The Effects Of In Vitro Fish Oil And Dietary Fish Meal Supplementation On Microdomains Within Bovine Luteal Cells, Michele Renae Plewes May 2018

The Effects Of In Vitro Fish Oil And Dietary Fish Meal Supplementation On Microdomains Within Bovine Luteal Cells, Michele Renae Plewes

Dissertations

Lipid microdomains are regions of plasma membrane rich in cholesterol and sphingolipids, ranging in size from 10–200 nm in diameter. These structures serve as platforms to facilitate co-localization of intracellular signaling proteins during agonistinduced signal transduction. The omega-3 polyunsaturated fatty acids are a distinct class of fatty acids that have been shown to incorporate into lipid microdomains, disrupting the structural integrity of domains, and possibly attenuation of cell signaling. Fish byproducts, such as fish oil or meal, are a rich source of omega-3 polyunsaturated fatty acids that can be supplemented into cell cultures and diets of breeding females as a …

Advancement Of Interpolyelectrolyte Complexes For The Delivery Of Genetic Material, Keith Hampton Parsons May 2018

Advancement Of Interpolyelectrolyte Complexes For The Delivery Of Genetic Material, Keith Hampton Parsons

Dissertations

This dissertation focuses on the development and advancement of interpolyelectrolyte complexes (IPECs) and block ionomer complexes (BICs) for the delivery of genetic material, namely RNA, to cells, both human and insect. RNA interference (RNAi) provides a powerful tool for disease treatment and the elimination of crop pests at the genetic level. Therefore, development of successful delivery vehicles for its effector molecules, small interfering and double stranded RNAs (siRNA and dsRNA), is imperative. IPECs and BICs show the most promise as RNAi vectors, and thus this work focuses on ascertaining the structure-property relationships affecting RNA delivery as well as applying such …

The Characterization Of The Transcription Factor Msab And Its Role In Staphylococcal Virulence, Justin Batte May 2018

The Characterization Of The Transcription Factor Msab And Its Role In Staphylococcal Virulence, Justin Batte

Dissertations

Staphylococcus aureus is a common human pathogen that is responsible for a wide range of infections, ranging from relative minor skin infections to life-threatening disease such as bacteremia, septicemia, and endocarditis. S. aureus possesses many different virulent factors that aid in its ability to cause this wide array of infections. One major virulence factor includes the production of capsular polysaccharide (CP). The production of CP plays a major role in the virulence response during infection specifically by providing S. aureus an antiphagocytic mechanism that allows the pathogen to evade phagocytosis during an infection. S. aureus has developed complex genetic regulatory …

Antimicrobial Materials Via Thiol-Ene Chemistry, Dahlia Amato May 2018

Antimicrobial Materials Via Thiol-Ene Chemistry, Dahlia Amato

Dissertations

With the increasing prevalence of antimicrobial resistance, the escalation of opportunistic/pathogenic infections is a looming global crisis. To avoid the pitfalls of conventional antibiotics, this dissertation focuses on developing macromolecular solutions to develop novel antimicrobial materials based on essential oils (thymol, carvacrol, and aldehydes). It is well established that essential oil derivatives exhibit high potency towards a wide range of pathogenic microbes. The rapid photopolymerization kinetics, limited by-products, and homogeneous network formation afforded by thiol-ene photopolymerization are utilized to either encapsulate essential oil derivatives or convert them into monomers which can subsequently be incorporated into new antimicrobial materials with new …

A Mechanistic Understanding Of Self-Propagating Amyloid-Β Oligomer Conformations In Alzheimer Disease, Dexter Nathanael Dean May 2018

A Mechanistic Understanding Of Self-Propagating Amyloid-Β Oligomer Conformations In Alzheimer Disease, Dexter Nathanael Dean

Dissertations

Alzheimer disease (AD) is a fatal neurodegenerative disorder characterized by the widespread deposition of proteinaceous plaques abundant in amyloid-β (Aβ) aggregates. Although the plaques mainly contain high molecular weight, insoluble Aβ fibrils, the low molecular weight soluble aggregates called oligomers have been shown as the primary toxic species responsible for synaptic dysfunction and neuronal loss in AD. The process of aggregation is nucleation-dependent, but also highly stochastic and inhomogeneous resulting in biophysically diverse assemblies. Recent advances in the field indicate a potential correlation between the phenotypic diversity observed in AD subtypes and aggregate polymorphism. Therefore, understanding the molecular mechanisms which …

Personality In California Sea Lions (Zalophus Californianus) And Harbor Seals (Phoca Vitulina): Methodological Convergence And Species-Specific Emotional Repertoires, Amber De Vere

Dissertations

Despite the wide acceptance of animal personality as a valid area of study, research on marine mammal personality remains remarkably scarce. What literature does exist predominantly focuses on bottlenose dolphins (Frick, 2016; Highfill & Kuczaj, 2007; Kuczaj, Highfill & Byerly, 2012; Lilley, de Vere, Yeatre & Kuczaj, 2018; Moreno, Highfill & Kuczaj, 2017). There is also strong evidence for individual differences in grey seals (Robinson et al., 2015; Twiss & Franklin, 2010; Twiss, Culloch & Pomeroy, 2011; Twiss, Cairns, Culloch, Richards & Pomeroy, 2012), and preliminary research has provided evidence of broad personality factors in pinniped species using behavioral coding …

