Erk-Mediated Timeless Expression Suppresses G2/M Arrest In Colon Cancer Cells, Beth K. Neilsen, Danielle E. Frodyma, Jaime L. Mccall, Kurt W. Fisher, Robert E. Lewis

Faculty & Staff Scholarship

The cell cycle is under circadian regulation. Oncogenes can dysregulate circadian-regulated genes to disrupt the cell cycle, promoting tumor cell proliferation. As a regulator of G2/M arrest in response to DNA damage, the circadian gene Timeless Circadian Clock (TIMELESS) coordinates this connection and is a potential locus for oncogenic manipulation. TIMELESS expression was evaluated using RNASeq data from TCGA and by RT-qPCR and western blot analysis in a panel of colon cancer cell lines. TIMELESS expression following ERK inhibition was examined via western blot. Cell metabolic capacity, propidium iodide, and CFSE staining were used to evaluate the effect of TIMELESS …

A new avenue of mining published genome-wide association studies includes the joint analysis of related traits. The power of this approach depends on the genetic correlation of traits, which reflects the number of pleiotropic loci, i.e. genetic loci influencing multiple traits. Here, we applied new meta-analyses of optic nerve head (ONH) related traits implicated in primary open-angle glaucoma (POAG); intraocular pressure and central corneal thickness using Haplotype reference consortium imputations. We performed a multi-trait analysis of ONH parameters cup area, disc area and vertical cup-disc ratio. We uncover new variants; rs11158547 in PPP1R36-PLEKHG3 and rs1028727 near SERPINE3 at genome-wide significance …

Abstract The human macula is more susceptible than the peripheral retina to developing blinding conditions such as age-related macular degeneration, diabetic retinopathy. A key difference between them may be the nature of their Mu¨ ller cells. We found primary cultured Mu¨ ller cells from macula and peripheral retina display significant morphological and transcriptomic differences. Macular Mu¨ ller cells expressed more phosphoglycerate dehydrogenase (PHGDH, a rate-limiting enzyme in serine synthesis) than peripheral Mu¨ ller cells. The serine synthesis, glycolytic and mitochondrial function were more activated in macular than peripheral Mu¨ ller cells. Serine biosynthesis is critical in defending against oxidative stress. …

Trade-Offs Relating To Grassland And Forest Mine Reclamation Approaches In The Central Appalachian Region And Implications For The Songbird Community, Eric L. Margenau, Petra B. Wood, Cathy A. Weakland, Donald J. Brown

Faculty & Staff Scholarship

Surface mining in the Appalachian region (USA) converts large areas of mature forest to early successional habitat. This shift in landscape structure has the potential to reduce habitat availability and suitability for forest-dwelling songbirds by reducing and fragmenting mature forest, but also to increase habitat availability for grassland- and shrubland-associated songbirds. We examined the influence of mountaintop mining/valley fill (MTMVF) reclamation habitats (grassland, shrubland, and remnant forest) on songbird community composition and abundance at three former MTMVF mines in southwestern West Virginia, relative to intact forest. We quantified the songbird community in 1999 and 2000 using point counts arranged throughout …

Coupling Simulation Model Between Mine Ventilation Network And Gob Flow Field, F.I. Wu, Y. Luo, X-T Chang Jan 2019

Coupling Simulation Model Between Mine Ventilation Network And Gob Flow Field, F.I. Wu, Y. Luo, X-T Chang

Faculty & Staff Scholarship

Many numerical simulation studies of coal spontaneous combustion have focused on the leakage flow field in the mine gob. However, most of these studies isolate the gob from the mine ventilation system, failing to consider the effects of air leakage on gob boundary conditions. A novel model coupling the mine ventilation network (MVN) and gob flow field (GFF) has been developed to simulate the overall mine ventilation system. The concept of network boundary node was proposed and the corresponding air flow rate balance equations were developed based on the node pressure method for MVN calculation and the finite element method …

From Statistical Correlations To Stochasticity And Size Effects In Sub-Micron Crystal Plasticity, Hengxu Song, Stefanos Papanikolaou Jan 2019

