The central element of this thesis is Grey Areas, a newly-composed dramatic piece in four movements for baritone and chamber ensemble. The work is highly personal in both its subject matter and its musical execution; it also contains a unique movement order that changes depending on the age of the baritone. Along with the written portion of this document and full score, I completed a studio recording of the entire composition as part of the project. Chronic hepatic damage can occur due to multiple causes including, alcohol, non-alcoholic steatohepatitis and chronic viral hepatitis. Liver fibrosis is a critical problem worldwide due to the extremely high incidence of alcohol associated liver damage and chronic viral hepatitis. Moreover, there is no FDA approved therapy for liver fibrosis. This thesis focuses on the intersection between sound and bodies as a way of re-envision the concept of human, using Octavia Butler’s Dawn as a case study. Specifically, this study contends that Butler’s re-envisioning is sonic, imagining the concept of self as it is understood by Jean-Luc Nancy’s idea of the “resonant subject,” in that sound embroils us within our environment. Heterocyclic amines (HCAs) are formed when cooking meats at high temperatures or for long periods of time, and their major metabolic pathway includes hepatic N‑hydroxylation by CYP1A2 followed by O-acetylation by N-acetyltransferase 2 (NAT2). NAT2 is a highly polymorphic gene involved in metabolism of drugs and xenobiotics, and its single nucleotide polymorphisms (SNPs) produce phenotypes characterized as slow, intermediate, or rapid acetylator. Recent studies have documented associations between HCAs and insulin resistance and between certain NAT2 phenotypes and insulin resistance. The purpose of this project is to investigate the less understood narrative of the Late Woodland peoples at the German site within the Lower Illinois River Valley. In order to determine the extent of Mississippian cultural interaction with the Jersey Bluff Phase, Late Woodland, peoples, I will examine the houses and artifacts found during the 2019 field season by the Centre for American Archeology. This thesis examines the media corporation PragerU as a parasite public, using the framework provided by Larson and McHendry (2019). Parasitic publics are “reactionary discursive spaces formed residual and institutionalized affectively through the invention, circulation and uptake of demagogic rhetorics” (Larson & McHendry, 2019, p. 517). The purpose of this thesis is to identify and analyze the argumentative tactics used by PragerU in the public sphere. I argue that PragerU utilizes its extensive network to resist counterpublicity and influence the dominant public sphere. Pathogens may enter surface waters as they are shed in human feces and potentially delivered to surface waters via sewer overflows, particularly in the eastern United States. This study examined of the presence of fecal indicators in two forks of Beargrass Creek in Louisville, Kentucky. Surface water grab samples (N=30), sediment samples, water quality, and a paired wastewater surveillance study were analyzed. SARS-CoV-2 was detected in a single stream sample despite ubiquitous presence of the virus within the area in wastewater and consistent evidence of a human fecal indicator. Butterfly populations are declining, and habitat degradation due to urbanization is a major contributing factor. Gardens represent a large proportion of land area in many cities, and thus may be important for conserving butterflies in urban environments. In this dissertation, I examine how garden features (ex: size, level of urbanization, plant diversity) affect adult butterfly diversity/abundance, behavior, and the predation risk faced by caterpillars. The frequency of harmful algal blooms (HABs) has been increasing across the globe, especially in river ecosystems. Although more common in lakes, more HABs have been reported in rivers in the past decade, particularly in the Ohio River. In 2019, a HAB stretched 200 to 300 miles throughout the Ohio River from September to October. Samples were collected for the duration of the 2019 bloom (August-October) in the McAlpine Pool in Louisville, KY to compare microcystis toxin (microcystin) concentrations on a temporal and spatial scale. This dissertation exposes one manifestation of the hidden curriculum of gender bias in the biological academe and explores the impacts of implicit essentialist perspectives on biology curricula and student understanding of an important evolutionary concept. Chapter one introduces the concept of gender essentialism and its relationship to the hidden curriculum of gender bias. I conclude the introductory chapter by reviewing tenets from queer curriculum theory and suggesting that applications of the theory may provide educators with the pedagogical tools required to counter implicit essentialist perspectives. Alphaviruses are positive sense, single strand, RNA viruses. These viruses occur on every populated continent. Alphaviruses are divided into two clades, the New-World and Old-World viruses. The New-World viruses include Eastern (EEEV), Western (WEEV), and Venezuelan equine encephalitis viruses (VEEV), and cause neuroinvasive disease. The Old-World viruses include Chikungunya (CHIKV) and Ross River viruses (RRV), and typically cause multijoint arthralgia. There are currently no