L1 Swahili speakers (N=12). THIS GARDEN is an audiovisual installation that tells the story of oyster farmers and place in Spring Creek, Florida. I use the oral history interviews and documentary photography I made there to explore aquaculturists’ deep relationship with the surrounding environment. To these farmers, the bay is a garden. They work within those often-unpredictable waters to cultivate new life. This accompanying thesis builds a chronological narrative of oyster aquaculture industry along Florida’s Forgotten Coast. In this study, a novel value-added dairy-based health formulation was developed using whey protein hydrolysate and probiotic organisms. In the first part of the study, different forms of whey proteins, concentrate WPC80, isolate WPI90, and hydrolysates WPH10, WPH15 and WPH20 were screened for bioactivities (antimicrobial, antioxidant, and antihypertensive activity). Hydrolysate WPH10, exhibiting the highest bioactivities was conjugated with maltodextrin. A batch of 2L conjugated solution was spray dried in a Niro drier with an inlet and outlet temperature of 200⁰C and 90⁰C, and alternatively, freeze dried at −80 °C under 50 mTorr vacuum. School gardening has a longstanding history in American education, dating as far back as the mid-1800s. Despite fluctuations in popularity throughout the twentieth century, school gardens have grown into an established method for hands-on teaching. This paper builds upon a historical framework of school gardening in American public education with particular focus on the connections between school gardens and agricultural education. This study offers a descriptive portrait of contemporary K-12 school gardens in South Dakota. Notch signaling is an evolutionarily conserved signaling pathway involved in cell fate decisions and cell renewal of multiple systems, including skeletal and muscle homeostasis. As the multifunctional nuclear complex of Notch1 is essential for its pathological function, identifying these interacting partners represents a potentially broad range of druggable vulnerabilities. We have completed a BioID proteomics study and found a large set of nuclear proteins associated with NOTCH1, including transcriptional regulators and epigenetic modulators, of which Histone Deacetylase 1 (HDAC1) is a promising interacting partner of Notch1.
