Organizational Support, Perceived Social Support, And Intent To Turnover Among Psychiatric Nurses: A Mixed Methods Study., Beverly Jean Myers
Organizational Support, Perceived Social Support, And Intent To Turnover Among Psychiatric Nurses: A Mixed Methods Study., Beverly Jean Myers
All ETDs from UAB
Nurse turnover compromises the quality of patient care and leads to an increase in organizational costs. Nurse turnover is defined as the intent to leave one's current job position either to transfer to another unit within the organization or to terminate employment with the organization. Few studies have examined the role social support received at work as well as at home, personal characteristics and work factors play in guiding psychiatric nurses' decisions to leave a current nursing position (turnover). The overall purpose of this mixed-methods sequential explanatory study was to explore factors that contribute to psychiatric nurses' intent to turnover. …

Transition metal doped II-VI semiconductor materials features excellent spectroscopic and lasing characteristics in the middle infrared (mid-IR). Among this class of laser material, the most impressive results have been obtained from chromium-doped zinc selenide (Cr2+:ZnSe) crystals. To date, Cr2+:ZnSe materials are arguably the best choice of laser gain material for the advancements of high power mid-IR lasers broadly tunable over the 2-3 μm spectral range. The dissertation objectives are to demonstrate technological advancements in the development Cr2+:ZnSe thin films by pulsed laser deposition (PLD) thus establishing a viable route for the synthesis of mid-IR laser gain media based on Cr2+:ZnSe …
Contributions Of The Atrial Natriuretic Peptide Gene System To The Relationship Between Pediatric Body Fat, Free Fatty Acids, And Blood Pressure, Amanda Willig
All ETDs from UAB
Elevated blood pressure and nonesterified fatty acids (NEFA) in children can lead to increased chronic disease risk during adulthood. The atrial natriuretic peptide precursor A (NPPA) and natiuretic peptide receptor A (NPR1) genes encode for atrial natriuretic peptide (ANP) and its cell receptor, respectively, and may influence levels of blood pressure and NEFA. Additionally, it is unclear how body composition, physical activity, physical fitness, and racial genetic ancestry influence pediatric NEFA and blood pressure. In order to understand the associations among these factors, five investigations were performed. For experimental aim 1, the association between blood pressure and body composition measures …

Small Molecule Inhibitors Of Acid Sensing Ion Channel-1, Yawar J. Qadri
All ETDs from UAB
Acid Sensing Ion Channel 1 is one of the many proteins in the Epithelial Sodium Channel/Degenerin family. The proteins in this family interact to form cation channels with unique biophysical properties and can all be inhibited by the small molecule amiloride. Their expression in many different cell types underlies their involvement in a large variety of physiological and pathophysiological processes. ASIC-1 containing channels, specifically, are an important therapeutic target for ischemic stroke, nociception, the invasiveness of glioblastoma cells, and many other processes including anxiety and memory formation. Of the members of the ENaC/Deg family, chicken ASIC-1 is the only protein …
Parental Separation Anxiety And Shared Responsibility For Diabetes-Related Tasks In Adolescents Diagnosed With Type 1 Diabetes Mellitus, Kathryn Anne Sheppard
Parental Separation Anxiety And Shared Responsibility For Diabetes-Related Tasks In Adolescents Diagnosed With Type 1 Diabetes Mellitus, Kathryn Anne Sheppard
All ETDs from UAB
Type 1 diabetes mellitus (T1D) is most commonly diagnosed in children and young adults, and complications may be life-long as well as life threatening. Management of T1D has proven to be very challenging during adolescence, and parental involvement in diabetes management has been shown to improve adherence and glycemic control in children and adolescents. Middle adolescents should become increasingly responsible for diabetes-related tasks under the supervision and guidance of parents (shared management). However, adolescents spend less time with their family as they get older, resulting in parents' being unable to validate effective self-management; this inability may lead to anxiety about …
The Comparison Of Mitochondrial Functions Of 2 Drosophila Species: Drosophila Ananassae And Drosophila Subobscura, Hye J. Shin
The Comparison Of Mitochondrial Functions Of 2 Drosophila Species: Drosophila Ananassae And Drosophila Subobscura, Hye J. Shin
All ETDs from UAB
Recently, it has been suggested that differences in mitochondrial function enabled Homo sapiens to migrate out of Africa by enabling humans to adapt to colder environments as they migrated to higher latitude areas. Through mitochondrial mutations that partially uncoupled oxidative phosphorylation, humans that moved to colder areas were able to generate more heat for better survival, while producing less energy. However, due to challenges in working with human populations, no specific studies have been able to show the existence of such differences in mitochondrial function within healthy humans. Because of numerous advantages of Drosophila as a developmental model, such as …

