Microfluidic technologies have enabled in vitro studies to mimic in vivo microvessel environment with sufficient complexity. However, there are still existing knowledge gaps and lack of convincing evidence to demonstrate and quantify key features of a functional microvessel. In this dissertation, a physiologically realistic microvessel model was developed with a stable and mature endothelium for studying complex vascular phenomena, such as endothelial cell signaling and barrier functions with the microscopic resolution at individual cellular levels. With advanced micromanufacturing and microfluidic technologies, two types of cost-efficient, easy to operate and reproducible microchannel network devices were fabricated, and the fabricated microchannels mimicked …

Alteration in vascular reactivity serves as a marker for cardiovascular disease. Enhancement of vasodilatory response through adenosine A2A receptor (A2AAR) signaling against high salt (HS) diet is speculated to be an adaptive response as salt-sensitive rats showed upregulation of A 2AAR and cyp-epoxygenases but salt-resistant rats did not. However, no direct evidence is available showing the compensatory role of A2A AR signaling upon salt loading. For this, understanding the functional and biochemical outcome of HS diet on vascular tone regulation and its underlying signaling mechanism in the presence and absence of A 2AAR is required. Poultry is one of the major contributors of inputs to U.S. environmental waters due to the large number of poultry grown per year in the U.S. and litter disposal practices. Two studies were conducted during this research: (1) to determine the temporal rate of deposition and growth or decay of microorganisms in poultry litter originating from poultry feces and (2) the release and transport of poultry litter associated microorganisms from soiled poultry litter applied as fertilizer through soil columns under simulated rainfall. Depressed smokers are a group of individuals whose health and overall functioning can benefit from interventions aimed to enhance well-being. Researchers have recently found significant relations between positive affect and smoking cessation success, and between low affect and smoking relapse, and unsuccessful abstinence (Cook et al., 2010; Leventhal et al., 2008, 2009; Niemiec, 2010). Aquaponics is the integration of aquaculture (the rearing of fish) and hydroponics (the conventional method of soilless farming). Effluent produced from the production of fish can be utilized as a source of nutrients by plants for growth in a hydroponic subsystem. For academic carrion insect succession studies to be applicable to forensic casework, the study must provide statistically defensible postmortem interval (PMI) estimates. Multiple statistical approaches have been used to describe insect succession, but no study to date has been able to determine a confidence interval about a succession based PMI estimate. The ability to reject PMI values, and thereby create a confidence interval is determined by statistical power, of which sample size is a positive correlate. Microfluidics continues to be of interest for analyzing chemical and biological samples because of the disposability, portability, low sample consumption, fast analysis time, and parallel analysis potential for multiple samples in a single device. To improve microfluidic device functionality, integrated systems-nanofluidic/microfluidic interfaces (NMIs) have been fabricated for concentrating samples and performing as molecular gates. Introduction: In sub-Saharan Africa, HIV/AIDS is one of the leading causes of mortality that disproportionately affects women and children. In this sub-region, 57% of those living with HIV are women and is home for 90% of children under the age of 15 that are living with HIV. The primary goal of this study was to evaluate the influence of adherence to nutrition guidelines and engaging in regular physical activity on the physical and psychological dimensions of well-being in a sample of middle-aged and older adults. All available sociodemographic indicators were also analyzed in order to investigate potential interactions. A structural equation model was constructed utilizing data from 348 middle-aged and older adults who completed the Health Behavior and Knowledge Survey. Results indicated the model was a good fit to the data, accounting for 40.4% of the variance in physical well-being and 26.3% of variance in psychological well-being. …

Peripheral vascular disease is a pathological disease state whereby the peripheral vascular system becomes progressively limited in its ability to adequately perfuse extremities despite increases in metabolic demand. One of the primary risk factors for development of peripheral disease is the Metabolic Syndrome. It is the presentation of three simultaneous comorbidities, with obesity being the most common denominator in most metabolic syndrome patients, but also including a pro-inflammatory state, pro-oxidant state, pro-thrombotic state, increased blood pressure, atherogenic dyslipidemia, or insulin resistance. Consumers of pastured hen eggs have justified paying an associated premium price because they perceive animal welfare, sustainability, and nutrition are enhanced compared to conventionally produced eggs. Microbes are capable of rapid genetic modification, enabling the habitation of a wide field of niches, including forming interdependent associations with macroscopic hosts. While ancient multipartite mutualisms have been shown to involve metabolic complementation, little is known concerning the early genomic adaptations leading towards co-residence within a novel host. There is limited biologic and epidemiologic evidence indicating an association between MRFs and cancer. Obesity causes a multitude of metabolic issues in the horse, yet stepwise alterations in glucose and lipid metabolism, mitochondrial capacity and oxidant status during weight loss have yet to be analyzed. Skeletal muscle contains subsarcolemmal (SSM) and interfibrillar (IFM) mitochondria, which respond differently to physiological stimuli, impacting tissue and whole body oxidant status. We hypothesized that 8 light-type horses would display improvements in skeletal muscle mitochondrial subpopulation function and decreases in circulating oxidant status markers during weight loss from an obese (7 to 8) to moderate (5) body condition score (BCS). Gastrointestinal nematode parasitism is the most widely reported health concern among sheep producers worldwide. The most concerning of which is Haemonchus contortus; a blood-sucking nematode found in the abomasum of sheep. Worldwide anthelminthic resistance has prompted exploration of alternative methods of parasite control. A study, conducted in two replicates, examined the effects of an artificial photoperiod on puberty in fall-born ewe lambs. Replicate I was initiated in December 2011 with prepubertal Dorset ewe lambs (n = 44). Replicate II was initiated beginning in December 2012 and included prepubertal Dorset, Suffolk x Dorset and Texel x Dorset ewe lambs (n = 52).
