Pharmacology and Toxicology Faculty Publications

Backgrounds/aims: Mesenchymal stromal cell-derived exosomes (MSC-EXs) could exert protective effects on recipient cells by transferring the contained microRNAs (miRs), and miR-132-3p is one of angiogenic miRs. However, whether the combination of MSC-EXs and miR-132-3p has better effects in ischemic cerebrovascular disease remains unknown. Methods: Mouse MSCs transfected with scrambler control or miR-132-3p mimics were used to generate MSC-EXs and miR-132-3p-overexpressed MSC-EXs (MSC-EXsmiR-132-3p). The effects of EXs on hypoxia/reoxygenation (H/R)-injured ECs in ROS generation, apoptosis, and barrier function were analyzed. The levels of RASA1, Ras, phosphorylations of PI3K, Akt and endothelial nitric oxide synthesis (eNOS), and tight junction proteins (Claudin-5 and …

Depressed Neuromuscular Transmission Causes Weakness In Mice Lacking Bk Potassium Channels, Xueyong Wang, Steven R.A. Burke, Robert J. Talmadge, Andrew A. Voss, Mark M. Rich

Neuroscience, Cell Biology & Physiology Faculty Publications

Mice lacking functional large-conductance voltage- and Ca2+-activated K+ channels (BK channels) are viable but have motor deficits including ataxia and weakness. The cause of weakness is unknown. In this study, we discovered, in vivo, that skeletal muscle in mice lacking BK channels (BK−/−) was weak in response to nerve stimulation but not to direct muscle stimulation, suggesting a failure of neuromuscular transmission. Voltage-clamp studies of the BK−/− neuromuscular junction (NMJ) revealed a reduction in evoked endplate current amplitude and the frequency of spontaneous vesicle release compared with WT littermates. Responses to 50-Hz stimulation indicated a reduced probability of vesicle release …

Potential For Wetlands To Remediate Harmful Pathogenic Fecal Coliform Bacteria From Streams, C. Ewing, Benjamin Strang, Bradley Axe, Jocelyn Birt, Brayden Kinney, Zachary Senger, Stephen J. Jacquemin

Lake Campus Research Symposium Abstracts and Posters

Wetlands are increasingly becoming a cornerstone of stream remediation in the highly eutrophic regions of the Midwestern United States. Wetlands have numerous advantages over other technologies as they incorporate natural biological process resultant from plants and bacteria while also providing an increase in wildlife habitat and greenspaces rather than relying on costly and technologically complex processes to treat waterways. The capacity for wetlands to remediate nutrients and improve water clarity is fairly well established. However, less is known about their potential to affect changes in the pathogenic microbial communities (such as E. coli) commonly associated with runoff in agricultural areas …

Nutrient Removal Potential Of Constructed Wetlands In Grand Lake St. Marys Watershed, Stephen J. Jacquemin, Jocelyn Birt, Benjamin Strang, Conner Ewing, Bradley Axe, T. Dirksen

Lake Campus Research Symposium Abstracts and Posters

Constructed wetlands are becoming an increasingly important management tool to reduce nutrient rich agricultural runoff in the Great Lakes region. The objective of this study was to assess the removal efficiency of two constructed wetlands operating on tributaries of Grand Lake St. Marys (Prairie Creek and Coldwater Creek) located in northwest Ohio. Water samples were collected weekly during 2019 year from inflow and outflow points where they were analyzed for nutrient (nitrate-N, total phosphorus, dissolved reactive phosphorus) concentrations following standard EPA colorimetric methods. Overall, while both wetlands experienced high mean nutrient inputs (concentrations in mg/L) across both fall and summer …

Continuous Water Quality Monitoring Platform For Grand Lake St Marys, Aaron Neikamp, Alex Lehman, Brandon Siefring, Jason Evers, Ryan M. Spicer, Shayna R. Petitjean

Lake Campus Research Symposium Abstracts and Posters

For the past decade, Grand Lake St. Marys (GLSM) has struggled to provide a stable and clean water source for the community affecting people and businesses alike. A safe level of microcystin –a toxin in the harmful algal blooms–is 20 ppb in recreational water, and GLSM has seen an excess of 82 ppb. As of now, there is no solution to continuously monitor the water quality; therefore, corrective actions are only based off intermittent samples taken by hand. A solution to this issue would be a water quality platform (WQP) that monitors parameters such as water and air temperature, conductivity, …

Productivity And Automation In Automotive Manufacturing: The Impact On Ohio Employment, James Olderham