From Statistical Correlations To Stochasticity And Size Effects In Sub-Micron Crystal Plasticity, Hengxu Song, Stefanos Papanikolaou

Faculty & Staff Scholarship

Metals in small volumes display a strong dependence on initial conditions, which translates into size effects and stochastic mechanical responses. In the context of crystal plasticity, this amounts to the role of pre-existing dislocation configurations that may emerge due to prior processing. Here, we study a minimal but realistic model of uniaxial compression of sub-micron finite volumes. We show how the statistical correlations of pre-existing dislocation configurations may influence the mechanical response in multi-slip crystal plasticity, in connection to the finite volume size and the initial dislocation density. In addition, spatial dislocation correlations display evidence that plasticity is strongly influenced …

Electrical Detection Of Current Generated Spin In Topological Insulator Surface States: Role Of Interface Resistance, C. H. Li, O. M. J. Van ‘T Erve, C. Yan, L. Li., B. T. Jonker Jan 2019

Electrical Detection Of Current Generated Spin In Topological Insulator Surface States: Role Of Interface Resistance, C. H. Li, O. M. J. Van ‘T Erve, C. Yan, L. Li., B. T. Jonker

Faculty & Staff Scholarship

Current generated spin polarization in topological insulator (TI) surface states due to spin-momentum locking has been detected recently using ferromagnet/tunnel barrier contacts, where the projection of the TI spin onto the magnetization of the ferromagnet is measured as a voltage. However, opposing signs of the spin voltage have been reported, which had been tentatively attributed to the coexistence of trivial two-dimensional electron gas states on the TI surface which may exhibit opposite currentinduced polarization than that of the TI Dirac surface states. Models based on electrochemical potential have been presented to determine the sign of the spin voltage expected for …

Synergy Of Physics-Based Reasoning And Machine Learning In Biomedical Applications: Towards Unlimited Deep Learning With Limited Data, Valeriy Gavrishchaka, Olga Senyukova, Mark Koepke

Faculty & Staff Scholarship

Technological advancements enable collecting vast data, i.e., Big Data, in science and industry including biomedical field. Increased computational power allows expedient analysis of collected data using statistical and machine-learning approaches. Historical data incompleteness problem and curse of dimensionality diminish practical value of pure data-driven approaches, especially in biomedicine. Advancements in deep learning (DL) frameworks based on deep neural networks (DNN) improved accuracy in image recognition, natural language processing, and other applications yet severe data limitations and/or absence of transfer-learning-relevant problems drastically reduce advantages of DNN-based DL. Our earlier works demonstrate that hierarchical data representation can be alternatively implemented without NN, …

Air Quality And Asthma Hospitalization: Evidence Of Pm2.5 Concentrations In Pennsylvania Counties, Elham Erfanian, Alan R. Collins

Regional Research Institute Working Papers

According to the World Health Organization, 235 million people around the world currently suffer from asthma, which includes approximately 25 million in the United States. There is substantial epidemiological evidence indicating linkages between outdoor air pollution and asthma symptoms, more specifically between concentrations of particulate matter and asthma. Using county level data for 2001-2014, a spatial panel framework is imposed based upon prevailing wind patterns to investigate the direct and indirect impacts of PM2.5 concentration levels on asthma hospitalization in Pennsylvania. This model controls for population density, precipitation, smoking rate, and population demographic variables. Results show that PM2.5 concentrations as …

A Regulated Charge Pump With Extremely Low Output Ripple, Mir Mohammad Navidi, David W. Graham

Faculty & Staff Scholarship

Abstract: In this paper, we present a regulated charge pump with extremely low output ripple (mV) that can be used for accurate programming of nonvolatile memory. We present a technique to include a low-drop-out regulator inside the charge-pump regulation loop to reduce the ripple. This charge pump was fabricated in a 0.35 µm standard CMOS process. The die area of this charge pump is 0.163 mm2 . While operating from a 2.5 V supply, this charge pump generates regulated voltages up to 10 V.

