The removal of mRNA transcript poly(A) tails by 3'-5' exonucleases is the rate-limiting step for controlled mRNA decay in eukaryotes. Poly(A)-specific ribonuclease (PARN) is one such exonuclease that degrades poly(A) tails, and although its in vitro activity is well-characterized, PARN’s patho-physiological roles in the cell are not well understood. Prior studies have found a possible role for PARN in cancer, in that PARN expression levels in human breast cancer tissues are often decreased compared to normal control tissues. Phospholipase D: Key Player In Macrophage-Mediated Inflammation And Resolution, Ramya Ganesan

Phospholipase D (PLD) is a cell membrane remodeling and signaling protein implicated in the pathology of chronic inflammation. As PLD is also central to macrophage cell migration, we investigated the molecular basis of PLD's involvement and regulation in macrophage-initiated inflammation (atherosclerosis) and resolution. We have found that PLD is associated with signaling proteins and positively affects cell movement, phagocytosis and NADPH-initiated release of Reactive Oxygen Species (ROS). We found that PLD2 but not PLD1 is important for foam cell formation that causes atherogenesis. We have also found a novel way of inducing macrophage class-switch (polarization) by PLD overexpression. PLD induces …

Stroke is a global burden that claims 6 million lives and permanently disables another 5 million each year. The risk of morbidity and mortality following stroke decreases when early diagnosis and treatment are achieved. However there is no blood test to diagnose ischemic stroke because biomarkers to date have not shown adequate sensitivity and specificity to be adopted into hospital protocols. The overall objective of this dissertation was to identify novel blood protein biomarkers of ischemic stroke. Allergy is a global health problem, affecting more than 40% of the world population. Allergy is often associated with a dysregulation of the immune system that becomes overactive against allergens. Several genes are known to be important for the proper homeostasis of the immune system in the epithelial tissues, and the loss of function of genes such as IKKß or SOX9 impacts the interaction between external stimuli and the host. Many of the allergic responses are triggered through the regulatory cascade governed by transcriptional factor NF-κB. – This study focuses on a central question: What key behavioral factors influence high school students’ compliance with preventative measures against the transmission of influenza? We use multilevel logistic regression to equate logit measures for eight precautions to students’ latent compliance levels on a common scale. Using linear regression, we explore the efficacy of knowledge of influenza, affective perceptions about influenza and its prevention, prior illness, and gender in predicting compliance. Hand washing and respiratory etiquette are the easiest precautions for students, and hand sanitizer use and keeping the hands away from the face are the most difficult. Perceptions of …

Trachyspermum ammi (L.) Sprague is an annual herbaceous plant belonging to the family Apiaceae and is mostly administered for stomach disorders. During the present research work, cold maceration and Microwave Assisted Extraction (MAE) of T. ammi Sprague was done by using methanol, dichloromethane and n-hexane separately as extracting solvents. For cold maceration, 2, 4, 6, 18 and 24 hours of time durations were used for each solvent. Maximum extract (50mg/g) was obtained at 18 hours of cold maceration with methanol, while maximum phenolics were also obtained from the same extract, which were calculated to be 170.89μg/g equivalent of Gallic acid. …

