Understanding The Host Immune Response Against Pseudomonas Aeruginosa To Develop Novel Therapeutics And Vaccines, Emel Sen Kilic Jan 2020

Understanding The Host Immune Response Against Pseudomonas Aeruginosa To Develop Novel Therapeutics And Vaccines, Emel Sen Kilic

Graduate Theses, Dissertations, and Problem Reports (ETD)

Pseudomonas aeruginosa is a Gram-negative opportunistic bacterium that causes a broad range of acute and chronic infections. The high adaptability and emergence of multidrug-resistant strains of this bacterium pose a significant threat to human health. Particularly, pneumonia caused by this pathogen is associated with high morbidity and mortality in immunocompromised patients. To prevent these infections, we aimed to develop novel vaccine strategies by characterizing the host immune response against P. aeruginosa. During respiratory infections, P. aeruginosa first contacts with epithelial cells along the respiratory tract. Using RNA-sequencing, we were able to characterize transcriptional changes of the epithelial cells in …

Haemonchus Contortus Antigen Identification And Use Of Tropomyosin In Ovine Vaccine Development, Brynnan P. Russ

Graduate Theses, Dissertations, and Problem Reports (ETD)

Globally, the small ruminant industry loses hundreds of millions of dollars due to parasitism by the gastrointestinal nematode Haemonchus contortus. This parasite feeds on blood in the host’s abomasum leading to disease including anemia, hypoproteinemia, lethargy, and death. There have been exhaustive efforts to manage this parasite including different management applications, anthelmintic treatment, and now integration of resistance genetics into a flock. Like antibiotics, overuse of anthelmintics has led to resistance of H. contortus to these drugs. Therefore, other methods of control are the main focuses of many research initiatives. St. Croix are a resistant breed of sheep that mount …

The Immunological Response To Chronic Stress In Obese Mice, Matthew Corey Parsley Jan 2020

The Immunological Response To Chronic Stress In Obese Mice, Matthew Corey Parsley

Graduate Theses, Dissertations, and Problem Reports (ETD)

Obesity and psychological stress are major risk factors for cardiovascular disease and have a positive correlation with vascular dysfunction. This functional deficit has been linked to a pro-inflammatory milieu, exacerbated by an upregulation of pro-inflammatory cytokines. Febuxostat, a xanthine oxidase inhibitor, has been shown to rescue vascular impairment in these models. The purpose of this study was to examine the individual and combined effects of obesity and chronic stress on neuroimmune cell populations, and assess if febuxostat is able mediate immune alterations.

Male mice (n=48) were assigned into either normal chow (lean) and high-fat diet (obese) groups. The lean animals …

Tree Growth And Water Use Efficiency During The Twentieth Century: From Global Trends To Local Drivers, Justin Michael Mathias Jan 2020

Tree Growth And Water Use Efficiency During The Twentieth Century: From Global Trends To Local Drivers, Justin Michael Mathias

Graduate Theses, Dissertations, and Problem Reports (ETD)

The frequency and severity of extreme environmental conditions will continue to increase under global environmental change. How terrestrial plants respond to prolonged, and often novel environmental stressors, will have profound impacts on, and feedbacks with, the Earth climate system at local to continental scales. Central to these feedbacks are plant stomata, actively regulated pores on the leaves of plants that act as a control valve over the fluxes of carbon dioxide (CO2) into the leaf during photosynthesis and water vapor (H2O) out of the leaf during transpiration. Importantly, changes in stomatal aperture do not affect the …

Hepatitis B In At-Risk Groups: Three Studies To Guide Education, Testing, And Vaccination Efforts In West Virginia, Stacy Tressler

Graduate Theses, Dissertations, and Problem Reports (ETD)

Background and Objectives: For over a decade, West Virginia’s (WV) rate of acute hepatitis B has been the highest in the United States (US). In 2017, WV’s rate was 11.7 per 100,000 population, almost 12 times higher than the national rate. The increase in acute hepatitis B cases is largely due to substance misuse, including injection drug use (IDU). Hepatitis B is a vaccine preventable disease yet many at-risk adults remain unvaccinated and susceptible to infection. The objectives of our studies are to 1) describe yearly changes in acute hepatitis B incidence in WV and assess county-level impact of the …

Impact Of Sire Selection And Breed On Parasite Resistance In Sheep, Andrew Ryan Weaver Jan 2020

Impact Of Sire Selection And Breed On Parasite Resistance In Sheep, Andrew Ryan Weaver

Graduate Theses, Dissertations, and Problem Reports (ETD)

Selection within and among breeds are strategies to mitigate the impact of parasitism given failing chemotherapeutics. While Texels have improved marketability compared to other parasite-resistant breeds, the mechanism by which Texels reduce fecal egg count (FEC) is unclear. The immune response to Haemonchus contortus (Hc) was compared in Texel, parasite-resistant St. Croix, and parasite-susceptible Suffolk sheep. Adult worms exposed to St. Croix- and Texel-derived peripheral blood mononuclear cells and serum in vitro had greater binding around the reproductive structures than Suffolk and worm egg release tended to be affected by breed (P = 0.09). Resistance in Texels …