Temporal Variations In Methane Emissions From An Unconventional Well Site, Derek Johnson, Robert Heltzel, Dakota Oliver Jan 2019

Temporal Variations In Methane Emissions From An Unconventional Well Site, Derek Johnson, Robert Heltzel, Dakota Oliver

Faculty & Staff Scholarship

Studies have aimed to quantify methane emissions associated with the growing natural gas infrastructure. Quantification is completed using direct or indirect methods—both of which typically represent only a snapshot in time. Most studies focused on collecting emissions data from multiple sites to increase sample size, thus combining the effects of geospatial and temporal variability (spatio-temporal variability). However, we examined the temporal variability in methane emissions from a single unconventional well site over the course of nearly 2 years (21 months) by conducting six direct quantification audits. We used a full flow sampling system that quantified methane mass emissions with an …

Depth Dependent Atomic Valence Determination In La0.7sr0.3mno3 Thin Films Using Synchrotron Techniques, Robbyn B. Trappen

Graduate Theses, Dissertations, and Problem Reports (ETD)

The valence of atoms often has a strong effect on the properties of materials, such as magnetism, conductivity, and superconductivity. The atomic valence is often perturbed at the surface and/or interface and this deviation may play a strong role in many physical phenomena such as interfacial coupling and dead layers, both magnetic and electric. In this dissertation, I present a non-destructive approach of combining two X-ray absorption detection modes, electron yield and fluorescence, with very different probing depths in conjunction with theory to map out the layer-by-layer valence of a thin film.

The weighted average Mn atomic valence as measured …

Stock Returns And Investor Sentiment: Textual Analysis And Social Media, Zachary Mcgurk, Adam Nowak, Joshua C. Hall

Economics Faculty Working Papers Series

The behavioral finance literature has found that investor sentiment has predictive ability for equity returns. This differs from standard finance theory, which provides no role for investor sentiment. We examine the relationship between investor sentiment and stock returns by employing textual analysis on social media posts. We find that our investor sentiment measure has a positive and significant effect on abnormal stock returns. These findings are consistent across a number of different models and specifications, providing further evidence against non-behavioral theories.

The Nac Family Transcription Factor Gmnac42–1 Regulates Biosynthesis Of The Anticancer And Neuroprotective Glyceollins In Soybean, Md Asraful Jahan, Brianna Harris, Matthew Lowery, Katie Coburn, Aniello M. Infante, Ryan J. Percifield, Amanda G. Ammer, Nik Kovinich Jan 2019

Faculty & Staff Scholarship

Background

Glyceollins are isoflavonoid-derived pathogen-inducible defense metabolites (phytoalexins) from soybean (Glycine max L. Merr) that have important roles in providing defense against pathogens. They also have impressive anticancer and neuroprotective activities in mammals. Despite their potential usefulness as therapeutics, glyceollins are not economical to synthesize and are biosynthesized only transiently and in low amounts in response to specific stresses. Engineering the regulation of glyceollin biosynthesis may be a promising approach to enhance their bioproduction, yet the transcription factors (TFs) that regulate their biosynthesis have remained elusive. To address this, we first aimed to identify novel abiotic stresses that enhance …

The Vwfa Is The Home Of Orthographic Learning When Houses Are Used As Letters, Lea Martin, Corrine Durisko, Michelle W. Moore, Marc N. Coutanche, Deborah Chen, Julie A. Fiez

Faculty & Staff Scholarship

Learning to read specializes a portion of the left mid-fusiform cortex for printed word recognition, the putative visual word form area (VWFA). This study examined whether a VWFA specialized for English is sufficiently malleable to support learning a perceptually atypical second writing system. The study utilized an artificial orthography, HouseFont, in which house images represent English phonemes. House images elicit category-biased activation in a spatially distinct brain region, the so-called parahippocampal place area (PPA). Using house images as letters made it possible to test whether the capacity for learning a second writing system involves neural territory that supports reading in …

Federal Regulations And U.S. Energy Sector Output, Joshua Hall, Shishir Shakya Jan 2019