approved antiviral therapeutics or vaccines for any alphavirus, making them a high priority for antiviral drug design and discovery. Breast cancer genetic susceptibility is modulated by high to low penetrance risk alleles. Population frequencies of high and moderate penetrant predisposing mutations are rare and responsible for a small percentage of population cases. Most genetic variation in breast cancer susceptibility is low penetrance common variants where causal genes and risk-associated mechanisms are largely unknown. Comparative genetics uses animal models to isolate and mechanistically evaluate genetic susceptibility to human disease. Rat strains have different susceptibility phenotypes to chemical-induced carcinogenesis. Shifting Sands is a re-exploration of the presentation of North Africans in colonial postcards, an examination of identity, and a critique of the modern Western museum. Since the inception of photography, colonizers used this medium- especially in the form of postcards- to categorize and exoticize Eastern peoples in order to more easily subjugate them. Competition with aggressive species is an important factor that shapes local community structure. Eavesdropping on the chemical communication systems of such behaviorally dominant species can help heterospecific species to avoid negative interactions. I review the chemical communication system and known ecological effects of the Neotropical canopy ant, Azteca chartifex/trigona (Chapter 1). Chronic arsenic exposure is a risk factor for the development of pulmonary diseases, including lung cancer, but the exact mechanism is not yet fully understood. Many previous studies have relied on acute arsenic exposure (i.e. 1-10 uM arsenic for 24 hours) to study arsenic toxicological effects. However, acute exposure does not reflect the levels or duration of arsenic associated with environmental exposure, or drinking of contaminated well water. Electroporation is widely practiced in research and clinical work for transfection of genetic sequences and drug molecule transport through the membrane barrier. However, a full theoretical explanation of the molecular mechanisms and thermodynamics responsible for pore formation, structure, and longevity does not yet exist. Alphaviruses are mosquito-borne arboviruses that have a positive sense, single-stranded RNA genome, which are capable of causing severe disease and thus pose a significant burden to public health. During alphaviral replication, significant quantities of viral genomic RNAs that lack the canonical alphaviral 5’ cap structure are produced and packaged into viral particles, despite the fact that these noncapped genomes cannot be translated and are essentially noninfectious. Voltage-gated potassium (Kv) channels control vascular resistance and facilitate the augmentation of myocardial blood flow in response to increases in cardiac workload (i.e., metabolic hyperemia). Native sarcolemmal Kv1 channels in arterial myocytes associate with a heteromeric auxiliary complex consisting of intracellular Kvβ1 and Kvβ2 proteins. The Kvβ proteins differentially regulate the function of heterologously expressed Kv channels, as well as native Kv1 channels in excitable cells of the cardiovascular and nervous systems. This dissertation explores the anticancer activity of Avaren-Fc (AvFc), a novel lectin-Fc fusion protein or “lectibody” targeting cancer and virus-associated high-mannose glycans. Previously, we have shown that AvFc recognizes a broad selection of established cancer cell lines from a wide array of tissue types, can potently induce antibody-dependent cell-mediated cytotoxicity (ADCC) against them, and exhibits anti-cancer activity in vivo. Despite the success of immunotherapy in many types of cancer, pancreatic adenocarcinoma (PDAC) has yet to benefit. Innate immune cells are critical to antitumor immunosurveillance and recent studies have revealed that these populations possess a form of memory, termed trained innate immunity, which occurs through transcriptomic, epigenetic, and metabolic reprograming. Though trained innate immunity has mostly been investigated in the context of infection, the induction of trained innate immunity could also protect against tumors, and specifically pancreatic tumors. Bacteriocins are narrow-spectrum antibiotics produced in nearly all lineages of bacteria, meaning that these antimicrobials target closely related individuals. The bacteriocins of Pseudomonas aeruginosa, called pyocins, are highly prevalent and diverse in populations of this species. Laboratory studies have shown that pyocins can function to mediate the outcome of interactions, often allowing for the coexistence of multiple strains, as no one pyocin genotype is competitively superior. This dissertation is a historical novel accompanied by a critical lyric essay. Through a combination of scholarly research and creative activity, the novel reimagines the lives of the women who lived as inmates and administrators in the Home for Friendless Women, a charity home that operated in Louisville in the late nineteenth century. The critical essay draws from my interdisciplinary background to combine personal experiences, archival and scholarly research, and literary analysis to connect the U.S. dress reform movement in the 1850s, and its failure to change public perception about what was acceptable attire for middle and upper class women, …
