Levansucrase (LS) is an exocellular fructosyltransferase catalyzing the hydrolysis of sucrose and polymerization of levan and levan-type fructo-oligosaccharides (LFOs) that have various potentials in pharmaceutical and biological applications such as prebiotics and anticancer agents. LFOs with the chain length of 5 (LFO5 or GF4) showed inhibition activity against the SW480 colorectal cell lines up to about 83%. This research aims to evaluate the photosynthesis-related parameters in chromosome segment substitution lines (CSSL), containing drought tolerant region from DH212 in a ‘KDML105’ genetic background. Screening at seedling stage indicated that CSSL4 was more tolerant to drought stress than ‘KDML105’ with the higher maximum quantum yield of Photosystem II (Fv/Fm). At vegetative stage, CSSL4 tended to have the higher level of light-saturated net photosynthetic rate (PNmax), stomatal conductance (gs), electron transport rate (ETR) and effective quantum yield of photosystem II photochemistry (FPSII), when compared to the ‘KDML105’ after 9 days of water withholding. ผักคะน้าจีนถูกนิยมนำมาใช้เป็นส่วนผสมในอาหารเอเชียตะวันออก ผักชนิดนี้จัดอยู่ในตระกูลกะหล่ำ ซึ่งผักในตระกูลนี้มีองค์ประกอบของสารต่อต้านอนุมูลอิสระสูง โดยเฉพาะอย่างยิ่งสารกลุ่มกลูโคซิโนเลตและไอโซไธโอซัยยาเนตซึ่งเป็นสารที่พบเฉพาะในผักตระกูลกะหล่ำ การศึกษาจำนวนมากแสดงให้เห็นว่าสารสกัดจากพืชตระกูลกะหล่ำมีฤทธิ์ต้านมะเร็ง อย่างไรก็ตามผู้คนส่วนใหญ่นิยมรับประทานผักคะน้าจีนด้วยการปรุงสุก โดยในงานวิจัยนี้มีวัตถุประสงค์เพื่อประเมินฤทธิ์ของน้ำผักคะน้าจีนต่ออัตราการมีชีวิตรอดและการเพิ่มจำนวนของเซลล์เม็ดเลือดขาว เซลล์ U937 ถูกเลี้ยงด้วยน้ำผักคะน้าแบบจีนแบบสดและที่ผ่านความร้อน ซึ่งถูกเตรียมโดยวิธีการนึ่ง (0.625, 1.25, 2.5 และ 5 %v/v) เพื่อเปรียบเทียบฤทธิ์ในการต้านมะเร็ง อัตราการมีชีวิตรอดมีชีวิตและการเพิ่มจำนวนของเซลล์ถูกทดสอบด้วยการใช้สีย้อมและวิธี XTT ที่ 24, 48 และ 72 ชั่วโมง ผลการวิจัยพบว่าที่ 72 ชม. การบ่มด้วยน้ำผักคะน้าจีนสด 5 %v/v ลดอัตราการมีชีวิตรอดของเซลล์ถึง 13 ± 11.9 % ในขณะที่การรักษาด้วยน้ำผักคะน้าจีนที่ผ่านความร้อน 5 %v/v ลดอัตราการมีชีวิตรอดของเซลล์ได้ 49 ± 8.3 % จำนวนเซลล์และการทดสอบ XTT แสดงให้เห็นว่าน้ำผักคะน้าจีนทั้งแบบสดและที่ผ่านความร้อนสามารถลดการเพิ่มจำนวนเซลล์ได้ตามระดับความเข้มข้นและเวลา วัฏจักรของเซลล์และการตายของเซลล์ที่เลี้ยงร่วมกับน้ำผักคะน้าจีนถูกวิเคราะห์โดยโฟลซัยโตมิตรี พบว่ามีประชากรของเซลล์เพิ่มขึ้นที่ระยะ sub G1 และเซลล์มีการตายแบบ apoptosis เพิ่มขึ้น โดยเฉพาะอย่างยิ่งที่ความเข้มข้นสูงและระยะเวลาที่มากขึ้น เมื่อเปรียบเทียบกับตัวควบคุม นอกจากนี้การวิเคราะห์ด้วยวิธี Western blot แสดงให้เห็นว่าการเกิดอะพอพโตซิสที่ถูกเหนี่ยวนำโดยน้ำผักคะน้าจีนนั้นเกี่ยวข้องกับการเพิ่มขึ้นของโปรตีน caspase 3 โดยรวมแล้วผลการวิจัยพบว่าน้ำผักคะน้าจีนสดมีประสิทธิภาพมากกว่าน้ำผักคะน้าจีนที่ผ่านความร้อนในการต้านมะเร็งเม็ดเลือดขาว อย่างไรก็ตามผักคะน้าจีนทั้งสองรูปแบบถูกใช้เป็นอาหารเสริมที่มีฤทธิ์ต้านมะเร็งเม็ดเลือดขาวได้

Use of reusable water container on Chulalongkorn University campus, though is pro-environmental, is currently limited among the students. The present study investigated antecedents of such behavior, using the theory of planned behavior (TPB) with health consciousness as an addition to the model. One-hundred and twenty Chulalongkorn freshmen filled out a TPB-based questionnaire with a health consciousness scale. Data analyses using LISREL reveal an excellent fit for the TPB model. Specifically, attitude, perceived norms, and perceived behavioral control significantly and positively predicted intention and the behavior. Health consciousness indirectly influence intention and behavior via attitude, perceived norms, and perceived behavioral control. …