Root Dynamics Of Crop Plants In A High Carbon Dioxide World: Effects Of Elevated Versus Ambient Carbon Dioxide Levels And No-Till Versus Conventional Agricultural Management, Charlotte Barker

Dissertations

Due to the continuing increases is atmospheric carbon dioxide levels and its potential effect on food sources, there is an interest in evaluating the effect of elevated CO2 concentration versus ambient CO2 concentration in agricultural crop plants although more studies have focused on the aboveground portions of plants rather than the roots. Additionally, the conservation agricultural method, no-till, has been widely suggested as a possible method of increasing soil organic carbon and increasing soil moisture in a hotter world.

This research involves two major agricultural plants, Sorghum bicolor (sorghum), and Glycine max (soybean) grown under four conditions including …

Systems Of Quantity Judgment In Various Species: A Meta-Analysis, Tiffany A. Woodard Baker May 2018

Systems Of Quantity Judgment In Various Species: A Meta-Analysis, Tiffany A. Woodard Baker

Dissertations

An abundance of behavioral and neuroimaging literature supports the presence of two cognitive systems for quantity judgments (Agrillo & Bisazza, 2014). In particular, small quantities are thought to be guided by the object-file system, a precise system that uses mental files to map onto real world objects, and large quantities by the approximate number system, an imprecise, estimation system (Dehaene, 1997). Evidence supporting both systems exists in a variety of species including nonhuman primates (Boysen & Hallberg, 2000), birds (Garland, Low, & Burns, 2012), amphibians (Uller, Jaeger, Guidry, & Martin, 2003), and fish (Agrillo, Dadda, Serena, Bisazza, 2009), but support …

Characterization Of Swine Production Using Measurements Collected Via Indoor Positioning System, Shaun Perisho May 2018

Characterization Of Swine Production Using Measurements Collected Via Indoor Positioning System, Shaun Perisho

Dissertations

Legislative and market initiatives are requiring that gestating sows move from individual housing to group settings. Little information is known about coping styles of individual sows in these more socially complex environments and thus the impact of different behavioral strategies on sow reproductive success and efficiency was investigated. The movements of 70 sows during periods of reintroduction into large pen gestational housing following insemination was measured with a commercially available system that tracked animal location and accelerations. Principle component analysis (PCA) was used to establish composite variables characterizing each animal’s behavioral response to social reintroduction and revealed the presence of …

Material Realism: A Systems Theory Of International Politics, Paul Burton May 2018

Material Realism: A Systems Theory Of International Politics, Paul Burton

Dissertations

Traditionally power has played a dominant role in all realist theories of international politics and little if any room was left for actor agency. Systemic outcomes were a function of system structure. System structure was the result of the positioning of states in the system according to their power. Conflict resulted from system structure. Actor agency was not a relevant consideration because system structure was deterministic. This paper presents a new theorization of realism that it calls material realism. Material realism hypothesizes globalization as a second independent variable alongside power. A longitudinal network model using all conflict events since 1992 …

Polymer Nanocomposites Containing High Aspect Ratio Particulates: Innovation In Co-Extruded Multilayer Barrier Films, Kevin Meyers May 2018

Polymer Nanocomposites Containing High Aspect Ratio Particulates: Innovation In Co-Extruded Multilayer Barrier Films, Kevin Meyers

Dissertations

Delaminated montmorillonite (MMT) clay/ maleic anhydride grafted LLDPE nanocomposite multilayer films with alternating layers of LDPE were produced through multilayer co-extrusion. The MMT concentration within the nanocomposite layers was increased through annealing the films in the melt due to a mismatch in interdiffusion rates of the polymer layers. Analysis of the nanocomposite layers upon annealing revealed that the platelets impinged upon one another resulting in significant improvement in oxygen barrier in the multilayer system, exceeding the results of bulk nanocomposites.

Model analysis demonstrated that increasing the nanoplatelet aspect ratio or initial concentration in the filled layers would lead to even …

Roles Of Phospholipases And Ribosomal S6 Kinase In Lipid Remodeling And Growth In Arabidopsis Response To Phosphate Deprivation, Yuan Su Apr 2018

Roles Of Phospholipases And Ribosomal S6 Kinase In Lipid Remodeling And Growth In Arabidopsis Response To Phosphate Deprivation, Yuan Su

Dissertations

Phosphate (Pi) is one of three macronutrients for plants, which is vital for plant growth and development. Understanding the mechanism by which plants respond and adapt to Pi deficiency not only unveils functions of genes and pathways involved, but also provides potential tools to manipulate crops to better stand Pi stress in low Pi-containing lands. One of the significant metabolic changes in plants under Pi starvation is the membrane lipid remodeling that converts Pi-containing lipids such as phospholipids to Pi-free lipids, such as glycolipids. To elucidate the metabolism and regulation of lipid remodeling, this dissertation characterizes the role of two …

