Development of latent HIV-infected CD4+ T lymphocytes is the major cause of HIV treatment failure. Adeno-associated virus (AAV) is an attractive vector for anti-HIV gene therapy due to its lack of pathogenicity, low immunogenicity, and persistent transgene expression. However, a major limitation for AAV gene transfer is the cell-specific tropism of each serotype. Only AAV serotypes 2 and 5 have been investigated in hematopoietic cells, both of these serotypes have shown low transduction efficiency. Since each of the currently described AAV serotypes demonstrate distinct tissue tropism, I hypothesized that other serotypes such as AAV1, 4, 8, or 9 may demonstrate …

Interleukin 2 (IL-2) is a member of the cytokine family and contributes to the proliferation, survival, and death of lymphocytes [1]. The interleukin-2 receptor (IL-2) is a tripartite receptor commonly expressed on the surfaces of many lymphoid cells and is composed of three non-covalently associated subunits, alpha (CD25), beta (CD122), and gamma (CD132) [2]. Our laboratory has previously described IL-2 receptor Beta expression by vasculature smooth muscle cells (VSMC) in mice and humans [3]. Five types of glucose transporters stimulate the transport of glucose from the bloodstream into cells such as muscle cells and adipocytes in response to insulin signaling or exercise. Skeletal muscle and adipose tissue are the main sites for glucose uptake and dispose in insulin action dependent pathway. In non-insulin stimulated condition, the GLUT4 storage vesicles stay away from the plasma membrane, so there is no GLUT4 translocation or GLUT4 mediated glucose uptake. However, in insulin stimulated state, insulin initiates insulin signaling cascade which results in rapid GLUT4 translocation and increased glucose uptake at the cell surface. Pathogen incidence rate prediction, which can be considered as time series modeling, is an important task for infectious disease incidence rate prediction and for public health. This paper investigates the application of a genetic computation technique, namely GEP, for pathogen incidence rate prediction. To overcome the shortcomings of traditional sliding windows in GEP-based time series modeling, the paper introduces the problem of mining effective sliding window, for discovering optimal sliding windows for building accurate prediction models. The ectoparasitic mite, Sarcoptes scabiei that burrows in the epidermis of mammalian skin has a long co-evolution with its hosts. Phenotypic studies show that the mites have the ability to modulate cytokine secretion and expression of cell adhesion molecules in cells of the skin and other cells of the innate and adaptive immune systems that may assist the mites to survive in the skin. The purpose of this study was to identify genes in keratinocytes and fibroblasts in human skin equivalents (HSEs) that changed expression in response to the burrowing of live scabies mites. Background: Cholera outbreaks result in significant morbidity worldwide. Transmission of the disease may be influenced by weather fluctuations, such as El Niño-Southern Oscillation (ENSO) and excessive rainfall, and socioeconomic status (SES), including income and education. El Niño events may influence many aspects of the climate, including the amount of rainfall occurring in a given year and surface water temperature. This study aimed to assess the influences of extreme weather and SES factors on cholera incidence in India during the seventh cholera pandemic (1961-2008).

In the United States there is a significant gap between Sexually Transmitted Infection (STI) rates of Caucasians and African Americans. This disparity is attributed to many causes including access to health care, socioeconomic status, and social dynamics. The objective of this research is to describe the racial demographics of STI infections in the Air Force and explore possible disparities based on military pay grades. The military enforces many of the recommendations on how to limit STI health disparities by offering easy access to free health care and mandatory regular checkups. Objective: To analyze the need for increased content on genomics in Public Health (PH) academic curriculum.
Methods: A literature synthesis was performed of various genetic science studies. Results were assessed with regard to the number and type of genomic studies published, cost to perform genetic analyses, health care provider and the public’s use and understanding of genetic testing capabilities. A search was conducted and assessment performed of public health genomics curriculum at accredited public health educational programs.

In Peru, treatment for latent tuberculosis infection (LTBI) with isoniazid is free for all people younger than 20 who have been in close contact with a person with active pulmonary tuberculosis (TB). Despite the availability of this drug therapy within the context of a TB control program that exceeds the international standards for TB care, very few children access, and even fewer complete a full course of treatment. Phylogenetic analyses can resolve historical relationships among genes, organisms or higher taxa. Understanding such relationships can elucidate a wide range of biological phenomena, including, for example, the importance of gene and genome duplications in the evolution of gene function, the role of adaptation as a driver of diversification, or the evolutionary consequences of biogeographic shifts. The community blood supply is a limited resource with a relatively short shelf-life. To ensure an adequate supply, a balance must be maintained between the numbers of donors providing blood and blood products required by hospitals to meet patient needs. Any change in the number of blood donors or in the volume of blood required by hospitals can create a blood shortage. During times of disaster, a blood shortage may be inevitable. Objective: As Americans consume more meals outside the home, food safety of restaurants is more critical. The purpose of this study is to examine the effect of food service establishment (FSE) inspection frequency on FSE performance. In this study, the U.S. Air Force (USAF) public health system is compared to Ohio local public health departments (Ohio LPHDs). The USAF inspects FSEs more frequently than Ohio LPHDs do. The goal is to determine if this increased frequency leads to higher food safety in the FSEs.
Methods: We reviewed 1482 reports from three USAF bases and 1909 reports from six Ohio LPHDs …

Introduction. International medical aid in the form of short-term medical missions (STTMs) receives more than $250 million in annual donations. Needs assessments prior to the STTM and outcomes analysis for improvement are often lacking. Objective. Describe the patient population attended to during a one-week STTM in Paraguay, South America, while examining “parasites” as a presenting concern, as a diagnosis by the providers, and the prescription of antiparasitic treatment. This analysis identified variance in intestinal parasite presentation and management by age, gender, and location. The administration frequency of antiparasitic medication was considered as prophylactic versus disease-treatment based.