The measurement of Near infrared (NIR) spectroscopy, combined with chemometric techniques, has been widely employed for quality control in food products. This study presents a methodology to optimize the calibration models, called "universal model" of NIR spectra of primary condition (glucose solutions) and maintain the accurate prediction of secondary conditions. การปลอมปนของสารสเตียรอยด์ชนิดเดกซาเมทาโซนและเพรดนิโซโลนในผลิตภัณฑ์สมุนไพรเป็นปัญหาสำคัญที่ก่อผลเสียร้ายแรงต่อสุขภาพของผู้ใช้ ในหลาย ๆ ประเทศทั่วโลก โดยเฉพาะอย่างยิ่งในประเทศกำลังพัฒนา งานวิจัยนี้ ประสบความสำเร็จในการพัฒนาอุปกรณ์สำหรับแยกและตรวจวัดปริมาณการปลอมปนของสารสเตียรอยด์ดังกล่าว โดยใช้อุปกรณ์ตรวจวิเคราะห์ฐานกระดาษและใช้หลักการทางเคมีไฟฟ้า (electrochemical paper-based analytical device หรือ ePAD) ซึ่งประกอบด้วยกระดาษ Whatman SG81 ตลับพลาสติกจากการพิมพ์สามมิติ และขั้วไฟฟ้าชนิด screen printed electrode โดยกระดาษ Whatman SG81 ใช้สำหรับแยกสารสเตียรอยด์ชนิดเดกซาเมทาโซนและเพรดนิโซโลน โดยใช้วัฏภาคเคลื่อนที่เป็นสารละลาย 60% ethyl acetate ใน cyclohexane ซึ่งผลการศึกษาพบว่าสามารถแยกสารสเตียรอยด์ดังกล่าวออกจากสิ่งเจือปนต่าง ๆ ได้ภายใน 7 นาที สารสเตียรอยด์ที่แยกออกจากกันบนกระดาษ นำมาวัดปริมาณด้วยเทคนิคเคมีไฟฟ้าชนิด differential pulse voltammetry โดยใช้ตลับพลาสติกจากการพิมพ์สามมิติที่ถูกออกแบบทำให้สารละลายอิเล็กโทรไลต์ถูกขังอยู่บนกระดาษ ทำให้สามารถตรวจวัดสัญญาณทางเคมีไฟฟ้าได้คงที่ ซึ่งอุปกรณ์ที่พัฒนาขึ้นสามารถตรวจวัดปริมาณปลอมปนของสารสเตียรอยด์ทั้งชนิดเดกซาเมทาโซนและ เพรดนิโซโลนได้ในช่วงความเข้มข้นระหว่าง 10 ถึง 500 µg/mL โดยมีค่าสัมประสิทธิ์สหสัมพันธ์เท่ากับ 0.988 และ 0.994 ตามลำดับ มีขีดจำกัดในการตรวจวิเคราะห์เชิงคุณภาพเท่ากับ 3.59 µg/mL และ 6.00 µg/mL ตามลำดับ และขีดจำกัดในการตรวจวิเคราะห์เชิงปริมาณมีค่าเท่ากับ 11.98 µg/mL และ 20.02 µg/mL ตามลำดับ การตรวจวัดปริมาณปลอมปนของสารสเตียรอยด์ทั้งสองชนิดในสิ่งตัวอย่างผลิตภัณฑ์สมุนไพรจริงด้วยวิธีที่พัฒนาขึ้นมีค่าสอดคล้องกับผลที่ตรวจด้วยวิธี HPLC และเป็นวิธีที่ใช้งานง่าย มีต้นทุนต่ำ รวดเร็ว และมีศักยภาพนำไปใช้ตรวจหาสิ่งตัวอย่างผลิตภัณฑ์สมุนไพรที่สงสัยว่ามีการปลอมปนของสารสเตียรอยด์ ในระดับภาคสนาม

One of the important strategies that enhance lipid accumulation; precursor of biodiesel production, is stress cultivation. Asiaticoside (AS) is active herbal compound isolated from Centella asiatica. It has the potential benefit in promoting type I collagen (COL I) synthesis and osteogenic differentiation in human periodontal ligament cells. However, it has low aqueous solubility which may hamper the bioavailability. Thus, the main objective of this study was to develop the thermoresponsive in situ gels containing AS by using cyclodextrin (CD). To determine the solubility enhancement of AS, phase-solubility profiles were investigated and revealed that CDs enhanced the aqueous solubility of AS. Background: Stevens-Johnson syndrome (SJS) usually causes ocular surface disease. 35% of these patients develop chronic ocular sequelae including symblepharon, limbal stem cell deficiency, etc. The sequelae from SJS may also affect the alteration of ocular microbiome. Subjects: 20 chronic SJS patients and 20 healthy subjects were enrolled for specimen collection from inferior conjunctiva for microbiome analysis by using conventional culture and next-generation sequencing(NGS) methods Methods: Prospective, age and sex matched analytical study Results: A significant higher proportion of culture-positive specimen was demonstrated in SJS group (SJS group 60%, healthy 10%, p-value=0.001). Durian (Durio zibethinus M.), a major economic fruit, actively produces infamous sulfur-containing compounds during ripening. However, sulfur metabolism which is important in this fruit is rarely investigated. In this study, the integration of metabolomics using CE-TOF/MS and transcriptomics using de novo RNA-seq assembly was performed to reveal metabolic changes involved in sulfur metabolism and chemical composition related to fruit quality. During ripening, the expression level of genes involved in sulfur metabolism is concurred with the reduced sulfate ion