Federal Regulations And U.S. Energy Sector Output, Joshua Hall, Shishir Shakya

Economics Faculty Working Papers Series

A large body of literature finds that the energy sector is important to economic growth and development. The production of energy has negative environmental impacts, however, which has resulted in the sector being highly regulated. While several studies examine the effect of particular regulations on the energy sector, in this study we use the recently developed measure of regulation called RegData to estimate the impact of federal regulations on the energy sector. We employ a panel ARDL model to find an inverted U-shaped relationship between federal regulations and U.S. energy sector output. Federal regulations appear to increase energy sector outputs …

The Efficient Corruption Hypothesis And The Dynamics Between Economic Freedom, Corruption, And National Income, Joshua Hall, John Levendis, Alexandre R. Scarcioffolo

Economics Faculty Working Papers Series

Income, economic freedom, and corruption interact in complex ways as all three variables are arguably endogenous. We explicitly model this endogeneity using a panel VAR framework. The pVAR models we estimate are able to explicitly model this endogeneity better than the single-equation panel data models previously used in the literature. Using data on corruption and income from the World Bank and economic freedom from the Fraser Institute, we provide evidence that corruption and the absence of economic freedom have a negative effect on national income.

Geographic Determinants Of Infant Health: The Impact Of Sports Facility Construction Projects, Brad Humphreys, Jane Ruseski Jan 2019

Geographic Determinants Of Infant Health: The Impact Of Sports Facility Construction Projects, Brad Humphreys, Jane Ruseski

Economics Faculty Working Papers Series

A large body of research examines determinants of infant health outcomes but little examines geographic variation generated by exogenous economic shocks. Linking birth data from the CDC Natality files to counties building new facilities from 1995-2002, we find that sports facility construction projects generate local negative externalities. Infants born during facility construction periods have lower birth weights than infants born in comparable counties where no new facility opened. Maternal prenatal health visits also fall in the post-facility-opening period. Mechanisms for this impact include local airborne particulate matter and reduced provision of government services affecting prenatal health.

Properties Of The Binary Neutron Star Merger Gw170817, B P. Abbott Jan 2019

Properties Of The Binary Neutron Star Merger Gw170817, B P. Abbott

Faculty & Staff Scholarship

On August 17, 2017, the Advanced LIGO and Advanced Virgo gravitational-wave detectors observed a low-mass compact binary inspiral. The initial sky localization of the source of the gravitational-wave signal, GW170817, allowed electromagnetic observatories to identify NGC 4993 as the host galaxy. In this work, we improve initial estimates of the binary’s properties, including component masses, spins, and tidal parameters, using the known source location, improved modeling, and recalibrated Virgo data. We extend the range of gravitational-wave frequencies considered down to 23 Hz, compared to 30 Hz in the initial analysis. We also compare results inferred using several signal models, which …

Contextual Interference In Speech Motor Learning Secondary To Similar Phonemes, Katelyn M. Bond Jan 2019

Contextual Interference In Speech Motor Learning Secondary To Similar Phonemes, Katelyn M. Bond

Capstones and Honors Theses

Purpose: The contextual interference (CI) effect is a motor learning phenomenon where learners experience difficulty during training resulting in poor performance; however, improved performance is observed in transfer conditions. Different variables elicit a CI effect, and the purpose of this study is to investigate whether phoneme (or sound) similarity may result in a CI effect during speech motor learning.

Method: The study included twenty-nine participants whose hearing and speech abilities were within the normal range. Participants were randomly assigned to one of two training sessions involving nonwords with either similar or dissimilar phonemes. Each training session included nonword repetition training …

An Application Of The Two-Stage Model Of Motor Learning To Speech Motor Control, Jacob W. Cahn Jan 2019

An Application Of The Two-Stage Model Of Motor Learning To Speech Motor Control, Jacob W. Cahn

Capstones and Honors Theses

Purpose: This study used a two-stage model of motor learning (Latash, 2012) to evaluate bad and good variance during a speech learning task. Stage 1 learning involves minimizing bad variance, or overall error, whereas Stage 2 learning involves changes in good variance while ceiling accuracy is maintained. Using two different variables to evaluate speech complexity (manner of articulation and syllable length), two hypotheses were evaluated: 1) A two-stage model exists during speech motor learning and 2) complexity of stimuli will alter the achievement and maintenance of each stage during learning.

Methods: Twelve native English speakers with normal speech and hearing …