Sclerotium rolfsii is a polyphagous soil borne fungus infecting more than 500 plant species across the world that is causing vast losses. Although the fungus is soil and seed borne, soil borne inoculum is most vital in infection-causing and syndrome development. Treating soil borne pathogens with fungicides is not reasonable due to very high costs. Environmental hazards are also involved. Few animals are known to lay eggs in the absence of ovulation or copulation, as it is presumably energetically wasteful and subjected to negative selection. Characterization of Smed-boule, a member of the DAZ family of germline RNA-binding proteins, revealed that egg capsule (or capsule) production and deposition occurs independently of the presence of gametes in the planarian flatworm Schmidtea mediterranea. Reduction of Smed-boule expression by RNA-interference (RNAi) causes ablation of spermatogonial stem cells and the inability of ovarian germline stem cells to undergo oogenesis. Ochratoxin A (OTA) is a mycotoxin produced by several fungi of the genera Aspergillus and Penicillium, principally P. verrucosum in temperate climate and A. ochraceous in warm regions. In poultry feed materials, mycotoxins are found most commonly in cereals and to a lesser degree, in meals. The presence of OTA in animal feed contributes significantly to health disorders and decreased production. In addition to aflatoxins, which is an ubiquitously distributed toxin, OTA is one of the reasons for economic losses in the poultry industry due to poor performance and immunosuppression. Background: Around 35% of adults with osteoarthritis (OA) have knee osteoarthritis (KOA), which generally progresses over several years; however, some individuals experience accelerated KOA (AKOA), a rapid progression to end-stage disease within 48-months. Purpose: To assess baseline differences among those who develop the different types of KOA, and to determine if baseline characteristics and measures can be used to predict an individual’s KOA status (Common KOA or AKOA) 48 months later.
Methods: Cross-sectional analysis of data from the Osteoarthritis Initiative (OAI) was completed. At baseline, 4,769 participants were enrolled. Data from individuals (n = 1,561) free of radiographic KOA (Kellgren-Lawrence …

Purpose: The purpose of this research is to determine smart practices that could be used by a public health department to have an efficient, productive and successful accreditation by the Public Health Accreditation Board (PHAB).
Methodology: Accreditation coordinators of eight accredited Ohio-based public health departments were interviewed in 2016. A qualitative study was conducted using semi-structured interviews with funneling. Results were generated using a hybrid model of analysis that included a content analysis to make valid inferences from the interview data.

This report describes the results of research performed with funds from the Sture Fredrik Anliot Grant Fund granted in March 2015. The primary aim of the project was to identify and measure various stormwater pollutant inputs to Yellow Springs Creek and Birch Creek into Glen Helen Nature Preserve and then into the Little Miami River in Greene County, Ohio from Spring 2015 to Winter 2016.

The cell cytoskeleton, a unique intracellular matrix found in all eukaryotes, is composed of three main protein structures, microtubules, microlaments and intermediate laments. The cytoskeleton maintains cell shape and internal structures providing mechanical support that facilitates intracellular transport (Parker et al., 2014). Many viruses such as the herpesviruses use the cytoskeleton system of the cell for infectivity (Henry Sum, M. S. 2015). Accurate analysis of specific microvesicles (MVs) from biofluids is critical and challenging. Here we described novel methods to purify and detect MVs shed from endothelial cells (ECs) and endothelial progenitor cells (EPCs) by combining microbeads with fluorescence quantum dots (Q-dots) coupled nanoparticle tracking analysis (NTA). In the in vitroscreening systems, we demonstrated that 1) anti-CD105 (EC marker) and anti-CD34 (EPC marker) conjugated-microbeads had the highest sensitivity and specificity for isolating respective MVs, which were confirmed with negative controls, CD41 and CD235a; 2) anti-CD144 (EC marker) and anti-KDR (EPC marker) conjugated-Q-dots exhibited the best sensitivity and specificity for their respective …

Background: In low and middle-income countries (LMICs), non-communicable diseases (NCDs) are rapidly emerging alongside a well-documented persistence of communicable diseases (CDs) in fragile health care systems. In sub-Saharan Africa, chronic NCDs are expected to overtake CDs as the leading cause of death. The aim of this study was to compare mortality rates due to CDs and NCDs between developed countries, developing countries, Burundi, Rwanda, and Uganda from 1990 to 2013.

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease caused by progressive degeneration of upper and lower motor neurons in the brain and spinal cord. Multiple mutations are found in some of the proteins associated with ALS, including superoxide dismutase (SOD1), fused in sarcoma (FUS) and trans-activation response DNA-binding protein (TDP-43). TDP-43 is a DNA and RNA binding protein, well conserved, and ubiquitously expressed in all tissues. TDP-43 resides in the nucleus and sometimes shuttles between nucleus and cytoplasm.
