Il-10 And Tgf-Beta Increase Connexin-43 Expression And Membrane Potential Of Hl-1 Cardiomyocytes Coupled With Raw 264.7 Macrophages, Cora B. Cox
Browse all Theses and Dissertations
Cardiomyocytes and macrophages have been found to interact via connexin-43 hemichannels. The role of connexin-43, however, is not fully understood. This study shows that these interactions aid in increasing the membrane potential of cardiomyocytes allowing contraction of the cells. HL-1 cardiomyocytes and RAW 264.7 macrophages in coculture increased expression of connexin-43 compared to cardiomyocytes alone. Co-cultures also increased the fluorescence of Di-8-ANEPPS potentiometric dye indicating an increase in cardiomyocyte membrane potential. Treatment with IL-10 and TGF-beta further increased connexin-43 expression and membrane potential. Treatment with SOCS3 inhibited the effects of TGF-beta and IL-10 while having no effect on its own. …

Co-infection is common among viruses and bacteria in the human respiratory system. Adenovirus (AdV) and Streptococcus pneumoniae are clinically relevant respiratory pathogens that cause morbidity and mortality in a variety of patient populations with the highest morbidity occurring among immunocompromised individuals, but also prevalent in infants and the elderly. Acute respiratory distress syndrome may become severe in healthy individuals when co-infection with S. pneumoniae and AdV occurs due to synergistic effects of the pathogens on the host. I hypothesized that S. pneumoniae infection decreases AdV transduction of airway epithelia. To test this hypothesis, we utilized the polarized immortalized airway epithelial …

Journal of Bioresource Management
Water shortage is one of the major limiting factors of crop productivity worldwide. This study was planned to explore the impact of Capparis decidua stem extract on wheat varieties (FSD-2008, 12066 and 13348) under water stress condition. Water deficit condition negatively impacted the seedling germination, growth, photosynthetic pigments and ions uptake capacity. Water limited condition significantly (p≤0.05) increased time to attain 50% germination, lipid peroxidation, H2O2 and enhanced the antioxidant mechanism to overcome the oxidative stress. Different concentrations of C. decidua extract i.e. Not Soaking (NS), 10%, 20%, 30% and 40% were used for seed …

Dermatobia hominis is one of many species of botflies whose larvae is parasitic to humans and is uniquely indigenous to Central and South America. The larvae of the human botfly gain nutrients to survive by feeding underneath mammal skin. As a consequence, within 24 hours, small 2- 3mm painful, erythematous, draining, and purulent papules with a classical air-pore develop as the initial cutaneous sign. At first, the lesions resemble that of an insect bite, but over time they enlarge to 10-35mm in diameter with a surrounding area of induration. The most common areas where the larvae are cutaneously deposited include …

Due to the lack of anthrax attacks in recent times, researchers have used naturally occurring events to assess their anthrax detection models, but these provide little information on how the models will perform in the context of an unannounced, intentional release of a bioterrorism agent, like anthrax. Therefore, it is important to develop a detection model using data surrounding real anthrax scares and events. We develop a methodology to detect an anthrax-related event on Twitter. We describe a process to separate the tweets concerning anthrax-related events from those not related so experts can address misconceptions and fears in real-time.Most tweets were …

Enhanced Expression Of Receptor Tyrosine Kinase Mer (Mertk) On Socs3-Treated Polarized Raw 264.7 Anti-Inflammatory M2c Macrophages, Sankhadip Bhadra
Celebration of Undergraduate & Graduate Research, Scholarship, and Creative Activities Materials
Macrophages are phagocytic cells located in tissues, organs and even circulated within our body as white blood cells. Based on the local cytokine milieu in tissue sites, macrophages may be polarized into pro-inflammatory M1 or anti-inflammatory M2 phenotypes. Receptor tyrosine kinase Mer (MERTK) helps in clearing dead neutrophils and other apoptotic cells from damaged tissue sites preventing chronic inflammation and autoimmune disorders. MERTK aids in the maintenance of tissue homeostasis and wound healing. Phosphatidylserine (PtdSer) present on the surface of apoptotic cells release “eat me” signals which are recognized by the two “bridging ligands” of MERTK receptor, Gas6 and ProS. …

