Development Of Polymerase Chain Reaction Enzyme Linked Immunosorbent Assay For Detection Of Human Papillomavirus 16 L1 Gene Methylation, Sasiprapa Liewchalermwong
Chulalongkorn University Theses and Dissertations (Chula ETD)
It has been well known that cervical cancer is caused by persistent infections with the high risk human papillomavirus (HR-HPV) especially HPV16 and 18. However, a minority of HR-HPV infected women developed cancer, therefore, triage test is necessary to particularly select some HR-HPV infected women who are at higher risk to progress to cervical cancer quickly for colposcopy examination. Since HPV16 L1 gene hypermethylation has been reported to be correlated well with high grade cervical lesions and cancer, the present study aims to develop PCR-ELISA for detection of HPV16 L1 gene methylation. Cervical cells were collected from women who referred …

The Role Of Chromatin Structure In Cellular Responses To Bacterial Cytolethal Distending Toxin In Saccharomyces Cerevisiae Model, Siriyod Denmongkholchai
Chulalongkorn University Theses and Dissertations (Chula ETD)
Cytolethal distending toxin subunit B (CdtB) is a DNase I-like bacterial genotoxin that causes DNA damage leading to cell cycle arrest and cell death. Previous reports demonstrated that CdtB has low level of DNase activity in vitro. Besides, a mutation at histone H2B phosphorylation site required for chromatin condensation during apoptosis (S10A) conferred partial resistance to CdtB in Saccharomyces cerevisiae model. Therefore, we hypothesized that chromatin structure may affect CdtB function. In this study, we identified chromatin regulators required for CdtB cytotoxicity in yeast. We found that deletions of certain components of SWR-complex (SWR1, SWC2, SWC5, SWC6, and ARP6), INO80-complex …

Bioengineered Regenerative Therapy For Diabetes Mellitus: Establishment Of Canine Mesenchymal Stem Cell-Derived Insulin Producing Cells, Watchareewan Rodprasert
Chulalongkorn University Theses and Dissertations (Chula ETD)
Trend of regenerative therapy for diabetes in human and veterinary practice has conceptually been proven according to Edmonton protocol and animal models. Establishing an alternative insulin-producing cell (IPC) resource is a challenge task for further clinical application. In this study, IPC generation from two practical canine mesenchymal stem cells (cMSCs), canine bone marrow-derived MSCs (cBM-MSCs) and canine adipose-derived MSCs (cAD-MSCs), was of interest. The results illustrated that cBM-MSCs and cAD-MSCs contained distinct pancreatic differentiation potential and required the tailor-made induction protocols. Generation of functional cBM-MSC-derived IPCs needed an integration of genetic and microenvironment manipulation using hanging-drop culture of PDX-1-transfected cBM-MSCs …

Chulalongkorn University Theses and Dissertations (Chula ETD)
Early childhood caries is a common oral disease found in children which could affect their general health. Streptococcus mutans is an important cariogenic pathogen since it possesses cariogenic properties, which are acid production, acid tolerance and biofilm formation. Recent studies showed that Candida albicans, which is frequently found together with S. mutans in plaques of children with dental caries, enhances cariogenicity. At the same time, increasing prevalence of non-albicans Candida species (NACs) in children and immune-compromised patients has been reported. Therefore, this study aims to investigate the effects of NACs when co-cultured with S. mutans on cariogenic properties and virulence …

Chulalongkorn University Theses and Dissertations (Chula ETD)
Atopic dermatitis (AD) is an inflammatory skin disease characterized by chronic and recurrent eczematous rash. AD occurs in both children and adults, but children are more affected. An important aggravating factor is caused by Staphylococcus aureus (S. aureus) infection. Therefore, topical (beta-lactam, macrolides, clindamycin) and systemic (mupirocin and fusidic acid) antibiotics are essential for treatment of AD. However, frequent use of antibiotics results in an increase in the incidence of antibiotic-resistant S. aureus. Methicillin resistance S. aureus (MRSA) becomes a problem in the treatment of skin diseases. It’s found that the change from penicillin binding protein (PBP) to PBP2a with …

Elastic Half-Plane Under Surface Excitations With Consideration Of Couple Stress And Surface Stress Effects, Toan Minh Le
Chulalongkorn University Theses and Dissertations (Chula ETD)
This thesis presents the modeling and analysis of an elastic half-plane under the surface loading and the frictionless contact of a rigid indentor by taking the influence of both surface and couple stresses into account. The corresponding boundary value problem is formulated by adopting the surface elasticity and couple stress theories. The method of Fourier integral transform is employed together with the boundary conditions on the top surface to derive the complete elastic field of the half-plane subjected to arbitrary surface loadings. The proposed solution scheme is also applied to construct a set of fundamental solutions associated with a normal …

Effect Of Chemical Cleaning Regimes On Ageing Of Microfiltration (Mf) And Ultrafiltration (Uf) Hollow-Fiber Membranes, Mariny Chheang
Chulalongkorn University Theses and Dissertations (Chula ETD)
Membrane fouling is a major obstacle in microfiltration (MF) and ultrafiltration (UF) in wastewater treatment and water reuse. Fouling and membrane cleaning have been extensively studied in batch experiments at a short filtration time, but long-term membrane property changes (e.g. ageing) are not fully explored. Thus, this dissertation focused on changes in physical and chemical properties of the PVDF membranes, and membrane ageing. Impacts of feed water composition (tannic acid and tannic acid+Fe2O3) and chemical cleaning regimes (NaOH, citric acid) on the membrane performance and property changes were investigated. UF and MF membranes rejected iron oxide particles perfectly. Whilst both …
Glycosylation Of 2-Nitro-1-Thioglycoside As Glycosyl Donor, Panachai Wongsrisupphakul
Glycosylation Of 2-Nitro-1-Thioglycoside As Glycosyl Donor, Panachai Wongsrisupphakul
Chulalongkorn University Theses and Dissertations (Chula ETD)
2-Nitro thioglycosides are versatile building blocks for amino sugar synthesis. Nitro moiety at C2 position acts as stable surrogate for amino group and can be converted at late state of the synthesis. In this work, chemical glycosylation of 2-nitro thioglycoside donors were investigated. 2-nitro thioglycosides were prepared from tri-O-acetyl-D-galactal which underwent functional group manipulations and nitration to obtain 2-nitrogalactal in 47% over 4 steps. Then, Michael additions of 2-nitrogalactal with various type of S-nucleophiles were performed by using NaDTC as activator to produce 2-nitrothioglycoside donors in 46–85% yield. Glycosylation between 2-nitro thioglycoside donors and methyl 2,3,4-tri-O-benzyl-α-D-glucopyranoside as glycosyl acceptor were …
Preparaton Of Selenium Nanoparticles Stabilized By Pullulan And Derivatives, Punnida Nonsuwan
Preparaton Of Selenium Nanoparticles Stabilized By Pullulan And Derivatives, Punnida Nonsuwan
Chulalongkorn University Theses and Dissertations (Chula ETD)
The inorganic/organic selenium nanoparticles (SeNPs)/pullulan derivative hybrid material was obtained using a simple and green strategy. The chemical structure of pullulan, folic acid decorated cationic pullulan (FA-CP) was designed for stabilizing SeNPs. The SeNPs stabilized by FA-CP were observed after the addition of a cysteine hydrochloride solution into the solution mixture of Na2SeO3 and FA-CP. The results suggested that the concentrations of cysteine and stabilizer were the key factors to control the shape and morphology of SeNPs. The spherical SeNP/FA-CP was found in ratio of Na2SeO3 to cysteine was 1:1. It exhibited low toxicity against normal cells while higher toxicity …