The Ecology Of Nest Cavity Use By Arboreal Ants In The Brazilian Cerrado: Resource Availability, Nest Modification, And Trophic Interactions, Galen Priest Apr 2018

The Ecology Of Nest Cavity Use By Arboreal Ants In The Brazilian Cerrado: Resource Availability, Nest Modification, And Trophic Interactions, Galen Priest

Dissertations

Arboreal ants in the Brazilian Cerrado rely on cavities in living trees as nest sites. These cavities are created by a community of, wood-boring beetles, which act as ecosystem engineers. Despite the importance of these cavities as a resource, little is known about their natural abundance and heterogeneity, how ants use and modify them as nest sites, and how this interaction between cavities and their ant occupants influences trophic interactions on cerrado trees. Here I use natural history observations and manipulative experiments to address these questions. In the first chapter I quantified the occurrence, heterogeneity, and use of beetle-created cavities …

Dna Binding Kinetics Of Large Antiviral Hairpin Polyamides, Jacquelyn Niederschulte

Dissertations

While vaccines exist for the some of the most problematic strains of human papillomavirus (HPV), a double stranded DNA virus, there is currently no cure. HPV remains one of the most commonly sexually transmitted infections and is responsible for virtually all cervical cancers and genital warts. Natural products Distamycin A and netropsin have inspired the hairpin Nmethylpyrrole (Py)/N-methylimidazole (Im) polyamides (PAs) studied here. The larger hairpin PAs, designed to bind to sites of 10 or more DNA bp, have been shown to be effective antivirals against oncogenic HPV strains 16, 18, and 31, while smaller hairpin PAs are not. Despite …

The Ecology And Feeding Behavior Of Mosquitoes In The Galapagos Islands, Samoa S. Asigau Apr 2018

The Ecology And Feeding Behavior Of Mosquitoes In The Galapagos Islands, Samoa S. Asigau

Dissertations

Mosquitoes remain important vectors in transmitting wildlife diseases. This dissertation aims to understand the role that mosquitoes play in transmitting wildlife diseases such as avian malaria, a protozoan parasite belonging to the genus Plasmodium. Using data from wild-caught mosquitoes captured in multiple years and across multiple islands on the Galapagos Archipelago, we describe distributional patterns of mosquitoes, their range limits and assess whether there exists a disease-free refuge as occurs in Hawaii. We show that altitudinal ranges for disease transmission of avian malaria may not be bounded by a stable disease-free refuge, since mosquitoes are found at all elevations, …

Steric Effects On The Formation Of Manganese Oxide Clusters And 2-Dimensional Ammonium Formate Architectures, Carl Oberle Apr 2018

Steric Effects On The Formation Of Manganese Oxide Clusters And 2-Dimensional Ammonium Formate Architectures, Carl Oberle

Dissertations

It has been demonstrated by the Beatty group that altering the identity of the ortho-substituent of a dianiline counterion affects the assembly and dimensionality of a cadmium-chloride layer from 2-D to 0-D. This work seeks to extend this finding to metal oxide and organic hydrogen-bonded materials. By systematically increasing the ortho-substituent’s size on the benzoate building block of reported manganese oxide clusters, of formula MnxOy(O2C-R)zLw (where R = Alkyl, L = neutral monodentate ligand), we aim to impact the self-assembly of these materials relative to their parent …

Interactions And Pathogen Transmission Between Carnivores In Madagascar, Fidisoa Rasambainarivo Apr 2018

Interactions And Pathogen Transmission Between Carnivores In Madagascar, Fidisoa Rasambainarivo

Dissertations

Introduced carnivores exert considerable pressure on native predators through predation, competition and disease transmission. Improved understanding of determinant factors of interactions and pathogen transmission between introduced and endemic wildlife may help to predict disease emergence, avoid pathogen spillover and help control outbreaks. Using non-invasive camera traps, I identified areas where transmission of pathogens might happen through records of shared space-use within a protected area in Eastern Madagascar. I showed that indirect interactions between animals were more likely to occur near the research station which may constitute a disease transmission hotspot for carnivores in the landscape. Secondly, I investigated the associations …

Evolving Specialization In An Agent-Based Model Without Task-Switching Costs, Shane Meyer Apr 2018

Evolving Specialization In An Agent-Based Model Without Task-Switching Costs, Shane Meyer

Dissertations

This work examines the possibility of evolving the phenotypic specialization associated with division of labor in an agent-based model without task-switching costs. The model examines two groups competing for vital resources, where members of one group are capable of sharing resources with other agents in their group. Agents attempt to collect resources which allow them to reproduce, with more resources leading to a greater number of offspring by asexual reproduction. Four variants of the model are examined, with combinations of one or two resources and the presence of a foraging risk. The presence of the foraging risk can lead to …