Student Papers in Local and Global Regional Economies

The supposed decline of the United States manufacturing industry has been a central topic of political concerns and research for decades. The classic story is that America’s might in this sector was seen as the backbone of the economy and a central factor in the nation’s economic dominance relative to the rest of the world. In light of this, it’s easy to see how a decline in the manufacturing base has become such an important issue. In recent years, two main theories have emerged as to what is to blame for the loss American manufacturing jobs over the last two …

Distributed Manufacturing In Dayton: Combating Covid-19 Through Public Service Employment, Joshua Pham, Grayson Thacker Apr 2020

Distributed Manufacturing In Dayton: Combating Covid-19 Through Public Service Employment, Joshua Pham, Grayson Thacker

Student Papers in Local and Global Regional Economies

With the decline in manufacturing in the United States following the 1980s, many once thriving cities began to fall into economic stagnation and decline. Dayton, Ohio is one of these cities, experiencing a mass exodus of its central industry, leaving behind a labor force with mismatched skills for the current job environment and a lack of opportunity. Such unemployment has only been exacerbated by the Great Recession and the more recent economic shutdown, stemming from the COVID-19 pandemic. To combat this decline and slow march to destitution, this paper proposes the implementation of a public service employment program (PSE) that …

Identifying Easy Indicators Of Dementia, Swati Padhee, Tanvi Banerjee, Valerie L. Shalin, Krishnaprasad Thirunarayan

Celebration of Undergraduate & Graduate Research, Scholarship, and Creative Activities Materials

Alzheimer’s Disease (AD) is a degenerative chronic neurodegenerative disease that affects millions of people and whose care costs billions of dollars. There is growing evidence that variations in speech and language may be early indicators of dementia. One of the most initial symptoms of dementia is speech impairment, including difficulty in finding words and changes to the grammatical structure. These early indicators can be detected by having the patients perform a picture description task, such as the Cookie Theft task from the Boston Diagnostic Aphasia Examination. However, much of the state-of-the-art NLP for dementia has been limited due to the …

Multi-Label Model For Toxicity Prediction, Xiu Huan Yap, Michael L. Raymer Apr 2020

Multi-Label Model For Toxicity Prediction, Xiu Huan Yap, Michael L. Raymer

Celebration of Undergraduate & Graduate Research, Scholarship, and Creative Activities Materials

Most computational predictive models are specifically trained for a single toxicity endpoint. Since more than 1300 toxicity assays have been reported in the TOXCAST dashboard, achieving high coverage over this growing number of toxicity endpoints remains challenging. Furthermore, single-endpoint models lack the ability to learn dependencies between endpoints, such as those targeting similar biological pathways, which may be used to boost model performance. In this study, we characterize the performance of 3 multi-label classification (MLC) models, namely Classifier Chains (CC), Label Powersets (LP) and Stacking (SBR), on Tox21 challenge data. These MLC models employ the Problem Transformation approach, which is …

Measuring Nomophobia And Exploration Of Consequences And Comorbidities, Sarah Marie Fryman, William L. Romine

Celebration of Undergraduate & Graduate Research, Scholarship, and Creative Activities Materials

Excessive use of smartphones has coined the term “Nomophobia”, or fear of not being able to use your smartphone. For many, these devices have become an extension of ourselves, which raises hesitation on whether or not society has become addicted to smartphones. Specific diagnostic criteria for smartphone addiction have yet to be settled, and even appropriate to use the word “addiction” when describing excessive usage of smartphones is controversial.

We therefore explore utilize current measures to explore the symptoms of nomophobia and their hierarchy, as well as comorbidities including social anxiety, self-esteem, distracted driving and sleep quality. A total of …

Predicting Alcohol Withdrawal In Intensive Care Units, Reza Sadeghi, Tanvi Banerjee, William L. Romine

Celebration of Undergraduate & Graduate Research, Scholarship, and Creative Activities Materials

Alcohol use disorder is a common health issue in older adults who are facing depression caused by retirement, loss of a spouse, pain, and sleep problems. The prolonged, heavy alcohol ingestion will lead to high alcohol dependency such that cessation or reduction of using alcohol causes alcohol withdrawal syndrome (AWS) in roughly 4 to 72 hours after the last drink. During the initial 8 hours, patients face anxiety, insomnia, nausea, and abdominal pain. This condition is followed by high blood pressure, increased body temperature, unusual heart rate, and confusion. If this syndrome does not receive any treatment, the patients will …