content in placenta and the up-accumulation of sulfur metabolism-related metabolites in pulps. A paper-based analytical devices (PADs) provides a great benefit in medical diagnostics, especially in the developing world. The first part of this dissertation describes the effect of sample viscosity on PADs for the measurement of pH and nitrite concentration in human saliva. Preliminary study found that viscosity had a significant effect on the calibration curve of nitrite and a consistent curve could be generated after added washing buffer. Nitrite measurement in artificial saliva with viscosity 1.87-5.10 mPa-s was tested on the proposed device and also showed a good correlation with the standard spectrophotometry method (p=0.8484, paired sample t-test, n=20). L-Phenylalanine (L-Phe) is one of the most important amino acids in food and pharmaceutical industries. It is widely used as a nutritional supplement and a precursor for the synthesis of food additives. The market of L-Phe has been stimulated by increasing demand for the low-calorie sweetener, aspartame. In Escherichia coli, the key enzyme which catalyzes the first committed step of aromatic amino acid biosynthesis pathway is 3-deoxy-D-arabino-heptulosonate-7- phosphate synthase (DAHP-synthase). L-pipecolic acid (L-PA) is a non-proteinogenic amino acid, however, it is an important precursor and a key ingredient of many pharmaceutically compound syntheses such as immunosuppressants and anesthetics. The production of amino acids via fermentation of microorganisms are getting attention, one of the widely used is an engineered E. coli. Angiotensin-I converting enzyme (ACE) is a key enzyme involved in regulating blood pressure. ACE converts angiotensin-I into angiotensin-II, which results in raised blood pressure. Inhibition of ACE leads to a decreased concentration of angiotensin-II and lowering of blood pressure. Food protein-derived ACE inhibitory peptides are considered to be a safer alternative to antihypertensive drugs. The aim of this research is to investigate the mechanical properties of polydimethylsiloxane reinforced with short silk fibers for suture pad application. The effect of aspect ratio and fibers content of the polydimethylsiloxane reinforced with silk fibers on the properties was evaluated. From the results, tensile modulus and tensile strength of polydimethylsiloxane reinforced with different aspect ratio were increased with increasing aspect ratio and fibers content. The composite with the addition of silk fiber produces higher tear resistance with suture thread compared with pure polydimethylsiloxane. An in-situ forming and sensing was developed as a high-throughput screening of fluorescent chemosensors for metal ions. The 45 salicylaldimines, formed in-situ from the condensation between 5 salicylaldehydes and 9 amines, in 96 well-plates were photographed under black light along with the 5 salicylaldehydes in the absence and presence of 18 metal ions. The library of fluorescence response images of these 50 chemosensors were processed into RGB values. Vietnam, like many developing countries, is undergoing a rapid pace of population ageing but within a resource limited context. Therefore, understanding the factors that are important to later life well-being has become a key policy issue. Using data from the 2011 Vietnam National Aging Survey, this thesis aims to investigate gender differences in the direct and indirect impacts of education on subjective well-being of older Vietnamese based on four domains: happiness, life satisfaction, loneliness, and depression (N=2,789, including 1,683 females and 1,106 males). It contributes new evidence to a controversial debate in the literature on long-term impacts of migration and of remittances on the children.