The duty to report certain conditions to public health or law enforcement authorities is one that falls on all physicians and other health care workers as part of their duty to protect the public from harm. In an open society, others, such as teachers, clergy, police officers, or simply neighbors, share the responsibility of protecting individuals at risk, often by reporting them to authorities. The emergency physician and others in the emergency department are uniquely positioned to identify people at risk or who pose a risk, and to report them as required or allowed under the law. In some circumstances, …

Aryl hydrocarbon receptor (AhR) mediates the immunosuppressive effects of 2,3,7,8 -tetrachlorodibenzodioxin (TCDD) in murine B cells. The effects of AhR activation on the regulation of expression of human immunoglobulin isotypes (μ, γ1-4, α1-2 and ε) and Ig secretion is unclear. Our previous results using CL-01 cell-line originating from a Burkitt’s lymphoma patient, demonstrated an inhibitory effect of TCDD on IgG expression but a surprising and marked loss of IgG secretion when the AhR was knocked out by siRNA or CRISPR/Cas9 gene editing. To determine if the AhR is a critical mediator of IgG expression, current study is focused on characterizing …

A Discrete Age Structured Model Of Hantavirus In A Rodent Reservoir In Paraguay, Megan A. Rúa
Biological Sciences Faculty Publications
Many rodent-borne hantaviruses are zoonotic pathogens that can cause disease in humans through inhalation of aerosolized rodent excreta. To evaluate the prevalence of Jaborâ virus (JABV) over time within its rodent reservoir, Akodon montensis, we formulated a mathematical model with multiple rodent age classes and unique infection class progression features. We then parameterized the model with data collected from a survey of JABV harbored by Akodon montensis in the Mbaracayú Reserve in Paraguay. Our model incorporates three types of infection over the lifetime of the rodent as well as a recovered class. A new feature of the model allows transition …

The goal of this investigation is to analyze data that influences premature death rates in Virginian counties. Specifically, I am looking at how the rates compare between rural and urban Virginian counties in 2020. In addition, I am analyzing how the premature death rates correlates with low birthweights, with air pollution, with preventable hospital stays, and with children in poverty and see how these variables predict the variance in premature death rates by county. I am using data from the County Health Rankings website. They gathered their data via surveys, the National Vital Statistics System, the CDC health tracking network, …

The prevalence of communicable diseases continues to be one of the continent's leading causes of deaths. Cholera is a waterborne disease triggered by toxigenic strains of the Gram-negative bacteria Vibrio Cholerae O1 strain and less common O139 strain. with symptoms such as severe acute watery diarrhea and vomiting leading to dehydration, progressing to hypovolemic shock and death if not treated timely due to its short incubation period (Pasetto et al., 2018). West African countries are largely portrayed as endemic to cholera, though the dynamics of outbreaks in these developing countries remain largely uncertain. The purpose of the study is to …

When people allow fear to drive their decision making, they often make decisions that do more harm than good. Examples of this include stocking up on ciprofloxacin, flooding doctors’ offices and buying black market antibiotics after the anthrax attacks of 2001. Therefore, it is important to be able to address what people are saying when another anthrax attack occurs. Supervised and unsupervised machine learning methodologies can be utilized to detect an event, classify the tweets by event, and to determine the main topics of discussion. Over the period of data collection, twenty events were detected. Three of these events concerned …

Birds are major vectors and reservoir hosts of pathogens across our globe. Pathogens can be spread through the dispersal and seasonal movements of migratory and resident wild birds. Although biogeographic barriers have the potential to restrict completely or in part gene flow in wild birds, variation in ecological drivers of dispersal likely influences the effectiveness of these barriers amongst different species. This dissertation seeks to enhance our knowledge of Southern Hemisphere waterfowl species’ dispersal, population genomics, and disease ecology using population genomic techniques (double-digest restriction-site associated DNA sequencing (ddRADseq)) and multi-variate statistics. First, we investigated the roles of dispersal behavior …