Alternative Media And Mulches In Organic Vegetable Production, Heather R. Griffith Jan 2020

Alternative Media And Mulches In Organic Vegetable Production, Heather R. Griffith

Graduate Theses, Dissertations, and Problem Reports (ETD)

Reducing or eliminating waste from organic vegetable production can conserve money as well as resources. Conventional greenhouse production of vegetable transplants often relies on abundant fertigation which produces large amounts of polluted runoff, and the field production of organic vegetables frequently relies on the use of polyethylene mulch. A greenhouse study was conducted to determine if organic transplants can be successfully produced in a greenhouse under reduced soil moisture in order to reduce leaching of nutrients and potential water pollution. To test this, we compared the effect of reduced irrigation volumes to the standard practice of irrigating with 120% of …

The State And War On Poverty: British Welfare Development And Its Legacies For Malawi, 1930s-1983, Gift Wasambo Kayira Jan 2020

The State And War On Poverty: British Welfare Development And Its Legacies For Malawi, 1930s-1983, Gift Wasambo Kayira

Graduate Theses, Dissertations, and Problem Reports (ETD)

This dissertation documents the struggles and dilemmas that the Malawian state endured as it attempted to achieve its developmental goals from the 1930s to 1983. It contributes to histories of development by focusing on the interventions both the colonial and postcolonial states made to improve the living standards of African rural communities, the ideas which shaped state programs, and the behavior of the state which such interventions reveal. Scholars typically argue that state policy in Malawi was necessarily destructive and limited the economic progress of the local communities. The state deliberately pursued land, market, and other agricultural policies that constrained …

The Host Gatekeeper: Using The Flagellar Pathway To Understand Symbiont Host Adaptation, Adam R. Pollio Jan 2020

The Host Gatekeeper: Using The Flagellar Pathway To Understand Symbiont Host Adaptation, Adam R. Pollio

Graduate Theses, Dissertations, and Problem Reports (ETD)

The acquisition of microbial partners is a strategy used by a diverse group of arthropods to overcome ecological barriers that might normally make certain niches uninhabitable. The unique phylogenetic opportunities attainable from the natural experiment of the Sodalis-allied clade allow for better understanding of how molecular structures evolve through time. Here, we focus on the evolution of the flagellar synthesis pathway, due to its complexity and ability to diverge in response to ecological pressures. We used this molecular pathway and natural experiment to show that normal evolutionary outcomes associated with symbiosis (i.e., genome reduction) do not explain the predicted conservation …

Allopurinol Alters The Expression Of Cytokines And Mediators Of Immune Cells In The Systemic 2 Tissue By Decreasing Plasma Urate., Lundrim S. Marku

Graduate Theses, Dissertations, and Problem Reports (ETD)

Diminished levels of urate have been linked to oxidative stress in birds and mammals. Urate, a major antioxidant that lowers reactive oxygen/nitrogen species (ROS/RNS), is thought to ameliorate the effects. The relationship between reduced urate, the immune system and the pathogenesis of the intestine, the liver or the brain has not been well characterized in avians. Moreover, recent studies have proposed that an increased permeability in the intestine due to some insult can induce inflammation in peripheral organs such as the brain. Allopurinol, a relatively toxic purine analogue that serves as a xanthine oxidase inhibitor, reduces urate levels which can …

Faculty & Staff Scholarship

Rates of adverse cardiovascular events have increased among middle-aged adults. Elevated ceramides have been proposed as a risk factor for cardiovascular events. Diet quality and weight status are inversely associated with several traditional risk factors; however, the relationship to ceramides is less clear. This study aimed to determine associations of adiposity and diet quality with circulating ceramides in middle-aged adults (n = 96). Diet quality was estimated using the Healthy Eating Index 2015 (HEI-2015). Serum ceramide concentrations were determined by liquid chromatography–mass spectrometry. A ceramide risk score was determined based on ceramides C16:0, C18:0, and C24:1 and their ratios to …

Clinical and Translational Science Institute

Stroke is the fifth leading cause of death in the U.S., with more than 100,000 deaths annually. There are a multitude of risks associated with stroke, including aging, cardiovascular disease, hypertension, Alzheimer’s disease (AD), and immune suppression. One of the many challenges, which has so far proven to be unsuccessful, is the identification of a cost-effective diagnostic or prognostic biomarker for stroke. Alkaline phosphatase (AP), an enzyme first discovered in the 1920s, has been evaluated as a potential biomarker in many disorders, including many of the co-morbidities associated with stroke. This review will examine the basic biology of AP, and …