All ETDs from UAB
The template for transcription and RNA replication for vesicular stomatitis virus (VSV) and other negative-strand RNA viruses is a ribonucleoprotein (RNP) complex consisting of the viral RNA genome and associated nucleocapsid (N) protein. The structure of the RNP-like complex of VSV showed that the RNA is sequestered between two lobes of the N protein, and adjacent N monomers are linked by an extensive network of interactions. Long-range interactions mediated by the N-terminal arm and C-terminal loop of the N protein stabilize the side-to-side contacts between adjacent N monomers, and are required for RNA encapsidation. It is unclear how the polymerase …

All ETDs from UAB
O-GlcNAcylation is an abundant and dynamic post-translational modification on serine and threonine residues of nuclear and cytoplasmic proteins. O-GlcNAc Transferase (OGT) and Nuclear Cytoplasmic O-GlcNAcase and Acetyltransferase (NCOAT) are the only two enzymes and major regulators in this process. In the past two decades, many studies have demonstrated its important modulation roles in nutrient sensing, protein turnover, cell cycle progression, transcription, translation as well as stress resistance. Among these biological functions, the understanding of O-GlcNAcylation dependent oxidative stress is relatively unknown. This study aims to examine the effects of O-GlcNAcylation on JNK1 and FOXO4 transcription factors under acute oxidative stress …

Role Of Epithelial Sodium Channel And Acid Sensing Ion Channel In Glioblastoma Multiforme, Niren Kapoor
All ETDs from UAB
Glioblastoma Multifrome is the most common and aggressive of the primary brain tumors. These tumors express multiple members of the Epithelial Sodium Channel (ENaC)/Degenerin (Deg) family, associated with a basally active amiloride sensitive cation current. We hypothesize that this glioma current is mediated by a hybrid channel composed of a mixture of Epithelial Sodium Channel (ENaC) and Acid Sensing Ion Channel (ASIC) subunits. To test this hypothesis we made dominant negative cDNAs for ASIC1, αENaC, and γENaC. D54-MG glioma cells transfected with the dominant negative constructs for ASIC1, αENaC, or γENaC showed reduced protein expression for each of the specific …

All ETDs from UAB
Cell culture models of oncogenesis that use cellular reprogramming to generate a neoplastic cell from a normal cell provide one of the few opportunities to study the early stages of breast cancer development. Human mammary epithelial cells (HMECs) were induced to undergo a neoplastic transformation using defined genetic elements to generate transformed HMECs (THMECs). We conducted proteomic and transcriptomic analysis of three points in the neoplastic progression of breast cancer. First we monitored alterations in the proteome that revealed a progressive trend towards a metastatic state. Next, we identified a set of genes from transcriptomic analysis that displayed this same …