Gun violence profoundly impacts the landscape of violence in the United States. The costs of gun injuries are, estimated to be $100 billion each year in the United States. Although many physicians and health professionals agree that they have a role to play in the prevention of violence (American Academy of Pediatrics [AAP], 2002), there is a distinct disconnect between perceived responsibility and actual practice (Sidelinger, Guerrero, Rodriguez-Frau, & Mirabal-Colon, 2005). Hypoxia-inducible factor-1 alpha (HIF-1a) is a critical component of the cellular oxygen-sensing machinery and is essential for placental formation and embryonic survival. In this study, we promoted prolonged expression of HIF-1a, by using a form that is insensitive to oxygen, denoted as CA-HIF-1a. In order to have continual placental specific expression of the CA-HIF-1a, lentiviral infection was performed on embryos at the blastocyst stage of development and transferred into pseudo-pregnant mothers. Placental analysis was performed via in situ hybridization on embryonic day 14.5 (E14.5) to determine the effects of CA-HIF-1a prolonged expression. In our current study the roles of natural killer (NK) cells in regulation of the acute phase of murine cytomegalovirus (MCMV) infection were demonstrated. NK cells utilize perforin and gamma interferon to kill MCMV infected cells. Activation of NK cells is controlled by the balance between inhibitory and activation receptors and modulated by cytokines produced from the infected cells. Type I interferons are produced mainly by plasmacytoid dendritic cells (PDCs). Type I interferons have a substantial role in enhancing NK cells cytotoxic activity and NK cells proliferation and maintain cell survival through STAT mediated signaling pathway. The placenta is a transient organ that develops upon initiation of pregnancy and is essential for fetal development and survival. The rodent placenta consists of three layers with predominantly analogous cell types in the human placenta. The labyrinth layer, which lies closest to the fetus, facilitates nutrient and waste exchange between mother and baby. Abnormalities of the placenta may occur as a result of cellular stress and have been associated with the pregnancy-associated disorders, intrauterine growth restriction and placental insufficiency. Reactive Oxygen Intermediates (ROI) can significantly affect B Lymphocyte function, but the effects on key signaling pathways and gene expression in activated and non-activated B lymphocytes are largely undefined. This study demonstrates a concentration dependent effect of exogenous hydrogen peroxide on the transcriptional regulation of a 3' regulatory region of the Immunoglobulin heavy chain gene (3'IghRR). Specifically, low μM hydrogen peroxide induces an enhancing effect while higher μM hydrogen peroxide (greater than or equal to 100 μM) is suppressive. The enhancing effects of hydrogen peroxide on 3'IghRR activity are dependent on the NFκB/Rel pathway, and appear to be most prominent …

Herpes simplex virus type 1 (HSV-1) challenges the host immune system through several mechanisms (Frey, et al., 2009). In vitro, U937 cells (human macrophage-like precursor cell line) are not susceptible to HSV-1 infection when they are not differentiating (Lopez-Guerrero and Alonso, 1997). Differentiation of these cells resistance can abrogates their resistance to HSV-1 (Tenney and Morahan, 1991). In this study, we examined the effect of HSV-1 infection on differentiated and polarized U937 cells. U937 cells are differentiated to M0 cells.
Then, M0 cells are polarized to distinct phenotypes, M1 or M2. M1 are proinflammatory macrophages while M2 are anti-inflammatory cells. …

A libraries from libraries combinatorial chemistry approach was employed to synthesize fluorinated derivatives of both oxindoles and isatins as potential pharmaceuticals or targeting agents for imaging purposes related to cancer or Alzheimer's disease. Synthesis for these fluorinated derivatives are described by routes involving, either: a) N-alkylation of 5-substituted isatins followed by Wolff-Kishner reduction to the corresponding oxindoles and final Knoevenagel condensation with aryl aldehydes, or; b) Wolff-Kishner reduction of the isatins followed by condensation and finishing with the N-alkylation of the aldol products. Herpes simplex type 1 (HSV-1) quiescent infection was established in L929 cells, murine fibroblasts, and in Neuro-2A (N2A) cells, mouse neuroblastoma, by treating them with the nucleoside analogue acyclovir (ACV) 24 hours before infection. Subsequent release of virus from the non-productive state was accomplished by treating the cells with the histone deacetylase inhibitor trichostatin A (TSA). The Na+K+2Cl- co-transporter-1 (NKCC1) effects the electroneutral movement of Na+, K+ and 2Cl- ions across plasma membranes. NKCC1 is considered a widely distributed, highly N-glycosylated ion transporter involved in the regulation of the intracellular chloride concentration ([Cl-]i), cell volume and salt secretion. NKCC1 locates in the plasma membrane of all cells studied so far, particularly on the basolateral side of most polarized cells. However, the majority of immunoreactive NKCC1 distributes in intracellular compartments of unknown nature whereas a small proportion of the transporter appears to be located in the plasma membrane.
