Neuroprotective Effects Of A Nicotinic Acetylcholine Receptor Agonist And Modulator In The Rodent Retina, Kazuhiro Iwamoto

The purpose of this dissertation is to assess the potential neuroprotective effect of an alpha7 nAChR agonist, PNU-282987, and modulator, PNU-120596, in an in vitro model of excitotoxicity and an in vivo model of acute glaucoma. In in vitro studies, retinas obtained from Long Evans rats were cultured using various concentrations of the PNU compounds to analyze neuroprotection against glutamate-induced excitotoxicity. After 3 days in culture, RGCs were identified using an antibody against Thy 1.1, visualized using a fluorescent dye and quantified. In culture, glutamate significantly decreased the number of RGCs. Recent estimates of the incidence and prevalence of autism spectrum disorders (ASDs) indicate substantial increases over the past 20 years (Centers for Disease Control and Prevention, 2009; Fombonne, 2009; Matson & Kozlowski, 2011; Schreibman & Koegel, 2005; Wing & Porter, 2002), in part because of the availability of significantly improved diagnostic assessment instruments (Lord & Corsello, 2005; Schreibman, 2005). Dosed Versus Prolonged Exposures: A Direct Comparison Of One-Session Treatments For Animal Phobias, Richard William Seim

It is widely accepted that for exposure-based therapies to be effective feareliciting stimuli must be presented continuously until there is a marked decrease in the client's anxiety (e.g., Eysenck, 1979; Foa & Kozak, 1986). However, an emerging body of research (cf. Seim, Waller, & Spates, 2010) suggests that a massed series of very brief exposures (< 150 sec) may be effective in the extinction of fear responses. The present study was designed to compare the efficacy and acceptability of two one-session treatments for animal phobias: one that utilized continuous, uninterrupted periods of exposure to a feared animal (Prolonged Exposures) and the other that utilized a massed series of brief (5-120 sec) exposure trials (Dosed Exposures). 24 adults (7 males, 17 females) between the ages of 18 and 57 years (M = 23.6) participated in this study. Each individual met DSM-IV criteria for a diagnosis of snake phobia or spider phobia. Participants were randomly assigned to one of two the two interventions. Both treatments required participants to gradually enter a room, approach, and eventually hold a live ball python or tarantula. Results from mixed model (between x within subjects) analyses of variance showed that the Dosed Exposure treatment performed equally well to Prolonged Exposures at decreasing behavioral avoidance, feelings of anxiety, perceptions of threat, and phobiaspecific cognitions from pre-treatment to post-treatment, and these gains were maintained at one-week follow-up. Although participants receiving Prolonged Exposures reported lower ratings of within-session anxiety, participants in the Dosed Exposure group had lower rates of treatment dropout, better compliance with procedures, and fewer safety-seeking behaviors during the treatment. These findings suggest that, contrary to popular belief, brief exposure trials can be effective in the extinction of phobic responses under certain conditions.

Trap grease offers a promising and less expensive alternative to food grade vegetable oils for biodiesel production, but its commercial marketability could be affected by its intense and offensive odor. Identification, quantitation and eventual elimination of the odorant compounds are vital challenges that need to be addressed before commercialization can be achieved. A rapid method using HS-SPME and GC/MS was developed in this study to identify and quantify the major volatile odorant compounds present in trap grease. The main odorant compounds were identified as short chain free fatty acids (SC-FFA) from C4 - C10.

Type 2 diabetes is mediated by insulin resistance, the inability of insulin to elicit a normal biological response in insulin responsive tissues. Several cellular models have been utilized to determine the mechanism of induction of insulin resistance but questions remain unanswered. One model, implicates the products of the Hexosamine Biosynthetic Pathway (HBP) in the induction of insulin resistance under hyperglycemia. The major end product of HBP, UDP-GlcNAc, is the substrate for O-GlcNAc transferase, an enzyme that catalyzes the O-GlcNAcylation of numerous proteins. Globalization reflects a shift in the workforce development paradigm. This descriptive qualitative case study was conducted at a Midwestern university with 12 student participants and three faculty members from three engineering programs. The purpose of this body of work was to explore the participants‘ perceptions of globalization, and to understand the implications of globalization on career and technical education (CTE) programs, program selection, and career choices, the training of the students enrolled in CTE programs, and the role of teaching faculty and the college in this process.