Transcriptional Regulation Of The Human Heme Oxygenase-1 Via Chromatin Looping In Renal Cells, Junghyun Kim
Transcriptional Regulation Of The Human Heme Oxygenase-1 Via Chromatin Looping In Renal Cells, Junghyun Kim
All ETDs from UAB
Heme oxygenase-1 (HO-1) is a critical enzyme catalyzing the degradation of heme and generating carbon monoxide, iron, and biliverdin. In addition to heme degradation, HO-1 expression is known to protect against various cellular insults and disease states including acute kidney injury, atherosclerosis, vascular restenosis, and others. Human HO-1 gene expression is tightly regulated at the level of transcription. The main goal of this study is to explore the transcriptional regulation of the human HO-1 gene in renal epithelial cells. Chromosome Conformation Capture (3C) demonstrates that multiple regulatory regions within the HO-1 promoter and enhancer regions are physically interacting with each …
Chronic Alcohol Consumption Promotes Opening Of The Mitochondrial Permeability Transition Pore And Increases Mitochondrial Injury In Liver, Adrienne Lester King
Chronic Alcohol Consumption Promotes Opening Of The Mitochondrial Permeability Transition Pore And Increases Mitochondrial Injury In Liver, Adrienne Lester King
All ETDs from UAB
Alcoholic liver disease is a serious public health concern. In particular, the mitochondrion is a specific target of ethanol toxicity and much of the damage can be related to unregulated Ca2+ homeostasis and oxidative stress which are key players in the induction of the mitochondrial permeability transition pore (MPTP) within the organelle. The mechanism behind the induction of the MPTP remains elusive. Therefore, this body of work will provide insight on what effects chronic alcohol consumption has on mitochondrial dysfunction with an emphasis on the MPTP. Chapter 2: Assessment of mitochondrial dysfunction arising from treatment with hepatotoxicants provides a description …

The Role Of Gli1 In Eralpha-Negative Breast Cancer: Promoting Survival, Migration, Invasion, And Metastasis, Yeon-Jin Kwon
All ETDs from UAB
Glioma-associated oncogene homolog 1 (Gli1) is a well-known oncogene and a transcription factor that mediates several signaling pathways important for tumor progression, such as hedgehog, TGFß, Ras, and EGFR. Although Gli1 is known to play an important role in cancers of brain, skin, prostate, and the pancreas, the role of Gli1 in breast cancer was not previously well-defined. Therefore, this dissertation focuses on defining the role of Gli1 and the mechanism underlying Gli1-mediated transcription in breast cancer. Interestingly, the major findings of the dissertation clearly indicate that Gli1 promotes cell survival and is predictive of a poor outcome in Estrogen …

Role Of Heme Oxygenase-1 In Acute Kidney Injury, Subhashini Bolisetty
All ETDs from UAB
Acute kidney injury (AKI), defined as the rapid loss of kidney function, is often seen in the setting of multiple organ failure in critically ill patients. Lack of established therapeutic approaches to overcome AKI has lead to unacceptably high incidence of morbidity and mortality in these patients. The molecular mechanisms that lead to AKI often have oxidative stress as a common pathogenic event. The kidney responds by prompt induction of its own anti-oxidant machinery including the highly inducible, anti-inflammatory and anti-apoptotic gene-heme oxygenase-1 (HO-1). This microsomal enzyme degrades pro-oxidant heme, which is released from heme proteins. The cytoprotective properties of …

Evolution Of Pneumococcal Serogroup 6, Preston Eugene Bratcher
Evolution Of Pneumococcal Serogroup 6, Preston Eugene Bratcher
All ETDs from UAB
Serogroup 6 of Streptococcus pneumoniae has been known to contain three serotypes, named 6A, 6B and 6C, with highly homologous capsule gene loci. The 6A and 6B capsule gene loci consistently differ from each other by only one nucleotide in the wciP gene. The 6A capsule gene locus has a galactosyltransferase (wciNα), which has been replaced with a glucosyltransferase (wciNß) in the 6C capsule gene locus. We considered that a new serotype, 6D, would be possible if the glucosyltransferase found in 6C strains was combined with the wciP gene from a 6B strain. We demonstrate that this gene combination yields …

It is estimated that 15 million migrants displaced by development projects every year in the world. However, research on health impacts of project-induced displacement is insufficient. The existing literature concerning the causal effect of migration on health is inconclusive due to the selective nature of migration and other methodological problems. This study employs the stress process model as the major theoretical framework, components from Cernea's IRR model as well as Scudder and Colson's framework are also incorporated to examine the short-term health impact of the displacement resulting from China's Three Gorges Dam Project. I also explore whether the displacement's effect …