Goal-free evaluation (GFE) is the process of determining the merit of an evaluand independent of the stated or implied goals and objectives, whereas goal achievement evaluation (GAE), as the most rudimentary form of goal-based evaluation, determines merit according to the evaluand’s level of accomplishment with regard to its goals. This study examines the utility of GAE and GFE from the perspective of the evaluation’s intended users. Splenic marginal zones are architecturally organized to generate a rapid response against blood- borne antigens entering the spleen. The marginal zone is a distinct anatomical micro-environment whose main components include the marginal zone macrophages, the marginal zone B cells, the marginal zone sinus, and the metallophilic macrophages. The long-term effects of stroke often include cognitive impairments, but other than cognitive rehabilitation, which is often not fully successful, there is no intervention to treat cognitive impairments in stroke survivors. Our laboratory has previously shown that immunotherapy directed against the Nogo-A protein, which is enriched on oligodendrocytes, improves recovery of skilled forelimb sensorimotor function in adult and aged rats after an ischemic stroke lesion to the sensorimotor cortex. Furthermore, this recovery was correlated with axonal sprouting from intact pathways to denervated areas, as well as dendritic sprouting and increased dendritic spine density in the contralateral sensorimotor cortex. Blockade of the oligodendrocyte-associated inhibitor Nogo-A has shown great promise in promoting neuronal regeneration, sprouting, and plasticity, as well as functional recovery in rodent and primate models of CNS injury. This dissertation addresses a simple question: Is an anencephalic child a person? These children are born with only a brain stem, and, as such, cannot experience any type of consciousness. If personhood is understood as an articulable moral category, particularly distinct from DNA membership, reasonable evidence would be required to attribute any such moral category in these cases. That is, to claim that children who may never think or feel are persons carries a philosophical burden that extends beyond mere Homo Sapiens membership. Diabetes leads to several alterations in cardiac structure, one of which is fibrosis of the ventricular myocardium. Myocardial fibrosis is a common underlying factor in most cardiac pathologies. Osteopontin (OPN) is a small phospho-protein that has been implicated in fibrotic tissue remodeling. In the heart, the expression of OPN protein is increased after acute and chronic pathologies. Upregulation of OPN coincides with a transition to heart failure and a direct role for OPN in the progression of diabetic cardiomyopathy to heart failure has been reported.

This dissertation approaches metaphysical and metaethical questions concerning the nature of the human person, the existence and nature of God, and the nature of moral judgment through contemporary neuroscience, cognitive science, scientific moral psychology, and analytic philosophy of mind. Contrary to proposals that seek a harmonious integration of "religion and science" this dissertation argues that contemporary bio-psychological sciences give one ample reason to be skeptical of many of the metaphysical and metaethical claims embedded in religious traditions like Christianity and Buddhism. Anxiety disorders are the most prevalent psychiatric disorders in childhood (Walkup & Ginsburg, 2002; Hirshfeld-Becker & Biederman, 2002), occuring in approximately twenty percent of the population (APA, 2000; Langley Lindsey, Bergaman & Piacentini, 2002). Children and adolescents with anxiety disorders often experience many detrimental effects such as low-self esteem, issues with social and family relationships, and a decrease in overall functioning, including academic performance. Host molecules have been synthesized to bind negative, positive, and neutral molecules such as proteins and enzymes, and have been used as optical sensors, electrochemical sensors, supramolecular catalysts, and in the pharmaceutical industry as anti-cancer agents.1 The field of nanoscience has exploited guest-host interactions to create optical sensors with colloidal gold and Dip-Pen nanolithography technologies. The history of vegetarianism in the United States has long been shrouded in myth, assumption and obfuscation. Vegetarianism as a vital ideological and political movement has often been presented--even by its proponents--as a product of twentieth century modernism, reflecting a rise in ethical consumer awareness. The historical record of the nineteenth century, however, tells a very different story. The notion that dietary choices could be connected with larger social and political goals was formulated during, and changed dramatically in the nineteenth century. The present dissertation attempts a comparative analysis of both the origin of death in the creation accounts and the divine causation of death in the main flood accounts in the ancient Near Eastern (ANE) literature and the Hebrew Old Testament (OT).
